alpha-Latrotoxin stimulates exocytosis by the interaction with a neuronal G-protein-coupled receptor.

alpha-Latrotoxin is a potent stimulator of neurosecretion. Its action requires extracellular binding to high affinity presynaptic receptors. Neurexin I alpha was previously described as a high affinity alpha-latrotoxin receptor that binds the toxin only in the presence of calcium ions. Therefore, the interaction of alpha-latrotoxin with neurexin I alpha cannot explain how alpha-latrotoxin stimulates neurotransmitter release in the absence of calcium. We describe molecular cloning and functional expression of the calcium-independent receptor of alpha-latrotoxin (CIRL), which is a second high af

[Gustave Roussy's (1874-1948) contribution to neuroendocrinology].

Gustave Roussy (1874-1948) is remembered as a distinguished French neuropathologist and a leader in the field of oncology. However, his original contribution to the study of hypothalamic functions and neuroendocrinology remains ignored. Among Roussy's discoveries is the first experimental demonstration of the hypothalamic origin of diabetes insipidus and adiposo-genital dystrophy. Also, based on his own histological work, he pioneered the concept of neurosecretion, which was termed by him "neuricrinie".

Effects of reducing agents on glutathione metabolism and the function of carotid body chemoreceptor cells.

Two current hypotheses of O2 sensing in the carotid body (CB) chemoreceptors suggest participation of oxygen reactive (ROS) species, but they are mechanistically opposed. One postulates that hypoxia decreases ROS levels; the other that hypoxia increases them. Yet, both propose that the ensuing alteration in the cellular redox environment is the key signal triggering hypoxic chemoreception. Since the glutathione redox pair is the main cellular buffer for ROS and the main determinant of the general redox environment of the cells, a way to test whether ROS participate in chemoreception is to dete

An opioid-neuropeptide-Y transmission line to luteinizing hormone (LH)-releasing hormone neurons: a role in the induction of LH surge.

We tested the hypothesis that a decrease in hypothalamic inhibitory opioid tone produced by naloxone (NAL) will activate neuropeptide-Y (NPY) neurosecretion in 17 beta-estradiol (E2)-primed ovariectomized (ovx) rats. NPY neurosecretion was assessed in two ways. First, we studied the effects of iv saline (controls) or NAL infusion (2 mg/h) between 1100-1400 h on NPY concentrations in seven microdissected sites in the medial basal hypothalamus (MBH) and preoptic area in association with the increase in the rate of LH secretion, and then we examined the effects of NAL on the in vitro release of N

[The mode of action of neurohormones on the central nervous system (author's transl)].

We tried to understand how hypophysiotropic polypeptides could be carried into some areas of the central nervous system. Granulations of neurosecretion in the ependymal cells and the presence of releasing hormones in the cerebrospinal fluid (CSF) suggest that CSF may transport these substances between the hypothalamus and other areas in the brain. Direct connections between the hypothalamus and other parts of the brain through secreting neurons are not necessary, the CSF transmitting to the cerebral hemispheres the hypothalamic polypeptides by the ventricles at the infundibulum.

The variation of the amount of the neurosecretory materials in the Corpus Allatum of Pimpla turionellae L. (Hymenoptera: ichneumonidae) during oocyte maturation.

The purpose of this study is to examine the effects of neurosecretory material (NSM) in Corpus Allatum (CA) of endoparasitic Pimpla turionellae L. on reproduction. For this purpose, the egg maturation of the insect and the amount of the NSM in CA of the serial section of the brain were studied. Egg maturation was determined by measuring the terminal oocyte length. In order to determine the amount of the NSM in CA the neurosecretory granules of which were stained by paraldehyde fuchsin are considered. The egg maturation of P. turionellae have performed twice. The NSM intensity in CA continued i

A signaling loop of REST, TSC2 and β-catenin governs proliferation and function of PC12 neural cells.

