Interaction of TACC3 and TSC2 at the nuclear envelope and mitotic structures.

Unspecified kidney donors (UKDs) are approached cautiously by some transplant professionals. The aim of this study was to interrogate the views of UK transplant professionals towards UKDs and identify potential barriers. A purposely designed questionnaire was validated, piloted and distributed amongst transplant professionals at each of the 23 UK transplant centres. Data captured included personal experiences, attitudes towards organ donation, and specific concerns about UKD. 153 responses were obtained, with representation from all UK centres and professional groups. The majority reported a p

The role of the nuclear envelope in Emery-Dreifuss muscular dystrophy.

The X-linked form of Emery-Dreifuss muscular dystrophy (X-EDMD) is caused by absence, or greatly reduced amounts, of the inner nuclear-membrane protein, emerin. The autosomal dominant form (AD-EDMD) is caused by missense mutations in lamins A and C, two components of the nuclear lamina that interact directly with emerin. Lamin A/C mutations also cause one form of dilated cardiomyopathy (CMD1A) and one form of limb-girdle muscular dystrophy (LGMD1B), both of which have clinical features in common with EDMD, as well as a rare, unrelated form of lipodystrophy (FPLD). Evidence is now emerging that

Aluminum exposure impairs nuclear envelope breakdown for mouse zygote formation.

Aluminum exposure ubiquitously presents in the environment and its toxicity elicits oxidative stress and metabolic disorders. In the present study, we reported that aluminum exposure disturbed zygote formation with mouse model. We found that aluminum-exposed mouse zygotes exhibited nuclear envelope breakdown defects, indicative of failed zygote development. Further analyses revealed that microtubules decreased with aberrant tubulin acetylation, and actin dynamics was disturbed, indicating the cytoskeleton impairment. This further leads to DNA damage and disturbed cell cycle in zygotes. Moreove

Nuclear envelope-limited chromatin sheets are part of mitotic death.

Nuclear envelope-limited chromatin sheets (ELCS) are enigmatic membranous structures of uncertain function. This study describes the induction of ELCS in p53 mutated Burkitt's lymphoma cell lines after treatment with irradiation or the microtubule inhibitor, SK&F 96365. Both treatments evoked similar mitotic death, involving metaphase arrest followed by extensive endopolyploidisation and delayed apoptosis, although the kinetics were different. We found that induction of ELCS and nuclear segmentation correlated with the amount and kinetics of M-phase arrest, mitosis restitution and delayed apop

Weakening the nuclear envelope: Lamin B receptor in melanoma metastasis.

Nuclear envelope fragility has been identified as a hallmark of the metastatic process. Indeed, cancer cells undertake a challenging journey through diverse environments during metastasis, notably squeezing through narrow spaces and dense collagen matrices. In their report, Baird et al. identify a key player in nuclear fragility in confined cancer cell migration: the Lamin B receptor. Its overexpression and clustering in specific regions of the nuclear envelope increase nuclear distortion and fragility, rendering nuclei more susceptible to rupture with consequent chromatin extrusion and genomi

Nuclear envelope rupture and resealing: mechanisms, consequences, and disease implications.

The nuclear envelope (NE) establishes an essential physical separation between the nucleoplasm and cytoplasm in eukaryotic cells. Its structural integrity ensures selective molecular exchange through nuclear pores, a fundamental feature of eukaryotic cells. For this reason, it has long been assumed that the NE remains intact. However, recent studies revealed localized rupture of the NE in diverse physiological and pathological contexts. These ruptures can be resealed by endogenous mechanisms, suggesting that rupture-resealing dynamics contribute to homeostasis and are altered in disease. In th

Nuclear envelope budding inhibition slows down progerin-induced aging process.

Progerin causes Hutchinson-Gilford progeria syndrome (HGPS), but how progerin accelerates aging is still an interesting question. Here, we provide evidence linking nuclear envelope (NE) budding and accelerated aging. Mechanistically, progerin disrupts nuclear lamina to induce NE budding in concert with lamin A/C, resulting in transport of chromatin into the cytoplasm where it is removed via autophagy, whereas emerin antagonizes this process. Primary cells from both HGPS patients and mouse models express progerin and display NE budding and chromatin loss, and ectopically expressing progerin in

TACC3-TSC2 maintains nuclear envelope structure and controls cell division.

