Spatial and temporal variation of five different pathogens and symbionts in Ixodes ricinus nymphs in the Netherlands.

The incidence of diseases caused by pathogens transmitted by the tick Ixodes ricinus vary over time and space through incompletely understood mechanisms. An important determinant of the disease risk is the density of infected ticks, which is the infection prevalence times the density of questing ticks. We therefore investigated the spatial and temporal variation of four pathogens and one of the most abundant symbionts in Ixodes ricinus in questing nymphs over four years of monthly collections in 12 locations in the Netherlands. The infection prevalence of all microbes showed markedly different

Unexpected failure of Ixodes scapularis nymphs to transmit a North American Borrelia bissettiae strain.

Globally, the Borrelia burgdorferi (sensu lato) complex comprises more than 21 species of spirochetes. Although the USA is home to a diverse fauna of Lyme disease group Borrelia species, only two are considered responsible for human clinical disease: Borrelia burgdorferi (sensu stricto) and Borrelia mayonii. However, evidence has implicated additional B. burgdorferi (s.l.) species in human illness elsewhere. While much research has focused on the B. burgdorferi (s.s.)-tick interface, tick vectors for most of the other North American Lyme disease group Borrelia species remain experimentally unc

Potential effects of blood meal host on bacterial community composition in Ixodes scapularis nymphs.

Tick microbiomes may play an important role in pathogen transmission. However, the drivers of microbiome variation are poorly understood, and this limitation has impeded mechanistic understanding of the functions of microbial communities for pathogen acquisition. The goal of this research was to characterize the role of the blood meal host in structuring the microbiome of Ixodes scapularis, the primary vector of Lyme disease in the eastern United States, and to determine if ticks that fed from different host species harbor distinct bacterial communities. We performed high-throughput 16S rDNA a

Developmental changes in salivary glands of nymphs and adults of the spinose ear tick Otobius megnini.

Otobius megnini has an autogenous 1 host life cycle, where larvae and nymphs stay attached inside the ear canal for long periods, but the adult tick is free living and can lay several egg batches without feeding. In order to obtain information about anatomical structures involved in this particular life cycle, nymphs and adults of O. megnini were dissected and salivary gland images were obtained in situ with the use of scanning electronic microscopy. Measurements of salivary alveoli were obtained with the use of ImageJ 1.40 g software. In the nymphs, the Type I alveoli are relatively small (me

Functional genomics tool: gene silencing in Ixodes scapularis eggs and nymphs by electroporated dsRNA.

BACKGROUND: Ticks are blood-sucking arthropods responsible for transmitting a wide variety of disease-causing agents, and constitute important public health threats globally. Ixodes scapularis is the primary vector of the Lyme disease agent in the eastern and central U.S. RNAi is a mechanism by which gene-specific double-stranded RNA (dsRNA) triggers degradation of homologous mRNA transcripts. Here, we describe an optimized protocol for effectively suppressing gene expression in the egg and nymphal stages of I. scapularis by electroporation. RESULTS: The genes encoding the putative Phospholip

Influence of elevation on Ixodes pacificus (Acari: Ixodidae) nymph seasonality, abundance, and Borrelia infection prevalence in the Sierra Nevada foothills.

The western blacklegged tick, Ixodes pacificus Cooley and Kohls, is a significant public health concern due to its capacity to vector Borrelia burgdorferi sensu stricto (Spirochaetales: Spirochaetaceae), the causative agent of Lyme disease. To determine the influence of elevation on the seasonality and abundance of Ixodes pacificus nymphs in the Sierra Nevada foothills of California, we used a standardized flagging procedure to sample nymphs from rocks and logs at 16 sites during 2018 to 2020. We sampled a total of 4,720 rocks and logs during 168 surveillance events and collected 1,469 I. paci

Oral toxicity of various Stemona collinsiae crude extracts against nymph and adult stages of American cockroach, Periplaneta americana (Dictyoptera: Blattodea).

