Synthesis and structure-activity relationship studies of 1,2,4-triazole-benzoxazepine hybrids as α-glucosidase inhibitors: molecular docking and biological evaluation.

A novel series of 1,2,4-triazole-benzoxazepine hybrid derivatives was designed, synthesized, and biologically evaluated as α-glucosidase inhibitors using an in vitro enzyme inhibition assay with acarbose as a reference standard. Several compounds demonstrated significant inhibitory activity, with compound 14 emerging as the most potent inhibitor (IC₅₀ = 49.26 µM), surpassing acarbose by approximately 3.6-fold, while compound 28 also showed notable activity (IC₅₀ = 72.14 µM). Structure-activity relationship analysis revealed that halogen substitution, particularly at the ortho position, plays a

Synthesis and structure-activity relationships of dual histamine H2 and gastrin receptor antagonists with modified benzodiazepine skeletons.

Three different types of dual histamine H2 and gastrin receptor antagonists, e.g. those bearing a benzazepine, benzoxazepine, or benzothiazepine skeleton instead of the benzodiazepine one as a gastrin receptor antagonistic moiety were synthesized to reduce high hydrophobicity of parent compounds and evaluated for the dual activities. These skeletal modifications significantly potentiated the binding affinity of dual antagonists with histamine H2 receptor but markedly diminished their binding affinity with the gastrin receptor and the gastrin versus CCK-A receptor selectivity. We evaluated in v

De Novo Design in an M1 Positive Allosteric Modulator Backup Program: The Discovery of VU6069863.

Recently, we disclosed our M1 PAM that had advanced into Phase I clinical studies, VU0467319, as well as a deuterated back-up compound, VU6045422, to address a major metabolite produced in man and improve tox profile. Ultimately, this series failed to overcome its liabilities and was terminated. However, the team was challenged with developing chemically distinct backup candidates without performing a new HTS campaign. De novo design strategies based upon the other two M1 ago-PAMs that had previously entered clinical development, MK-7622 and TAK-071, led to the discovery of a fundamentally new

Dipeptidyl peptidase-1 inhibition with brensocatib reduces the activity of all major neutrophil serine proteases in patients with bronchiectasis: results from the WILLOW trial.

BACKGROUND: Brensocatib is an oral, selective, reversible inhibitor of dipeptidyl peptidase-1 (DPP-1), responsible for activating neutrophil serine proteases (NSPs) including neutrophil elastase (NE), proteinase 3 (PR3), and cathepsin G (CatG). In chronic inflammatory lung diseases such as non-cystic fibrosis bronchiectasis (NCFBE), neutrophils accumulate in the airways resulting in excess active NSPs that cause damaging inflammation and lung destruction. METHODS: The 24-week WILLOW trial (NCT03218917) was a randomized, double-blind, placebo-controlled, parallel-group trial in patients with N

Degradation caused by incompatibility between sodium stearyl fumarate (PRUV) and AZD7986 in the drug product.

During compatibility study of the AZD7986 project, a peak of 3 area% at the tail (RRT 1.03) of the active pharmaceutical ingredient (API) was discovered for all tablets containing sodium stearyl fumarate (PRUV) under humid condition (e.g. 50 °C/75% RH), regardless of choice of disintegrant or filler combination. The degradant was needed to be identified to understand the corresponding reaction mechanism and help the final formulation design. Structure elucidation was therefore done by analysis using high resolution mass spectrometry. The degradant was found to be a Michael addition product of

Analysis of Drug-Induced Gastrointestinal Obstruction and Perforation Using the Japanese Adverse Drug Event Report Database.

Drug-induced gastrointestinal obstruction (DIGO) and gastrointestinal perforation (DIGP) may be the result of gastrointestinal hypomotility and severe constipation, which may lead to potentially fatal complications of bowel ischemia, sepsis and perforation. We evaluated the onset profile of DIGs (DIGO and DIGP) associated with prescription drugs by analyzing data in the Japanese Adverse Drug Event Report (JADER) database. We selected 161 DIG-related drugs and categorized them into 19 classes based on the Anatomical Therapeutic Chemical (ATC) Classification System. Finally, we focused on 58 dru

Structure-based optimization of GSK2982772-derived RIPK1 inhibitors reveals the dispensability of the methylene linker and identifies a potent anti-necroptosis candidate for further optimization.