Studies of the role of tuberous sclerosis complex (TSC) proteins (TSC1/TSC2) in pathology have focused mainly on their capacity to regulate translation and cell growth, but their relationship with alterations of cellular structures and the cell cycle is not yet fully understood. The transforming acidic coiled-coil (TACC) domain-containing proteins are central players in structures and processes connected to the centrosome. Here, TACC3 interactome mapping identified TSC2 and 15 other physical interactors, including the evolutionary conserved interactions with ch-TOG/CKAP5 and FAM161B. TACC3 and

Optimized detection of circulating anti-nuclear envelope autoantibodies by immunofluorescence.

BACKGROUND: Antinuclear antibodies are useful diagnostic tools in several autoimmune diseases. However, the routine detection of nuclear envelope autoantibodies using immunofluorescence (IF) is not always easy to perform in patients' sera because of the presence of autoantibodies to other nuclear and cytoplasmic components which could mask the characteristic rim-like pattern of nuclear envelope autoantibodies. This is particularly common in sera from patients with primary biliary cirrhosis (PBC), which generaly have high titres of anti-mitochondrial antibodies. Therefore, we have assayed a num

Autoantibodies against nuclear envelope-associated proteins in primary biliary cirrhosis.

An antinuclear immunofluorescence pattern displaying a thin ring confined to the nuclear envelope was assessed in sera from 38 patients with primary biliary cirrhosis and in sera from a control group of 277 patients with other antinuclear antibody-positive diseases. This rim-like antinuclear reactivity was present in sera from 20 primary biliary cirrhosis patients (52.6%) but in only two patients from the control group (0.7%) (p less than 0.001). Furthermore, this autoantibody was present in three of four primary biliary cirrhosis patients without antimitochondrial antibodies. Presence of this

Nuclear Deformities Minimally Affect Fiber-Type-Specific Disease Progression in Murine Models of Nuclear Envelope Myopathy.

Abnormalities in nuclear morphology are specific features of the nuclear envelopathies, including Emery-Dreifuss muscular dystrophy (EDMD). The presence of abnormally shaped nuclei in the skeletal muscles of EDMD patients and murine models has been reported both in vivo and in vitro; however, how the presence of nuclear architectural abnormalities affects disease development and progression remains unclear. In this study, we analyzed slow-twitch soleus (SOL) muscles and fast-twitch extensor digitorum longus (EDL) muscles from the following EDMD model mice: emerin knockout (Emd), LmnaH222P/H222

Bidirectional increase in permeability of nuclear envelope upon poliovirus infection and accompanying alterations of nuclear pores.

Poliovirus and some other picornaviruses trigger relocation of certain nuclear proteins into the cytoplasm. Here, by using a protein changing its fluorescence color with time and containing a nuclear localization signal (NLS), we demonstrate that the poliovirus-triggered relocation is largely due to the exit of presynthesized nuclear protein into the cytoplasm. The leakiness of the nuclear envelope was also documented by the inability of nuclei from digitonin-permeabilized, virus-infected (but not mock-infected) cells to retain an NLS-containing derivative of green fluorescent protein (GFP). The human cytomegalovirus nuclear egress complex unites multiple functions: Recruitment of effectors, nuclear envelope rearrangement, and docking to nuclear capsids.

BACKGROUND: Nuclear replication represents a common hallmark of herpesviruses achieved by a number of sequentially unrolled regulatory processes. A rate-limiting step is provided by nucleo-cytoplasmic capsid export, for which a defined multiregulatory protein complex, namely, the nuclear egress complex (NEC), is assembled comprising both viral and cellular components. The NEC regulates at least 3 aspects of herpesviral nuclear replication: (1) multimeric recruitment of NEC-associated effector proteins, (2) reorganization of the nuclear lamina and membranes, and (3) the docking to nuclear capsi

Nuclear lipid droplets in Caco2 cells originate from nascent precursors and in situ at the nuclear envelope.

