Carbocyanine labeled LDL for optical imaging of tumors.

RATIONALE AND OBJECTIVES: The purpose of this study was to define and characterize carbocyanine labeled low-density lipoprotein (LDL) to be used in the optical imaging of LDL receptor (LDLr)-overexpressing tumor models. MATERIALS AND METHODS: 1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate (DiI) was used to label LDL (DiI-LDL). Scatchard plots were generated to determine the maximum binding capacity B(max) and dissociation constants K(D) of DiI-LDL in B16 melanoma (B16) and hepatoblastoma G(2) (HepG(2)) cell lines. Selective uptake of DiI-LDL into both tumor cells and corre

Lacrimal Canaliculus Imaging Using Optical Coherence Tomography Dacryography.

Lacrimal canaliculus (LC) has a key role in tear drainage, but it is difficult to evaluate the LC in detail, using the existing examinations. In this study, our novel LC imaging technique provided the high-resolution images of LC in a non-invasive manner. Three-dimensional images of LC were acquired via the palpebral conjunctiva from 20 healthy volunteers (20 eyes) and 10 patients with various lacrimal disorders (10 eyes), using optical coherence tomography (OCT) dacryography (OCTD). The LC images showed morphological differences between the vertical and horizontal segments. The function of LC

Magneto-optical nanoparticles for cyclic magnetomotive photoacoustic imaging.

Photoacoustic imaging has emerged as a highly promising tool to visualize molecular events with deep tissue penetration. Like most other modalities, however, image contrast under in vivo conditions is far from optimal due to background signals from tissue. Using iron oxide-gold core-shell nanoparticles, we have previously demonstrated the concept of magnetomotive photoacoustic (mmPA) imaging, which is capable of dramatically reducing the influence of background signals and producing high-contrast molecular images. Here, we report two significant advances toward clinical translation of this tec

Alternative optical concept for electron cyclotron emission imaging.

The implementation of advanced electron cyclotron emission imaging (ECEI) systems on tokamak experiments has revolutionized the diagnosis of magnetohydrodynamic (MHD) activities and improved our understanding of instabilities, which lead to disruptions. It is therefore desirable to have an ECEI system on the ITER tokamak. However, the large size of optical components in presently used ECEI systems have, up to now, precluded the implementation of an ECEI system on ITER. This paper describes a new optical ECEI concept that employs a single spherical mirror as the only optical component and explo

Optical coherence tomography in imaging of macular diseases.

OCT (opitcal coherence tomography), is a diagnostic method that enables the analysis of the retinal structures by means of high-resolution tomographic cross-sections of the retina. Whereas fluorescein and indocyanine angiography allow visualization of the retinal epithelium layer and chorioretinal vessels, OCT may help in diagnosing and monitoring the condition of many internal retinal layers. Ultrasound B-mode examinations have a resolution of about 150 microm while OCT provides a resolution of 10 microm. OCT makes it possible to detect and measure morphological changes, retinal thickness, re

Optical imaging of molecular signatures in pulmonary inflammation.

Biomedical imaging has become an important tool in the study of "-omics" fields by allowing the noninvasive visualization of functional and molecular events using in vivo staining and reporter gene approaches. This capacity can go beyond the understanding of the genetic basis and phenotype of such respiratory conditions as acute bronchitis, adult respiratory distress syndrome (ARDS), chronic obstructive pulmonary disease (COPD), and asthma and investigate the development of disease and of therapeutic events longitudinally and in unperturbed environments. Herein, we show how the application of

Near infrared fluorescent optical imaging for nodal staging.

Current techniques to assess lymph node metastases in cancer patients include lymphoscintigraphy after administration of a nonspecific radiocolloid in order to locate and resect lymph nodes for pathological examination of harbored cancer cells. Clinical trials involving intradermal or subcutaneous injection of antibody-based nuclear imaging agents have demonstrated the feasibility for target-specific, molecular imaging of cancer-positive lymph nodes. The basis for employing near-infrared (NIR) optical imaging for assessing disease is evidenced by recent work showing functional lymph imaging in

Imaging of dysthyroid optic neuropathy.

Dysthyroid optic neuropathy (DON) is a sight-threatening complication of thyroid eye disease and can lead to permanent vision loss if not treated early. Imaging with computed tomography (CT) or magnetic resonance imaging (MRI) can aid in the diagnosis and early recognition of DON. A number of quantitative and qualitative imaging features have been associated with DON. This article summarises the definition, prevalence, and utility of these radiological findings in the diagnosis of DON.

The site of the optical image in the eye.

