The influence of Nematospiroides dubius on subsequent Nippostrongylus brasiliensis infections in mice.

The fecundity and longevity of Nippostrongylus brasiliensis was prolonged in mice previously infected with Nematospiroides dubius only when the former developed from the larval stage in those mice. Such worms appeared to be less immunogenic than worms which developed in mice never exposed to N. dubius. It is proposed that prolonged fecundity and longevity resulted from an adaptation undertaken by the worms in the face of host antibodies which had been developed against the pre-existing N. dubius infection.

Protective immunity to Nippostrongylus brasiliensis in the rat. II. Adaptation by worms.

Adapted worms are the adult worms found in immune rats reinfected with Nippostrongylus brasiliensis. They are adapted because, when transferred from the small intestines of donor rats to the intestines of normal rats, they persist for a much longer period than normal worms transferred from initially infected donors. When transferred, the internal structure of adapted worms is disorganized and they are permanently stunted but these factors do not account for their longevity in this experimental situation. The adaptation occurs during the larval stages of parasitic life beginning at the L3/L4 mo

Microtopography of Nippostrongylus brasiliensis (Nematoda: Heligmosomatidae): parasitic larval stages and adults.

Specimens of the rat hookworm, Nippostrongylus brasiliensis (Nematoda) were recovered from lungs (third- and fourth-stage larvae) and intestine (fourth-stage larvae and adults). The following features were studied in the different stages by scanning electron microscopy: cephalic structures, especially sense organs, synlophe, cervical region, and caudal part. The main differences between the third and fourth stages concerned the lip-like structures around the oral aperture, the appearance of the cephalic space with the presence of a cephalic cap in fourth-stage larvae, the pattern of longitudin

Nippostrongylus (Nematoda) in protein-malnourished rats: host mortality, morbidity and rehabilitation.

The synergistic relationship between host nutrition and the effects of Nippostrongylus was investigated experimentally in rats fed a diet containing 1% (w/w) protein. Rats were infected with 10 doses of N. brasiliensis varying from 0.35 to 99 larvae/g and their daily food intake and weekly body mass were compared with those uninfected rats fed the same diet for a period of 9 weeks. The deterioration of the uninfected rats observed as a result of protein-energy malnutrition was found to be accelerated in a dose-dependent manner in the rats infected with N. brasiliensis. Analysis of food intake

Immunologic properties of mast cells from rats infected with Nippostrongylus brasiliensis.

The concentration of IgE in the serum of Sprague-Dawley rats increased after infection with Nippostrongylus brasiliensis (NB). The IgE concentration in normal rats was less than 1 mug/ml. After re-infection with NB, the concentration increased in 100 to 300 mug/ml. Mast cells were purified from peritoneal cells of both normal and NB-infected animals. Purified mast cells from the infected animals released histamine upon exposure to NB antigen. The antibody specific for IgE released histamine from purified mast cells of both normal and infected animals. Dose-reponse curves of histamine release s

Antecedent Nippostrongylus infection alters the lung immune response to Plasmodium berghei.

In endemic regions, it is not uncommon for patients to be co-infected with soil-transmitted helminths and malaria. Although both malaria and many helminth species use the lungs as a site of development, little attention has been paid to the impact that pulmonary immunity induced by one parasite has on the lung response to the other. To model the consequences of a prior hookworm exposure on the development of immunity to malaria in the lungs, mice were infected with Nippostrongylus brasiliensis and 2 weeks later challenged with Plasmodium berghei. We found that a pre-existing hookworm-induced t

Partial cross-resistance between Strongyloides venezuelensis and Nippostrongylus brasiliensis in rats.