Intestinal epithelial cells convert excess fatty acids into triglyceride (TAG) for storage in cytoplasmic lipid droplets and secretion in chylomicrons. Nuclear lipid droplets (nLDs) are present in intestinal cells but their origin and relationship to cytoplasmic TAG synthesis and secretion is unknown. nLDs and related lipid-associated promyelocytic leukemia structures (LAPS) were abundant in oleate-treated Caco2 but less frequent in other human colorectal cancer cell lines and mouse intestinal organoids. nLDs and LAPS in undifferentiated oleate-treated Caco2 cells harbored the phosphatidate ph

Roles of DNA Damage Response Pathway in the Regulation of the Nuclear Envelope.

The nuclear envelope (NE) functions as a barrier between the cytoplasm and nucleus. Over the past decade, NE has revealed unexpectedly divergent structural alterations. NE rupture triggers the uncontrollable exchange of macromolecules across the NE and potentially causes DNA damage. Conversely, a recent study demonstrated that DNA damage induces NE rupture and that one of the major kinases in the DNA damage response (DDR) pathway, ataxia telangiectasia and Rad3-related protein, ATR, is a key molecule in these events. Here, we review the role of the DDR pathway in NE regulation, with a focus ma

Mislocalization to the nuclear envelope: an effect of the dystonia-causing torsinA mutation.

Primary dystonia is a disease characterized by involuntary twisting movements caused by CNS dysfunction without underlying histopathology. DYT1 dystonia is a form of primary dystonia caused by an in-frame GAG deletion (DeltaE302/3) in the TOR1A gene that encodes the endoplasmic reticulum luminal protein torsinA. We show that torsinA is also present in the nuclear envelope (NE), where it appears to interact with substrate, and that the DeltaE302/3 mutation causes a striking redistribution of torsinA from the endoplasmic reticulum to the NE. In addition, DeltaE302/3-torsinA recruits WT torsinA t

Primary biliary cirrhosis and the molecular cell biology of the nuclear envelope.

I hope I have demonstrated how basic research on the molecular cell biology of the nuclear envelope has provided information about the autoimmune disease PBC. I have given several examples of how highly specific immunologic reagents, obtained from patients with this disease, have been of value in experiments on the basic cell biology of the nuclear envelope. Continued work should provide further clues on how autoimmunity underlies the pathophysiology of PBC and should also provide additional reagents to study the processes of nuclear protein targeting and cell division.

Nuclear envelope rupture initiation in confined migration: A geometric-kinetic framework.

Nuclear envelope rupture during confined migration varies widely even at matched constriction size, indicating that pore diameter alone is an incomplete predictor of rupture initiation. This review proposes a geometric-kinetic framework that organizes rupture risk along two coupled axes. Geometric stress focusing is driven by high curvature at the leading nuclear apex, and rate-limited remodeling emerges when deformation outpaces effective nuclear relaxation. Each axis is captured by a measurable, workflow-anchored coordinate, placing diverse confinement events on a shared operating-point map.

PP2A-B55SUR-6 promotes nuclear envelope breakdown in C. elegans embryos.

Nuclear envelope (NE) disassembly during mitosis is critical to ensure faithful segregation of the genetic material. NE disassembly is a phosphorylation-dependent process wherein mitotic kinases hyper-phosphorylate lamina and nucleoporins to initiate nuclear envelope breakdown (NEBD). In this study, we uncover an unexpected role of the PP2A phosphatase B55SUR-6 in NEBD during the first embryonic division of Caenorhabditis elegans embryo. B55SUR-6 depletion delays NE permeabilization and stabilizes lamina and nucleoporins. As a result, the merging of parental genomes and chromosome segregation

Using nuclear envelope mutations to explore age-related skeletal muscle weakness.

Skeletal muscle weakness is an important determinant of age-related declines in independence and quality of life but its causes remain unclear. Accelerated ageing syndromes such as Hutchinson-Gilford Progerin Syndrome, caused by mutations in genes encoding nuclear envelope proteins, have been extensively studied to aid our understanding of the normal biological ageing process. Like several other pathologies associated with genetic defects to nuclear envelope proteins including Emery-Dreifuss muscular dystrophy, Limb-Girdle muscular dystrophy and congenital muscular dystrophy, these disorders c
