A few axonal proteins distinguish ventral spinal cord neurons from dorsal root ganglion neurons.

A series of proteins putatively involved in the generation of axonal diversity was identified. Neurons from ventral spinal cord and dorsal root ganglia were grown in a compartmented cell-culture system which offers separate access to cell somas and axons. The proteins synthesized in the neuronal cell somas and subsequently transported into the axons were selectively analyzed by 2-dimensional gel electrophoresis. The patterns of axonal proteins were substantially less complex than those derived from the proteins of neuronal cell bodies. The structural and functional similarity of axons from dif

Differentiation of Human Neurons and Neuron-Like Cells to Study VZV Neuronal Infection.

Varicella zoster virus (VZV) is a neurotropic human herpesvirus that causes varicella upon primary infection and establishes latency in neurons of the peripheral nervous system. The reactivation of latent neuronal infection results in herpes zoster and, in rare cases, severe neurological complications. VZV is human specific, and the investigation of VZV-neuron interactions requires human neuronal models, presenting challenges due to limited accessibility and ethical constraints. Here, we describe approaches to generate mechanoreceptive sensory neurons from small-molecule-derived neuronal precu

New neurons flatten social hierarchies.

As complex individualized behaviors in mice arise even when genes and environment are held constant, emerging social group structures should also depend on individual behavior. Adult hippocampal neurogenesis drives individual behavior and is involved in certain social behaviors. Would thus the emergence of social structure depend on adult-born neurons? To answer this question, we co-housed cohorts of female mice with intact and genetically diminished general adult neurogenesis in a large-scale automated tracking system for three months and analyzed the development of social behaviors over time

Optogenetics studies of kisspeptin neurons.

Optical systems and genetic engineering technologies have made it possible to control neurons and unravel neuronal circuit behavior with high temporal and spatial resolution. The application of optogenetic strategies to understand the physiology of kisspeptin neuronal circuits has evolved in recent years among the neuroendocrine community. Kisspeptin neurons are fundamentally involved in controlling mammalian reproduction but also are implicated in numerous other physiological processes, including but not limited to feeding, energy expenditure, core body temperature and behavior. We conducted

Analysis of Airway Vagal Neurons.

Internal organs, including the airway, are innervated by neurons of the autonomic and sensory nervous systems. The airway-innervating sensory neurons primarily originate from the vagus nerve, whose cell bodies are found, in rodents, in the jugular and nodose ganglia complex (JNC). About half of these sensory neurons expressed the heat-sensing ion channel TRPV1 and evolved to limit tissue damage by detecting chemical, mechanical, or thermal threats and to initiate protective airway reflexes such as coughing and bronchoconstriction. They also help monitor the host homeostasis by sensing nutrient

Mitochondria, motor neurons and aging.

While the role of mitochondria in aging has been well characterized, their involvement in motor neuron aging remains poorly understood. Thus, we performed an exhaustive ultrastructural study of mitochondria in motor neurons from aged rats that revealed dramatic alterations in the mitochondria of axon terminals at neuromuscular junctions, characterized by swelling, mitochondrial fusion and the presence of megamitochondria. These alterations were not observed in ventral horn motor neurons in the spinal cord of aged rats, which were only altered by the appearance of electron-dense bodies in the d

Apraxia, metaphor and mirror neurons.

Ideomotor apraxia is a cognitive disorder in which the patient loses the ability to accurately perform learned, skilled actions. This is despite normal limb power and coordination. It has long been known that left supramarginal gyrus lesions cause bilateral upper limb apraxia and it was proposed that this area stored a visual-kinaesthetic image of the skilled action, which was translated elsewhere in the brain into the pre-requisite movement formula. We hypothesise that, rather than these two functions occurring separately, both are complementary functions of chains of "mirror neurons" within

Orexinergic neurons and barbiturate anesthesia.

Orexins (OXs) regulate sleep with possible interactions with brain noradrenergic neurons. In addition, noradrenergic activity affects barbiturate anesthesia. As we have also recently reported that OXs selectively evoke norepinephrine release from rat cerebrocortical slices we hypothesized that barbiturate anesthesia may result from of an interaction with central orexinergic systems. To test this hypothesis, we performed a series of in vivo and in vitro studies in rats. In vivo, the effects of i.c.v. OX A, B and SB-334867-A (OX1 receptor antagonist) on pentobarbital, thiopental or phenobarbital

Genetic manipulation of noradrenergic neurons.

