Herd-level seroprevalence of Fasciola hepatica and Ostertagia ostertagi infection in dairy cattle population in the central and northeastern Poland.

BACKGROUND: Fasciola hepatica and Ostertagia ostertagi infections are widespread in cattle population of Europe, however data on their prevalence in Poland are only fragmentary. Therefore, the cross-sectional study was carried out to determine the herd-level seroprevalence of F. hepatica and O. ostertagi infection in dairy cattle population in the central and north-eastern provinces Poland, and to identify basic local risk factors for these infections. In total, 598 herds were enrolled, 394 (65.9%) in the north-eastern province and 204 (34.1%) in the central province. In each herd the question

Apparent prevalence of and risk factors for infection with Ostertagia ostertagi, Fasciola hepatica and Dictyocaulus viviparus in Swiss dairy herds.

Infections with helminth parasites can negatively affect performance of dairy cows. Knowledge on infection intensity, spatial distributions and risk factors are key to develop targeted treatment strategies. Canada and most EU countries have conducted large investigations, but respective data for Switzerland were missing. We now performed a bulk tank milk serosurvey for Ostertagia ostertagi, Fasciola hepatica, and Dictyocaulus viviparus on a total of 1036 voluntarily participating dairy herds that were sampled at confinement periods, i.e. in winter 2014/15 or 2015/16, respectively. All samples

Measuring the effect of avermectins and milbemycins on somatic muscle contraction of adult Haemonchus contortus and on motility of Ostertagia circumcincta in vitro.

The mechanism of anthelmintic resistance against the widely used macrocyclic lactones (MLs) is still not fully understood. Pharyngeal, somatic body muscles and the ovijector have been proposed as putative sites of action as well as resistance. In the present study the effects of three avermectins and three milbemycins on adult parasitic nematodes were evaluated in vitro. The Muscle Transducer system was used to investigate the effects of MLs on muscle contraction in female Haemonchus contortus and effects on motility were measured in Ostertagia (Teladorsagia) circumcincta using the Micromotili

Transcriptomic Analysis of Peripheral Blood Mononuclear Cells During Ostertagia ostertagi Infection in Cattle Highlights a Generalized Host Immune Reaction.

One of the most concerning ruminant infections is the parasite Ostertagia ostertagi. Known commonly as the brown stomach worm, it is ingested by grazing cattle where it then progresses its life stages, occupying the host abomasum and then the intestine, causing illness. This results in lower commercial production and at worst, death of young calves. Over time, anthelmintic treatment has become less efficacious against cattle nematodes. As a result, alternative control strategies are needed. Our study looked to elucidate mechanisms underlying attenuation of the host immune response by examining

The effects of a concurrent infection of Trichostrongylus colubriformis and Ostertagia circumcincta on calcium, phosphorus and magnesium transactions along the digestive tract of lambs.

Twelve lambs, paired on the basis of live weight, were cannulated in the abomasum, proximal jejunum and terminal ileum. Six were infected with 3000 T. colubriformis and 3000 O. circumcincta larvae per day for 18 weeks. The other six lambs were pair-fed to the infected lambs. All animals were offered fresh ryegrass and white clover pasture, cut daily. Dry matter intake, live weight and plasma Ca, P and Mg concentrations were measured throughout the experiment. During weeks 7 and 17 post-infection, digesta flow along the gastrointestinal tract of infected and control animals was measured. Parasi

Serological evidence of Ostertagia ostertagi infection in dairy cows does not impact the efficacy of rabies vaccination during the housing period.

Nematode infections modulate the immune reaction of humans and livestock and may impair immune responses to non-parasitic antigens such as those present in vaccines. In this study, the relationship between antibodies directed against Ostertagia ostertagi, the economically most important nematode infection of cattle in temperate regions, and the magnitude and the kinetics of the antibody response to rabies vaccination was investigated in a commercial dairy herd of 46 cows. During the stabling period, all animals received a single intramuscular administration with a commercial inactivated rabies

Evaluation of immunization with gut membrane glycoproteins of Ostertagia ostertagi against homologous challenge in calves and against Haemonchus contortus in sheep.

