Isolation and characterization of a duck reovirus strain from mature ducks in China.

In 2018, a disease characterized by splenic hemorrhage and necrosis killed ducks in a duck farm in Guangxi province, China. A duck reovirus strain was isolated from the tissues of the dead ducks by inoculating duck embryos and BHK-21 cells. Electron microscopy of the cultured the isolate showed that the viral particles were nearly round in shape and approximately 70 nm in diameter, and they were designated DRV-GL18. Sequence analysis showed that the GL18 strain viral genome was 23,419 nt in length and had 10 dsRNA segments. Phylogenetic analysis of cDNA amplicons of segments encoding the prote

Review of Viral Arthritis in Canada.

Viral arthritis/tenosynovitis, a disease caused by avian reovirus (ARV), leads to great economic losses for the chicken industry worldwide. Since autumn 2011, the poultry industries in the United States and Canada have sustained significant economic losses in the progeny of broiler breeders vaccinated with classic strains of ARV. Vaccination failure has been caused by field challenge with variant ARVs. The variant field ARVs are refractory to the immunity stimulated by classic vaccines and have become the prevalent challenge in the field. Because all genotypes described in the literature have

Postvaccinal reovirus infection with high mortality in breeder chicks.

A broiler breeder flock was subcutaneously vaccinated at the hatchery with a live avian orthoreovirus (ARV) vaccine against viral arthritis. Chicks began to die at 3 days of age and postmortem examination revealed massive subcutaneous hemorrhages and edema on the dorsal aspect of the neck at the site of vaccination, a severe necrotic hepatitis, and pulmonary edema. Microscopically, the main lesion was a multifocal vacuolar degeneration and necrosis of randomly distributed small groups of hepatocytes with presence of apoptotic and multinucleated syncytial cells. Necrotic foci were also found in

Genetics and biological characteristics of duck reoviruses isolated from ducks and geese in China.

The emergence and circulation of duck reovirus have caused severe threats to domestic waterfowl production because of the lethal infections they cause in ducks and geese. However, the evolution of circulating duck reoviruses and their replication and pathogenicity in domestic birds have not been fully investigated. In this study, we identified and isolated six duck reoviruses from clinical samples of sick or deceased farmed ducks and geese and sequenced their full genomes. Phylogenetic analysis revealed the evolutionary landscape of duck reoviruses and the complex reassortment of these circula

Mortality associated with avian reovirus infection in a free-living magpie (Pica pica) in Great Britain.

BACKGROUND: Avian reoviruses (ARVs) cause a range of disease presentations in domestic, captive and free-living bird species. ARVs have been reported as a cause of significant disease and mortality in free-living corvid species in North America and continental Europe. Until this report, there have been no confirmed cases of ARV-associated disease in British wild birds. CASE PRESENTATION: Sporadic individual magpie (Pica pica) mortality was detected at a single site in Buckinghamshire, England, April-September 2013. An adult female magpie was found moribund and subsequently died. Post-mortem e

Central nervous system signs in chickens caused by a new avian reovirus strain: a pathogenesis study.

The present study describes the pathogenesis of infection of chicks with a new avian reovirus strain, belonging to the so-called enteric reovirus strains (ERS) that is capable of causing central nervous system signs in SPF white leghorns. After intramuscular (IM) or oral inoculation birds were either observed for clinical signs or sacrificed for macroscopic, histological and virological examination for 21 days. Virus isolation was performed on the brain, leg muscle, hock joint, liver and spleen. For the detection of viral antigen the immunohistochemistry (IHC) technique was performed on the ca

Fowl adenovirus strains 1/A and 11/D isolated from birds with reovirus infection.

Inclusion body hepatitis (IBH) is, in some cases, a fatal disease affecting fowl by adenovirus strains which are subdivided into 5 species (A-E). In the current study, we investigated sequences from the Loop L1 region of the hexon gene of sequences of adenovirus field stains 1/A and 11/D isolated from a poultry flock co-infected with IBH and avian reoviruses ARVs. In early 2021, an epidemiologic survey highlighted the coinfection adenoviruses with other viruses (orthoreovirus infection) as being particularly deleterious within the poultry industry. Here, we investigated the Loop L1 HVR1-4 regi

Essential role of the D domain of linc000889 in inhibiting avian reovirus replication.

