Direct nitrogen fixation at the edges of graphene nanoplatelets as efficient electrocatalysts for energy conversion.

Nitrogen fixation is essential for the synthesis of many important chemicals (e.g., fertilizers, explosives) and basic building blocks for all forms of life (e.g., nucleotides for DNA and RNA, amino acids for proteins). However, direct nitrogen fixation is challenging as nitrogen (N₂) does not easily react with other chemicals. By dry ball-milling graphite with N₂, we have discovered a simple, but versatile, scalable and eco-friendly, approach to direct fixation of N₂ at the edges of graphene nanoplatelets (GnPs). The mechanochemical cracking of graphitic C--C bonds generated active carbon spe

Spatiotemporal co-transcriptomics uncovers coordinated rhizobia-legume stress programs supporting nitrogen fixation in Medicago.

Rhizobial nitrogen fixation in legume nodules requires precise spatial and temporal coordination between host and bacterial gene expression. Here, we applied spatial enhanced resolution omics sequencing (Stereo-seq) to profile Medicago nodules across five developmental stages, generating a co-transcriptomic atlas of nodule development. Using a mixed-probe capture strategy, we simultaneously detected host and rhizobial transcripts within the same tissue sections, enabling analysis of host-microbe transcriptional coordination during symbiosis. We identified dynamic changes in host cell compositi

The preprotein translocase YajC mediates biofilm formation and symbiotic nitrogen-fixation in Mesorhizobium huakuii.

UNLABELLED: In bacteria, the auxiliary complex SecDF-YajC participates in the Sec system, facilitating post-translational translocation of preproteins across the membrane through the SecYEG channel. Although YajC homologs are strongly conserved in rhizobial strains and may play important roles in many processes, the functions and mechanisms by which they are involved in the interaction between rhizobia and host legumes are unknown. By constructing Mesorhizobium huakuii yajC mutant, we observed an increase in the strain growth rate, but a reduction in both the biomass and average thickness of t

Elevated nitrate levels inhibit nitrogen fixation in Azotobacter chroococcum via an ammonium-independent pathway.

Nitrate impairs both symbiotic and free-living biological nitrogen fixation (BNF). While nitrate-induced phosphorylation signalling has been implicated in the inhibition of symbiotic BNF, the suppression of free-living BNF has generally been attributed to ammonium generated during nitrate assimilation. However, whether nitrate can inhibit free-living BNF independently of ammonium feedback regulation remains unclear. Here, an ammonium-deregulated mutant of Azotobacter chroococcum (A4) was used to investigate whether nitrate inhibits nitrogen fixation independently of ammonium regulation. Despit

Unveiling full mechanistic picture of Mo-catalysed nitrogen fixation driven by SmI2-H2O system.

Ammonia is an essential chemical that underpins modern agriculture and industry, but the direct conversion of dinitrogen to ammonia under mild conditions remains highly challenging. To this end, molybdenum nitride complexes bearing 1,3-bis(di-tert-butylphosphinomethyl)benzimidazole-2-ylidene pincer ligands have been shown to catalyse ammonia formation from dinitrogen using samarium diiodide and water. However, the catalytic mechanism remains poorly understood owing to limited information on the key intermediates and the solution-state speciation of the samarium reductant. Here we show a full c

Light-Driven Nitrogen Fixation to Ammonia over Aqueous-Dispersed Mo-Doped TiO2 Colloidal Nanocrystals.

Photocatalytic nitrogen fixation to ammonia and nitrates holds great promise as a sustainable route powered by solar energy and fed with renewable energy resources (N2 and H2O). This technology is currently under deep investigation to overcome the limited efficiency of the process. The rational design of efficient and robust photocatalysts is crucial to boost the photocatalytic performance. Widely used bulk materials generally suffer from charge recombination due to poor interfacial charge transfer and difficult surface diffusion. To overcome this limitation, this work explores the use of aque

Railroad derived nitrogen and heavy metal pollution does not affect nitrogen fixation associated with mosses and lichens at a tundra site in Northern Sweden.

