[Immunoreactive somatostatin in the peripheral olfactory system and nasal mucosa of the human].

Immunohistochemically, the cyclic retradecapeptide somatostatin was demonstrated in periglomerular cells and fibres of the glomerular layer (Fig. 3) of the human olfactory bulb as well as in the superficial granule layer and fibres (Fig. 2) of the olfactory tract. Additionally, somatostatin was found in endothelial cells of subepithelial arterioles and capillaries of the regio olfactoria (Fig. 4a, b) and superior turbinate. We assume that somatostatin has a paracrine transmitter function in the peripheral olfactory system and a regulative function on the blood flow of the superior nasal mucosa

[Pharmacologic study of the inhibition of the glomerular input to the olfactory tubercle of the frog induced by a puff of air on the olfactory mucosa].

The effects of 4-aminopyridine (10(-2) mol/l), aminooxyacetic acid (AOAA, 10(-4)-10(-3) mol/l), beta-alanine (10(-3)-10(-2) mol/l) and bicuculline (10(-5), 10(-4) mol/l) applications on the frog olfactory bulb (OB) were studied in vivo. The suppression of the orthodromic potential postsynaptic components evoked by a puff on the olfactory mucosa (OB input inhibition), or by single electrostimulation of the olfactory nerve (postsynaptic inhibition) was evaluated. 4-aminopyridine greatly reduced the OB input inhibition and strongly increased the postsynaptic inhibition. AOAA and bicuculline incre

Regeneration of olfactory mucosa in mice after inhalation exposure to perchloroethylene.

An experimental group of 20 male pure-bred mice was exposed to perchloroethylene gas at 300 ppm for 6 h daily for 5 days. Histopathological study of the nasal mucosa, particularly the olfactory mucosa, was performed sequentially 2 weeks to 3 months after exposure, to clarify the process of regeneration. The tissue damage due to perchloroethylene gas was more persistent in the nasal mucosa of the olfactory region than in the respiratory region. Two weeks after exposure, ciliated epithelial cells, as well as normal pseudostratified nonciliated columnar epithelium, began to appear in the area pre

Histological and histochemical studies of the secretory components of the salamander olfactory mucosa: effects of isoproterenol and olfactory nerve section.

Secretory components of the salamander olfactory mucosa, sustentacular cells (SC), and Bowman's glands (BG), were examined histologically and histochemically. In the aquatic larval salamander, SC in sensory grooves contained secretory granules; the submucosa contained a single layer of homogeneous, ductless glands. In the land-dwelling adult salamander, SC spanning a flat epithelial sheet contained vesicles. Subjacent to the epithelium in both dorsal and ventral mucosae lay BG whose ducts opened at the surface of the epithelium. In the ventral mucosa, two additional layers of olfactory glands

Immunocytochemical localization of copper-zinc superoxide dismutase in mouse olfactory mucosa.

Copper-zinc superoxide dismutase (Cu-Zn SOD) has been localized in mouse nasal mucosa. Immunocytochemical staining using polyclonal antibody against Cu-Zn SOD revealed endogenous Cu-Zn SOD in sustentacular cells in the olfactory area, and in ciliated epithelial cells in the respiratory area of the olfactory mucosa. Since these cells are located in the superficial portion of the olfactory mucosa, Cu-Zn SOD may serve as an intracellular antioxidant.

Expression of growth factors in fetal human olfactory mucosa during development.

The olfactory epithelium is one of the unique sites in the mammalian nervous system at which there is continual neurogenesis. Constant turnover of primary sensory neurons in the periphery results in remodeling of neuronal circuits and synapses in the olfactory bulb throughout life. Most of the specific mechanisms and factors that control this process are still unknown. Recent studies suggest that growth factors and their receptors may play a significant role in the development and continuous regeneration of olfactory neurons. In this study, standard immunohistochemical methods were used to det

Glutathione and glutathione S-transferase in the rainbow trout olfactory mucosa during retrograde degeneration and regeneration of the olfactory nerve.