Stemona collinsiae exhibits insecticidal resistance against various pests and insect vectors. However, insecticidal activity of S. collinsiae roots has not been tested for some insect vectors, including the American cockroach, Periplaneta americana. The synanthropic insect P. americana is a reservoir of pathogenic and non-pathogenic microorganisms and a cause of infectious diseases and cockroach allergy. This important vector transmits microorganisms to animals and humans to cause vector-borne diseases. This research involved detection of the nymphicidal and adulticidal activities of S. collin

ON THE NYMPH AND PROSOPON OF THE TSUTSUGAMUSHI, LEPTOTROMBIDIUM AKAMUSHI, N. SP. (TROMBIDIUM AKAMUSHI BRUMPT), CARRIER OF THE TSUTSUGAMUSHI DISEASE.

Nomenclature. As mentioned above, the prosopon and the nymph of the tsutsugamushi have many characteristics which distinguish it from the other trombidia. These are the pale color and small size of the body, delicacy of the skin, the conspicuous constriction between the cephalothoracic region and the abdomen, rudimentary eyes and tracheal openings, absence of tracheal capillaries, etc. The fact that the thumb or the appendage of the palpus does not start from the side of the fourth joint of the latter but from its end, is another charac teristic which does not occur in other trombidia. Identification of soil-dwelling predators of tick nymphs (Acari: Ixodidae) by stable isotope labeling.

Ticks (Family Ixodidae) spend most of their life cycle as immature stages in the soil and litter, and as any other soil invertebrates, are likely to be controlled top-down by soil-dwelling predators. To date, the ability of soil invertebrate predators to control ixodid tick population remains little known, partly due to methodological difficulties. In the current study, we developed and successfully tested a novel method of labeling live Ixodes ricinus (L., 1758) (Ixodida: Ixodidae) nymphs with a 15N isotope label. Labeled ticks were used in a small-scale 8-day-long microcosm experiment to rev

Red and fallow deer determine the density of Ixodes ricinus nymphs containing Anaplasma phagocytophilum.

BACKGROUND: The density of Ixodes ricinus nymphs infected with Anaplasma phagocytophilum is one of the parameters that determines the risk for humans and domesticated animals to contract anaplasmosis. For this, I. ricinus larvae need to take a bloodmeal from free-ranging ungulates, which are competent hosts for A. phagocytophilum. METHODS: Here, we compared the contribution of four free-ranging ungulate species, red deer (Cervus elaphus), fallow deer (Dama dama), roe deer (Capreolus capreolus), and wild boar (Sus scrofa), to A. phagocytophilum infections in nymphs. We used a combination of ca

The Botanical Monoterpenes Linalool and Eugenol Flush-Out Nymphs of Triatoma infestans (Hemiptera: Reduviidae).

Monoterpenes are the main components of essential oils. Some members of this chemical family present insecticidal activity. Triatoma infestans (Klug) is the main vector of Chagas disease in Argentina, Bolivia, Paraguay, and Perú. The objective of this work was to evaluate the effect of six monoterpenes (1,8-cineole, eugenol, linalool, menthol, α-terpineol, and thymol) on the locomotor and flushing out activity of T. infestans. A video tracking technique was used to evaluate the locomotor activity of nymphs exposed to different concentrations of these chemicals applied as films on filter paper.

A nasopharyngeal human infestation caused by Linguatula serrata nymphs in Van province: a case report.

The reason for the presentation of this case is that Linguatula serrata, a parasite rarely encountered in humans, was found in a patient in Van province. The patient, who was 26 years old, lived in Erciş a town in Van province, East Turkey. She was admitted to the Outpatient Clinic of Infectious Diseases of Erciş Government Hospital with a complaint of coughing a few worms about 4 cm long from the oral cavity, and also sore throat, partial voice loss and vomiting. The polyclinic doctor suspected that the worm was a parasite but he could not make a diagnosis, and the patient was referred to Hea

Occurrence of multiple infections with different Borrelia burgdorferi genospecies in Danish Ixodes ricinus nymphs.

