OECD 414 supplementary prenatal developmental toxicity study of sodium molybdate dihydrate in the rat and benchmark dose evaluation.

In a continuing investigation of the potential for reproductive and developmental toxicity of molybdenum (Mo), consequent to the previous published OECD studies [1,2] and as directed by the European Chemicals Agency [3], a supplemental rat GLP-compliant Prenatal Developmental Toxicity (PNDT) study was conducted to investigate higher dose levels of sodium molybdate dihydrate (SMD) in an identical study design (OECD 414)[4] to Murray et al. 2014a [1], at dietary concentrations calculated to provide target Mo levels of 80 and 120 mg/kg bw/day (the maximum-tolerated dose). There was no effect on p

How to resolve inconclusive predictions from defined approaches for skin sensitisation in OECD Guideline No. 497.

In June 2021 the Organisation for Economic Co-operation and Development published Guideline No. 497 on Defined Approaches for Skin Sensitisation (DASS GL). There are two DAs published, known as the 2o3 and the ITS. The 2o3 uses two concordant results from either the DPRA, KeratinoSens™, or the h-CLAT assays to predict hazard (sensitiser/non-sensitiser). The ITS applies a score to results from the DPRA, the h-CLAT and an in silico model to predict United Nations Globally Harmonized System (GHS) sub-categories (1A/1B/Not Classified). The ITS can use Derek Nexus as the in silico model (known as I

Using effect biomarker thresholds in regulatory risk assessment: an OECD-based framework for assessing cumulative exposures to chemical mixtures.

The international initiative titled: "Using Adverse Outcome Pathways (AOP) to address combined exposures to chemicals with relevant effect-biomarkers", was coordinated by the OECD's Working Parties on Exposure Assessment (WPEA) and Hazard Assessment (WPHA). It brought together over 90 experts from 25 countries to advance the regulatory and scientific use of effect biomarkers for assessing exposures to chemical mixtures and their associated biological and health-relevant effects. Effect biomarkers provide biologically integrated information that can complement traditional single-substance appro

Towards EU regulatory hazard assessment of metabolic endocrine disrupters: Integrating new biomarkers into OECD test guidelines.

Exposure to 'metabolic disrupting chemicals' (MDCs) are increasingly implicated in obesity, diabetes and/or fatty liver disease; indeed, these metabolic changes may play a role in the global metabolic disorders' epidemic. To better assess and manage the health risks of MDCs, improved hazard identification is needed. This review describes how current in vivo OECD Test Guidelines (TGs) can better capture MDC effects. The biological and clinical evidence to support the inclusion of promising human relevant biomarkers, blood parameters, endpoints and relevant tissues for MDCs for potential inclusi

Predicting points of departure and potency categories for fragrance ingredients by integrating OECD in vitro models.

Continuous potency assessment is crucial for conducting quantitative risk assessment (QRA) of sensitizers. Quantitative regression models, based on in vitro methods, have been developed to calculate points of departure for use in skin sensitization QRA. These models calculate a point of departure as a predicted value for Local Lymph Node Assay (LLNA) EC3 or potency value (PV), integrating data from the kinetic Direct Peptide Reactivity Assay (kDPRA), KeratinoSens (KS) assay, and human Cell Line Activation Test (h-CLAT). The goal of this study was to determine how in vitro predicted EC3s and PV

Assessment of skin sensitization properties of few-layer graphene and graphene oxide through the Local Lymph Node Assay (OECD TG 442B).

Skin contact is one of the most common exposure routes to graphene-based materials (GBMs) during their small-scale and industrial production or their use in technological applications. Nevertheless, toxic effects in humans by cutaneous exposure to GBMs remain largely unexplored, despite skin contact to other related materials has been associated with adverse effects. Hence, this in vivo study was carried out to evaluate the cutaneous effects of two GBMs, focusing on skin sensitization as a possible adverse outcome. Skin sensitization by few-layer graphene (FLG) and graphene oxide (GO) was eval

Investigation of the potential of bisphenol A substitutes to induce allergic contact sensitization using OECD defined approaches.

Bisphenol A is a high production volume chemical used extensively in the manufacture of polycarbonate plastics, epoxy resins, and thermal printer paper with a high potential for occupational and post-production dermal exposure. Bisphenol A-containing plastics were commonly used in food packaging, resulting in significant public exposure through leaching into foodstuff. The public is also at risk of dermal exposure due to environmental contamination. Due to public health concerns regarding the potential for endocrine disrupting effects, efforts have been applied to replace bisphenol A with sa

What makes contact tracing successful? An empirical analysis of COVID-19 data from 38 OECD countries.

BACKGROUND: Despite widespread adoption of contact tracing during the COVID-19 pandemic, countries exhibited marked differences in tracing performance. Existing studies have documented policy variation but lack standardized, comparable outcome indicators of tracing effectiveness. Recent reviews emphasize the need for quantitative, cross-national, outcome-based measures and broader frameworks that account for contextual factors such as governance, education, and culture. To address this gap, this study aims to validate the time lag (τ) between peaks in confirmed cases and testing as a proxy for

An in-depth assessment of diabetes-related lower extremity amputation rates 2000-2013 delivered by twenty-one countries for the data collection 2015 of the Organization for Economic Cooperation and Development (OECD).

BACKGROUND: International comparisons of diabetes-related lower extremity amputation rates are still hampered by different criteria used for data collection and analysis. We aimed to evaluate trends and variation of major/minor amputations, using agreed definitions adopted by the Organization for Economic Cooperation and Development in 2015. METHODS: Direct age-sex standardized rates were calculated per 100,000 subjects per year between 2000 and 2013, using major/minor amputations with diabetes diagnosis as numerators and the total population or number of people with diabetes as denominators.