Peanut and ConA lectins were used as ligands to isolate glycoproteins from detergent extracts of adult Ostertagia ostertagi membranes. As judged by their profiles following SDS-PAGE, these fractions closely resembled the equivalents from Haemonchus contortus which are derived from the nematode intestinal cell microvillar membranes and which are highly protective when used as antigens. Groups of calves were immunized with the peanut and ConA binding fractions of Ostertagia, either as separate or pooled antigens mixed with QuilA as adjuvant. All calves, including controls immunized with adjuvant

Exposure to Ostertagia ostertagi in Swedish dairy cattle: Insights from a 20-year comparison based on BTM-ELISA and questionnaire analysis.

Gastrointestinal parasites, especially Ostertagia ostertagi - the stomach worm - are known to affect the health and productivity of cattle. This study investigates the level of O. ostertagi antibodies on Swedish dairy farms and aims to investigate differences in the parasitic exposure over 20 years. Bulk-tank milk (BTM) samples were taken from 509 farms across Sweden, and the results were analysed along a north-south gradient in relation to three main country regions. Antibody levels against the stomach worm were quantified using a standardised and validated enzyme linked immunosorbent assay (

An improved model for the population dynamics of cattle gastrointestinal nematodes on pasture: parameterisation and field validation for Ostertagia ostertagi and Cooperia oncophora in northern temperate zones.

Gastrointestinal nematodes (GIN) are amongst the most important pathogens of grazing ruminants worldwide, resulting in negative impacts on cattle health and production. The dynamics of infection are driven in large part by the influence of climate and weather on free-living stages on pasture, and computer models have been developed to predict infective larval abundance and guide management strategies. Significant uncertainties around key model parameters limits effective application of these models to GIN in cattle, however, and these parameters are difficult to estimate in natural populations

A calcium-activated nucleotidase secreted from Ostertagia ostertagi 4th-stage larvae is a member of the novel salivary apyrases present in blood-feeding arthropods.

Apyrases (ATP-diphosphohydrolase) comprise a ubiquitous class of glycosylated nucleotidases that hydrolyse extracellular ATP and ADP to orthophosphate and AMP. One class of newly-described, Ca2+-dependent, salivary apyrases known to counteract blood-clotting, has been identified in haematophagous arthropods. Herein, we have identified a gene (Oos-apy-1) encoding a protein that structurally conforms to the Ca2+-activated apyrase from the bed bug, Cimex lectularius, by immunologically screening an Ostertagia L4 cDNA expression library. The expressed protein (rOos-APY-1) was biochemically functio

The use of bulk-tank milk ELISAs to assess the spatial distribution of Fasciola hepatica, Ostertagia ostertagi and Dictyocaulus viviparus in dairy cattle in Flanders (Belgium).

Fasciola hepatica, Ostertagia ostertagi and Dictyocaulus viviparus are helminth parasites with a wide distribution and an important economic impact in cattle in temperate climates. This paper describes the spatial distribution of F. hepatica, O. ostertagi and D. viviparus in dairy herds in Flanders (Belgium). One thousand eight hundred herds were selected at random from the Flemish dairy population (n=7002), stratified on community level to obtain a sample representative for the entire study area. From each herd, a bulk milk sample collected in autumn 2006 was analysed with previously describe

Multinomial logistic regression based on neural networks reveals inherent differences among dairy farms depending on the differential exposure to Fasciola hepatica and Ostertagia ostertagi.

Fasciola hepatica and Ostertagia ostertagi are cattle parasites with worldwide relevance for economic outcome as well as animal health and welfare. The on-farm exposure of cattle to both parasites is a function of host-associated, intrinsic, as well as environmental and farm-specific, extrinsic, factors. Even though knowledge on the biology of both parasites exists, sophisticated and innovative modelling approaches can help to deepen our understanding of key aspects fostering the exposure of dairy cows to these pathogens. In the present study, multiple multinomial logistic regression models we

A molecular assessment of Ostertagia leptospicularis and Spiculopteragia asymmetrica among wild fallow deer in Northern Ireland and implications for false detection of livestock-associated species.

