Systemic induction of a Capsicum chinense nitrate reductase by the infection with Phytophthora capsici and defence phytohormones.

The mRNA differential display technique was used to identify genes from Habanero pepper (Capsicum chinense Jacq.) seedlings whose expression is modified systemically by infection with the oomycete Phytophthora capsici L. Experiments with different oligonucleotide primer combinations revealed that no single gene was synthesised de novo. Instead, the quantitative accumulation of multiple transcripts was found. From these transcripts, levels of a nitrate reductase (Capsicum chinense nitrate reductase, CcNR), which has a high percentage of identity with other Solanaceae NRs, showed a consistent in

[Multicenter evaluation of the indirect nitrate reductase assay for the rapid detection of multidrug-resistant tuberculosis].

Multidrug-resistant tuberculosis (MDR-TB) is defined as resistance to at least isoniazid (INH) and rifampicin (RIF), and it complicates the implementation of tuberculosis control programmes. The rapid detection of MDR-TB is crucial to reduce the transmission of disease. The nitrate reductase assay (NRA) is one of the colorimetric susceptibility test methods for rapid detection of MDR-TB and based on the ability of reduction of nitrate to nitrite by Mycobacterium tuberculosis. The aim of this study was to evaluate the performance of the NRA for the rapid detection of MDR-TB. A total of 237 M.tu

Differential regulatory role of nitric oxide in mediating nitrate reductase activity in roots of tomato (Solanum lycocarpum).

BACKGROUND AND AIMS: Nitric oxide (NO) has been demonstrated to stimulate the activity of nitrate reductase (NR) in plant roots supplied with a low level of nitrate, and to affect proteins differently, depending on the ratio of NO to the level of protein. Nitrate has been suggested to regulate the level of NO in plants. This present study examined interactive effects of NO and nitrate level on NR activity in roots of tomato (Solanum lycocarpum). METHODS: NR activity, mRNA level of NR gene and concentration of NR protein in roots fed with 0.5 mM or 5 mM nitrate and treated with the NO donors,

Involvement of nitrate reductase in nitric oxide generation and in the induction of acclimation responses to phosphorus restriction in soybean plants.

Nitrate reductase (NR) is an essential enzyme because of its role in nitrogen metabolism and in key signaling events through the generation of the reactive nitrogen species, nitric oxide (NO). In this work, we evaluated changes in endogenous NO levels during the onset of P-restriction in soybean plants (Glycine max), focusing on the possible pathways involved in its generation, namely NR and NO synthase like activity, NOSlike, and the subsequent role of NR during low P-acclimation. During the first 96 h of P-starvation NO levels increased in the leaves. The treatment of plants with L-NAME (a k

Nitrate, nitrite and nitric oxide reductases: from the last universal common ancestor to modern bacterial pathogens.

The electrochemical gradient that ensues from the enzymatic activity of cytochromes such as nitrate reductase, nitric oxide reductase, and quinol oxidase contributes to the bioenergetics of the bacterial cell. Reduction of nitrogen oxides by bacterial pathogens can, however, be uncoupled from proton translocation and biosynthesis of ATP or NH4(+), but still linked to quinol and NADH oxidation. Ancestral nitric oxide reductases, as well as cytochrome c oxidases and quinol bo oxidases evolved from the former, are capable of binding and detoxifying nitric oxide to nitrous oxide. The NO-metabolizi

Post-Translational Control of Nitrate Reductase Under Elevated CO2 in Solanum lycopersicum: Carbon-Nitrogen Signaling and Photosynthetic Acclimation.

Rising atmospheric CO2 is altering carbon-nitrogen interactions in C3 crops, with tomato (Solanum lycopersicum L.) showing enhanced carbon assimilation but frequently reduced nitrogen acquisition and assimilation. Nitrate reductase (NR), the rate-limiting enzyme in nitrate reduction, plays a central role by integrating nitrate assimilation with carbon metabolism and nitric oxide (NO) signaling. This review summarizes current knowledge of NR regulation in tomato under elevated CO2 (eCO2), focusing on post-translational mechanisms and their contribution to photosynthetic acclimation. Elevated CO

NIN-like protein 2 regulation of nitrite reductase mediates vacuole integrity in Medicago truncatula nodules under high nitrate.

The NIN-like protein 2 (NLP2) transcription factor is highly expressed in the infected cells of the N2 fixation zone of mature nodules in Medicago truncatula. In these cells, NLP2 directly activates the expression of leghemoglobin genes and is required for the full expression of nitrite reductase (NiR). Histological examination of nodules formed under different nitrate regimes revealed that the infected cells of nlp2 display abnormal vacuole morphology under high nitrate (5 mm KNO3). This phenotype is associated with starch accumulation, higher expression of starch biosynthesis genes, and incr

EPR-detected NO biosynthesis via nitrate reductase in non-climacteric pepper fruit: downregulation by ripening, peroxynitrite and H2S.

This study presents the first direct evidence of nitric oxide (•NO) production in ripening pepper fruits (Capsicum annuum L.) and identifies the specific enzyme responsible for its generation. By clarifying the enzymatic origin of •NO, this research enhances our understanding of the redox signaling mechanisms involved in the development of non-climacteric pepper fruits. Using electron paramagnetic resonance (EPR) spectroscopy combined with a spin-trapping technique, •NO production was directly detected at various stages of ripening. Enzymatic assays confirmed that the nitrite- and NADH (reduce

Nitrate reductase-mediated NO production enhances Cd accumulation in Panax notoginseng roots by affecting root cell wall properties.

