Survival of Cryptosporidium parvum oocysts under various environmental pressures.

The survival of various isolates of Cryptosporidium parvum oocysts under a range of environmental pressures including freezing, desiccation, and water treatment processes and in physical environments commonly associated with oocysts such as feces and various water types was monitored. Oocyst viability was assessed by in vitro excystation and by a viability assay based on the exclusion or inclusion of two fluorogenic vital dyes. Although desiccation was found to be lethal, a small proportion of oocysts were able to withstand exposure to temperatures as low as -22 degrees C. The water treatment

New rapid staining methods of Cryptosporidium oocysts in stools.

Two new extemporaneous negative-staining methods are proposed to detect Cryptosporidium oocysts in stools, using light-green and merbromine in 1% and 2% aqueous solution, respectively. A Ziehl-Neelsen stain as modified by Henriksen and Polhenz was used as a reference technique. A comparison between these two new stains and the reference, a modified Ziehl-Neelsen mod. method gave almost identical sensitivity and specificity. We propose their use in routine diagnosis for enteric cryptosporidiosis.

Shedding of oocysts in immunocompetent individuals infected with Cryptosporidium.

In June 1985, we investigated an outbreak of cryptosporidiosis at a day-care center in Florida. Through day-care center-wide stool screening, 33% (28/84) of the children and 22% (4/18) of the staff members submitting stool specimens were found to have cryptosporidiosis. Children 12-35 months of age and their teachers were at highest risk. All but 1 of the Cryptosporidium-positive patients reported diarrhea, which lasted between 1-44 days. Serial stool specimens were obtained from 8 infected individuals (5 children and 3 adults). Duration of oocyst shedding ranged from 8 to more than 50 days an

Effect of Glycosaminoglycans on Cryptosporidium Oocyst Attachment and Excystation.

Cryptosporidium causes severe gastrointestinal disease resulting from the ingestion of oocysts, followed by oocyst excystation in the small intestine and the release of infective sporozoites. An understudied strategy for Cryptosporidium inactivation is purposeful oocyst excystation, as sporozoites do not survive long in the environment. This study showed that C. parvum oocyst excystation was induced by direct contact with various glycosaminoglycans (GAGs), including heparin (Hep), chondroitin sulfate A (CSA), and hyaluronan (HA), assembled on polydopamine (PD)-functionalized surfaces. Dynamics of Toxoplasma gondii Oocyst Phagocytosis by Macrophages.

Oocysts are the environmentally resistant stage of the protozoan parasite Toxoplasma gondii. They are responsible for foodborne infections in humans and animals worldwide. Infectious oocysts contain sporozoites that have to exit the sporocyst and oocyst walls to initiate replication of the parasite within the host tissues. Given their robustness and resistance to chemical degradation, it is still unclear how the oocyst and sporocyst walls release the sporozoites. This process called excystation is thought to occur in the small intestine as a result of the combined action of digestive agents, y

Antigen incorporation on Cryptosporidium parvum oocyst walls.

Cryptosporidium parvum oocysts are the infective stages responsible for transmission and survival of the organism in the environment. In the present work we show that the oocyst wall, far from being a static structure, is able to incorporate antigens by a mechanism involving vesicle fusion with the wall, and the incorporation of the antigen to the outer oocyst wall. Using immunoelectron microscopy we show that the antigen recognized by a monoclonal antibody used for diagnosis of cryptosporidiosis (Merifluor(R), Meridian Diagnostic Inc.) could be found associated with vesicles in the space betw

Cryptosporidium parvum--propagation of oocyst in neonatal calves.

Shedding of Cryptosporidium parvum oocyst was studied in experimentally infected Jersey-Sindhi cross bred calves. Three 7 day old bull calves housed in isolation were orally infected with 10(8) oocysts of Cryptosporidium parvum. The prepatent and patent period of the experimental infection were 5 and 4 days respectively. Maximum oocyst output [2 x 10(5) oocyst per gram (OPG) was observed on the 7th day post inoculation (PI). The mean total oocyst output was 2.5 x 10(7). Diarrhoea started on the second day of oocyst shedding.

Prevalence of Cryptosporidium oocysts in livestock in Trinidad and Tobago.

Faecal samples from 683 diarrhoeic and non-diarrhoeic piglets, calves, lambs and goat kids were examined for Cryptosporidium oocysts. The prevalence of infection in piglets and lambs was 19.6% and 20%, respectively, and these rates were significantly higher than those detected in calves (8.7%). Amongst the four animal species studied, the detection rates were higher in diarrhoeic than in non-diarrhoeic animals and in animals under extensive and semi-intensive husbandry systems. However, these differences were not statistically significant.

Cryptosporidiosis--oocyst shedding and infection in household contacts.

The incidence of cryptosporidiosis in children with acute diarrhoea when compared with age-matched controls was 6.1% in a paediatric unit. Studies were made on eight immunocompetent children with cryptosporidiosis to determine the period of oocyst shedding after cessation of diarrhoea. The majority (75%) shed oocysts for less than ten days after cessation of diarrhoea. There was considerable variation among patients, some eliminating oocysts more quickly than others. This was not related to the severity or duration of diarrhoea. The sucrose floatation technique was found to be more sensitive t

Recognition and laboratory characteristics of an atypical oocyst of Cryptosporidium.

