Excitatory and inhibitory effects of noradrenaline on synaptic transmission in the rat olfactory cortex slice.

An investigation has been made of the effects of noradrenaline on excitatory transmission at the lateral olfactory tract (LOT)-superficial pyramidal cell synapse of the rat olfactory cortex slice by measuring the effects of bath-applied noradrenaline on the amplitudes and latencies of the field potentials evoked on LOT stimulation. Low concentrations of noradrenaline (0.1-5 microM) facilitate transmission whereas higher doses (20-250 microM) depress transmission. Both these effects were completely blocked by non-selective alpha- and beta-adrenoceptor antagonists, by 2-amino-5-phosphonovaleric

Effects of nicotine and tobacco smoke on the electrical activity of the cerebral cortex and olfactory bulb.

1. Effects of nicotine, cigarette smoke and carbon monoxide have been compared in the cat encéphale isolé preparation, exhibiting a synchronized electrocorticogram (ECoG) and behavioural sleep.2. 2 ml samples of smoke, containing approximately 7 mug nicotine (approximately 2 mug nicotine/kg for a 3 kg cat), introduced into the lungs at 30 s intervals from a smoking simulator, caused desynchronization of the ECoG and behavioural arousal.3. Effects of smoke were matched in the same experiments by intravenous injections of nicotine, 2 mug/kg every 30 s.4. The use of specific nicotine antagonists,

Functional heterogeneity in human olfactory cortex: an event-related functional magnetic resonance imaging study.

Studies of patients with focal brain injury indicate that smell perception involves caudal orbitofrontal and medial temporal cortices, but a more precise functional organization has not been characterized. In addition, although it is believed that odors are potent triggers of emotion, support for an anatomical association is scant. We sought to define the neural substrates of human olfactory information processing and determine how these are modulated by affective properties of odors. We used event-related functional magnetic resonance imaging (fMRI) in an olfactory version of a classical cond

Olfactory inputs modulate respiration-related rhythmic activity in the prefrontal cortex and freezing behavior.

Respiration and airflow through the nasal cavity are known to be correlated with rhythmic neural activity in the central nervous system. Here we show in rodents that during conditioned fear-induced freezing behavior, mice breathe at a steady rate (~4 Hz), which is correlated with a predominant 4-Hz oscillation in the prelimbic prefrontal cortex (plPFC), a structure critical for expression of conditioned fear behaviors. We demonstrate anatomical and functional connections between the olfactory pathway and plPFC via circuit tracing and optogenetics. Disruption of olfactory inputs significantly r

Interaction between phorbol dibutyrate and anaesthetics on synaptic responses from olfactory cortex of rat.

Phorbol esters have been shown to enhance the release of transmitters and to potentiate the effect of local anaesthetics in olfactory cortex of the rat. This work examined the interaction between phorbol dibutyrate and a range of substances, which act on axonal conduction and synaptic transmission. Synaptically-evoked field responses were elicited by stimulation of the lateral olfactory tract of slices of olfactory cortex, maintained in vitro. Ketamine (0.2-1.0 mM), benzocaine (0.2-1 mM), atropine (0.1-2 mM) and tetrodotoxin (20-200 nM) depressed synaptic transmission and, in the presence of p

The trajectory of mitral cell axons in the rabbit olfactory cortex revealed by intracellular HRP injection.

The projection of the axon and axon collaterals of mitral cells to the olfactory cortex was studied in the rabbit by intracellular staining with horseradish peroxidase (HRP). The stained mitral cell axons were reconstructed from the soma to the most caudal portion of the anterior piriform cortex (aPC). Single mitral cells projected to cytoarchitectonically different areas of the olfactory cortex, i.e., the anterior olfactory nucleus (AON), the aPC, and the olfactory tubercle (OT). All the stained mitrall cells projected to both the AON and the aPC, and about one-fourth of the mitral cells proj

Impaired Odor Perception in Autism Spectrum Disorder Is Associated with Decreased Activity in Olfactory Cortex.