Based on good empirical evidence it is generally said that the layer of the retina containing the rod and cone cells is the site where the optical image of the external world falls, and whence the neural signal is transmitted to the brain. The historical path to this idea is traced from the ancient Greeks, through Kepler to modern times. The newer hypothesis suggests that the retinal pigment epithelial (RPE) layer behind the rods and cones may be the actual photoreceptive apparatus. It is supported by clinical pictures, such as retinal detachment and Vitamin A deficiency, and the physical law

Compressive optical image encryption.

An optical image encryption technique based on compressive sensing using fully optical means has been proposed. An object image is first encrypted to a white-sense stationary noise pattern using a double random phase encoding (DRPE) method in a Mach-Zehnder interferometer. Then, the encrypted image is highly compressed to a signal using single-pixel compressive holographic imaging in the optical domain. At the receiving terminal, the encrypted image is reconstructed well via compressive sensing theory, and the original image can be decrypted with three reconstructed holograms and the correct k

Functional magnetic resonance imaging and optical imaging for dominant-hemisphere perisylvian arteriovenous malformations.

OBJECTIVE: In this study, we developed an a priori system to stratify surgical intervention of perisylvian arteriovenous malformations (AVMs) in 20 patients. We stratified the patients into three categories based on preoperative functional magnetic resonance imaging (fMRI) language activation pattern and relative location of the AVM. METHODS: In Group I (minimal risk), the AVM was at least one gyrus removed from language activation, and patients subsequently underwent asleep resection. In Group II (high risk), the AVM and language activation were intimately associated. Because the risk of pos

Optical coherence tomography imaging of optic disc cavernous haemangioma.

Optic disc cavernous haemangiomas are either found incidentally or after presentation with vitreous haemorrhage. They are characterised by a cluster of grapes appearance to the multiple vascular saccules that make up the tumour. They are more often found in the retinal periphery but rarely occur at the optic disc. Optical coherence tomography (OCT) imaging may be a useful non-invasive imaging modality to follow-up these lesions. We present the case of an asymptomatic 60-year-old lady referred from her optometrist with a lesion overlying the optic disc and the ensuing diagnosis of cavernous hae

Diffusion tensor imaging of the optic radiations after optic neuritis.

Trans-synaptic degeneration could exacerbate neurodegeneration in multiple sclerosis (MS). We aimed to assess whether anterograde trans-synaptic degeneration could be identified in the primary visual pathway in vivo. Diffusion tensor imaging (DTI) was used to assess the optic radiations in 15 patients with previous optic nerve inflammation and 9 healthy volunteers. A probabilistic atlas of the optic radiations was created from healthy diffusion tractography data. Lengthwise profiles for DTI parameters (axial [λ(||) ], radial [λ(⟂) ] and mean diffusivity [MD], fractional anisotropy [FA] and the

Non-mydriatic handheld optical coherence tomography for panretinal imaging.

We present a handheld, non-mydriatic optical coherence tomography (OCT) system engineered for panretinal imaging across both pediatric and adult patients. The prototype achieves a 140° visual angle, enabling detailed visualization from the posterior pole to the peripheral retina without the need for pharmacologic dilation or scleral depression. A customized optical design minimizes beam wander while maintaining diffraction-limited resolution throughout the field of view. The system employs a 400 kHz VCSEL light source, facilitating high-speed image acquisition along with an extended axial imag

Optical Imaging Technologies and Clinical Applications in Gastrointestinal Endoscopy.

Optical imaging technologies expand gastrointestinal endoscopy beyond white-light endoscopy (WLE), improving visualization of mucosal, vascular, and subsurface features. They are applied to the detection of neoplastic and premalignant lesions, inflammatory diseases, and small bowel and pancreatic disorders, though their validation and readiness for routine practice vary. This review critically evaluates both guideline-endorsed and investigational optical imaging techniques across major gastrointestinal indications, highlighting diagnostic performance, level of validation, current guideline rec

Tissue optical clearing and 3D imaging of virus infections.

Imaging pathogens within 3D environment of biological tissues provides spatial information about their localization and interactions with the host. Technological advances in fluorescence microscopy and 3D image analysis now permit visualization and quantification of pathogens directly in large tissue volumes and in great detail. In recent years large volume imaging became an important tool in virology research helping to understand the properties of viruses and the host response to infection. In this chapter we give a review of fluorescence microscopy modalities and tissue optical clearing met

Choroidal imaging using optical coherence tomography: techniques and interpretations.

The choroid is vascularized membranous tissue that supplies oxygen and nutrients to the photoreceptors and outer retina. Choroidal vessels underlying the retinal pigment epithelium are difficult to visualize by ophthalmoscopy and slit-lamp examinations. Optical coherence tomography (OCT) imaging made significant advancements in the last 2 decades; it allows visualization of the choroid and its vasculature. Enhanced-depth imaging techniques and swept-source OCT provide detailed choroidal images. A recent breakthrough, OCT angiography (OCTA), visualizes blood flow in the choriocapillaris. Howeve
