Optogenetic approaches to vision restoration.

Inherited retinal disease (IRD) affects about 1 in 3000 to 1 in 5000 individuals and is now believed to be the most common cause of blindness registration in developed countries. Until recently, the management of such conditions had been exclusively supportive. However, advances in molecular biology and medical engineering have now seen the rise of a variety of approaches to restore vision in patients with IRDs. Optogenetic approaches are primarily aimed at rendering secondary and tertiary neurons of the retina light-sensitive in order to replace degenerate or dysfunctional photoreceptors. Suc

Purkinje Cells as Gatekeepers of Seizure Susceptibility: Insights From Optogenetics.

Purkinje cells (PCs), the principal output neurons of the cerebellar cortex, are classically implicated in motor coordination via inhibitory projections to the deep cerebellar nuclei (DCN). Emerging evidence suggests their influence extends to seizure susceptibility, yet the underlying mechanisms remain unclear. Here, we investigated the functional role of PCs in locomotion and seizure-like activity in zebrafish larvae. Using the UAS/Gal4 system, we selectively expressed light-sensitive ion channels in PCs: Channelrhodopsin-2 (ChR2) to activate, and Anion ChannelRhodopsin-2 (ACR2) to inhibit n

Atypical collective oscillatory activity in cardiac tissue uncovered by optogenetics.

Many biological processes emerge as frequency-dependent responses to trains of external stimuli. Heart rhythm disturbances, that is cardiac arrhythmias, are important examples as they are often triggered by specific patterns of preceding stimuli. In this study, we investigated how ectopic arrhythmias can be induced by external stimuli in cardiac tissue containing a localised area of depolarisation. Using optogenetic in vitro experiments and in silico modelling, we systematically explored the dynamics of these arrhythmias, which are characterised by local oscillatory activity, by gradually alte

Restoring Function After Severe Spinal Cord Injury Through BioLuminescent-OptoGenetics.

UNLABELLED: The ability to manipulate specific neuronal populations of the spinal cord following spinal cord injury (SCI) could prove highly beneficial for rehabilitation in patients through maintaining and strengthening still existing neuronal connections and/or facilitating the formation of new connections. A non-invasive and highly specific approach to neuronal stimulation is bioluminescent-optogenetics (BL-OG), where genetically expressed light emitting luciferases are tethered to light sensitive channelrhodopsins (luminopsins, LMO); neurons are activated by the addition of the luciferase

Optogenetic approaches for neural tissue regeneration: A review of basic optogenetic principles and target cells for therapy.

Optogenetics has revolutionized the field of neuroscience by enabling precise control of neural activity through light-sensitive proteins known as opsins. This review article discusses the fundamental principles of optogenetics, including the activation of both excitatory and inhibitory opsins, as well as the development of optogenetic models that utilize recombinant viral vectors. A considerable portion of the article addresses the limitations of optogenetic tools and explores strategies to overcome these challenges. These strategies include the use of adeno-associated viruses, cell-specific

Using Optogenetics to Reverse Neuroplasticity and Inhibit Cocaine Seeking in Rats.

This protocol demonstrates the steps needed to use optogenetic tools to reverse cocaine-induced plasticity at thalamo-amygdala circuits to reduce subsequent cocaine seeking behaviors in the rat. In our research, we had found that when rats self-administer intravenous cocaine paired with an audiovisual cue, synapses formed at inputs from the medial geniculate nucleus of the thalamus (MGN) onto principal neurons of the lateral amygdala (LA) become stronger as the cue-cocaine association is learned. We hypothesized that reversal of the cocaine-induced plasticity at these synapses would reduce cue

Near-Infrared-Light Activatable Nanoparticles for Deep-Tissue-Penetrating Wireless Optogenetics.

Optogenetics has been developed to control the activities and functions of cells with high spatiotemporal resolution, cell-type specificity, and flexibility. However, current optogenetic tools generally rely on visible light (e.g., blue or yellow) with shallow tissue penetration ability that does require invasive fiber-optic probes to deliver visible light into organs and animal tissues. This often results in a series of side effects, such as tissue damage and unwanted inflammation. Fortunately, the emerging wireless optogenetic tools that can respond to deep-tissue-penetrating near-infrared (

IoT-enabled wireless neural implant for chronic, programmable neuropharmacology and optogenetics.

Wireless in vivo neuropharmacology and optogenetics offer a powerful way to link molecular signaling, defined neural populations, and behavior in freely moving animals. However, existing implantable wireless systems developed for this purpose often suffer from imprecise dosing, backflow contamination, and nonrefillable reservoirs, and their reliance on experimenter presence can itself alter neural activity and behavior. Here, we introduce a remotely actuated and programmable implant for drug delivery and optical stimulation (RAPIDO), which integrates a refillable, replaceable, and backflow-fre

Labeled quantitative proteomics dataset of optogenetics induced axon regeneration in mice.

This labeled quantitative proteomics dataset was collected from a transgenic channel rhodopsin mouse model (Chr2) subjected to light stimulation after traumatic optic nerve crush (ONC). Protein extraction was performed by careful mincing of the tissue in extraction buffer (TEAB, NaCl and SDS). Protein amounts were normalized across samples using dot blot densitometry and ImageJ software. Samples were labeled for quantification using a modified TMTpro™ 16plex Label Reagent Set (Thermo Scientific™) after performing an overnight trypsin digestion. Optogenetics for visual restoration: From proof of principle to translational challenges.

Degenerative retinal disorders are a diverse family of diseases commonly leading to irreversible photoreceptor death, while leaving the inner retina relatively intact. Over recent years, innovative gene replacement therapies aiming to halt the progression of certain inherited retinal disorders have made their way into clinics. By rendering surviving retinal neurons light sensitive optogenetic gene therapy now offers a feasible treatment option that can restore lost vision, even in late disease stages and widely independent of the underlying cause of degeneration. Since proof-of-concept almost

New era of optogenetics: from the central to peripheral nervous system.