Autism Spectrum Disorders (ASDs) are characterized by atypical sensory functioning in the visual, tactile, and auditory systems. Although less explored, olfactory changes have been reported in ASD patients. To explore these changes on a neural level, 18 adults with ASD and 18 healthy neurotypical controls were examined in a 2-phase study. Participants were first tested for odor threshold and odor identification. Then, (i) structural magnetic resonance (MR) images of the olfactory bulb were acquired, and (ii) a functional MR imaging olfaction study was conducted. ASD patients exhibited decrease

[Effect of hydrocortisone on synaptic transmission in surviving slices of the olfactory cortex from the rat brain].

A study was made of the effect of bath-applied hydrocortisone (10(-4) M) on excitatory transmission of the lateral olfactory tract (LOT) - superficial pyramidal cell synapse of the rat olfactory cortex slice. Population EPSP and IPSP were depressed for 6-12 min., then they were increased (60 min.) by hydrocortisone. Recovery of evoked potentials was long (40-60 min.) at washing of slices hy a normal incubation medium. Habituation of synaptic potentials during LOT repeating stimulation was deteriorated by hydrocortisone application in perfusion solution. The data obtained were considered to be

Effects of barbiturates on responses evoked by excitatory amino acids in slices of rat olfactory cortex.

A study has been made of the effects of ranges of concentrations of phenobarbitone, pentobarbitone and thiopentone on responses evoked by gamma-aminobutyric acid (GABA), L-glutamate, L-aspartate, N-methyl-D-aspartate, kainate and quisqualate in slices of olfactory cortex of the rat. All three barbiturates affected GABA-evoked depolarizations similarly in that responses were potentiated by small doses but markedly inhibited at greater concentrations. Responses to L-aspartate and L-glutamate were little affected except at the largest dose of phenobarbitone tested (10 mM). The responses evoked by

Trajectory of olfactory cortex degeneration from normal cognition to Alzheimer's disease: Insights from multimodal neuroimaging.

OBJECTIVE: The olfactory cortex is among the earliest brain regions affected by Alzheimer's disease (AD), with olfactory deficits frequently preceding cognitive decline. This study aimed to characterize the functional and structural degeneration trajectory of the olfactory cortex from normal cognition (NC) to mild cognitive impairment (MCI) and eventually to AD using multimodal neuroimaging techniques. MATERIALS AND METHODS: A total of 105 participants (28 with NC, 35 with MCI, and 42 with AD) were subjected to olfactory [University of Pennsylvania Smell Identification Test (UPSIT)] and cogni

Classification of Prefrontal Cortex Activity Based on Functional Near-Infrared Spectroscopy Data upon Olfactory Stimulation.

The sense of smell is one of the most important organs in humans, and olfactory imaging can detect signals in the anterior orbital frontal lobe. This study assessed olfactory stimuli using support vector machines (SVMs) with signals from functional near-infrared spectroscopy (fNIRS) data obtained from the prefrontal cortex. These data included odor stimuli and air state, which triggered the hemodynamic response function (HRF), determined from variations in oxyhemoglobin (oxyHb) and deoxyhemoglobin (deoxyHb) levels; photoplethysmography (PPG) of two wavelengths (raw optical red and near-infrare

The distribution of axon collaterals from the olfactory bulb and the nucleus of the horizontal limb of the diagonal band to the olfactory cortex, demonstrated by double retrograde labeling techniques.

Three different pairs of double retrograde axonal tracers have been used to study the distribution of axon collaterals from individual cells in the olfactory bulb and the nucleus of the horizontal limb of the diagonal band: (1) horseradish peroxidase (HRP) and tritiated apo-HRP (3H-HRP), (2) HRP and 125I-wheat germ agglutinin (I-WGA), and (3) the fluorochromes true blue (TB) and bisbenzimide (BB) or nuclear yellow (NY). With each combination of tracers, paired injections were made into different parts of the olfactory system, and the olfactory bulb and the nucleus of the diagonal band were exa

Morphological changes in secondary, but not primary, sensory cortex in individuals with life-long olfactory sensory deprivation.

