Photomutagenicity of N-nitrosoproline dissolved in non-aqueous solvent, oleic acid.

In the present study, we investigated the genotoxicity of the active products formed from N-nitrosoproline (NPRO) dissolved in oleic acid following ultraviolet A (UVA) irradiation, bypassing the need for metabolic activation. We previously demonstrated the photomutagenicity of NPRO dissolved in a phosphate-buffered solution. It has been suggested that the association of the nitrosamine group with acid ions facilitates rapid photodissociation and photoactivation. We hypothesized that NPRO's inherent carboxyl group may mimic an acid, inducing photodissociation and photomutagenicity, even in a no

Measuring functional residual capacity in normal and oleic acid-injured lungs.

Functional residual capacity (FRC) is an important oxygen reserve that is often depleted in acute respiratory failure. Recent interest in the mechanisms of liquid ventilation and limited experience in measuring FRC in paralyzed, mechanically ventilated, normal and lung-injured animal models have mandated development of accurate laboratory techniques. Eight sheep, from 17 to 27 kg, were anesthetized and instrumented to provide a tracheostomy, a pulmonary artery catheter, and carotid arterial line. They were randomized to two groups, one of which received 0.07 ml/kg of intravenous oleic acid to

Elucidating the pH Effects on Oleic Acid and Interactions with Lipid Membranes.

The interplay between fatty acids (FAs) and lipid membranes under varying pH conditions is important for deciphering fundamental cellular processes and advancing the design of responsive applications. Endowed with membrane incorporation, fatty acids (FAs) can induce curvature on membranes, resulting in alterations in fluidity, permeability, and stability. Despite the wide amount of research on membrane effects induced by FAs, the pH effect on their interaction behaviors remained unclear. Considering the varying pH environments in vivo, the study investigates how pH modulates the aggregation be

Results of fibre and toner flotation depending on oleic acid dosage.

The literature was reviewed with respect to deinking flotation methods with toner samples, specifically emphasizing the speciation of copy machine and laser printing, which produce an increasing quantity of paper that is difficult to recycle. Speciation here refers to the physical-chemical characteristics of the toner, which change because of the polymerization (fusion) and oxidation process, due to exposure to heat, light and oxygen (air) during the printing process. To simulate the deinking flotation, after the ideal disintegration process, samples of toner were prepared in order to provide

Eradication of drug resistant Staphylococcus aureus by liposomal oleic acids.

Staphylococcus aureus (S. aureus) represents a major threat to a broad range of healthcare and community associated infections. This bacterium has rapidly evolved resistance to multiple drugs throughout its antibiotic history and thus it is imperative to develop novel antimicrobial strategies to enrich the currently shrinking therapeutic options against S. aureus. This study evaluated the antimicrobial activity and therapeutic efficacy of oleic acid (OA) in a liposomal formulation as an innate bactericide against methicillin-resistant S. aureus (MRSA). In vitro studies showed that these OA-loa

Oleic acid activates TGFβ-Smad3 signaling to promote ovarian cancer progression.

BACKGROUND: Ovarian cancer represents the most aggressive and lethal gynecological cancer, frequently demonstrating a distinct propensity for abdominal metastasis. Malignant ascites caused by abdominal metastasis provide a tumor microenvironment (TME) in ovarian cancer. Notably, oleic acid is abundant in ovarian cancer ascites, though its functional significance in TME modulation and tumor metastatic regulation remains poorly characterized. Stearoyl-CoA desaturase 1 (SCD1) is the key enzyme in the synthesis of oleic acid. Our study systematically explores the pathological role of oleic acid an

Enzymatic synthesis and characterization of novel lipophilic inotodiol-oleic acid conjugates.