Insights for dementia risk reduction among lower SES adults in OECD countries: scoping review of interventions targeting multiple common health risk factors.

The number of people living with dementia is expected to rise to 153 million cases globally by 2050. This will come at a high economic and human cost to societies with disproportionate effects on socioeconomically disadvantaged groups who experience greater exposures to- and fewer protections from- the environmental, social and behavioural drivers of dementia risk. Almost half (45%) of dementia incidence could theoretically be prevented or delayed by addressing 14 modifiable risk factors. While several studies have demonstrated the feasibility of multidomain dementia risk reduction interventio

Integrated skin sensitization assessment based on OECD methods (III): Adding human data to the assessment.

Skin sensitizer potency assessment based on new approach methodologies is key to deriving a point of departure (PoD) for risk assessment. Regression models to predict a PoD based on OECD-validated in vitro tests and trained on local lymph node assay (LLNA) data were previously presented, and results from human tests were recently compiled. To integrate both data sources, the Reference Chemical Potency List (RCPL), which provides potency values (PV) for 33 chemicals integrating LLNA and human data in a structured weight-of-evidence approach, was developed. When calculating regression models vs

Is fish embryo test (FET) according to OECD 236 sensible enough for delivering quality data for effluent risk assessment?

Over the past few years, the fish embryo test (FET) has become widely accepted as an animal-friendly protocol for ecotoxicological research. As Organisation for Economic Co-operation and Development (OECD) 236, the FET has been widely applied for simple mixture exposures under the Registration, Evaluation, Authorisation, and Restriction of Chemicals regulation of the European Union; and now its use is spreading worldwide as a supposedly reliable whole-effluent test (i.e., the testing of complex mixture exposures). However, comparative peer-reviewed data regarding the FET's efficiency for whole

A scoping review of vaccine certificate implementation in Canada and OECD countries during the COVID-19 pandemic: Outcomes and lessons learned.

Vaccine certificates were introduced during the COVID-19 pandemic to document vaccination status, promote uptake and enable safer reopening of society. While these policies supported public health efforts, their implementation raised operational, ethical and legal concerns, sparking debate about their future use in health emergencies. We conducted a scoping review to synthesize the implementation processes, challenges and outcomes of vaccine certificate systems in Canada and other OECD countries. An academic search was conducted in August 2024, across Ovid MEDLINE, Embase and Scopus using cont

Comparison of two human cell-based in vitro eye hazard assessment methods: the cytotoxicity test for evaluation of eye irritation (CEI) and the OECD 492B method.

Lately, we have developed a cytotoxicity test for evaluation of eye irritation (CEI), which is able to classify a substance as serious eye damage, eye irritant or not requiring classification. The OECD 492B method can also classify substances into three categories. The aim of the present study was to compare two methods for classifying chemicals into eye hazard categories: OECD 492B and CEI. The first was performed according to the OECD 492B guideline on SkinEthic human corneal epithelium and the second according to our internal protocol on the HDFn cell line. Eighteen substances were tested (

Backlogs, waiting times and waiting lists of elective surgeries across OECD countries.

BACKGROUND: Many OECD countries suspended elective (non-emergency) care during the pandemic to divert efforts towards COVID-19 patients, which generated a backlog of patients. OBJECTIVE: This study measures the extent to which waiting times and volume changed over time before and after COVID-19 in OECD countries (between 2016-2023). We test whether COVID-19 had a different effect on the waiting time of the patients on the list versus the wait of patients treated, two common measures of waiting times. It discusses how waiting times and volume can be used as measures of health system resilience

Development and validation of an OECD reproductive toxicity test guideline with the mudsnail Potamopyrgus antipodarum (Mollusca, Gastropoda).

Mollusks are known to be uniquely sensitive to a number of reproductive toxicants including some vertebrate endocrine disrupting chemicals. However, they have widely been ignored in environmental risk assessment procedures for chemicals. This study describes the validation of the Potamopyrgus antipodarum reproduction test within the OECD Conceptual Framework for Endocrine Disrupters Testing and Assessment. The number of embryos in the brood pouch and adult mortality serve as main endpoints. The experiments are conducted as static systems in beakers filled with artificial medium, which is aerat

Advancing Skin Sensitization Potency Categorization Using U-SENS™ in OECD TG 497.

OECD Guideline 497 includes defined approaches (DAs) that combine new approach methodologies (NAMs) to predict skin sensitization hazard and categorize potency according to the United Nations Globally Harmonized System of Classification and Labelling of Chemicals (UN GHS). To increase flexibility, the OECD allows the substitution in the DAs of NAMs addressing the same key events (KEs) in the adverse outcome pathway (AOP). This study evaluated the implementation of the U-SENS™ assay for dendritic cell activation (KE3) within two integrated testing strategy (ITS) DAs. First, EC150 threshold valu

Integrated skin sensitization assessment based on OECD methods (I): Deriving a point of departure for risk assessment.

Three guidelines covering key events in the skin sensitization adverse outcome pathway are endorsed by the Organisation for Economic Co-operation and Development (OECD). A recent guideline covers defined approaches (DA) to combine data from these tests for regulatory (sub)classification. The guideline methods provide continuous data that could charac-terize the sensitization potency on a more granular scale beyond (sub)classifications. We assembled a comprehensive database of in vitro and in vivo results from OECD guideline tests. Building on a previous approach using regression models, we pro