Traffic derived nitrogen (N) and heavy metal pollution is a well-known phenomenon, but little explored in otherwise pristine ecosystems such as subarctic tundra. Here, the main source of N input to the ecosystem is via N2 fixation by moss- and lichen-associated bacteria. While inhibitory effects of N deposition on moss-associated N2 fixation have been reported, we still lack understanding of the effects of traffic derived N and heavy metal deposition on this ecosystem function in an otherwise pristine setting. To test this, we established a distance gradient (0-1280 m) away from a metal pollut

Copper causes reduced nitrogen fixation but does not accumulate in the nodules of the legume Lotus japonicus.

Copper can be a soil contaminant at concentrations that are toxic to plants, particularly because of copper-induced oxidative stress. The legume-rhizobia partnership that allows for biological nitrogen fixation is sensitive to oxidative stress, and this study investigates if copper acts directly on the machinery of nitrogen fixation, or indirectly via toxicity to the entire plant. When Lotus japonicus inoculated with its rhizobial partner Mesorhizobium loti was exposed to 300 or 450 µM of copper, biomass was reduced by 30-40% in the shoots, 40-55% in the roots, and 40-60% in the nodules relati

Active nitrogen fixation by Crocosphaera expands their niche despite the presence of ammonium - A case study.

Unicellular nitrogen fixer Crocosphaera contributes substantially to nitrogen fixation in oligotrophic subtropical gyres. They fix nitrogen even when significant amounts of ammonium are available. This has been puzzling since fixing nitrogen is energetically inefficient compared with using available ammonium. Here we show that by fixing nitrogen, Crocosphaera can increase their population and expand their niche despite the presence of ammonium. We have developed a simple but mechanistic model of Crocosphaera based on their growth in steady state culture. The model shows that the growth of Croc

Simultaneous bioprecipitation of cadmium to cadmium sulfide nanoparticles and nitrogen fixation by Rhodopseudomonas palustris TN110.

This study investigated the abilities of a purple non-sulfur bacterium, Rhodopseudomonas palustris TN110 to bioremediate cadmium through the biosynthesis of CdS nanoparticles and to fix nitrogen simultaneously. Under microaerobic-light conditions, R. palustris TN110 synthesized CdS nanoparticles. The produced CdS nanoparticles had a spherical shape and an average size of 4.85 nm. The Fourier transform infrared spectrum of the nanoparticles reveals the carbonyl groups, bending vibrations of the amide I and II bands of proteins, and CN stretching vibrations of aromatic and aliphatic amines. Promoting nitrogen fixation by threonine synthesis in co-culture of Azotobacter vinelandii and Escherichia coli.

Amino acid production directly from N2 is a big challenge in green biomanufacturing. Merging biological nitrogen fixation (BNF) with target amino acid biosynthesis in co-culture system offers a promising solution. Here, we constructed a Nitrogen Fixation-Utilization Coupling (NFUC) system that enabled threonine production directly from N2 and ethanol, by co-culturing engineered Azotobacter vinelandii for BNF and Escherichia coli for threonine biosynthesis. Systematic metabolic engineering increased threonine titer 3.4-fold over the initial co-culture through optimizing NH4+ release, threonine

Silicon Nanoparticles Alter Soybean Physiology and Improve Nitrogen Fixation Potential Under Atmospheric Carbon Dioxide (CO2).

The interactive effects between nano-silicon dioxide (n-SiO2) and elevated CO2 (eCO2; 645 ppm) on soybean physiology, nitrogen fixation, and nutrient dynamics under climate stress remain underexplored. This study elucidates their combined effects under ambient (aCO2; 410 ppm) and eCO2 conditions. eCO2 + n-SiO2 synergistically enhanced shoot length (30%), total chlorophyll (112.15%), and photosynthetic rate (103.23%), alongside improved stomatal conductance and intercellular CO2 (17.19%), optimizing carbon assimilation. Nodulation efficiency increased, with nodule number and biomass rising by 4

Foliar Application with Iron Oxide Nanomaterials Stimulate Nitrogen Fixation, Yield, and Nutritional Quality of Soybean.