In the peripheral olfactory organ, continual olfactory receptor neuron (ORN) turnover exposes neighboring cells to potentially damaging cellular debris such as free radicals. These, in turn, may be inactivated by binding directly onto glutathione (GSH) or by enzymatic conjugation with glutathione S-transferase (GST). In this study, we have investigated GSH and GST during retrograde degeneration and regeneration of the olfactory nerve in rainbow trout. In these fish, prolonged ORN physiological activity and structural integrity following transection of the olfactory nerve may be mediated by GSH

Protocol for postmortem bedside endoscopic procedure to sample human respiratory and olfactory cleft mucosa, olfactory bulbs, and frontal lobe.

We present a protocol for the rapid postmortem bedside procurement of selected tissue samples using an endoscopic endonasal surgical technique that we adapted from skull base surgery. We describe steps for the postmortem collection of blood, cerebrospinal fluid, a nasopharyngeal swab, and tissue samples; the clean-up procedure; and the initial processing and storage of the samples. This protocol was validated with tissue samples procured postmortem from COVID-19 patients and can be applied in another emerging infectious disease. For complete details on the use and execution of this protocol, p

HIV-1 detection in the olfactory mucosa of HIV-1-infected participants.

OBJECTIVE: HIV infection chronically affects the central nervous system (CNS). Olfactory mucosa is a unique site in the respiratory tract that is directly connected to the CNS; thus we wanted to evaluate olfactory mucosa as a surrogate of CNS sampling. DESIGN: We conducted a preliminary study examining HIV populations and susceptible cells in the olfactory mucosa. METHODS: Olfactory mucosa was sampled by minimally invasive brushing. Cerebrospinal fluid (CSF) analyses were performed as per routine clinical procedures. Olfactory marker protein, CD4+, CD8+, and trans-activator of transcription

Cytochrome P450 and steroid hydroxylase activity in mouse olfactory and vomeronasal mucosa.

The aims of this study are to identify the sex steroid-metabolizing cytochrome P450 enzymes of the vomeronasal organ (VNO) and to determine the activities of VNO microsomes to metabolize estradiol, progesterone, and testosterone. Several P450 isoforms, including CYP1A2, CYP2A, CYP2B, CYP2C, CYP2G1, and CYP3A, NADPH P450-reductase, and microsomal epoxide hydrolase were detected in mouse VNO, although their expression levels were much lower than those in the main olfactory epithelium. VNO microsomes were active toward the three steroid hormones, producing metabolite profiles similar to those see

Glutathione and gamma-glutamyl transpeptidase are differentially distributed in the olfactory mucosa of rats.

Components of the gamma-glutamyl cycle, including thiols, glutathione (GSH) and gamma-glutamyl transpeptidase (gamma-GT), were localized in the nasal mucosae of rats using histochemical and immunohistochemical methods. In olfactory mucosa, thiols were widely distributed, with intense staining in the mucociliary complex (MC), basal cells, acinar cells of Bowman's glands (BG), and olfactory nerve bundles, and with moderate staining in olfactory receptor neurons (ORNs). GSH was localized in MC, BG acinar cells, nerve bundles and, to a lesser extent, in ORNs. gamma-GT immunoreactivity was restrict

Novel genes for potential ligand-binding proteins in subregions of the olfactory mucosa.

Odorant detection is specifically mediated via receptor neurons in the olfactory mucosa but is a complex process involving a number of different cell types producing proteins of differing function. We have used the technique of subtractive hybridization cDNA cloning to identify novel genes expressed exclusively in the olfactory mucosa which may play a role in olfaction. Ten distinct groups of cDNA clones were identified which corresponded to mRNA transcripts highly expressed in rat olfactory mucosa but undetectable in thymus, kidney, lung, brain, spleen and liver. Some of these clones identify

The cyanide-metabolizing enzyme rhodanese in rat nasal respiratory and olfactory mucosa.

Hydrogen cyanide is a commonly occurring and highly toxic air pollutant. Inhalation of hydrogen cyanide would expose the nasal tissues to its toxic affects unless a detoxicating mechanism were available. Experiments with rat nasal tissues showed that the cyanide-metabolizing enzyme, rhodanese, is present in high concentrations, particularly in the olfactory region. The olfactory tissues had nearly 7-fold more rhodanese on a per mg mitochondrial protein basis than did the liver. These experiments show that nasal metabolism of cyanide may have an important influence on the toxicity of inhaled cy
