The malaria parasite Plasmodium falciparum undergoes closed mitosis, which occurs within an intact nuclear envelope, and differs significantly from its human host. Mitosis is underpinned by the dynamics of microtubules and the nuclear envelope. To date, our ability to study P. falciparum mitosis by microscopy has been hindered by the small size of the P. falciparum nuclei. Ultrastructure expansion microscopy (U-ExM) has recently been developed for P. falciparum, allowing the visualization of mitosis at the individual nucleus level. Using U-ExM, three intranuclear microtubule structures ar

PURPOSE: To study the ultrastructure of nuclear chromatin changes of conjunctival epithelial cells in imprints of dry-eye patients.
METHODS: Imprints with nuclear chromatin changes of the conjunctival epithelium from dry-eye patients were first selected by light microscopy (LM). The latter allowed processing material with special interest (chromatin changes) for electron microscopy (EM). Ultrathin sections were cut tangential to the filter surface to obtain an large overview of the collected imprints. For scanning electron microscopy (SEM), selected specimens were dehydrated in ethanol and dr

Little is known about the cytoarchitecture of human myometrial cells in pregnancy, and whether or not this may be influenced by maternal characteristics such as age, parity and body mass index (BMI). The aim of this study was primarily to evaluate human myometrial smooth muscle cell (SMC) and nuclear volume in the third trimester of human pregnancy, and secondarily to investigate if these parameters are altered in relation to the maternal characteristics outlined above. Myometrial biopsies were obtained from 30 women undergoing elective caesarean delivery at term. One-micrometer sections were

Dientamoeba fragilis is a pathogenic trichomonad found in the gastrointestinal tract of humans and is implicated as a cause of diarrhoea. Despite its discovery over a century ago, there has been no recent thorough description of this parasite by microscopy. Scanning electron microscopy, transmission electron microscopy, confocal and light microscopy were therefore used to characterise D. fragilis populations growing in xenic culture. Two different populations - smooth and ruffled cells - were identifiable by scanning electron microscopy. No flagella, pelta structures, undulating membrane or ps

Epstein-Barr virus nuclear antigen 1 (EBNA-1) is essential for the maintenance of latent EBV plasmids, and is also a transcriptional regulator. Nuclear lamins, components of the nuclear lamina, have also been found in the nucleoplasm. We report here that EBNA-1 coincided with lamin B1 in the nucleoplasm and around the nuclear rim during S-phase by confocal microscopy of cells transfected with EBNA-1 in the absence of EBV plasmids. Lamin B1, which is rarely detected in nuclear soluble fractions, was detected in chromatin and nuclear matrix fractions of the EBNA-1-expressing cells. These observa

BACKGROUND: Nuclear import, dependent on the transporter importin α (IMPα), is a drug target for apicomplexan parasites Plasmodium falciparum and Toxoplasma gondii. Indeed, a panel of small molecule inhibit interactions between IMPα and nuclear localization signals (NLSs) in vitro and the growth of rapidly dividing stages (P. falciparum blood stages and T. gondii tachyzoites) in culture.
OBJECTIVES: As new drugs targeting multiple life cycle stages of both parasites are required, the panel of IMPα inhibitors was tested for their ability to inhibit nuclear transport in the rapidly dividing sta

Multiple skeletal components, including actin and associated proteins, are located in both the cytoplasm and the nucleus. Here, we describe a methodology to study nuclear actin and its associated proteins through a comprehensive approach. First, we focus on nuclear isolation as a singular step to separate and distinguish skeletal components within the nucleus. Nuclear actin organization can be regulated by using pharmacological agents (e.g., latrunculin, jasplakinolide) or genetic tools, such as mutants of nuclear actin (NLS-actinR62D). These approaches allow us to analyze the nuclear morpholo

The advance in biochemical and microscopy techniques has revealed the complexity and intricate nucleoplasm structure. Several subcompartments were identified in nucleus and the importance of these subcompartments in processes crucial for normal nuclear activity has been demonstrated. In this mini-review, we will give an overview about the composition, function, and importance of the major nuclear subcompartments. Also, we will show the impact that perturbing these structures can cause in normal nuclear activity, and how these can contribute to the development of some human diseases.

The nuclear pore complex (NPC) is a complex macromolecular structure comprised of multiple copies of ~30 different nucleoporin proteins (Nups). Collectively, these Nups function to regulate genome organization, gene expression, and nucleocytoplasmic transport (NCT). Recently, defects in NCT and alterations to specific Nups have been identified as early and prominent pathologies in multiple neurodegenerative diseases, including Amyotrophic Lateral Sclerosis (ALS), Alzheimer's Disease (AD)/Frontotemporal Dementia (FTD), and Huntington's Disease (HD). Advances in both light and electron microscop

Leukoplakia, lichen planus and carcinoma in situ of human oral mucosa were analysed histometrically by light microscopy. Leukoplakia lesions showed a greater total nuclear density than lichen planus lesions. They had a similar basal nuclear density to carcinomas in situ. The total and basal nuclear areas were notably greater in carcinomas in situ, indicating the presence of larger nuclei in comparison to those of other lesions. We believe it to be possible to make a differential diagnosis based on numerical variables.