Avian reovirus (ARV) is a double-stranded RNA virus that can cause immunosuppression, irregular bleeding, spleen necrosis, and other symptoms in ducks, posing a serious threat to the poultry industry. Previous studies have confirmed that the duck-derived linc000889 can inhibit ARV replication. In this study, qRT-PCR revealed that linc000889 could promote the expression of interferon-β (IFN-β) and its downstream genes Mx and OASL. Subsequently, using RNA pull-down and protein mass spectrometry, we identified that the host protein NLRX1 associates with the sense strand of linc000889. This bindin

Synergistic pathogenicity of novel duck Orthoreovirus and salmonella typhimurium in ducks.

Co-infection with Novel Duck Orthoreovirus (N-DRV) and Salmonella Typhimurium (S. Typhimurium) poses significant threats to duck health. The disease associated with N-DRV is characterized by epizootic outbreaks featuring hepatosplenomegaly, hemorrhage, and necrosis. Ducklings infected with STM primarily exhibit symptoms such as fibrinous pericarditis, perihepatitis, and intestinal hemorrhage. In recent years, China has experienced outbreaks involving mixed infections of these two pathogens in various regions, leading to more complex and atypical clinical manifestations. This study investigated

Novel multigene molecular characterization of avian reovirus strains and associated embryonic pathogenicity.

UNLABELLED: Avian reovirus (ARV) infection causes a variety of health problems in poultry, including tenosynovitis/arthritis, hepatitis, malabsorption, etc., leading to huge economic losses. Therefore, regular monitoring of ARV strains is crucial for detecting new variants to mitigate clinical disease. Traditional classification approaches have focused primarily on a single gene, σC, which encodes the viral spike protein that elicits neutralizing antibodies. However, additional capsid proteins could play a role by inducing a host antiviral immune response. In this study, the λC, μB, σC, and σB

Nanovaccine based on chitosan oligosaccharide-polyethyleneimine packaged circRNA encoding sigma C proteins elicits protective immunity against novel duck reovirus in ducks.

The promise of mRNA vaccines lies in their design flexibility and efficient immune activation. Emerging as a next-generation platform, circular RNAs (circRNAs) offer superior molecular stability and lower immunogenicity than their linear mRNA counterparts. However, the feasibility of a circRNA vaccine against avian viral diseases remains unexplored. To address this, we devised a novel nanovaccine based on the natural biomacromolecule chitosan oligosaccharide conjugated polyethylenimine (CS-PEI) packaged circRNA platform for avian species and evaluated its efficacy and protective potential. We

Single dose of recombinant baculovirus vaccine expressing sigma B and sigma C genes provides good protection against novel duck reovirus challenge in ducks.

The novel duck reovirus (NDRV) disease causes high economic losses, resulting in substantial economic losses in waterfowl industry. However, currently, no commercial vaccines are available to alleviate NDRV infection throughout the world. Here, we developed two subunit vaccine candidates for NDRV based on the insect cell-baculovirus expression system (IC-BEVS). Two recombinant viruses, namely rBac-σB and rBac-σC, were successfully generated based on the consensus sequence of NDRV. Then, the σB and σC subunit vaccine candidates were prepared by directly inactivating the recombinant virus infect

Isolation and Pathogenicity Analysis of a Novel Orthoreovirus Caused the Outbreak of Duck Viral Arthritis in China.

Novel duck orthoreovirus can infect various hosts, mainly causing spleen necrosis and hemorrhagic necrotizing hepatitis, leading to immunosuppression and growth impairment. Infectious diseases characterized by swollen hock joints, movement disorders, and impaired growths have recently occurred in significant duck breeding areas in China. Infected ducks from Shandong Province were selected, and N-DRV-LY20 was isolated from the swollen joints. Next-generation sequencing (NGS) of N-DRV-LY20 and homology analysis showed that all ten fragments had the highest homology to N-DRV and the closest affin

Hypericin-loaded graphene oxide protects ducks against a novel duck reovirus.

