Establishment of an in vitro culture and regeneration protocol for the native Chilean grass Polypogon australis Brong.

Polypogon australis Brong. is a native Chilean grass frequently found colonizing metal-rich mine tailings, yet it lacks an established in vitro regeneration system to support controlled physiological and biotechnological studies. Here, we report a reproducible protocol for seed germination, callus induction, and plant regeneration using coleoptile-mesocotyl explants. Surface-sterilized seeds were germinated on Murashige and Skoog (MS) medium supplemented with sucrose, achieving a cumulative germination percentage of 47.67 ± 3.15% after 15 days. The coleoptile-mesocotyl explant proved highly re

Shoot-derived miR2111 controls legume root and nodule development.

Legumes control their nodule numbers through the autoregulation of nodulation (AON). Rhizobia infection stimulates the production of root-derived CLE peptide hormones that are translocated to the shoot where they regulate a new signal. We used soybean to demonstrate that this shoot-derived signal is miR2111, which is transported via phloem to the root where it targets transcripts of Too Much Love (TML), a negative regulator of nodulation. Shoot perception of rhizobia-induced CLE peptides suppresses miR2111 expression, resulting in TML accumulation in roots and subsequent inhibition of nodule o

Improved cotton transformation protocol mediated by Agrobacterium and biolistic combined-methods.

The combined Agrobacterium- and biolistic-mediated methods of cotton transformation provide a straightforward and highly efficient protocol for obtaining transgenic cotton. Cotton (Gossypium spp.) is the most important crop for natural textile fiber production worldwide. Nonetheless, one of the main challenges in cotton production are the losses resulting from insect pests, pathogens, and abiotic stresses. One effective way to solve these issues is to use genetically modified (GM) varieties. Herein, we describe an improved protocol for straightforward and cost-effective genetic transformation

Effect of Rhazya stricta on the developing rat fetus.

Rhazya stricta is a medicinal plant traditionally used in the treatment of diabetes mellitus, inflammation, and helminthiasis. Our objective was to determine if the plant extract has any effect on fetal development in the rat. A lyophilized extract of the plant was administered daily on three consecutive gestation days (GD) covering the period of preimplantation and organogenesis. The fetuses were examined on GD 20. Higher doses (5.0 or 8.0 g/kg) of R. stricta generally caused a reduction in maternal weight gain, compared to controls, whereas the lower doses (0.5 to 2.0 g/kg) did not. Treatmen

Heritable Phytohormone Profiles of Poplar Genotypes Vary in Resistance to a Galling Aphid.

Insect galls are highly specialized structures arising from atypical development of plant tissue induced by insects. Galls provide the insect enhanced nutrition and protection against natural enemies and environmental stresses. Galls are essentially plant organs formed by an intimate biochemical interaction between the gall-inducing insect and its host plant. Because galls are plant organs, their development is likely to be governed by phytohormones involved in normal organogenesis. We characterized concentrations of both growth and defensive phytohormones in ungalled control leaves and galls

Single-Cell Transcriptomics Reveals that A Conserved GA-GASA1-ROS Module Promotes Lateral Root Founder Cell Specification.

The specification and commitment of lateral root founder cells (LRFCs) from postembryonic pericycle cells are critical steps in LR development, yet their earliest molecular determinants remain unclear. Using single-cell transcriptomics, we resolved transcriptional transitions guiding LRFC fate in Arabidopsis thaliana and identified five distinct phases of transcriptional progression during LRFC specification. Through differentially expressed gene analysis and phenotypic observation, we showed that application of exogenous GA (20 and 50 μM) promoted the nuclear migration and division in LRFCs i

Polyamine Metabolism Is Involved in the Direct Regeneration of Shoots from Arabidopsis Lateral Root Primordia.

Plants can be regenerated from various explants/tissues via de novo shoot meristem formation. Most of these regeneration pathways are indirect and involve callus formation. Besides plant hormones, the role of polyamines (PAs) has been implicated in these processes. Interestingly, the lateral root primordia (LRPs) of Arabidopsis can be directly converted to shoot meristems by exogenous cytokinin application. In this system, no callus formation takes place. We report that the level of PAs, especially that of spermidine (Spd), increased during meristem conversion and the application of exogenous

Parasitic-Plant Parasite Rewires Flowering Pathways to Induce Stem-Derived Galls.

Gall-inducing insects manipulate host plant development, redirecting cellular fate and physiological processes to form novel structures. This phenomenon is even more intriguing when the host itself is a holoparasitic plant with minimal photosynthetic capacity. In the stem of Cuscuta campestris, the weevil Smicronyx madaranus forms galls that unexpectedly activate photosynthesis, in contrast to the typical suppression of photosynthetic activity observed in leaf-derived galls. This reversal of the usual source-to-sink transition highlights a unique form of insect-induced organogenesis. To elucid

TaANT11-4A enhances grain yield by orchestrating photosynthesis and carbohydrate metabolism through direct transcriptional activation.