A mildly macerated, hydropic fetus was delivered spontaneously at 25 weeks gestation by a multigravidous black mother. At autopsy, gross and microscopic evidence suggested fetal anemia, and excessive extramedullary erythroblastosis was present generally. Erythroid precursor cells in the pulmonary capillary circulation contained eosinophilic nuclear inclusions consistent with human parvovirus B19 infection. Transmission electron microscopy on osmicated Epon lung sections showed lucent, granular chromatin corresponding to the inclusions. In rare foci only these sections contained paracrystalline

UNLABELLED: Kinetochores facilitate interaction between chromosomes and the spindle apparatus. The formation of a metazoan trilayered kinetochore is an ordered event in which inner, middle, and outer layers assemble during disassembly of the nuclear envelope during mitosis. The existence of a similar strong correlation between kinetochore assembly and nuclear envelope breakdown in unicellular eukaryotes is unclear. Studies in the hemiascomycetous budding yeasts Saccharomyces cerevisiae and Candida albicans suggest that an ordered kinetochore assembly may not be evolutionarily conserved. Here,

Mechanical stress on eukaryotic nucleus has been implicated in a diverse range of diseases including muscular dystrophy and cancer metastasis. Today, there are very few non-perturbative methods to quantify nuclear mechanical properties. Interferometric microscopy, also known as quantitative phase microscopy (QPM), is a powerful tool for studying red blood cell biomechanics. The existing QPM tools, however, have not been utilized to study biomechanics of complex eukaryotic cells either due to lack of depth sectioning, limited phase measurement sensitivity, or both. Here, we present depth-resolv

Caviomonas mobilis was collected from the caecum of mice harbouring a controlled fauna. Phase contrast and immunofluorescence microscopy using an anti-tubulin antibody and electron microscopy demonstrated the presence of one basal body bearing a flagellum and a second barren basal body, both inserted in the face of two cup-like depressions in the nuclear surface, as in other enteromonad/diplomonad genera. Three microtubular fibres arise close to the main basal body: the first, composed of three microtubules cross-linked with a dense structure, lies within a groove above the nuclear surface; th

Chicken erythrocytes are nucleated blood cells that provide a useful model for assessing oxidative stress-related membrane injury, but the conventional hemolysis assays do not resolve single-cell nanoscale mechanical alterations. Here, we established an atomic force microscopy (AFM)-based workflow to characterize structural and nanomechanical alterations associated with 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH) exposure and curcumin-associated protection in chicken erythrocytes. Hemolysis assays were used to select a moderate injury condition and a protective curcumin dose. Confocal

In mammals, circadian rhythms are generated by delayed negative feedback, in which period (PER1-PER3) and cryptochrome (CRY1, CRY2) proteins gradually accumulate in the nucleus to suppress the transcription of their own genes. Although the importance of nuclear import and export signals for the subcellular localization of clock proteins is well established, little is known about the dynamics of these processes as well as their importance for the generation of circadian rhythms. We show by pharmacological perturbations of oscillating cells that nuclear import and export are of crucial importanc

Immunoflourescent (IF) microscopic studies were done on a patient with bullous pemphigoid who had a high titer of antinuclear antibodies (ANAs). Direct IF microscopy showed deposition of lgG and C3 at the basement membrane zone (BMZ), while, by indirect IF microscopy, ANAs (but not circulating pemphigoid antibodies) were detected. The 2M sodium chloride (NaCl) solution-treated substrates, in which nuclear antigens were eluted, uncovered pemphigoid antibodies. Treatment of the substrates with NaCl solution unmasked BMZ fluorescence in another set of experiments in which a large amount of serum

Electron microscopy study of muscle biopsies from patients with autosomal-dominant Emery-Dreifuss muscular dystrophy revealed nuclear alterations in about 10% of the preserved muscle fibers. The major findings consisted of peripheral heterochromatin loss or detachment from the nuclear envelope, and of interchromatin texture alterations. These abnormalities are similar to those reported in an animal model of the disease and to those found in the X-linked form of Emery-Dreifuss muscular dystrophy. These results suggest that an abnormal ultrastructural arrangement of the nuclear periphery is a co

Huntington's disease (HD) is caused by a polyglutamine expansion of the huntingtin protein, resulting in the formation of polyglutamine aggregates. The mechanisms of toxicity that result in the complex HD pathology remain only partially understood. Here, we show that nuclear polyglutamine aggregates induce nuclear envelope (NE) blebbing and ruptures that are often repaired incompletely. These ruptures coincide with disruptions of the nuclear lamina and lead to lamina scar formation. Expansion microscopy enabled resolving the ultrastructure of nuclear aggregates and revealed polyglutamine fibri

T lymphocyte activation plays a pivotal role in adaptive immune response and alters the spatial organization of nuclear architecture that subsequently impacts transcription activities. Here, using stochastic optical reconstruction microscopy (STORM), we observe dramatic de-condensation of chromatin and the disruption of nuclear envelope at a nanoscale resolution upon T lymphocyte activation. Super-resolution imaging reveals that such alterations in nuclear architecture are accompanied by the release of nuclear DNA into the cytoplasm, correlating with the degree of chromatin decompaction within