Novel duck reovirus (NDRV) disease is a serious infectious disease for poultry, for which no effective therapy has been established. Therefore, development of novel antivirals against NDRV is urgently needed. In present study, we developed a complex wherein hypericin (HY), which shows broad-spectrum antiviral activity, was loaded onto graphene oxide (GO), which has a high drug-loading capacity and low cytotoxicity. The antiviral activity of the complex (GO/HY) was studied in DF-1 cells and in ducklings infected with the NDRV TH11 strain. GO/HY showed a dose-dependent inhibition of NDRV replica

Comparative analysis of viral biological characteristics and pathogenicity of representative prevalent avian reovirus strains from genotypes I to V.

The variable pathogenicity of avian reovirus (ARV), driven by genotypic diversity, poses significant control challenges to the global poultry industry. However, biological and pathogenic distinctions among genotypes remain poorly characterized. We performed a comprehensive comparison of 14 ARV strains (genotypes I-V), evaluating viral biological characteristics and pathogenicity indicators. Phylogenetic analysis of these isolates and field strains in China based on the σC gene revealed co-circulation of multiple genotypes (predominantly I and II), with epidemic strains actively evolving and ge

A σC-protein-based indirect enzyme-linked immunosorbent assay for clinical detection of antiavian reovirus antibodies.

Avian reovirus (ARV) is the causative agent of avian viral arthritis and causes significant economic losses to the global poultry industry. For clinical diagnosis, detecting ARV-specific antibodies is crucial. We successfully expressed the ARV-σC protein in insect cells using the baculovirus expression vector system, achieving an expression level of approximately 200 mg/L. We developed an indirect enzyme-linked immunosorbent assay (iELISA) using the ARV-σC protein as a coating antigen to detect antibodies against it. The inter-batch and intrabatch coefficients of iELISA variation were less tha

Maternal antibodies reduce costs of an immune response during development.

Young vertebrates are dependent primarily on innate immunity and maternally derived antibodies for immune defense. This reliance on innate immunity and the associated inflammatory response often leads to reduced growth rates after antigenic challenge. However, if offspring have maternal antibodies that recognize an antigen, these antibodies should block stimulation of the inflammatory response and reduce growth suppression. To determine whether maternal and/or offspring antigen exposure affect antibody transmission and offspring growth, female Japanese quail (Coturnix japonica) and their newly

Genotypic characterization and molecular evolution of avian reovirus in poultry flocks from Brazil.

Avian reovirus (ARV) is one of the main causes of infectious arthritis/tenosynovitis and malabsorption syndrome (MAS) in poultry. ARVs have been disseminated in Brazilian poultry flocks in the last years. This study aimed to genotype ARVs and to evaluate the molecular evolution of the more frequent ARV lineages detected in Brazilian poultry-producing farms. A total of 100 poultry flocks with clinical signs of tenosynovitis/MAS, from all Brazilian poultry-producing regions were positive for ARV by PCR. Seventeen bird tissues were submitted to cell culture and ARV RNA detection/genotyping by two

Evidence for vertical transmission of avian reovirus in chickens.

Avian reovirus (ARV) has emerged as a highly prevalent pathogen in Chinese poultry, characterized by its high mutation rate, environmental resilience, and dual vertical-horizontal transmission routes, leading to rising chick morbidity and substantial economic losses. To evaluate vertical transmission, 945 dead embryos and 58 weak chicks from a Guangdong hatchery between January 2023 and December 2024 were tested, yielding a 9.6 % (96/1003) ARV positivity rate. Three strains (GD/SB/202301, WMC06, LN02) were successfully isolated. Phylogenetic analysis classified GD/SB/202301 and WMC06 as genoty
