D2AAK1 derivatives as neuroprotective agents: Synthesis and structural and pharmacological evaluation.

This study describes 14 novel D2AAK1 derivatives exhibiting enhanced neuroprotective properties. These derivatives were tested for their inhibitory effects on AChE and MAO-B, as well as their effect on cell viability under normal conditions and under oxidative stress. D2AAK1 derivatives show strong cytoprotective effects, increasing cell viability by up to 80% under normal conditions and up to 60% under oxidative stress. These effects may involve modulation of MAPK p38 and Nrf2 pathway interactions, known to promote antioxidant and anti-apoptotic responses. In vivo studies indicated a benefici

Imidazolylacetophenone oxime-based multifunctional neuroprotective agents: Discovery and structure-activity relationships.

Alzheimer's disease (AD) possesses a complex pathogenetic mechanism. Nowadays, multitarget agents are considered to have potential in effectively treating AD via triggering molecules in functionally complementary pathways at the same time. Here, based on the screening (∼1400 compounds) against neuroinflammation, an imidazolylacetophenone oxime ether (IOE) was discovered as a novel hit. In order to obtain SARs, a series of imidazolylacetophenone oxime derivatives were constructed, and their C=N bonds were confirmed as the Z configuration by single crystals. These derivatives exhibited potential

Exogenous Ketone Bodies as Promising Neuroprotective Agents for Developmental Brain Injury.

Ketone bodies are a promising area of neuroprotection research that may be ideally suited to the injured newborn. During normal development, the human infant is in significant ketosis for at least the first week of life. Ketone uptake and metabolism is upregulated in the both the fetus and neonate, with ketone bodies providing at least 10% of cerebral metabolic energy requirements, as well as being the preferred precursors for the synthesis of fatty acids and cholesterol. At the same time, ketone bodies have been shown to have multiple neuroprotective effects, including being anticonvulsant, d

Rat hippocampal slice culture models for the evaluation of neuroprotective agents.

Organotypic slices cultured for weeks in vitro represent an extremely valuable strategy for the investigation of the long-term properties of neuronal circuits under physiological and pathological conditions. Here, we describe how to prepare rat organotypic hippocampal slice cultures and how to expose them for appropriate periods of time to excitotoxic agents or to oxygen and glucose deprivation conditions, in order to mimic the pattern of pyramidal cell damage which is observed in vivo and in other in vitro models. This preparation is very useful not only to study synaptic plasticity or the pa

Tetramethylpyrazine nitrone: a multifaceted neuroprotective agent in neurodegenerative disorders.

Neurodegenerative disorders such as Alzheimer's disease (AD), Parkinson's disease (PD), and amyotrophic lateral sclerosis (ALS) share key pathological features, including oxidative stress, mitochondrial dysfunction, and impaired protein homeostasis, yet remain without effective disease-modifying therapies. Tetramethylpyrazine nitrone (TBN), a synthetic derivative of tetramethylpyrazine bearing a free radical-scavenging nitrone moiety, has emerged as a promising multi-target neuroprotective agent. This review synthesizes preclinical and clinical data supporting TBN's therapeutic potential in AD

Saffron: A Multitask Neuroprotective Agent for Retinal Degenerative Diseases.

Both age related macular degeneration (AMD) and light induced retinal damage share the common major role played by oxidative stress in the induction/progression of degenerative events. Light damaged (LD) rats have been widely used as a convenient model to gain insight into the mechanisms of degenerative disease, to enucleate relevant steps and to test neuroprotectants. Among them, saffron has been shown to ameliorate degenerative processes and to regulate many genes and protective pathways. Saffron has been also tested in AMD patients. We extended our analysis to a possible additional effect r

Neuroprotection by immunomodulatory agents in animal models of Parkinson's disease.

Parkinson's disease (PD) is an age-related neurodegenerative disease for which the characteristic motor symptoms emerge after an extensive loss of dopamine containing neurons. The cell bodies of these neurons are present in the substantia nigra, with the nerve terminals being in the striatum. Both innate and adaptive immune responses may contribute to dopaminergic neurodegeneration and disease progression is potentially linked to these. Studies in the last twenty years have indicated an important role for neuroinflammation in PD through degeneration of the nigrostriatal dopaminergic pathway. C

New agents promote neuroprotection in Parkinson's disease models.

