Decidual vascularization during organogenesis after perigestational alcohol ingestion.

Perigestational alcohol consumption up to early organogenesis can produce abnormal maternal vascularization via altered decidual VEGF/receptor expression. CF-1 female mice were administered with 10% ethanol in drinking water for 17 days prior to and up to day 10 of gestation. Control females received water without ethanol. Treated females had reduced frequency of implantation sites with expanded vascular lumen (P < 0.05), α-SMA-immunoreactive spiral arteries in proximal mesometrial decidua, reduced PCNA-positive endothelial cells (P < 0.01) and diminished uterine NK cell numbers (P < 0.05) in

Organogenesis of endocrine pancreas from transplanted embryonic anlagen.

The number of donor human pancreas organs that can be transplanted directly or used for islet of Langerhans isolation is limited. We and others have shown that it is possible to 'grow' new pancreatic tissue in situ by transplanting embryonic organ-specific pancreatic precursor cells. This technology takes advantage of the fact that selective development of islets takes place post transplantation of embryonic pancreas and that the developing organ can attract its blood supply from an appropriate vascular bed post transplantation, enabling the transplantation of pancreas in 'cellular' form. "Osteo-Organogenesis Niche" Hyaluronic Acid Engineered Materials Directing Re-Osteo-Organogenesis via Manipulating Macrophage CD44-MAPK/ERK-ETV1/5-MRC1 Axis.

The strategy of re-organogenesis provides an optimal framework for restoring complex organ structures and functions in adult damage. While the focus has often been on restoring organogenesis stem cells, there is limited investigations of reverting the environmental niche to support this approach. The guiding principle of "Nature selects the fittest to survive" drives the intricate dynamic changes in cellular events within the niche environment, especially through immune surveillance. The extracellular matrix (ECM) serves as the "self-associated molecular patterns" of the niche, containing exte

Spatiotemporal role of SETD2-H3K36me3 in murine pancreatic organogenesis.

Pancreas development is tightly controlled by multilayer mechanisms. Despite years of effort, large gaps remain in understanding how histone modifications coordinate pancreas development. SETD2, a predominant histone methyltransferase of H3K36me3, plays a key role in embryonic stem cell differentiation, whose role in organogenesis remains elusive. Here, by combination of cleavage under targets and tagmentation (CUT&Tag), assay for transposase-accessible chromatin using sequencing (ATAC-seq), and bulk RNA sequencing, we show a dramatic increase in the H3K36me3 level from the secondary transitio

Neuron navigator 3a regulates liver organogenesis during zebrafish embryogenesis.

Endodermal organogenesis requires a precise orchestration of cell fate specification and cell movements, collectively coordinating organ size and shape. In Caenorhabditis elegans, uncoordinated-53 (unc-53) encodes a neural guidance molecule that directs axonal growth. One of the vertebrate homologs of unc-53 is neuron navigator 3 (Nav3). Here, we identified a novel vertebrate neuron navigator 3 isoform in zebrafish, nav3a, and we provide genetic evidence in loss- and gain-of-function experiments showing its functional role in endodermal organogenesis during zebrafish embryogenesis. [Organogenesis of developing metanephroi allografted into nonimmunosuppressed adult rats].

OBJECTIVE: To explore novel methods of possible donor organ supply and immunologic tolerance induction of organ transplantation. METHODS: Whole metanephroi from d14-19 (E14-E19) embryos of pregnant rats were grouped and allografted into the omenta or near remnants of renal vessels of nonimmunosupressed adult rats. At the time of implantation, host rats underwent unilateral nephrectomy. Four weeks after implantation, allografted metanephroi in host rats were removed for gross, biochemical and histopathological examination. RESULT: Four weeks post-implantation, (1) E19 and E18 metanephroi had

Dmxl1 is required for survival in the mouse to organogenesis stages of development.

Cleft lip and cleft palate are among the most common congenital anomalies and are the result of incomplete fusion of embryonic craniofacial processes or palatal shelves. Genetic factors are known to play a large role in these anomalies, but the list of known causal genes is far from complete. As part of a larger effort to sequence patients with micrognathia and cleft palate, we identified candidate pathogenic variants in dmx-like 1 (DMXL1). We used genome editing to create an allelic series of Dmxl1 in the mouse: a small deletion and the two orthologous missense variants. We do not find eviden

Conserved and Divergent Features of Human and Mouse Kidney Organogenesis.

Human kidney function is underpinned by approximately 1,000,000 nephrons, although the number varies substantially, and low nephron number is linked to disease. Human kidney development initiates around 4 weeks of gestation and ends around 34-37 weeks of gestation. Over this period, a reiterative inductive process establishes the nephron complement. Studies have provided insightful anatomic descriptions of human kidney development, but the limited histologic views are not readily accessible to a broad audience. In this first paper in a series providing comprehensive insight into human kidney f

Spatiotemporal regulation of lateral root organogenesis in Arabidopsis by cytokinin.

The architecture of a plant's root system, established postembryonically, results from both coordinated root growth and lateral root branching. The plant hormones auxin and cytokinin are central endogenous signaling molecules that regulate lateral root organogenesis positively and negatively, respectively. Tight control and mutual balance of their antagonistic activities are particularly important during the early phases of lateral root organogenesis to ensure continuous lateral root initiation (LRI) and proper development of lateral root primordia (LRP). Here, we show that the early phases of

The role of Hoxa3 gene in parathyroid gland organogenesis of the mouse.