The neurotransmitter norepinephrine has been the focus of intense investigation for nearly a century. With advances in technology come novel approaches for testing hypotheses about the physiological roles of norepinephrine and the genes involved in norepinephrine (NE) biosynthesis, metabolism, and noradrenergic signaling. Homologous recombination techniques, which generate mice deficient in specific gene products, aid the integrated physiologist and pharmacologist in the evaluation of protein function. Mouse models lacking proteins involved in NE biosynthesis or metabolism provide tools to exp

Imitation, mirror neurons and autism.

Various deficits in the cognitive functioning of people with autism have been documented in recent years but these provide only partial explanations for the condition. We focus instead on an imitative disturbance involving difficulties both in copying actions and in inhibiting more stereotyped mimicking, such as echolalia. A candidate for the neural basis of this disturbance may be found in a recently discovered class of neurons in frontal cortex, 'mirror neurons' (MNs). These neurons show activity in relation both to specific actions performed by self and matching actions performed by others,

Resonance behaviors and mirror neurons.

This article is subdivided into two parts. In the first part we review the properties of a particular class of premotor neurons, the "mirror" neurons. With this term we define neurons that discharge both when the monkey makes a particular action and when it observes another individual (monkey or human) making a similar action. The second part is an attempt to give a neurophysiological account of the mechanisms underlying behaviors where an individual reproduces, overtly or internally, movements or actions made by another individual. We will refer to these behaviors as "resonance behaviors". We

Genetic disorders of motor neurons.

Disorders of the motor neuron are etiologically and clinically heterogeneous and cause serious disability and death. Whereas mendelian inheritance can be demonstrated in a subset of these disorders, the genetic contribution to the sporadic forms of motor neuron degeneration are not well understood. In families with spinal muscular atrophy, Kennedy disease and amyotrophic lateral sclerosis, genetic linkage analysis and positional cloning have proven to be extremely productive. The genetics of these neurodegenerative disorders are reviewed.

Does lead damage motor neurons?

It has been suggested that lead (Pb) may damage motor neurons selectively after retrograde axonal transport from the neuromuscular junction. This has reinforced suspicions that Pb could be a cause of some sporadic motor neuron diseases. However, the long-term effect of Pb on spinal motor neurons is not known. To investigate this, we injected mice in the right hindlimb with 0.1 ml of 2.5% Pb nitrate. Nine months later we measured the sizes and counted the numbers of motor neuron cell bodies in the anterior horns of the L4 spinal cord. There was no significant difference in sizes or numbers of m

Neonatal destruction of dopaminergic neurons.

Rats treated as neonates with 6-hydroxydopamine are proposed to model the dopamine deficiency associated with Lesch-Nyhan syndrome (LNS). To understand the neurobiological basis of specific behaviors in LNS, investigations were undertaken in these neonatally lesioned rats. Several new findings resulted from these studies. The first was that D1-dopamine receptors are essential for the action of D2-dopamine receptors, a phenomenon called "coupling" of receptor function. Another finding was that D1-dopamine receptors must be repeatedly stimulated before maximal behavioral sensitivity can be obser

Ultrastructure of hypothalamic paraventricular neurons.

The hypothalamic paraventricular nucleus (PVN) plays a pivotal role in the regulation of endocrine processes and the modulation of autonomic functions. The multi-channel outputs of the nucleus are directed toward the anterior and posterior lobes of the pituitary, autonomic centers, and limbic structures. Counterbalancing the wide spectrum of efferent projections, the nucleus receives humoral signals from endocrine glands and neuronal afferents from several loci of the brain. The multiple functions of the nucleus are executed by neurons that are organized in distinct subnuclei and display compl

[Neurons in the hyperchromic state].

The paper summarizes findings concerning the ultrastructure and cytochemistry of the cortical hyperchromic neurons which appear under different experimental conditions (in intact animals, in mechanical traumatization of neuronal tissue, during formation of a local focus of destruction, upon the administration of aminazine, and in the process of autolysis). Characteristics of the development of the hyperchromic status in each of the above experimental situations are specified. The authors consider the questions concerning the life-time development of neuronal hyperchromia, a relationship of hyp

Nociceptor neurons shape antiviral immunity.

The nervous and immune systems cooperate to protect the host, yet the contribution of sensory neurons to antiviral immunity remains incompletely defined. Nociceptor neurons do more than relay pain: they detect viral products and inflammatory cues through pattern-recognition receptors (PRRs), including Toll-like receptors (TLRs) and RIG-I-like receptors (RLRs), and respond to mediators such as type I interferons (IFNs), tumor necrosis factor (TNF), and interleukin-1β (IL-1β). Upon activation, these fibres release neuropeptides and neurotransmitters, including calcitonin gene-related peptide (CG