Individuals with congenital sensory deprivation usually demonstrate altered brain morphology in areas associated with early processing of the absent sense. Here, we aimed to establish whether this also applies to individuals born without a sense of smell (congenital anosmia) by comparing cerebral morphology between 33 individuals with isolated congenital anosmia and matched controls. We detected no morphological alterations in the primary olfactory (piriform) cortex. However, individuals with anosmia demonstrated gray matter volume atrophy in bilateral olfactory sulci, explained by decreased c

EMX1 homeoprotein is expressed in cell nuclei of the developing cerebral cortex and in the axons of the olfactory sensory neurons.

We analyzed the distribution of EMX1 during mouse development. EMX1 is a homeoprotein encoded by Emx1, a regulatory homeobox gene expressed in the developing forebrain. Its distribution essentially overlaps the expression domains of Emx1 transcripts. The EMX1 protein is present in the developing dorsa telencephalon, that is in the cerebral cortex, olfactory bulb and hippocampus. In the cerebral cortex EMX1 is present in nuclei of proliferating, differentiating and most mature neurons belonging to all cortical layers. In the olfactory bulb it is present in all proliferating cells during develop

Olfactory neuroepithelium transplanted onto the parietal cortex of rats: electroolfactogram in absence of connections with the host brain.

A technique, allowing continuous electrophysiological monitoring of the functional properties of transplanted olfactory neuroepithelium is described. Viability of isolated parts of nasal mucosa obtained from adult rats was tested in vitro. Pieces of olfactory neuroepithelium (2 X 2 mm) were transplanted onto the exposed parietal cortex of adult rats and protected by plastic wells covered with a transparent lid. At one-week intervals the lid was removed and the transplanted epithelium was tested for electrical responses to amyl acetate vapours. Reliable electroolfactograms appeared 3 weeks afte

Monosynaptic and disynaptic activation of pyriform cortex neurons by synchronous lateral olfactory tract volleys in the rabbit.

To elucidate the organization of synaptic inputs to pyriform cortex neurons, intracellular and extracellular responses of single units were analyzed in urethane-anesthetized rabbits. The lateral olfactory tract (LOT) or the olfactory bulb (OB) was electrically stimulated. Intracellular recordings revealed two types of cells (type I and type II cells), according to the types of EPSP evoked by the LOT or OB shock. The EPSP in the type I cells had shorter latencies (0.0 to 0.9 ms) from the onset of the component 2 (C2) wave of the field potential (which signals the onset of the synaptic depolariz

Polysynaptic activation of the dentate gyrus of the hippocampal formation: an olfactory input via the lateral entorhinal cortex.

The possibility that olfactory input is transmitted to specific subregions of the hippocampal formation via the entorhinal cortex was investigated electrophysiologically by analyzing the laminar profiles of potentials evoked in the hippocampal formation by stimulation of the lateral olfactory tract (LOT). LOT stimulation resulted in long latency (14--20 ms) evoked responses in the dentate gyrus of the hippocampal formation ipsilateral to the stimulation. The variable long latency of these responses and their inability to follow stimulus rates of 40/s suggested that these potentials reflected p

The effects of lesions to the rat hippocampus or rhinal cortex on olfactory and spatial memory: retrograde and anterograde findings.

The role of the hippocampal system in retrograde and anterograde amnesia was investigated by using a novel olfactory-guided paradigm and a traditional test of spatial learning. In the retrograde study, rats were trained on a sequence of two-choice olfactory discriminations in the weeks prior to receiving neurotoxic lesions of the hippocampus or aspiration lesions of the perirhinal-entorhinal cortex. Memory tests for preoperatively learned discriminations revealed no statistical impairment for subjects with damage to the hippocampus on a problem learned remote in time from surgery (i.e., 4 week