Mice with a targeted deletion of the Hoxa3 gene have defects of derivatives of the third branchial arch and pouch. To address the role of the Hoxa3 gene in parathyroid organogenesis, we examined the third pharyngeal pouch development by immunohistochemistry (IHC) using the secretory protein (SP)-1/chromogranin A antiserum, which recognizes the parathyroid from its initial formation onward. At embryonic day (E) 11.5, the SP-1/chromogranin A-immunoreactive primary rudiment of the parathyroid appeared in the cranial region of the third pharyngeal pouch of wild-type embryos. In Hoxa3-null mutants,

Hierarchical chromatin polyvalency governs robust gene regulation and organogenesis.

Precise temporal control of gene expression is fundamental for embryonic development, yet the epigenetic and chromatin basis governing transcriptional timing remain poorly understood. The bivalency model, characterized by coexistence of H3K4me3 and H3K27me3, has been proposed to mark a poised state ready for activation. However, this model has been challenged by lacking of rapid gene activation in response to H3K27me3 depletion, suggesting that the H3K27me3 mark is not responsible for the silencing. Here, through temporal epigenomic profiling of post-implantation mouse embryos and use of the p

Embryonic Circadian Rhythm Establishment, Homeostasis, and Dysfunction During Organogenesis.

Background: Circadian rhythms play a crucial role in the management of the temporal organization of various physiological and cellular processes in mammalian cell types. These rhythms are involved in the regulation of the cell cycle and metabolism and have implications for pathogenesis and physiological homeostasis. Therapeutic approaches that target circadian regulation are emerging for the treatment of digestive disorders, metabolic diseases, and cancer. The proper coordination of cellular clocks is essential for tissue homeostasis and metabolic health. Methods: The exact mechanisms governin

Hypothalamic-pituitary organoid generation through the recapitulation of organogenesis.

This paper overviews the development and differentiation of the hypothalamus and pituitary gland from embryonic stem (ES) and induced pluripotent stem (iPS) cells. It is important to replicate the developmental process in vivo to create specific cells/organoids from ES/iPS cells. We also introduce the latest findings and discuss future issues for clinical application. Neuroectodermal progenitors are induced from pluripotent stem cells by strictly removing exogenous patterning factors during the early differentiation period. The induced progenitors differentiate into rostral hypothalamic neuron

Sonic Hedgehog Signaling in Organogenesis, Tumors, and Tumor Microenvironments.

During mammalian embryonic development, primary cilia transduce and regulate several signaling pathways. Among the various pathways, Sonic hedgehog (SHH) is one of the most significant. SHH signaling remains quiescent in adult mammalian tissues. However, in multiple adult tissues, it becomes active during differentiation, proliferation, and maintenance. Moreover, aberrant activation of SHH signaling occurs in cancers of the skin, brain, liver, gallbladder, pancreas, stomach, colon, breast, lung, prostate, and hematological malignancies. Recent studies have shown that the tumor microenvironment

Brown fat organogenesis and maintenance requires AKT1 and AKT2.

OBJECTIVE: Understanding the signaling mechanisms that control brown adipose tissue (BAT) development is relevant to understanding energy homeostasis and obesity. The AKT kinases are insulin effectors with critical in vivo functions in adipocytes; however, their role in adipocyte development remains poorly understood. The goal of this study was to investigate AKT function in BAT development. METHODS: We conditionally deleted Akt1 and Akt2 either individually or together with Myf5-Cre, which targets early mesenchymal precursors that give rise to brown adipocytes. Because Myf5-Cre also targets

Osr1 Interacts Synergistically with Wt1 to Regulate Kidney Organogenesis.

Renal hypoplasia is a common cause of pediatric renal failure and several adult-onset diseases. Recent studies have associated a variant of the OSR1 gene with reduction of newborn kidney size and function in heterozygotes and neonatal lethality with kidney defects in homozygotes. How OSR1 regulates kidney development and nephron endowment is not well understood, however. In this study, by using the recently developed CRISPR genome editing technology, we genetically labeled the endogenous Osr1 protein and show that Osr1 interacts with Wt1 in the developing kidney. Whereas mice heterozygous for

Parasympathetic innervation maintains epithelial progenitor cells during salivary organogenesis.

The maintenance of a progenitor cell population as a reservoir of undifferentiated cells is required for organ development and regeneration. However, the mechanisms by which epithelial progenitor cells are maintained during organogenesis are poorly understood. We report that removal of the parasympathetic ganglion in mouse explant organ culture decreased the number and morphogenesis of keratin 5-positive epithelial progenitor cells. These effects were rescued with an acetylcholine analog. We demonstrate that acetylcholine signaling, via the muscarinic M1 receptor and epidermal growth factor re

Xenobiotic kidney organogenesis: a new avenue for renal transplantation.

Currently many efforts are being made to apply regenerative medicine to clinical renal diseases. It has been suggested that some renal diseases which maintain renal structure can be treated by infusion of stem cells isolated from the bone marrow or adult kidney. However such cell-based therapy cannot be applied to the treatment of chronic renal disease, in which renal structure, including the kidney scaffold, is totally disrupted. Therefore, absolute kidney regeneration is needed to rebuild a whole functional kidney de novo and eliminate the requirement for dialysis. However, due to the anatom