Panax notoginseng (Burk) F. H. Chen is a traditional medicinal herb in China. However, the high capacity of its roots to accumulate cadmium (Cd) poses a potential risk to human health. Although there is some evidence for the involvement of nitric oxide (NO) in mediating Cd toxicity, the origin of Cd-induced NO and its function in plant responses to Cd remain unknown. In this study, we examined NO synthesis and its role in Cd accumulation in P. notoginseng roots. Cd-induced NO production was significantly decreased by application of the nitrate reductase inhibitor tungstate but not the nitric o

Nitrate Reductase Genes AtNIA1 and AtNIA2 Confer Heat Stress Resilience via ROS Homeostasis and HSP Expression in Arabidopsis.

Heat stress is a key environmental factor that adversely affects plant growth, development, and productivity. Nitrate reductase (NR), encoded by AtNIA1 and AtNIA2, plays a crucial role in nitric oxide (NO) biosynthesis, which mediates stress responses in plants. In this study, we investigated the roles of AtNIA1 and AtNIA2 in regulating plant heat stress tolerance. Under heat stress conditions, Arabidopsis thaliana plants maintained higher relative water content and chlorophyll levels, whereas atnia1 and atnia2 mutants exhibited greater physiological damage. Oxidative stress markers such as MD

How iron-bearing minerals affect the biological reduction of Sb(V): A newly discovered function of nitrate reductase.

As a toxic element of global concern, the elevated concentration of antimony (Sb) in the environment has attracted increasing attention. Microorganisms have been reported as important driving forces for Sb transformation. Iron (Fe) is the most important metal associated element of Sb, however, how Fe-bearing minerals affect the biological transformation of Sb is still unclear. In this study, the effects of Fe-bearing minerals on biological Sb(V) reduction were investigated by employing a marine Shewanella sp. CNZ-1 (CNZ-1). Our results showed that the presence of hematite, magnetite and ferrih

Magnesium-induced transporters remodel the ionome of mulberry leaves by promoting the nitrate reductase (NIA) gene in nitrogen metabolism.

Magnesium (Mg) is fundamental for diverse physiological, biochemical, and molecular processes in higher plants because its deficiency or toxicity severely restricts development. The supply of Mg remodels the plant ionome and transporter profiles while simultaneously activating the nitrate reductase enzyme, which catalyzes nitrate reduction to nitrite and enhances nitrogen (N) assimilation. However, the specific mechanisms involving ion interactions and transporters in Mg metabolism that improve N metabolism in mulberry remain largely unexplored. To address this research gap, we employed physio

Nitrate reductase activity of needles of Norway spruce fumigated with different mixtures of ozone, sulfur dioxide, and nitrogen dioxide.

Four-year-old spruce clones (Picea abies (L.) Karst.) cultivated in sand and provided with a complete nutrient solution, or a solution deficient in magnesium and calcium, were exposed to the pollutant mixtures SO(2)/NO(2), O(3)/NO(2), and O(3)/SO(2)/NO(2), at realistic concentrations for 32 weeks. Fumigation caused a slight increase of total N contents in current year needles, whereas in one-year-old needles N concentrations did not change. The response of nitrate reductase activity to pollutant stress depended on needle age and nutrient supply, respectively. In one-year-old needles fumigation

Nitrate reductase BoNIA2b responds to nitric oxide to positively regulate the accumulation of Glucoraphanin and Sulforaphane in broccoli hairy roots.

Glucoraphanin (GRA) and sulforaphane (SF) are potent anticancer compounds. Understanding their accumulation mechanisms is crucial for increasing their levels in broccoli. This study aimed to investigate the role of nitric oxide (NO) in GRA and SF accumulation and identify key genes involved in this process. Low NO concentrations were found to promote GRA and SF accumulation. Transcriptome sequencing and Weighted Gene Co-expression Network Analysis (WGCNA) identified BoNIA2b, a nitrate reductase (NR) gene, as a key regulator of NO-mediated GRA and SF accumulation. Silencing BoNIA2b reduced endo

Evaluation of direct microplate nitrate reductase assay as a rapid method for the detection of multiple and extensively tuberculosis drug resistance.

INTRODUCTION: Reports of Mycobacterium tuberculosis resistant to multiple drugs are increasing globally and laboratories are becoming increasingly aware of the need for drug susceptibility testing. In recent years, due to the long time required by conventional drug susceptibility testing, new approaches have been proposed for faster detection of drug resistance, such as the nitrate reductase assay, considered fast and inexpensive, making it a good diagnostic tool for low resource countries. OBJECTIVE: The present study proposed a fast direct colorimetric drug susceptibility testing method in

Isolation, purification, and characterization of nitrate reductase from a salt-tolerant Rhodotorula glutinis yeast strain grown in the presence of tungsten.

The salt-tolerant Rhodotorula glutinis yeast strain grew in medium containing nitrate, 1 mM tungsten, and trace amounts of molybdenum (as impurities from the reagents used). Isolation of electrophoretically homogenous preparation of nitrate reductase from the Rh. glutinis cells grown under these growth conditions is described. The isolated nitrate reductase is a molybdenum-containing homodimer with molecular mass of 130 kD, containing 0.177 mol of Mo per mol of the enzyme. The activity of the enzyme is maximal at pH 7.0 and 35-45 degrees C and is inhibited by low concentrations of azide and cy
