Sterol Composition in the Lichens Lobaria pulmonaria and Lobaria retigera: Does Photobiont Matter?

The lipid composition of the mycobint and photobiont symbiotic partners of lichenized ascomycetes varies greatly. The aim of this study was to compare the profile of the major sterols in two closely related lichens from the genus Lobaria with different photobionts. The three-component lichen Lobaria pulmonaria has two photobionts. While the main photobiont is the chlorophycean alga Symbiochloris reticulata, this lichen contains small amounts of the cyanobacterium Nostoc. By contrast, the cyanobacterium Nostoc is the main photobiont in Lobaria retigera. Relatively loosely bound sterols were ext

Single-Enzyme Conversion of Tryptophan to Skatole and Cyanide Expands the Mechanistic Competence of Diiron Oxidases.

Skatole is a pungent heterocyclic compound derived from the essential amino acid l-tryptophan by bacteria in the mammalian digestive tract. The four-step anaerobic conversion of tryptophan to skatole is well-established; though, to date, no aerobic counterpart has been reported. Herein, we report the discovery of the oxygen-dependent skatole synthase SktA that single-handedly converts 5-bromo-l-tryptophan to 5-bromoskatole, obviating the need for a multienzyme process. SktA is part of a three-gene biosynthetic gene cluster (BGC) in the cyanobacterium Nostoc punctiforme NIES-2108 and functions

Production of the neurotoxin BMAA by a marine cyanobacterium.

Diverse species of cyanobacteria have recently been discovered to produce the neurotoxic non-protein amino acid beta-methylamino-L-alanine (BMAA). In Guam, BMAA has been studied as a possible environmental toxin in the diets of indigenous Chamorro people known to have high levels of Amyotrophic Lateral Sclerosis/ Parkinsonism Dementia Complex (ALS/PDC). BMAA has been found to accumulate in brain tissues of patients with progressive neurodegenerative illness in North America. In Guam, BMAA was found to be produced by endosymbiotic cyanobacteria of the genus Nostoc which live in specialized cyca

Interaction of cyanobacteria and microplastics polystyrene spiked with pharmaceutical drug-paracetamol.

The microplastics polymer-polystyrene, polyethylene, polyvinyl chloride, etc. are now recognized as potent threats to the aquatic system due to the Trojan horse effect i.e., they adsorb other pollutants such as pharmaceutical drugs, organic solvents, metals etc. and act as a vector or carrier. Polystyrene (PS) is one of the most usable plastics worldwide that abundantly contaminates the aquatic body. However, to date, only a few studies have focused on the eco-toxic effects of polystyrene in combination with other pollutants. Therefore, in the present study, the effect of polystyrene (pristine

2-Deoxy-4-epi-scyllo-inosose (DEI) is the Product of EboD, a Highly Conserved Dehydroquinate Synthase-like Enzyme in Bacteria and Eustigmatophyte Algae.

A cryptic cluster of genes, known as the ebo cluster, has been found in a variety of genomic contexts among bacteria and algae. In Pseudomonas fluorescens NZI7, the ebo cluster (a.k.a. EDB cluster) is involved in the bacterial repellent mechanism against nematode grazing. In cyanobacteria, the cluster plays a role in the transport of the scytonemin monomer from the cytosol to the periplasm. Despite their broad distribution and interesting phenotypes, neither the pathway nor the functions of the enzymes are known. Here we show that EboD proteins from the ebo clusters in Nostoc punctiforme and S

Sulfur is in the Air: Cyanolichen Marriages and Pollution.

Cyanolichens are symbiotic organisms involving cyanobacteria and fungi (bipartite) or with the addition of an algal partner (tripartite). Cyanolichens are known for their heightened susceptibility to environmental pollution. We focus here on the impacts on cyanolichens due to rising air pollution; we are especially interested in the role of sulfur dioxide on cyanolichen biology. Cyanolichens due to air pollution including sulfur dioxide exposure, show symptomatic changes including degradation of chlorophyll, lipid membrane peroxidation, decrease in ATP production, changes in respiration rate,

An automated high-throughput lighting system for screening photosynthetic microorganisms in plate-based formats.

The capacity of photosynthetic microorganisms to fix carbon dioxide into biomass positions them as promising cell factories for sustainable biomanufacturing. However, limitations in screening throughput hinder the identification of enzymes, strains, and growth conditions needed to realize this potential. Here we present a microplate-based high-throughput cultivation system that can be integrated into existing automation infrastructure and supports growth of both prokaryotic and eukaryotic photosynthetic microorganisms. We validate this system by optimizing BG-11 medium compositions for Synecho

Mechanisms of sludge reduction in Nostocoides-driven enhanced biological phosphorus removal process.