Although researchers are pursuing "disease modifying" medications to slow or stop Parkinson's disease (PD) progression, a myriad of agents with protective properties in cell cultures and animal models have yielded few treatments in clinical practice. Developing safe and effective treatments with disease-modifying/neuroprotective mechanisms of action and identifying patients in the pre-motor phase will be a challenge. The implication of tyrosine hydroxylase (TH), the enzyme that catalyzes the formation of L-3,4-dihydroxyphenylalanine, in the pathogenesis of PD at different levels makes it a pro

Pharmacological properties of BN82451: a novel multitargeting neuroprotective agent.

BN82451 belongs to a new family of small molecules designated as multitargeting or hybrid molecules. BN82451 is orally active, has good central nervous system penetration, and elicits potent neuronal protection and antiinflammatory properties. Neuronal protection is due to Na+ channel blockade, antioxidant properties, and mitochondria-protecting activity, whereas inhibition of cyclooxygenases is mostly responsible for its antiinflammatory activity. BN82451 has been shown to exert a potent neuroprotective effect in various in vitro and in vivo animal models. BN82451 was found to exert a signifi

Cryptoxanthin as a multitarget neuroprotective agent: mechanistic and in silico perspectives.

OBJECTIVES: Neurodegenerative diseases such as Parkinson's disease (PD), Huntington's disease (HD), Alzheimer's disease (AD), and Amyotrophic lateral sclerosis (ALS) are complex disorders driven by multiple pathological processes, including oxidative stress, mitochondrial dysfunction, protein misfolding, and neuroinflammation. Due to this multifactorial nature, there is a growing interest in identifying natural compounds with multi-targeted neuroprotective properties. This review aims to evaluate the therapeutic potential of β-cryptoxanthin, a naturally occurring xanthophyll carotenoid, as a c

Naringenin as a multi-target neuroprotective agent in neurodegenerative diseases.

Neurodegenerative diseases (ND) such as Alzheimer's disease (AD), Parkinson's disease (PD), Huntington's disease (HD), Amyotrophic lateral sclerosis (ALS), and Multiple sclerosis (MS) are characterized by progressive neuronal loss driven by complex and multifactorial pathogenic mechanisms. Naringenin (NAR), a citrus-derived flavanone, has attracted considerable interest as a neuroprotective molecule due to its pleiotropic pharmacological activities such as antioxidant, anti-inflammatory and ability to modulate multiple cellular targets. This review provides a comprehensive overview of NAR phar

Metformin as a potential neuroprotective agent that influence Parkinson's disease.

Parkinson's disease (PD) is a significant neurodegenerative complication that impacts around 2% of individuals over the age of 65. The disease is characterized by a gradual decline of nigrostriatal dopaminergic neurons, which leads to physical impairments in individuals. Unfortunately, no drugs are currently available that can halt the progression and alleviate the symptoms of PD. Recently, there has been growing evidence to suggest a link between PD and type-2 diabetes (T2DM) which may provide a pharmacological explanation for this relation. In this review, we examine the literature that supp

Axon degeneration is an early event and pathological in neurodegenerative conditions and nerve injuries. To discover agents that suppress neuronal death and axonal degeneration, we performed drug screens on primary rodent neurons and identified the pan-kinase inhibitor foretinib, which potently rescued sympathetic, sensory, and motor wt and SOD1 mutant neurons from trophic factor withdrawal-induced degeneration. By using primary sympathetic neurons grown in mass cultures and Campenot chambers, we show that foretinib protected neurons by suppressing both known degenerative pathways and a new pa

Design, Synthesis, and Evaluation of Coumarin-Rasagiline Hybrids as Multifunctional Neuroprotective Agents.

The design of hybrid molecules with multifunctional profiles has emerged as a promising strategy in medicinal chemistry, particularly for tackling complex disorders such as neurodegenerative diseases. There is an urgent need for therapeutic agents capable of acting on multiple pathological targets to both control neuronal death and support the recovery of already compromised neuronal populations. In this study, we synthesized and evaluated a series of hybrid compounds combining a coumarin scaffold with an alkynylamine moiety inspired by rasagiline. These molecules were studied for their potent