Rats were immunized through an initial infection with 1,000 filariform larvae (L3) of Nippostrongylus brasiliensis and after complete expulsion of worms they were challenged with 1,000 L3 of Strongyloides venezuelensis to investigate whether cross-resistance developed against a heterologous parasite. Nippostrongylus brasiliensis-immunized rats developed a partial cross-resistance against S. venezuelensis migrating larvae (MSL3) in the lungs and adult worms in the small intestine. The population of MSL3 in the lungs were significantly lower (P < 0.05) in immunized rats (22.0 +/- 7.4) compared w

Endocrine responses of protein-malnourished rats infected with Nippostrongylus brasiliensis (Nematoda).

During the course of a primary infection of Nippostrongylus brasiliensis in protein-malnourished rats, plasma concentrations of corticosterone were found to decrease from 0.89 +/- 0.03 at the start of the infection to 0.28 +/- 0.08 mumol/l, 9 days post-infection (p.i.). Similarly, adreno-corticotrophic hormone levels were also observed to fall from 62.09 +/- 12.06 to 20.54 +/- 1.81 by 5 days p.i. and to 35.12 +/- 16.61 nmol/l by 9 days p.i. However, no significant changes were seen in the dry weight of the adrenal glands other than those expected in relation to changes in body weight. Concentr

FVB/N mice are highly resistant to primary infection with Nippostrongylus brasiliensis.

Nippostrongylus brasiliensis larvae are particularly susceptible to immunological attack during the pre-lung stage of primary and secondary infections in mice. Whilst most of the common laboratory strains of mice are permissive hosts for the parasite, in this study we report for the first time, the strong resistance of naive FVB/N mice to N. brasiliensis. Damage to larvae is evident within the first 24 h of infection and this may be critical to later larval development and reproductive success. Inflammatory responses in the skin, and larval escape from this tissue were comparable in susceptibl

The effect of vaccination with a recombinant Nippostrongylus brasiliensis acetylcholinesterase on infection outcome in the rat.

Nippostrongylus brasiliensis, the rodent hookworm, is a commonly used model of gastrointestinal nematode infection. This parasite, hookworms and several livestock nematode parasites of importance secrete distinct forms of acetylcholinesterases (AChE) that have been ascribed a putative parasite protective function. We tested the hypothesis that vaccination with the secreted enzyme would be deleterious to the parasite. Rats were immunised with a recombinant AChE isoform B via the subcutaneous, intra-peritoneal and intra-nasal routes using different adjuvants dependent on the mode of delivery and

IgE antibody responses in bronchoalveolar spaces of rats infected with Nippostrongylus brasiliensis.

IgE levels in the bronchoalveolar lavage fluids (BALF) of rats increased significantly following infection with Nippostrongylus brasiliensis. This increase corresponded with a concurrent increase in serum IgE levels. However, a comparison of IgE to albumin ratio in both BALF and serum suggested local accumulation and/or production of IgE in the bronchoalveolar spaces rather than leakage from serum. Subsequent analysis of BALF showed presence of heat-labile PCA activity with highest anti-worm titer (1:64) on Days 11-16 postinfection (pi). Secondary infection resulted in up to a fourfold increas

Protection of rats against nippostrongylus brasiliensis with worm antigens by oral administration.

Different types of antigens collected from adult Nippostrongylus brasiliensis were used to vaccinate rats against this parasite: somatic worm homogenate, secretory antigens and whole fixed worms. After challenge infection there was a significant reduction of worm number in immunized rats. However, vaccination with secretory antigens only induced a high level of immunity, indicating that protective antigens were probably worm metabolites.

The cellular transfer to immunity to Nippostrongylus brasiliensis in inbred rats (Lewis strain).

Mesenteric lymph node cells obtained from highly inbred donor rats (Lewis strain), resistant to Nippostrongylus brasiliensis infection, were syngeneically transferred by intravenous injection into previously uninfected recipients. The adoptively immunized recipients were then challenged with either 1500 or 3000 third stage N. brasiliensis larvae on the day of cell transfer. The degree of resistance transferred was assessed by monitoring daily faecal egg output, differential worm burdens on days 6 and 10 of infection and the number of eggs per uterus in gravid worms. The syngeneic transfer of 1