Sustainable strategies for the management of iron deficiency in agriculture are warranted because of the low use efficiency of commercial iron fertilizer, which confounds global food security and induces negative environmental consequences. The impact of foliar application of differently sized γ-Fe2O3 nanomaterials (NMs, 4-15, 8-30, and 40-215 nm) on the growth and physiology of soybean seedlings was investigated at different concentrations (10-100 mg/L). Importantly, the beneficial effects on soybean were size- and concentration-dependent. Foliar application with the smallest size γ-Fe2O3 NMs

Growth-promoting Sphingomonas paucimobilis ZJSH1 associated with Dendrobium officinale through phytohormone production and nitrogen fixation.

Growth-promoting Sphingomonas paucimobilis ZJSH1, associated with Dendrobium officinale, a traditional Chinese medicinal plant, was characterized. At 90 days post-inoculation, strain ZJSH1 significantly promoted the growth of D. officinale seedlings, with increases of stems by 8.6% and fresh weight by 7.5%. Interestingly, the polysaccharide content extracted from the inoculated seedlings was 0.6% higher than that of the control. Similar growth promotion was observed with the transplants inoculated with strain ZJSH1. The mechanism of growth promotion was attributed to a combination of phytohorm

Long-Term Warming Suppresses Nitrogen-Fixation Gene Transcription in the Rhizosphere of Invasive Plant Solidago canadensis.

Climate warming can enhance the growth performance of invasive plants, potentially increasing their nitrogen (N) demand. However, whether this additional demand is supported by free-living N fixation remains unclear. Here, we used a 12-year warming experiment to determine how warming affected diazotrophic abundance, community composition, and nifH transcription in the roots and rhizosphere of the invasive plant Solidago canadensis during the growing and flowering stages. While warming increased plant biomass, it did not increase diazotrophic abundance or nifH transcription in either roots or r

Climate Change and Nitrogen-Fixing Legumes: Investigating Stress-Modulated Dynamics of Carbon Fixation and Root Nodulation.

In the Anthropocene era, climate change is increasingly subjecting the crops to overlapping abiotic stressors such as drought, elevated temperatures, and air pollution, thereby disrupting their physiological integrity and functional performance. This review synthesises current knowledge on responses of N2-fixing plants to such stressors, focusing on core physiological processes and symbiotic nitrogen fixation via nodulation. The intricate interdependence between these traits is explored through the lens of altered source-sink relationships, which are highly sensitive to multifactorial environm

Soil water and inorganic nitrogen contents drive soil microbial carbon fixation during wetland reclamation and restoration.

Wetlands are critical terrestrial carbon sinks, playing a vital role in mitigating global warming by fixing substantial atmospheric carbon dioxide. To explore how wetland reclamation and restoration affect soil microbial carbon fixation, we sampled soils from five land-use types in western Jilin Province, China: undisturbed natural reed wetland, reclaimed rice paddy and upland field, restored agricultural drainage wetland and naturally restored wetland after farmland abandonment. We analyzed the effects of wetland reclamation and restoration on microbial communities, metabolic pathways, and ca

Overestimated natural biological nitrogen fixation translates to an exaggerated CO2 fertilization effect in Earth system models.

CO2 fertilization of the terrestrial biosphere is limited by nitrogen. Biological nitrogen fixation (BNF) is the dominant natural nitrogen source to the terrestrial biosphere and can alleviate nitrogen limitation but is poorly constrained in Earth system models (ESMs). Here, we compare terrestrial BNF from an ensemble of ESMs of the 6th Coupled Model Intercomparison Project to a new global synthesis of observations across natural and agricultural biomes. We find that compared to observations, ESMs underestimate agricultural BNF but overestimate natural BNF in the present day by over 50%. Natur

Hydrogenase genes from Rhizobium leguminosarum bv. viciae are controlled by the nitrogen fixation regulatory protein nifA.

Rhizobium leguminosarum bv. viciae expresses an uptake hydrogenase in symbiosis with peas (Pisum sativum) but, unlike all other characterized hydrogen-oxidizing bacteria, cannot express it in free-living conditions. The hydrogenase-specific transcriptional activator gene hoxA described in other species was shown to have been inactivated in R. leguminosarum by accumulation of frameshift and deletion mutations. Symbiotic transcription of hydrogenase structural genes hupSL originates from a -24/-12 type promoter (hupSp). A regulatory region located in the -173 to -88 region was essential for prom
