Anti-parasitic activity of pelleted sainfoin (Onobrychis viciifolia) against Ostertagia ostertagi and Cooperia oncophora in calves.

BACKGROUND: Increasing anthelmintic-resistance in nematodes of ruminants emphasises the need for sustainable parasite control. Condensed tannin-containing legume forages such as sainfoin (Onobrychis viciifolia) have shown promising anthelmintic properties in small ruminants but this has never been explored in cattle. Therefore, our aim was to examine the efficacy of sainfoin against cattle nematodes in vivo. METHODS: Fifteen Jersey male calves (2-4 month-old) were allocated into two groups and fed isoproteic and isoenergetic diets mainly composed of sainfoin pellets (Group SF; n = 9, three pe

Breed-dependent associations of production characteristics with on-farm seropositivity for Ostertagia ostertagi in dairy cows.

BACKGROUND: Pasture-borne parasites like Ostertagia ostertagi have a negative effect on dairy cow health and productivity. The aim of the present study was to assess potential breed-dependent associations of O. ostertagi seropositivity with dairy cow production traits, i.e. milk yield, milk fat and milk protein. METHODS: We describe these associations in German Holstein (GH) cows, a specialised dairy breed, compared with a dual-purpose breed, i.e. German Simmental (SIM). Data from 560 farms across Germany housing 93,030 dairy cows were included. Of the 560 farms, 383 farms housed GH cows and

Behavioural response to gastrointestinal parasites of yearling dairy calves at pasture.

AIMS: To investigate the association between gastrointestinal parasites (GIP) and animal behaviour in dairy calves under New Zealand pastoral conditions, using animal-mounted, accelerometer-based sensors. METHODS: Thirty-six, 5-6-month-old, Friesian-Jersey, heifer calves fitted with animal activity sensors to track behaviour were randomly allocated to one of two treatment groups. Half the animals were challenged with an oral dose of 20,000 larvae of Ostertagia ostertagi and Cooperia oncophera once a week for 3 weeks and half were unchallenged. Five weeks after the last dose, seven infected an

Multinomial logistic regression based on neural networks reveals inherent differences among dairy farms depending on the differential exposure to Fasciola hepatica and Ostertagia ostertagi.

Fasciola hepatica and Ostertagia ostertagi are cattle parasites with worldwide relevance for economic outcome as well as animal health and welfare. The on-farm exposure of cattle to both parasites is a function of host-associated, intrinsic, as well as environmental and farm-specific, extrinsic, factors. Even though knowledge on the biology of both parasites exists, sophisticated and innovative modelling approaches can help to deepen our understanding of key aspects fostering the exposure of dairy cows to these pathogens. In the present study, multiple multinomial logistic regression models we

Phylogenetic characterization of β-tubulins and development of pyrosequencing assays for benzimidazole resistance in cattle nematodes.

Control of helminth infections is a major task in livestock production to prevent health constraints and economic losses. However, resistance to established anthelmintic substances already impedes effective anthelmintic treatment in many regions worldwide. Thus, there is an obvious need for sensitive and reliable methods to assess the resistance status of at least the most important nematode populations. Several single nucleotide polymorphisms (SNPs) in the β-tubulin isotype 1 gene of various nematodes correlate with resistance to benzimidazoles (BZ), a major anthelmintic class. Here we descri

An attempt to produce Ostertagia circumcincta infections in Mongolian gerbils.

Adult infections of Ostertagia circumcincta were successfully established in gerbils, although worm growth was retarded compared with infections in sheep. Chronic administration of beta-methasone to the host enhanced development of the parasite reproductive system but did not increase infection rate or prevent worm rejection. Infection rates achieved with single infections were not increased by using daily or weekly trickle infection regimes with or without prior exsheathment of larvae. Examination of stomachs from infected gerbils demonstrated pathological changes in the pyloric region which

Studies on the effect of treatment with anthelmintics on weight gain in calves, naturally infected with gastrointestinal nematodes.

To avoid disease outbreaks caused by gastro-intestinal nematodes different prophylactic measures are recommended. In this connection pasture hygienic achievements and anthelmintics are used. In practice it is often difficult to realize pasture hygienic steps as move the animals to parasite free areas and to avoid that they come back to the infected. In many cases the parasite control is limited to routine deworming. Our intention was to study if anthelmintic treatment without other measures had any effect on weight gain and number of eggs in faeces in calves naturally infected with Ostertagia

Teladorsagia circumcincta resistance to moxidectin and multiple anthelmintic groups in ewes following use of the persistent drug before lambing.

This paper describes the nematode control strategy adopted by a Scottish hill sheep farmer. It provides an example of the limited use of anthelmintic drugs, targeted towards control of the periparturient rise in faecal nematode egg output in ewes and exploitation of grazing management, resulting in limited exposure of naive sheep to infective larvae on pasture. Resistance to benzimidazole, imidazothiazole and macrocyclic lactone anthelmintics, and to a combination of a macrocyclic lactone and imidazothiazole anthelmintic drugs was diagnosed. The targeted use of a persistent anthelmintic drug t

Persistent efficacy of doramectin against experimental nematode infections in calves.

Three studies were conducted involving cattle exposed to experimental nematode infections. These studies were designed to investigate the prophylactic activity of a single subcutaneous treatment of doramectin at 200 micrograms kg-1 body weight against infections of Ostertagia ostertagi, Cooperia oncophora and Dictyocaulus viviparus. For each study, parasite-naive calves were randomly allocated to either a treated or a matched control group. One group received doramectin and the other received doramectin and the other received either no treatment or an injection of saline at 1 ml per 50 kg body

The prophylactic effect of ivermectin treatment of calves, three weeks after turnout, on gastro-intestinal helminthiasis.

