Influenza A viruses can infect a wide range of hosts, creating opportunities for zoonotic transmission, i.e., transmission from animals to humans, and placing the human population at constant risk of potential pandemics. In the last hundred years, four influenza A virus pandemics have had a devastating effect, especially the 1918 influenza pandemic that took the lives of at least 40 million people. There is a constant risk that currently circulating avian influenza A viruses (e.g., H5N1, H7N9) will cause a new pandemic. Vaccines are the cornerstone in preparing for and combating potential pand

The 1918 influenza pandemic spread rapidly around the globe, leading to high mortality and social disruption. The countermeasures available to mitigate the pandemic were limited and relied on nonpharmaceutical interventions. Over the past 100 years, improvements in medical care, influenza vaccines, antiviral medications, community mitigation efforts, diagnosis, and communications have improved pandemic response. A number of gaps remain, including vaccines that are more rapidly manufactured, antiviral drugs that are more effective and available, and better respiratory protective devices.

INTRODUCTION: Nipah virus (NiV) is a highly pathogenic zoonotic virus associated with a high case fatality rate and human-to-human transmission, and it is listed as a priority pathogen by the World Health Organization (WHO) due to the lack of approved therapeutics or vaccines. NiV is classified as a biosafety level 4 (BSL-4) pathogen, which severely limits experimental studies in the absence of specialized containment facilities. Therefore, the establishment of pseudovirus-based surrogate models is essential for NiV vaccine research.
METHODS: In this study, single-antigen mRNA-lipid nanoparti

Influenza A viruses pose a significant threat to global health, impacting both humans and animals. Zoonotic transmission, particularly from swine and avian species, is the primary source of human influenza outbreaks. Notably, avian influenza viruses of the H5N1, H7N9, and H9N2 subtypes are of pandemic concern through their global spread and sporadic human infections. Preventing and controlling these viruses is critical due to their high threat level. Vaccination remains the most effective strategy for influenza prevention and control in humans, despite varying vaccine efficacy across strains.

When we talk about 20-20 it goes without saying that we are talking about Indian Premier League matches. But when we talk about 2020, we get goosebumps even remembering the memories of the coronavirus disease 2019 (COVID-19) pandemic! It was just about that time when the National Medical Commission (NMC) rolled out the Competency-based Medical Education (CBME) curriculum for the first Bachelor of Medicine, Bachelor of Surgery (MBBS) batch admitted in the 2019-2020 academic year. Who knew that the timetable so well planned would go for a toss due to the global pandemic? It was indeed a learning

The Paramyxoviridae family includes established human pathogens such as measles virus, mumps virus, and the human parainfluenza viruses; highly lethal zoonotic pathogens such as Nipah virus; and a number of recently identified agents, such as Sosuga virus, which remain poorly understood. The high human-to-human transmission rate of paramyxoviruses such as measles virus, high case fatality rate associated with other family members such as Nipah virus, and the existence of poorly characterized zoonotic pathogens raise concern that known and unknown paramyxoviruses have significant pandemic poten

The success of nucleic acid-based vaccines during the COVID-19 pandemic positioned these technologies at the forefront of global preparedness. A retrospective analysis, however, demonstrates that while vaccines play a central role, they do not provide universal protection. Therapeutic interventions, such as monoclonal antibodies (mAbs), are also critical for an effective pandemic response, as they ensure both prophylaxis and treatment of vulnerable populations, prevent overload of healthcare systems, and safeguard the continuity of critical infrastructures. Despite the proven efficacy of antiv

Children and adults experienced the COVID-19 pandemic differently. While the impact on children was significant, it was different from what had originally been expected. Here, we outline key developments and learnings from the COVID-19 pandemic's effects on children and draw lessons for pandemic preparedness.

Smallpox is an acute, febrile, contagious disease caused by the Variola virus, which is a member of the Poxviridae family. Until the 1970s, smallpox had been a pandemic disease for more than 3000 years, endemic in tropical and developing areas and periodically epidemic worldwide. The World Health Organization declared smallpox to be completely eradicated in 1980 as the result of global vaccination efforts. At that time, all routine vaccination programs were terminated, given the success of thismonumental eradication. Although smallpox remains fully eradicated, uncertainty exists regarding the

The COVID-19 pandemic caused global disruption that reverberated across Africa and revealed deep systemic vulnerabilities. Nonetheless, the scientific response to SARS-CoV-2 catalyzed unprecedented global scientific collaboration, established new collaborative networks, and accelerated local capacity building across Africa. This legacy extends beyond COVID-19, informing responses to subsequent mpox and Ebola outbreaks, and establishes a framework to respond to future infectious disease threats in Africa.

This study was designed to assess factors associated with a high level of knowledge about influenza among displaced persons and labor migrants in Thailand. We conducted a cross-sectional study of 797 documented and undocumented migrants thought to be vulnerable to influenza during the early stages of the 2009 H1N1 pandemic. Data were collected on socio-demographic factors, migration status, health information sources, barriers to accessing public healthcare services and influenza-related knowledge using a 201-item interviewer-assisted questionnaire. Among the different types of influenza, part

A major challenge during viral pandemics is the ability to develop therapeutics whose efficacy can withstand viral genetic evolution. During the COVID-19 pandemic, five SARS-CoV-2 monoclonal antibody (mAb) therapeutics were rendered ineffective within a period of 2 years, leading to the U.S. FDA revoking their emergency use authorization. Here, we describe ReconnAb-multimers, a new therapeutic design that broadly and potently neutralize all tested betacoronaviruses that use host ACE2 as their receptor to enter cells. These ReconnAb-multimers have potent neutralization efficacy via avidity, enh

The SARS-CoV-2 pandemic that causes COVID-19 respiratory syndrome has caused global public health and economic crises, necessitating rapid development of vaccines and therapeutic countermeasures. The world-wide response to the COVID-19 pandemic has been unprecedented with government, academic, and private partnerships working together to rapidly develop vaccine and antibody countermeasures. Many of the technologies being used are derived from prior government-academic partnerships for response to other emerging infections.

Vaccination against influenza is considered to be one of the key interventions in case of a pandemic. Unfortunately, shortages in vaccine supplies will occur because of the substantial increase in vaccine demands worldwide and the limited available supply resources. The recommended use of monovalent--instead of current trivalent--vaccines containing 15 micro g hemagglutinin (HA) per dose can theoretically triple vaccine volumes but is unlikely to meet the demand. Furthermore, previous experiences demonstrated that one dose of 15 micro g HA will not be sufficient to elicit protective antibody l

Throughout history, pandemics and their aftereffects have spurred society to make substantial improvements in healthcare. After the Black Death in 14th century Europe, changes were made to elevate standards of care and nutrition that resulted in improved life expectancy.1 The 1918 influenza pandemic spurred a movement that emphasized public health surveillance and detection of future outbreaks and eventually led to the creation of the World Health Organization Global Influenza Surveillance Network.2 In the present, the COVID-19 pandemic exposed many of the pre-existing problems within the US h
