Human papillomavirus vaccination in males.

Human papillomavirus (HPV) is one of the most common sexually transmitted diseases (STDs) and is frequently presented clinically as anogenital warts in both males and females. There is a high rate of transmission of HPV in female partners of men with pre-existing penile warts, and HPV infection in men has been shown to contribute to HPV infection and subsequent cervical disease in women. HPV is associated with significant morbidity and represents a substantial healthcare burden. Data indicate that males experienced a longer duration of genital warts and incurred greater treatment costs than wo

A quadrivalent vaccine for human papillomavirus.

Cervical cancer is responsible for at least 13,500 deaths a year in the European Union and more than 30 times that worldwide. DNA from a group of human papillomaviruses (HPVs) is found in virtually all cervical carcinoma cells, and these so-called high-risk HPV types are acknowledged to be a necessary cause of this disease. Vaccines that target these HPV types have the potential to dramatically reduce the incidence of cervical carcinoma. Low-risk HPV types cause genital warts, a common sexually transmitted disease, and vaccines that target these HPV types and the high-risk HPV types, are there

Human papillomavirus vaccine development.

The greatest successes in combating important viral infections have been achieved using vaccines. A vaccine to prevent genital tract human papillomavirus (HPV) infections, especially those types associated with genital tract malignancy, could significantly reduce morbidity and mortality from cervical and other genital tract cancers. However, several barriers currently stand in the way of HPV vaccine development. The immunological response to natural HPV infection is incompletely understood and there is uncertainty about which viral antigen(s) should be included in a candidate vaccine. It is cl

Toward a human papillomavirus vaccine.

Much progress has been made over the past 10 years with regard to development of a vaccine active against HPV. Successful protection has been achieved in animal models using species-specific papillomaviruses; however, HPV diversity may delay the development of successful HPV vaccines, especially those designed as therapeutic vaccines for the treatment of HPV-induced carcinoma and high-grade dysplasia of the cervix. Difficulties arise with regard to assessment of efficacy of potential vaccines. The impetus for long-term studies on vaccine efficacy will initially stem from in vitro evidence of r

[Human papillomavirus prophylactic vaccine].

Human papillomavirus causes viral-dependent cancers, including cervical, anal, vulvar, penile, vaginal, and oropharyngeal, and condyloma acuminata. In the last decade, HPV prophylactic vaccine has been developed and spread worldwide after many large-scale clinical studies. These studies demonstrate significant clinical efficacy for prevention of HPV16/18/6/11-related diseases. In particular, prevention of cervical cancer should be the most important role in the world. In Japan, incidence of cervical cancer does not increase, but the peak of age of the patients at 2005 is 25-45 years old and be

Adverse event monitoring of the human papillomavirus vaccines in Scotland.

BACKGROUND: Human papillomavirus (HPV) vaccines are currently utilised globally in national immunisation programmes. While evidence from clinical trials and epidemiological studies suggest that the HPV vaccines are both effective and safe, concerns about the safety of the vaccine and scientifically unproven associations with severe adverse events following immunisation have led to dramatic decreases in vaccine uptake in Japan and acceptance issues in other countries. AIM: In Scotland, we utilised hospital admissions data to assess the impact of the HPV immunisation programme on the incidence

A review of clinical trials of human papillomavirus prophylactic vaccines.

End of study analyses of the phase III trials of prophylactic human papillomavirus (HPV) virus-like particle (VLP) vaccines in young women are now largely completed. Two distinct vaccines were evaluated, Gardasil(®) (Merck & Co., Whitehouse Station, NJ USA) a quadrivalent vaccine containing VLPs of types 6, 11, 16 and 18 and Cervarix(®) (GlaxoSmithKline Biologicals, Rixensart, Belgium), a bivalent vaccine containing VLPs of types 16 and 18. Both vaccines exhibited excellent safety and immunogenicity profiles. The vaccines also demonstrated remarkably high and similar efficacy against the vacci

Age-appropriate use of human papillomavirus vaccines in the U.S.

Cervical infections by approximately 15 cancer-associated (carcinogenic) human papillomavirus (HPV) genotypes cause virtually all cervical cancer and its immediate precursor lesions worldwide. Prophylactic vaccines against human papillomavirus (HPV) types HPV16 and HP18, which cause 70% of cervical cancer worldwide, hold great promise for reducing the burden of cervical cancer worldwide. However, current HPV vaccines prevent future infections and related cervical abnormalities and do not treat pre-existing HPV infections. In the U.S., HPV vaccine introduction should be considered in the contex

Impact of prophylactic human papillomavirus vaccines on dermatology and venereology.

Prophylactic HPV L1 VLP quadrivalent and bivalent vaccines have a great importance for patients seen by dermatologists and venereologists. Both vaccines protect against HPV16- and HPV 18-associated anogenital cancers, as well as cancers of the mouth, the upper respiratory tract and skin, especially of the fingers and periungual region. The quadrivalent HPV6, 11, 16, 18 vaccine also prevents anogenital warts (condylomata acuminata) which are the most common benign tumors of this body region. HPV-vaccination (Gardasil) has been approved in Germany and other European Countries since October 2006

Progress in prophylactic and therapeutic vaccines for human papillomavirus infection.