In two grazing experiments carried out in 1982 and 1983 the prophylactic effect on gastro-intestinal helminthiasis of a single ivermectin treatment of calves 3 weeks after turnout was studied in animals which were turned out early on contaminated pasture and in calves which were turned out late on mown pasture. A single ivermectin treatment 3 weeks after early turnout did not prevent heavy helminth infections. However, such single treatment of calves 3 weeks after a late turnout on mown pasture appears to be a promising way of preventing heavy infections. Late turnout on mown pasture without a

Modelling the consequences of targeted selective treatment strategies on performance and emergence of anthelmintic resistance amongst grazing calves.

The development of anthelmintic resistance by helminths can be slowed by maintaining refugia on pasture or in untreated hosts. Targeted selective treatments (TST) may achieve this through the treatment only of individuals that would benefit most from anthelmintic, according to certain criteria. However TST consequences on cattle are uncertain, mainly due to difficulties of comparison between alternative strategies. We developed a mathematical model to compare: 1) the most 'beneficial' indicator for treatment selection and 2) the method of selection of calves exposed to Ostertagia ostertagi, i.

Economic value and course of infection after treatment of cattle having a low level of nematode parasitism.

To determine whether it is economically advantageous to treat calves having inapparent parasitism, we conducted experiments from 1971 to 1976, involving more than 1,800 calves from 30 pens or lots, using formulations of thiabendazole, levamisole, and crufomate (ruelene). Differential egg counts, cultured larvae, and cultured parasitic stages were used to estimate the kind and degree of nematode parasitism. Differentiation of infective larvae consistently established Cooperia as the predominating (%) genus in all fecal samplings. Bunostomum, when initially present, decreased or disappeared, whe

The effect of mixed grazing of sheep and cattle on worm burdens in lambs.

Lambs from ewes grazed without cattle at 4.9, 7.4 and 9.9 ewes and lambs per hectare and from ewes run with steers at an equivalent median stocking rate at four ratios in which sheep comprised 30%, 40%, 50% and 70% of the animal equivalents were killed and examined for worms. Time-dependent RNA transcriptional profiling of abomasal mucosa in cattle infected with Ostertagia ostertagi.

In this study, we infected Holstein calves with Ostertagia ostertagi stage 3 larvae (L3) and determined gene expression profiles of abomasal fundic and pyloric mucosa by RNA sequencing (RNA-seq) at 3-5, 7-9, 10, and 21 days post-infection (dpi), which represent late L3 and early L4 (between 3-5 dpi), mid to late L4 (between 7-9 and 10 dpi) and adult stages (21 dpi) of the parasitic stage. Bioinformatics analyses were performed to profile the transcriptomic changes over time as well as between cattle abomasal tissues. The results will help understand the gastric responses of the host, especiall

The effect of repeated, four-weekly eprinomectin treatment on milk production in pasture-based, seasonally-calving dairy cattle.

A randomised clinical trial from the North Island of New Zealand was conducted to assess the effect of repeated anthelmintic treatment on milk production, and to assess factors that affect treatment response. Nine hundred and twenty three multiparous, lactating dairy cattle from three pasture-based, spring-calving dairy herds were enrolled in this trial. Within each herd, cattle were stratified on age and calving date, and were randomly allocated to treatment (n=319) or control (n=604) groups. The treatment group received ≥ 0.05 mg/kg of topical eprinomectin every 28 days for eight treatments

In vivo passage through calves of nematophagous fungi selected for biocontrol of parasitic nematodes.

The experiment was designed to test the survival and performance of stress selected nematophagous fungi after passage through the gastro-intestinal tract of cattle. Ruminating calves were fed daily with a fixed amount of fungal material grown on barley grains. The excreted dung was collected on days four and five after the start of the feeding experiment. Barley grains were washed out of the excreted dung and incoculated on water-agar plates. After incubation for one week, nine of ten fungal isolates were re-isolated from these plates. The predatory capacity of the fungi in the excreted faeces

Development of Bovine Gastric Organoids as a Novel In Vitro Model to Study Host-Parasite Interactions in Gastrointestinal Nematode Infections.

Gastro-intestinal nematode (GIN) parasites are a major cause of production losses in grazing cattle, primarily through reduced growth rates in young animals. Control of these parasites relies heavily on anthelmintic drugs; however, with growing reports of resistance to currently available anthelmintics, alternative methods of control are required. A major hurdle in this work has been the lack of physiologically relevant in vitro infection models that has made studying precise interactions between the host and the GINs difficult. Such mechanistic insights into the infection process will be valu

Rapid selection for β-tubulin alleles in codon 200 conferring benzimidazole resistance in an Ostertagia ostertagi isolate on pasture.

Resistance to benzimidazoles (BZs) is widespread in sheep nematodes and increasing in those of cattle. Several reasons including the predominant use of pour-on anthelmintics and lack of scales in field conditions lead to under-dosing of cattle and therefore to increased selection pressure. In an field experiment the frequency of BZ-resistance associated allele (TAC) in codon 200 in the β-tubulin isotype 1 gene of Ostertagia ostertagi was monitored over one grazing season (approximately 30 weeks). Group 1, consisting of four calves, was experimentally infected with a pure O. ostertagi populatio