In this study, we establish an efficient enzymatic approach for producing novel inotodiyl-oleates (IOs) from pure inotodiol and oleic acid to improve the properties of inotodiol. For the esterification between inotodiol and oleic acid, CALA and n-hexane were the optimal biocatalyst and solvents for forming IOs with 80.17% conversion yield. These IOs comprised two distinct monoesters, the C3 or C22 ester forms of inotodiol. Intriguingly, no diesters were detected. The IOs had a melting point of 53.48 °C, much lower than that of inotodiol (192.06 °C). The in vitro digestion rate of IOs (25-28%)

Novel antibiofilm strategies against Pseudomonas aeruginosa NG4 using oleic acid and indole-3-butyric acid.

Biofilm-associated infections caused by Pseudomonas aeruginosa are highly tolerant to conventional antibiotics, driving the search for novel agents capable of disrupting biofilm formation and associated virulence traits. In this study, the antibiofilm, anti-motility, and growth inhibitory effects of indole-3-butyric acid (IBA) and oleic acid (OA) were evaluated against pathogenic P. aeruginosa NG4. Biofilm biomass was quantified by crystal violet staining. The swarming and swimming motility were assessed on semi-solid agar, and planktonic growth was measured spectrophotometrically. Confocal la

Oleic acid restores the impaired antitumor immunity of γδ-T cells induced by palmitic acid.

Dietary fatty acids (FAs) are associated with the therapeutic intervention under various health conditions. Human γδ-T cells are indispensable for immunosurveillance toward malignant cells. However, their impact on γδ-T cell metabolism and function remains poorly unexplored. Here, we applied targeted metabolomics analysis to serum FAs among cancer patients undergoing γδ-T cell therapy and discovered that palmitic acid (PA) or oleic acid (OA) levels were associated with the efficacy of Vγ9Vδ2-T cell therapy. We further elucidated that PA suppresses the antitumor activity of Vγ9Vδ2-T cells by di

Microwave-driven oleic acid esterification over chlorosulfonic acid-treated hydroxyapatite: synergism for intensified biodiesel production.

Treating hydroxyapatite (HAP) with sulfonic acid without structural destruction remains challenging owing to the sensitivity of HAP to acidic pH. In this work, natural derived HAP was prepared using natural phosphate via a dissolution/precipitation process. Notwithstanding the challenge, the prepared HAP was treated with three concentrations of chlorosulfonic acid in dichloromethane to prepare HAP-S1, HAP-S2 and HAP-S3 depending on the acid content under carefully controlled conditions. The treatment of HAP with the lowest acid concentration led to the preservation of the apatite framework wit

Hepatic-portal oleic acid inhibits feeding more potently than hepatic-portal caprylic acid in rats.

In several human and animal studies, medium-chain triglycerides decreased food intake more than did long-chain triglycerides. It is possible that faster uptake and metabolism of medium-chain fatty acids in the liver is responsible for this difference. To test this hypothesis we compared the feeding effects of hepatic portal vein (HPV) infusion of the medium-chain fatty acid caprylic acid (CA) with those of the long-chain fatty acid oleic acid (OA). Contrary to our expectation, six-h HPV infusion of 14 microg/min (50 nmol/min) OA robustly inhibited feeding, whereas infusion of 22 or 220 microg/

Oleic acid and peanut oil high in oleic acid reverse the inhibitory effect of insulin production of the inflammatory cytokine TNF-alpha both in vitro and in vivo systems.

BACKGROUND: Chronic inflammation is a key player in pathogenesis. The inflammatory cytokine, tumor necrosis factor-alpha is a well known inflammatory protein, and has been a therapeutic target for the treatment of diseases such as Rheumatoid Arthritis and Crohn's Disease. Obesity is a well known risk factor for developing non-insulin dependent diabetes melitus. Adipose tissue has been shown to produce tumor necrosis factor-alpha, which has the ability to reduce insulin secretion and induce insulin resistance. Based on these observations, we sought to investigate the impact of unsaturated fatty

Oleic Acid Levels in HSALR Mouse Model of Myotonic Dystrophy Type 1.