Feces from some patients with clinically unremarkable cryptosporidiosis contained an unusual variety of oocyst not previously recognized. These atypical oocysts were shown by electron microscopy to have a distinctive three-layered outer coat and, by immunofluorescence with a monoclonal antiserum, to lack an antigen present on the surface of typical oocysts. In contrast to typical oocysts, the atypical variety is very fragile and quickly collapses when suspended in solutions of high osmotic pressure or in lipid solvents. Atypical oocysts cannot be stained by methods used to stain typical oocyst

Eimeria tenella: further studies on the development of the oocyst.

AIM: The present study investigated the processes of macrogametogenesis and oocyst formation of Eimeria tenella (Xiamen strain), including the formation of wall-forming body1 (WFB1) and wall-forming body 2 (WFB2), the club-shape body and the origin of the residual body during the transformation from a macrogamete to an oocyst. METHOD: Transmission electron microscopy was used to follow ultrastructural changes of the organelles during parasite development. Frozen section techniques and special staining were used to determine the chemical composition of the club-shape body. RESULTS: Electron l

[The efficacy of water ozonation on the inactivation of oocysts of Cryptosporidium].

Cryptosporidium oocysts are currently found in surface waters. Their incomplete removal by sand filtration and their high resistance to chlorinated disinfectants make possible waterborne cryptosporidiosis outbreaks after drinking treated water. Because ozone is an alternative to chlorine for final drinking water disinfection the aim of our study was to determine the ozonation conditions able to eliminate the infectivity of oocysts. Feces from horses naturally infected by C. parvum were mixed with distilled water. The obtained suspension was clarified by passing through a graded series of four

Timing of symptoms and oocyst excretion in human cryptosporidiosis.

To determine the timing of symptoms and oocyst excretion after the acquisition of cryptosporidium infection, we used a screening parasitologic stool examination to identify patients and then contacted them for the collection of retrospective histories and follow-up stool specimens. The study included 68 otherwise healthy patients with an identifiable source and time of infection. All 68 had diarrhea, 61 had abdominal pain, most also had other gastrointestinal symptoms, 33 had fever, and all recovered spontaneously. Among the 50 patients who submitted follow-up stool samples, more than 90 perce

The effects of selected gases on excystation of coccidian oocysts.

The mechanism of CO2 action in changing coccidian oocyst wall permeability was indirectly studied by substituting NO, NO2, N2O, H2S, SO2, CH4, NH3, and 8M urea in place of CO2 in an established excystation procedure. Changes in oocyst wall permeability of Eimeria stiedai, E. bovis, and E. tenella were determined by incubation in test gases and cysteine HCl followed by attempted activation of sporozoites by trypsin and bile and staining of intraoocyst components with methylene blue. The gases CH4, NO2, and N2O were negative for all 3 tests, as were SO2, NH3, and 8M urea which, in addition, were

Techniques for inactivating Toxoplasma gondii oocysts: a systematic review.

The oocyst, a resistant form of Toxoplasma gondii, plays an important role in the transmission of this protozoan. The objective of this review was to report the methods capable of inactivating oocysts through a systematic review of the literature carried out in the Scientific Electronic Library Online, Web of Science, Science Direct, PubMed and Scopus databases. The keywords searched were (((effects OR infectivity OR resistance) AND Toxoplasma) AND oocyst). We selected 16 articles that described 309 different treatments. Among all the protocols evaluated, 35.60% (110/309) were effective in ina

rTgOWP1-f, a specific biomarker for Toxoplasma gondii oocysts.

Toxoplasma gondii oocyst wall protein 1 (TgOWP1) integrates a family of seven proteins, consensually assumed as specific antigens of Toxoplasma gondii oocyst stage, located in the outer layer of the oocyst wall. Herein, we notice the expression of a recombinant antigen, rTgOWP1-f, derived from a fragment selected on basis of its structural homology with Plasmodium MSP1-19. Rabbit polyclonal antibodies anti-rTgOWP1-f evidence ability for specific identification of environmental T. gondii oocysts. We assume, rTgOWP1-f, as a possible biomarker of oocysts. In addition, we present findings supporti

Ultrastructural changes in Cryptosporidium parvum oocysts by gamma irradiation.

Cryptosporidium parvum is known as one of the most highly resistant parasites to gamma irradiation. To morphologically have an insight on the radioresistance of this parasite, ultrastructural changes in C. parvum sporozoites were observed after gamma irradiation using various doses (1, 5, 10, and 25 kGy) following a range of post-irradiation incubation times (10 kGy for 6, 12, 24, 48, 72, and 96 hr). The ultrastructures of C. parvum oocysts changed remarkably after a 10-kGy irradiation. Nuclear membrane changes and degranulation of dense granules were observed with high doses over 10 kGy, and

Modelling Cryptosporidium oocysts transport in small ungauged agricultural catchments.

Cryptosporidium is an environmentally robust pathogen that has caused severe waterborne disease outbreaks worldwide. The main source of zoonotic Cryptosporidium parvum oocysts in human drinking water is likely to be from farm animals via catchment pathways with water as the main transport vector. The vast majority of small agricultural catchments are ungauged therefore it is difficult to use a process model to predict and understand the mechanisms and activities that regulate the risk of surface water contamination from agricultural areas. For this study, two ungauged agricultural catchments i
