Vacuolation of the retinal pigment epithelium in the foveomacular region of aging rhesus monkey eyes was re-examined with the eyes initially fixed in osmium tetroxide to retain the alcohol-soluble lipid. All vacuoles were found to be completely occupied by lipid material. Similar cells were positive to oil red O. The vacuolation of the pigment epithelial cell is considered to be a form of lipoidal degeneration.

We compare the ultrastructure of the gerbil sublingual gland as seen after cryofixation followed by substitution with osmium tetroxide, with the more familiar appearance of material processed by glutaraldehyde-osmium chemical fixation. In previous work of our laboratory it was demonstrated that collidine (2,4,6-trimethylpyridine) abolishes the core osmiophilia and chromaffin reaction from rat pinal gland and vas deferens nerves. This abolition was apparent when tissues were briefly incubated in collidine or when they wer fixed in glutaraldehyde or osmium tetroxide using collidine as a buffer substance. Nigerian Echis carinatus venom incorporated into sphingomyelin-cholesterol liposomes stabilized with osmium tetroxide produced a rapid, sustained and protective immune response when injected i.v. into sheep. The response was similar to that reported earlier in mice. The implications of the findings are discussed in relation to the production of cheaper and more effective antivenoms. The possible use of immunostimulants for increasing the protective effect of antisera raised using this method are also considered.

Serotonin-like immunoreactivity (5-HT-LI) has been localized at the ultrastructural level in enterochromaffin (EC) cells of rat gastrointestinal tract. Ultra-thin sections of tissues embedded in epoxy resin were incubated with 5-HT antisera and antibody binding sites were visualized with protein A-gold. During fixation of fleas infected with plague agent (with preliminary cut heads, limbs and posterior part of the abdomen) in 2.5% glutaric dialdehyde or 2% osmium tetroxide their disinfection was obtained in two days. After double fixation with 2-hour exposure in each of these fixators disinfection of the material was acheived only after additional 2-day maintenance in 70% ethyl alcohol. Fleas, which had been placed into fixators or in 70% ethyl alcohol after double fixation without preliminary dissection, were disinfected completely only in 10 days.

Fifty-two adults and 31 children, who were treated with osmic acid for one knee joint, were observed for 8-18 years. No other local treatment of the knee joints was given.