The AINTEGUMENTA (ANT) transcription factor family plays important roles in floral organogenesis and plant development; however, the mechanisms by which ANT proteins regulate yield remain unclear. Here, we conducted a genome-wide search and identified 64 ANT genes in the wheat genome. Association analysis of yield-related traits in 403 wheat accessions revealed that ANT family genes were significantly associated with grain size and weight. Among these genes, TaANT11-4A showed a strong association with yield-related traits. Three single-nucleotide polymorphisms (SNPs) near TaANT11-4A modulated

KNOX1 Transcription Factors in Plants with a Special Focus on Horticultural Crops: A Review.

Class I KNOX1 (KNOTTED1-like homeobox 1) transcription factors integrate gibberellin (GA), cytokinin (CK), and auxin (IAA) signaling to maintain shoot apical meristem identity and coordinate plant organogenesis. This review examines the structural conservation, evolutionary dynamics, and regulatory architecture of KNOX1 genes across horticultural crops, drawing essential mechanistic context from model species. We synthesize KNOX1 functions in six agronomic domains, including plant architecture and branching, leaf morphogenesis and ornamental traits, floral development and sex determination, fr

The phytoplasma effector, phyllogen, structurally mimics host plant MADS transcription factors.

Phytoplasmas are obligate plant pathogens that include species causing severe developmental abnormalities in diverse plant families. Phytoplasma infections may result in dwarfism, witches' broom, virescence, and phyllody, the conversion of reproductive organs into leaf-like structures. Phyllody is due to the activity of phytoplasma effector proteins, called phyllogens, that interact with the host MADS transcription factor (MTF) network. This interaction was believed to be specific for members of the AP1/SEP/AGL6 superclade of tetrameric MTFs, key regulators of floral meristem identity and orga

GhWIP2, a WIP zinc finger protein, suppresses cell expansion in Gerbera hybrida by mediating crosstalk between gibberellin, abscisic acid, and auxin.

Cell expansion is a key determinant for the final size and shape of plant organ, and is regulated by various phytohormones. Zinc finger proteins (ZFPs) consist of a superfamily involved in multiple aspects of organ morphogenesis. However, little is known about WIP-type ZFP function in phytohormone-mediated organ growth. Using reverse genetics, RNA-seq and phytohormone quantification, we elucidated the role of a new WIP-type ZFP from Gerbera hybrida, GhWIP2, in controlling organ growth via regulation of cell expansion. GhWIP2 localizes to the nucleus and acts as a transcriptional repressor. Con

Curcumin-rich plant material production through a robust and reproducible micropropagation protocol.

Curcuma longa Linné is a widely valued spice due to its nutritional and therapeutic properties. Its production methods, vulnerability to diseases, and increasing anthropogenic pressure contribute to the limited availability of planting materials. This study aims to develop a micropropagation protocol that ensures the production of curcumin-rich planting stocks. Rhizomes were either pre-treated or not with Copper hydroxide (8%), disinfected with Mercuric chloride (HgCl₂ at 0.5%, 1%, or 2%), then treated with Ethanol (70%), followed by disinfection with Sodium hypochlorite (NaClO at 30%, 40%, or

SSR Marker-Assisted Selection and Monitoring of High-Metabolite Cell Lines in Plant Tissue Culture: Protocols and Somaclonal Variation Assessment.

Simple Sequence Repeat (SSR) marker-assisted selection has proven to be the most powerful tool for genetic monitoring and selection in in vitro culture systems. It is widely used in detecting somaclonal variation and producing true-to-type in vitro plantlets with superior quality. Genetic fidelity is the critical point in plant tissue culture, and SSR markers act as a supporter to pass this critical point through locus specificity, codominancy and high polymorphism. SSR markers allow the precise detection of genetic changes during callus formation, organogenesis, somatic embryogenesis, and lon

Metabolomics and transcriptomics combined with physiology reveal key metabolic pathway responses in tobacco roots exposed to NaHS.

Hydrogen sulfide (H2S) has emerged as a novel endogenous gas signaling molecule, joining the ranks of nitric oxide (NO) and carbon monoxide (CO). Recent research has highlighted its involvement in various physiological processes, such as promoting root organogenesis, regulating stomatal movement and photosynthesis, and enhancing plant growth, development, and stress resistance. Tobacco, a significant cash crop crucial for farmers' economic income, relies heavily on root development to affect leaf growth, disease resistance, chemical composition, and yield. Despite its importance, there remains
