Infection of cynomolgus monkeys with microfilariae (Mf) of Onchocerca lienalis was studied as a model for human ocular onchocerciasis. Normal monkeys and immunized monkeys were given intracorneal/subconjunctival, intracameral, or intravitreal injections of Mf or bovine serum albumin (control). Selected animals were given diethylcarbamazine citrate (DEC) orally (15 mg/kg) daily after the ocular infection. Following intravitreal challenge, living Mf and fibrinous exudates were visible by slit-lamp in both the anterior chamber and vitreous. Eosinophils and macrophages surrounded the Mf in the vit

A study of the relationship between microfilaruria and onchocercal eye lesions has been carried out among 619 persons in 5 villages of North Benin. There is evidence of a significant relationship between microfilaruria and irreversible eye lesions in men aged 15-34 years. The association of microfilaruria with high numbers of microfilariae in the anterior segment of the eye occurs more in older men.

Onchocerciasis is a leading cause of blindness in equatorial Africa and in endemic regions in Central America. Understanding of the pathologic processes involved in onchocercal eye disease and of the role of immunopathologic mechanisms in its development has been substantially limited by the shortage of eyes for histologic study and by the lack of a naturally occurring animal model. The inoculation of microfilariae of Onchocerca species into the eyes of laboratory animals may reproduce selected aspects of onchocercal eye disease, such as punctate keratitis. Studies in these models support the

Ocular immunopathologic responses of inbred guinea pigs infected with Onchocerca microfilariae from domesticated animals were studied as a laboratory model of human ocular onchocerciasis. A single intracorneal infection of normal guinea pigs with microfilariae produced only minimal ocular lesions. In contrast, intracorneal infection of guinea pigs previously immunized by systemic infection with microfilariae produced intense corneal and uveal inflammation. Transfer of splenic lymphocytes from immunized donors to syngeneic normal recipients substituted effectively for the active immunization. C

Onchocerca volvulus is a spirurid nematode that mainly affects the rural poor of Sub-Saharan Africa, Yemen and parts of Central and South Africa. River blindness, caused by Onchocerca volvulus, is considered to be the second commonest infectious cause of blindness worldwide. We report a rare case of ocular onchocerciasis where a live adult worm was extracted from the eye of a patient from a nonendemic region. The worm was identified as Onchocerca volvulus based on morphological features. The patient was treated with Ivermectin (0.2 mg/kg). At six months follow-up she had complete remission of

Onchocerciasis is caused by the parasitic worm Onchocerca volvulus, which releases millions of offspring (microfilariae). Microfilariae migrate through the skin and can enter the anterior or posterior regions of the eye. While alive, the microfilariae appear to cause little or no inflammation, being in the anterior chamber. However, when they die, either by natural attrition or after chemotherapy, the host response to degenerating worms can result in ocular inflammation (keratitis, uveitis, chorioretinitis, neuritis of the optic nerve) that causes progressive loss of vision and ultimately lead

The development of an onchocercal chorioretinopathy from the first detectable signs to a full blown oncho fundus is not fully understood. We investigated the intraocular humoral immune response against Onchocerca volvulus, human S-antigen, IRBP and crude retinal extract (using an ELISA) by examining paired aqueous humour and serum samples obtained from onchocerciasis patients (without [n = 10] and with ocular symptoms [n = 8]) and endemic controls [n = 14] from Sierra Leone (West Africa). A local intraocular anti-retinal IgG antibody production could not be demonstrated in onchocerciasis patie

Despite its importance as a cause of blindness, there is still much to be learned about the way in which onchocerciasis damages ocular tissues. Much of the difficulty encountered in investigating the problem is due to the lack of an adequate laboratory model amenable to study outside the endemic areas of O. volvulus infection. An attempt has been made to reproduce the ocular effects of O. volvulus in humans by infecting rabbits and guinea-pigs and bovine (O. gutturosa) and equine (O. cervicalis) strains of Onchocerca. The subconjunctival inoculation of rabbits with microfilariae of O. volvulus

Involvement of the outer eye in Onchocerca volvulus infection is characterized by an inflammatory process around degenerating microfilariae. Individual microfilariae in the cornea may be associated with localized punctate lesions or, especially when present in large numbers, may provoke a sclerosing keratitis due to invasion by fibrovascular pannus from the limbal conjunctiva. Anterior uveitis develops in some patients and can be either granulomatous, probably as a result of direct microfilarial invasion of the iris and ciliary body, or non-granulomatous, in which case the inflammation is like

In a population survey in 1970/72 the prevalence of head nodules was found to be 1.7% in 1098 cases of onchocerciasis in the rain-forest and 0.6% in 1128 cases in the Sudan-savanna of Cameroon. In a follow-up survey in the same villages 3-4 years later more attention was given to the detection of head nocules, and the corresponding prevalences were 5.5 and 5.0%. In the follow-up survey a strong associated was demonstrated between the presence of head nodules and lesions of both the anterior and posterior segments of the eye. The relative risks of having eye lesions in patients with head nodule