The RE-1-specific silencing transcription factor (REST or NRSF) is a transcription repressor that orchestrates differentiation and also operates in differentiated neurons and neurosecretory cells (neural cells). Its role in proliferation has been investigated so far only in rapidly growing tumors, with conflicting results: suppression in non-neural tumors, stimulation in medulloblastomas. Working with two clones of chromaffin-neuronal PC12 cells, which express different levels of REST, and using genetic complementation and knockdown approaches, we show that REST also promotes proliferation in

Mitochondrial complex III: an essential component of universal oxygen sensing machinery?

Oxygen is necessary for the survival of mammalian cells. In order to maintain adequate cellular oxygenation, mammals have evolved multiple acute and long-term adaptive responses to hypoxia. These include hypoxic increases in erythropoiesis, pulmonary vasoconstriction and carotid body neurosecretion. Collectively, these responses help maintain oxygen homeostasis as oxygen levels remain scarce. There are multiple effectors proposed to underlie these diverse responses to hypoxia including PHD2, AMPK, NADPH oxidases, and mitochondrial complex III. Here I propose a model wherein complex III is inte

Effect of CO on VO2 of carotid body and chemoreception with and without Ca2+.

This study was done using high PCO (> 500 Torr at PO2 of 120 Torr) in the carotid body perfusate in vitro, and recording simultaneously the activity of the whole carotid sinus nerve (CSN) and VO2 of the carotid body. In the cascade of excitation of CSN by high PCO in the dark [light eliminated the excitation; S. Lahiri, News Physiol. Sci. 9 (1992) 161-165], Ca2+ effects occur at the level of neurosecretion after the level of oxygen consumption, according to the following scheme: CO-hypoxia-->VO2 decrease-->K+ conductance decrease-->cell depolarization-->cytosolic Ca2+ rise-->neurosecretion-->n

Naloxone stimulates comparable release of luteinizing hormone-releasing hormone from tissue fragments from ovariectomized, estrogen-treated young and middle-aged female rats.

Endogenous opioids play a role in the regulation of LH-RH neurosecretion throughout the estrous cycle and during the preovulatory LH surge on proestrus. Experimental evidence suggests that opioid influence may be altered with age, and it has been hypothesized that these alterations may contribute to the loss of regular estrous cyclicity in aging female rats. The present study utilizes an in vitro perifusion paradigm to compare the ability of opiate receptor blockade with naloxone to stimulate LH-RH release from tissue fragments from ovariectomized, estrogen-treated young and middle-aged female

Substance P immunoreactivity exhibits frequent colocalization with kisspeptin and neurokinin B in the human infundibular region.

Neurons synthesizing neurokinin B (NKB) and kisspeptin (KP) in the hypothalamic arcuate nucleus represent important upstream regulators of pulsatile gonadotropin-releasing hormone (GnRH) neurosecretion. In search of neuropeptides co-expressed in analogous neurons of the human infundibular nucleus (Inf), we have carried out immunohistochemical studies of the tachykinin peptide Substance P (SP) in autopsy samples from men (21-78 years) and postmenopausal (53-83 years) women. Significantly higher numbers of SP-immunoreactive (IR) neurons and darker labeling were observed in the Inf of postmenopau

[Mechanism of hyocorticosteronemia action of Strongyloides ratti in the rat].

In order to explain the hypocorticosteronemia which establishes itself in rats with Strongyloides ratti parasites, we are proceeding with a functional exploration of the hypothalamo-hypophyso-corticosurrenal axis in experimentally infested rodents. The adreno-cortico-tropic-hormone test (A.C.T.H.), stress to ether and the Lysin-8-Vasopressin and Metopirone tests lead to concordant results: -- the hypothalamo-pituitary-corticosurrenal present a functional integrity which allows it to react normally to direct stimulation or to feed-back; -- the deficiency seems to relate to the hypothalamic secr

Vasculature of the Suprachiasmatic Nucleus: Pathways for Diffusible Output Signals.

Transplant studies demonstrate unequivocally that the suprachiasmatic nucleus (SCN) produces diffusible signals that can sustain circadian locomotor rhythms. There is a vascular portal pathway between the SCN and the organum vasculosum of the lamina terminalis in mouse brain. Portal pathways enable low concentrations of neurosecretions to reach specialized local targets without dilution in the systemic circulation. To explore the SCN vasculature and the capillary vessels whereby SCN neurosecretions might reach portal vessels, we investigated the blood vessels (BVs) of the core and shell SCN. T

Effects of low glucose on carotid body chemoreceptor cell activity studied in cultures of intact organs and in dissociated cells.