The pathogen Borrelia burgdorferi causes Lyme Borreliosis in human and animals world-wide. In Europe the pathogen is transmitted to the host by the vector Ixodes ricinus. The nymph is the primary instar for transmission to humans. We here study the infection rate of five Borrelia genospecies: B. burgdorferi sensu stricto, B. afzelii, B. garinii, B. valaisiana, B. lusitaniae in nymphs, by IFA and PCR. 600 nymphs were collected in North Zealand of Denmark. Each nymph was first analysed by IFA. If positive for spirochaetal infection, the genospecies was determined by PCR. The infection rate of B.

Spatiotemporal patterns of host-seeking Ixodes scapularis nymphs (Acari: Ixodidae) in the United States.

The risk of Lyme disease for humans in the eastern United States is dependent on the density of host-seeking Ixodes scapularis Say nymphal stage ticks infected with Borrelia burgdorferi. Although many local and regional studies have estimated Lyme disease risk using these parameters, this is the first large-scale study using a standardized methodology. Density of host-seeking I. scapularis nymphs was measured by drag sampling of closed canopy deciduous forest habitats in 95 locations spaced among 2 degrees quadrants covering the entire United States east of the 100th meridian. Sampling was don

Babesia bigemina: sporozoite isolation from Boophilus microplus nymphs and initial immunomolecular characterization.

It has been hypothesized that babesial sporozoites express specific antigens that induce protective immunologic responses in cattle. However, they remain uncharacterized, partly for lack of research on the sporozoite stage of Babesia spp. This field suffers from complete knowledge of parasite development in the tick salivary gland; limited amounts of sporozoites from ticks, and a lack of protocols for induction and purification of sporozoites. In this work, Boophilus microplus larvae infected with B. bigemina were fed on susceptible cattle. Nymphs were collected and macerates were separated by

[The reaction of taiga ticks to an attractant. 1. The experimental contact of nymphs with attractant-acaricidal granules].

Four experimental variants used the following: 1) granules only from a filler (control); 2) those from a filler and an attractant; 3) and 4) those from a filler, an attractant, and a toxicant (permethrin or lambda cigalothrin, respectively). Each experiment used 20 nymphs of the first laboratory generation. 90 and 25% were in contact with attractive and control granules, respectively. Out of the nymphs in experiments 3 and 4 granules, 94 and 100% died. The contacts of 4 sec or more duration led to death. The experimental findings suggest that the designing and application of attractive acarici

Influence of climatic factors on Ixodes ricinus nymph abundance and phenology over a long-term monthly observation in Switzerland (2000-2014).

BACKGROUND: One of the major public health challenges in the field of communicable diseases consists of being able to predict where and when a population is at risk of being infected by a pathogen. In the case of vector-borne diseases, such predictions often require strong ecological knowledge of the vector life-cycle and the environmental conditions promoting or preventing its establishment and maintenance. In this study, we analyse how climatic factors influence the abundance and phenology of the Lyme borreliosis vector Ixodes ricinus in a Swiss temperate forest, based on a long-term monthly

First evidence of conspecific hyperparasitism in Dermacentor marginatus nymphs feeding on a rabbit under experimental conditions.

Hyperparasitism in ticks, particularly in nymphs of the order Ixodidae, is a rare phenomenon. In our laboratory tick colony, female rabbits are used as a blood source for the ticks, housing them under controlled conditions. Feeding Dermacentor marginatus nymphs monitoring was performed daily, and engorged and detached ticks were collected and stored in desiccators at constantly 24 ± 2 °C and 85-90% relative humidity (16 h light/8 hours dark). Nymphs suspected of conspecific hyperparasitism were preserved in ethanol for analysis and imaging. This study presents the first documented case of hype