Optogenetics has recently gained recognition as a biological technique to control the activity of cells using light stimulation. Many studies have applied optogenetics to cell lines in the central nervous system because it has the potential to elucidate neural circuits, treat neurological diseases and promote nerve regeneration. There have been fewer studies on the application of optogenetics in the peripheral nervous system. This review introduces the basic principles and approaches of optogenetics and summarizes the physiology and mechanism of opsins and how the technology enables bidirectio

[Optogenetic reactivation of dormant cone photoreceptors].

Optogenetics enable the restoration of light sensitivity in various retinal cell types. In degenerated retinas, dormant cones whose cell bodies are intact but no longer respond to light can be optogenetically reactivated. This restores all cone-dependent signalling pathways. In RP mouse models the hyperpolarizing opsins eNpHR and Jaws led to the optogenetic reactivation of dormant photoreceptors, with measurable light responses. This was accompanied by improvements in behavioral tests. The transplantation of optogenetically modified photoreceptors also shows functional integration into the ret

A Step-by-Step Protocol for Optogenetic Kindling.

Electrical kindling, repeated brain stimulation eventually resulting in seizures, is widely used as an animal model of epileptogenesis and epilepsy. However, the stimulation electrode used for electric kindling targets unknown neuronal populations and may introduce tissue damage and inflammation. Optogenetics can be used to circumvent these shortcomings by permitting millisecond control of activity in genetically defined neurons without gross injury or inflammation. Here we describe an easy step-by-step protocol for optogenetic kindling - optokindling - by which seizures are eventually elicite

All-optical closed-loop control of human cardiomyocyte networks exploiting holographic optogenetics.

Heart diseases can trigger life-threatening arrhythmias by disrupting cardiac contraction wavefront propagation. In contrast to conventional treatments like electrical pacemakers, optogenetics offers high spatial and temporal resolution for non-invasive interrogation and modulation of cellular activity. However, combining sensing and actuation into a real-time control loop remains challenging. A major obstacle is the data load associated with dense sensor arrays typically required for spatially resolved wavefront monitoring. To overcome this, we present an all-optical, closed-loop control stra

Pushing the Frontiers: Optogenetics for Illuminating the Neural Pathophysiology of Bipolar Disorder.

Bipolar disorder (BD), a disabling mental disorder, is featured by the oscillation between episodes of depression and mania, along with disturbance in the biological rhythms. It is on an urgent demand to identify the intricate mechanisms of BD pathophysiology. Based on the continuous progression of neural science techniques, the dysfunction of circuits in the central nervous system was currently thought to be tightly associated with BD development. Yet, challenge exists since it depends on techniques that can manipulate spatiotemporal dynamics of neuron activity. Notably, the emergence of opto

Self-adaptive cardiac optogenetics device based on negative stretching-resistive strain sensor.

Precision medicine calls for high demand of continuous, closed-loop physiological monitoring and accurate control, especially for cardiovascular diseases. Cardiac optogenetics is promising for its superiority of cell selectivity and high time-space accuracy, but the efficacy of optogenetics relative to the input of light stimulus is detected and controlled separately by discrete instruments in vitro, which suffers from time retardation, energy consumption, and poor portability. Thus, a highly integrated system based on implantable sensors combining closed-loop self-monitoring with simultaneous

Analysis of Prefrontal-Amygdala Synaptic Functions Using Optogenetics and Patch-Clamp Recording.

Duchenne muscular dystrophy (DMD) is an X-linked neuromuscular disease characterized by progressive muscle wasting caused by a mutation in the DMD gene encoding the dystrophin protein. Dystrophin is highly expressed in the central nervous system (CNS), and DMD patients are often diagnosed with abnormalities in emotional and social functions. However, the pathophysiology of the CNS in DMD patients is not fully understood. The DMD mouse models show abnormalities in anxiety, fear and social behavior, and synaptic dysfunction in the amygdala, which is involved in emotional and social processing. T

[Optogenetics and cell replacement in retinology : Regenerative ophthalmology-What we can do!].

For many degenerative retinal diseases that progressively lead to blindness, no treatment options are available so far. In recent years, several innovative therapies have been experimentally explored, which are promising because they are independent of the genetic cause of the degenerative disease. One of these is optogenetics, which involves light-sensitive proteins that selectively act as ion channels or ion pumps to control the potential of the treated cell. Thus, these cells can be stimulated or inhibited by light, quasi functionally remote controlled. In this way artificial photoreceptors

Battery-free, lightweight, injectable microsystem for in vivo wireless pharmacology and optogenetics.

Pharmacology and optogenetics are widely used in neuroscience research to study the central and peripheral nervous systems. While both approaches allow for sophisticated studies of neural circuitry, continued advances are, in part, hampered by technology limitations associated with requirements for physical tethers that connect external equipment to rigid probes inserted into delicate regions of the brain. The results can lead to tissue damage and alterations in behavioral tasks and natural movements, with additional difficulties in use for studies that involve social interactions and/or motio

Optogenetic Therapy for Restoring Aspects of Visual Function.

BACKGROUND: Retinitis pigmentosa is an inherited degenerative retinal disease that can lead to irreversible blindness. Optogenetic therapy has shown potential for restoring visual function at late stages of the disease. METHODS: In this open-label study, we evaluated the safety of ganglion cell-directed optogenetic therapy in 10 participants with blindness due to advanced retinitis pigmentosa. Each participant received a single intravitreal injection of an adeno-associated viral (AAV) vector encoding the red-shifted channelrhodopsin ChrimsonR in the worse-seeing eye. The primary outcome was s