The participation of the carotid body (CB) in glucose homeostasis and evidence obtained in simplified cultured CB slices or dissociated cells have led to the proposal that CB chemoreceptor cells are glucoreceptors. However, data generated in intact, freshly excised organs deny CB chemoreceptor cells' glucosensing properties. The physiological significance of the contention has prompted the present study, performed in a newly developed preparation of the intact CB organ in culture that maintains chemoreceptor cells' microenvironment. Chemoreceptor cells of intact CBs in culture retained their c

Ultrastructure of the neurohypophysis.

This review summarizes our current knowledge of the ultrastructure of the human neurohypophysis and includes comments on its anatomy, physiology, and embryology. The neurohypophysis represents a unique tissue having neural and endocrine characteristics and possessing ultrastructural features distinct from those of conventional endocrine organs such as the anterior pituitary, thyroid, pancreatic islets, etc. In contrast to these glands, the neurohypophysis is composed of the processes of mature neurons. As such, it is not capable of synthesizing hormones but only of their storage and release. N

Novel cell types, neurosecretory cells, and body plan of the early-diverging metazoan Trichoplax adhaerens.

BACKGROUND: Trichoplax adhaerens is the best-known member of the phylum Placozoa, one of the earliest-diverging metazoan phyla. It is a small disk-shaped animal that glides on surfaces in warm oceans to feed on algae. Prior anatomical studies of Trichoplax revealed that it has a simple three-layered organization with four somatic cell types. RESULTS: We reinvestigate the cellular organization of Trichoplax using advanced freezing and microscopy techniques to identify localize and count cells. Six somatic cell types are deployed in stereotyped positions. A thick ventral plate, comprising the m

An analytical study of the neurosecretory cells of the brain of the mature larvae and diapause pupae of Amsacta collaris Hampson (Lepidoptera: Arctiidae) under different temperature regimes.

The neurosecretory cells (NSCs) of the brain of the mature larvae and 3 months old diapause pupae of Amsacta collaris Hampson (Lepidopera: arctiidae) have been studied under different temperature regimes (0, 7, 18, 37, and 42 degrees C), using paraldehyde fuchsin and performic acid resorcin fuchsin staining techniques. The 0 degree C is fatal to the larvae, and at 7 degrees C, within 24 h the quantity of NSM reduced considerably in all the subtypes of A-cells; activity started 3rd day onwards, and on 5th day, the A1-cells appeared loaded with NSM, the A2-cells moderately filled, but no change

Synganglial and neurosecretory morphology of the chicken mite Dermanyssus gallinae (DeGeer) (Mesostigmata: Dermanyssidae).

Histological techniques and paraldehyde-fuchsin (PAF) staining were used to study the synganglion and to locate neurosecretory regions and neurosecretion within the synganglion of the chicken mite, Dermanyssus gallinae. The synganglion, which is formed internally by neuropilar ganglia, gives rise to a single esophageal and paired cheliceral, palpal, pedal (I-IV), and opisthosomal nerves. The neuropilar ganglia are interconnected by commissures and connectives within the synganglion. Twelve PAF-positive neurosecretory regions are present in unfed protonymphs, unfed deutonymphs, virgin males and

Distribution of opioid peptides in the pituitary: a new hypothalamic-pars nervosa enkephalinergic pathway.

The distribution of the endorphins, beta-endorphin and enkephalin (Met5-enkephalin and Leu5-enkephalin), was determined in the pars distalis, intermedia, and nervosa of the rat pituitary using both immunocytochemical and radioimmunological methods. Immunoreactive (ir) beta-endorphin was found in pars distalis and pars intermedia. On gel filtration of the pars distalis extracts, beta-endorphin immunoreactivity was eluted in three peaks corresponding to pro-opiocortin (5%), beta-lipotropin (75%), and beta-endorphin (20%). beta-Endorphin was the only component in the pars intermedia.