Myotonic dystrophy type 1 (DM1) is characterized by altered RNA processing, muscle wasting and metabolic dysregulation. In a previous in vitro study, we found reduced endogenous oleic acid (OA; 18:1n-9) in myogenic cells from DM1 patients, and short-term OA supplementation rescued key disease readouts. Here, we asked whether lipid pathways are perturbed in a DM1 in vivo background. We employed the widely used HSALR mouse model, which expresses human skeletal alpha-actin transcripts harbouring expanded CTG repeats and recapitulates key DM1 phenotypes. RNA-seq-derived functional enrichment acros

Oleic acid fuels cisplatin-resistant ovarian cancer through FABP4-driven lipid uptake.

BACKGROUND: Ovarian cancer (OC) depends on lipids as fuel for metastasis and growth. We previously showed that cisplatin resistant (Pt-R) OC cells uptake higher amounts of fatty acids (FAs) compared to sensitive (Pt-S) cells, a process which facilitates cancer cell survival under cisplatin-induced oxidative stress. METHODS: Isogenic pairs of Pt-S and Pt-R OC cell lines were cultured in low serum conditions supplemented with either 50 μM oleic acid (OA, unsaturated) or 50 μM palmitic acid (PA, saturated) and used for viability assays, RNA-Sequencing, and cell cycle analysis. Self-assembled leuprolide-oleic acid nanoparticles encapsulating docetaxel for synergistic drug delivery.

In the present study, an amphiphilic leuprolide acetate (LEU)-oleic acid (OA) conjugate (LOC) was synthesized by conjugating the peptide drug LEU with OA to form self-assembled nanoparticles (LON). This may maximize the synergistic effectiveness of fatty acids and anticancer drugs in prostate cancer. Docetaxel (DTX)-encapsulating LON (LOND) have also been developed for triple-synergistic drug delivery. LON and LOND retained a uniformly spherical morphology and had particle sizes ranging 130.6-159.1 ± 3.1 nm, depending on the freeze drying or redispersion process. Neither LON nor encapsulation

Oleic acid prevents erythrocyte death by preserving haemoglobin and erythrocyte membrane proteins.

Haemolysis of erythrocytes upon exposure to haemato-toxic phenylhydrazine (PHZ), makes it an experimental model of anaemia and a partial model of β-thalassaemia, where oxidative stress (OS) was identified as principal causative factor. Oleic acid (OA) was evidenced to ameliorate such stress with antioxidative potential. Erythrocytes were incubated in vitro using 1 mM PHZ, 0.06 nM OA. Erythrocyte membrane protein densities and haemoglobin (Hb) status were examined. Any interaction of Hb with PHZ/OA was checked by calorimetric and spectroscopic analysis using pure molecules. Oleic acid ameliorates amyloidosis in cellular and mouse models of Alzheimer's disease.

Several lines of evidence support protective as well as deleterious effects of oleic acid (OA) on Alzheimer's disease (AD) and other neurological disorders; however, the bases of these effects are unclear. Our investigation demonstrates that amyloid precursor protein (APP) 695 transfected Cos-7 cells supplemented with OA have reduced secreted amyloid-beta (Aβ) levels. An early-onset AD transgenic mouse model expressing the double-mutant form of human APP, Swedish (K670N/M671L) and Indiana (V717F), corroborated our in vitro findings when they were fed a high-protein, low-fat (18% reduction), ch

Cardiac dysfunction during acute lung injury induced by oleic acid in dogs.

Circulatory abnormalities are common during the adult respiratory distress syndrome and contribute importantly to the high mortality seen with this illness, but what causes them is not well characterized. Oleic acid-induced acute lung injury is an experimental model of the human syndrome in which hemodynamic changes are also common. To characterize a possible link between lung injury and cardiac dysfunction in this model, we evaluated cardiac function in mongrel dogs after oleic acid administration. Stroke volume index decreased, the radionuclide ventriculographically determined ejection fract