Necroptosis is closely associated with a variety of inflammatory diseases, with receptor-interacting serine/threonine-protein kinase 1 (RIPK1) serving as a key regulator of this signaling pathway. RIPK1 inhibitors based on the 5-benzyl-N-(5-methyl-4-oxo-2,3,4,5-tetrahydrobenzo[b][1,4]oxazepin-3-yl)-1H-1,2,4-triazole-3-carboxamide scaffold have demonstrated potent anti-inflammatory activity, and two representatives, GSK2982772 and GSK3145095, advanced into Phase II clinical trials. However, their clinical development encountered significant challenges, highlighting the need for structurally nov

Eleclazine Suppresses Ventricular Fibrillation in Failing Rabbit Hearts with Ischemia-Reperfusion Injury Undergoing Therapeutic Hypothermia.

INTRODUCTION: Eleclazine is a highly selective late sodium current inhibitor, possibly effective in reducing ventricular fibrillation (VF) in heart failure (HF) with ischemia-reperfusion (IR) injury. The electrophysiological effects of eleclazine at therapeutic hypothermia (TH) are unknown. We investigated the effects of eleclazine in suppressing VF in failing rabbit hearts with IR injury undergoing TH. METHOD: HF was induced by right ventricular pacing. An IR model was created using coronary artery ligation for 60 min, followed by reperfusion for 30 min. Hearts were excised and Langendorff-p

Repositioning of Tak-475 In Mevalonate Kinase Disease: Translating Theory Into Practice.

BACKGROUND: Mevalonate Kinase Deficiency (MKD, OMIM #610377) is a rare autosomal recessive metabolic and inflammatory disease. In MKD, defective function of the enzyme mevalonate kinase, due to a mutation in the MVK gene, leads to the shortage of mevalonate- derived intermediates, which results in unbalanced prenylation of proteins and altered metabolism of sterols. These defects lead to a complex multisystem inflammatory and metabolic syndrome. OBJECTIVE: Although biologic therapies aimed at blocking the inflammatory cytokine interleukin- 1 can significantly reduce inflammation, they cannot

Electrooxidation potential as a tool in the early screening for new safer clozapine-like analogues.

The chemical modification of clozapine (1) has permitted the finding of new analogues, e.g., olanzapine (2), quetiapine (3), 5-(4-methylpiperazin-1-yl)-8-chloropyrido[2,3-b][1,5]benzoxazepine fumarate (9), with a clinical or psychopharmacological profile similar to that of clozapine. However, when developing new derivatives, the designers are discouraged by the development of clozapine-induced agranulocytosis. Different researchers have raised the role played by the oxidizability of the molecule in such a deleterious effect. In the present paper, we examined the oxidation profile (direct scave

Targeting prostaglandin E2 EP1 receptors prevents seizure-associated P-glycoprotein up-regulation.

Up-regulation of the blood-brain barrier efflux transporter P-glycoprotein in central nervous system disorders results in restricted brain access and limited efficacy of therapeutic drugs. In epilepsies, seizure activity strongly triggers expression of P-glycoprotein. Here, we identified the prostaglandin E2 receptor, EP1, as a key factor in the signaling pathway that mediates seizure-induced up-regulation of P-glycoprotein at the blood-brain barrier. In the rat pilocarpine model, status epilepticus significantly increased P-glycoprotein expression by 92 to 197% in the hippocampal hilus and gr

Pharmacologic Features, Clinical Applications, and Drug Safety Evaluation of Inavolisib in the Treatment of Metastatic Breast Cancer.

The clinical benefit of available treatments in metastatic hormone receptor (HR)+/HER2-breast cancer (BC) remains limited due to the onset of resistance mechanisms, hence the need for increasingly personalized therapies targeting specific molecular alterations. In these cancers, the estrogen receptor, CDK4/6, and the phosphoinositide 3-kinase pathway are key drivers of tumor growth and survival. In preclinical studies, targeting these pathways together has been shown to enhance treatment response and delay resistance; in clinical practice, however, implementation of this combination has been l