Nostocoides, a key phosphate-accumulating organism (PAOs), is abundant in wastewater treatment plants (WWTPs); however, the mechanism of sludge reduction in biological phosphorus removal systems under its dominance remains largely unknown. This study employed a lab-scale, exploratory multiparameter approach to examine extracellular polymeric substance (EPS) profiles, intracellular metabolites, microbial community, membrane integrity, and physiological activity to elucidate how Nostocoides-enriched enhanced biological phosphorus removal (EBPR) configurations accomplished sludge reduction. Native Metabolomics Unveils Suomilide Analogs with Potent Trypsin Inhibitory Activity.

Cyanobacteria are well-known for their ability to produce a diverse array of serine protease inhibitors and thus are a promising source of pharmaceutical lead structures. Here, native metabolomics was employed with trypsin to rapidly screen whole cyanobacterial metabolomes and uncover new potent inhibitors. In combination with subsequent correlative LC-HR-MS/MS-based analysis, this led to the fast discovery of the well-characterized natural product family of suomilides from Nostoc sp. KVJ20 as highly potent trypsin inhibitors. Notably, two of the six putative binders were recognized as potenti

Cyanobacterial Polyhydroxybutyrate (PHB): Screening, Optimization and Characterization.

In modern life petroleum-based plastic has become indispensable due to its frequent use as an easily available and a low cost packaging and moulding material. However, its rapidly growing use is causing aquatic and terrestrial pollution. Under these circumstances, research and development for biodegradable plastic (bioplastics) is inevitable. Polyhydroxybutyrate (PHB), a type of microbial polyester that accumulates as a carbon/energy storage material in various microorganisms can be a good alternative. In this study, 23 cyanobacterial strains (15 heterocystous and 8 non-heterocystous) were scr

Selective human trypsin inhibitors from cyanobacteria.

AIMS: Aberrant activity of human trypsin isoenzymes has been linked to diseases like cancer. The high structural and catalytic similarity among the three human trypsin isoenzymes makes selective inhibition of individual trypsin isoenzymes particularly challenging. As cyanobacteria are a prolific source of structurally diverse serine protease inhibitors with potential pharmaceutical applications, we studied them as producers of selective human trypsin inhibitors. METHODS AND RESULTS: Here, we evaluated aqueous extracts of 505 strains of cyanobacteria, isolated from the Baltic Sea and Finnish l

The Effect of Abiotic Factors on Abundance and Photosynthetic Performance of Airborne Cyanobacteria and Microalgae Isolated from the Southern Baltic Sea Region.

Cyanobacteria and microalgae present in the aquatic or terrestrial environment may be emitted into the air and transported along with air masses over long distances. As a result of staying in the atmosphere, these organisms may develop a greater tolerance to stressful factors, but this topic is still relatively unknown. The main aim was to show an autecological characteristic of some airborne microalgae and cyanobacteria strains by a factorial laboratory experiment approach, including changes in irradiance, temperature, and salinity conditions. The additional purpose of this work was also to p

Aeruginosin 525 (AER525) from Cyanobacterium Aphanizomenon Sp. (KUCC C2): A New Serine Proteases Inhibitor.

Aeruginosins (AERs) are one of the most common classes of cyanobacterial peptides synthesised through a hybrid non-ribosomal peptide synthase/polyketide synthase pathway. They have been found in Microcystis, Nodularia spumigena, Oscillatoria/Plantothrix, and Nostoc. The presence of AER in Aphanizomenon isolated from the Curonian Lagoon was reported for the first time in our previous work. Here, the structure of aeruginosin 525 (AER525), isolated from Aphanizomenon sp. KUCC C2, was characterised based on high-resolution mass spectrometry. This new AER variant shows potent activity against throm

Biodegradation and utilization of organophosphorus pesticide malathion by Cyanobacteria.

Three strains of filamentous Cyanobacteria were used to study their growth and utilization of organophosphorus pesticide malathion. A sharp decrease in the growth of the algal strains was observed by increasing the concentration of malathion. Amongst them Nostoc muscorum tolerated different concentrations and was recorded as the highest efficient strain for biodegradation (91%) of this compound. Moreover, carbohydrate and protein content of their cells overtopped the other strains especially at higher concentrations. The algal strains were further subjected to grow under P-limitation in absenc