BACKGROUND: Wild deer populations utilizing livestock grazing areas risk cross-species transmission of gastrointestinal nematode parasites (GINs), including GINs with anthelmintic resistance (AR) traits. Wild deer have been shown to carry problematic GIN species such as Haemonchus contortus and Trichostrongylus species in the UK, but the presence of livestock GINs in Northern Ireland deer populations is unknown. Also, is it not known whether AR traits exist among GINs of deer such as Ostertagia leptospicularis and Spiculopteragia asymmetrica in pastureland where anthelmintics are heavily used.

Integration of ITS-2 rDNA nemabiome metabarcoding with Fecal Egg Count Reduction Testing (FECRT) reveals ivermectin resistance in multiple gastrointestinal nematode species, including hypobiotic Ostertagia ostertagi, in western Canadian beef cattle.

A large-scale Fecal Egg Count Reduction Test (FECRT) was integrated with ITS-2 rDNA nemabiome metabarcoding to investigate anthelmintic resistance in gastrointestinal nematode (GIN) parasites in western Canadian beef cattle. The study was designed to detect anthelmintic resistance with the low fecal egg counts that typically occur in cattle in northern temperate regions. Two hundred and thirty-four auction market-derived, fall-weaned steer calves coming off pasture were randomized into three groups in feedlot pens: an untreated control group, an injectable ivermectin treatment group, and an in

A model for the development of the free-living stages of Ostertagia leptospicularis, used in conjunction with on-farm egg count data, to estimate sources of pasture contamination on New Zealand red deer (Cervus elaphus) farms.

Farmed red deer face challenges from nematode parasites, primarily the pulmonary species Dictyocaulus eckerti and the complex of Ostertagiinae nematodes in the abomasum. Previous investigations on New Zealand deer farms identified limited seasonality in faecal egg and larval output in all stock classes, however, this does not indicate the key times of year those eggs develop into infective-stage larvae, and subsequently contribute to infection risk. A simple temperature-driven model was developed for the free-living stages of a representative deer-specific Ostertagiinae species; Ostertagia lep

Multispecies helminth parasitism of grazing dairy cows in Germany and Austria, examined in the housing period.

Helminth composition and burden data for dairy cows have not been reported for >40 years for Germany and even less information is available for Austria. In the context of two recent studies, helminth parasitism was studied in 32 cows (23 from six farms in Bavaria and Tyrol; 9 from one farm in Saxony) from pasture-based dairy farms necropsied during the housing period. Helminths were enumerated and identified based on morphological characters (all helminths but rumen flukes) or molecular techniques (rumen flukes). Thirteen species of gastrointestinal nematodes and two species each of liver fluk

Efficacy of the morantel sustained release trilaminate bolus against gastrointestinal nematodes and its influence on immunity in calves.

An experiment was conducted in calves to investigate the efficacy of a morantel sustained release trilaminate bolus (MSRT) to control gastrointestinal parasitism and to assess the development of immunity during the use of MSRT. Two groups (M and U) of four calves each were infected three times a week with a mixed Ostertagia ostertagi and Cooperia oncophora infection for 12 weeks. Calves of Group M received an MSRT at the start of the experiment. Twenty weeks after the start of the experiment, all animals, including a previously uninfected control group (C), received a challenge with 100,000 Os

Anthelmintic resistance of gastrointestinal nematodes in dairy calves within a pasture-based production system of south West Western Australia.

The objective of this study was to determine the prevalence of gastrointestinal nematodes among post-weaned calves aged between 4 and 12 months old within a pasture-based system of south west Australia and quantify the level of anthelmintic resistance. Pre-treatment FECs were monitored on 14 dairy farms. Anthelmintic resistance was assessed on 11 of the farms. Control FECs were compared with anthelmintic FECs at 14 days post-treatment with doramectin (injectable), levamisole (oral), fenbendazole (oral) and a levamisole/abamectin combination (pour-on). Results demonstrate a strong level of anth

Gastrointestinal nematode populations in stabled ewes of Rimouski region.

Nematode populations is stabled ewes of the Rimouski region were studied by means of fecal worm egg counts, fecal culture of larvae, and worm counts at necropsy. It was found that during the winter strongyle egg counts were low, Trichostrongylus eggs being most numerous, The stronglye egg counts increased following lambing and reached peak in June. Ostertagia spp was the principal contributor to this "spring-rise", with substantial contribution from Trichostrongylus and Haemonchus contortus.