Virus-like particle (VLP) subunit vaccines composed of the major capsid protein L1 of the genital human papillomaviruses (HPVs) are now in Phase III clinical trials. The vaccines are immunogenic and safe and early results indicate efficacy. VLPs induce strong cell-mediated as well as humoral immune responses and chimeric VLPs including an HPV early protein may have therapeutic potential. Polynucleotide and recombinant viral vaccines encoding nonstructural viral proteins show therapeutic and prophylactic efficacy in animal models and are candidate immunotherapies for established low-grade benig

[The first vaccine against cancer: the human papillomavirus vaccine].

The last 20 years is one of the most remarkable periods in the fight against cancer, with the realization that some human papillomaviruses are causally related to cancer and with the development of the vaccine against human papillomavirus infections. This is a historical event in medicine and the prophylactic human papillomavirus vaccines have provided powerful tools for primary prevention of cervical cancer and other human papillomavirus-associated diseases. This is very important as human papillomavirus infection is probably the most common sexually transmitted infection worldwide, and over

Human papillomavirus vaccine trials and tribulations: Vaccine efficacy.

As of December 2014, there were 3 approved vaccines for human papillomavirus (HPV): bivalent Cervarix (GlaxoSmithKline, New York, NY), quadrivalent Gardasil (Merck and Co, Kenilworth, NJ), and 9-valent Gardasil-9 (Merck and Co). The average cost per dose is $120, with a recommended 3-dose course. The quadrivalent vaccine is the most widely administered worldwide. As with the bivalent and 9-valent vaccines, the vaccine is considered safe, although concerns have been raised. In addition to immunization against the targeted HPV types, there is evidence that there is cross protection against other

The receipt of the human papillomavirus vaccine's influence on future human papillomavirus vaccine recommendations by medical and dental students.

BACKGROUND: With a growing vaccine-hesitant population and with the increase in oral cancers, particularly in a younger population, there is a heightened need for dental and medical providers to advocate for vaccination, including the human papillomavirus (HPV) vaccine. This study aimed to assess HPV vaccine knowledge, personal immunization status, and attitudes and opinions of dental students (DSs) and medical students (MSs). METHODS: Students from 1 medical and 4 dental schools in the United States completed an anonymous online survey assessing knowledge and attitudes and opinions about the

Deciphering Knowledge and Opinions of Human Papillomavirus and Human Papillomavirus Vaccination for Facilitation of Point-of-Care Vaccination in Adults.

IMPORTANCE: Human papillomavirus (HPV) vaccination rates remain significantly below rates for other common childhood vaccines, which has implications for future rates of HPV-associated oropharyngeal squamous cell carcinoma (OPSCC). OBJECTIVE: To assess whether individuals who were aware of the association between HPV and OPSCC would be more likely to have been previously vaccinated. DESIGN, SETTING, AND PARTICIPANTS: This survey study included patients aged 18 to 45 years who sought routine outpatient care at the otolaryngology clinic at Boston Medical Center from September 1, 2020, to May 1

[Research progress regarding the clinical evaluation on recombinant human papillomavirus vaccines].

Human papillomavirus (HPV) is the main cause for cervical cancer, anogenital cancers and genital warts. Three HPV vaccines have been licensed abroad. Data from clinical trials showed high efficacy of the HPV vaccines in young women with 90%-100% vaccine-related HPV diseases prevented. Though efficacy of the vaccine appears lower in older women, this population can still benefit from vaccination. Immunobriging trials show that the two-dose schedule in 9-14 years old girls elicits non-inferior immune response than the three-dose one in young adults. In addition, HPV vaccines can reduce the recur

Translational mini-review series on vaccines: Monitoring of human papillomavirus vaccination.

Persistent infection with oncogenic human papillomavirus (HPV) is a necessary cause of cervical cancer. Moreover, HPV type 16 (and to a lesser degree HPV type 18) is linked with more rare cancers, namely cancer of the vulva, vagina, penis, anus, oropharynx and larynx. Effective prophylactic vaccines have been developed. In this review, we briefly address immunological aspects of HPV infection and the results of HPV vaccination trials. Internationally standardized monitoring and evaluation of prophylactic HPV vaccination programmes will be essential for arriving at the most (cost-)effective str

Exceeding expectations: factors contributing to the potency of human papillomavirus vaccines.

The commercial prophylactic vaccines for human papillomavirus (HPV) are subunit vaccines composed of spontaneously assembled virus-like particles formed from 360 copies of the L1 major virion protein. The vaccines have exceeded expectations in immunogenicity, clinical trials efficacy, and effectiveness in national immunisation programmes. Booster doses are not required to generate these outcomes. In this Review, we discuss the immunological and virological considerations that can help with explaining the high potency of HPV vaccines. These considerations provide biological plausibility for HPV

Prophylactic immunization with human papillomavirus vaccines induces oral immunity in mice.

OBJECTIVE: Although it has been shown that prophylactic vaccination can induce genital immunity, there is inadequate information on human papillomavirus (HPV) vaccine-induced oral immunity, which is of particular interest due to HPV-associated oropharyngeal malignancies and recurrent respiratory papillomatosis. Therefore, we assessed the efficacy of various HPV vaccines against oral HPV pseudovirus (PsV) infection in mice. STUDY DESIGN: Preclinical scientific investigation. METHODS: C57BL/6 mice were vaccinated three times at 2-week intervals with either Gardasil (Merck, Kenilworth, NJ) (50

Vaccinating HIV patients: focus on human papillomavirus and herpes zoster vaccines.

Vaccination has been one of our most powerful tools to decrease morbidity and mortality from infectious diseases in the last century. It is critical to understand the evolving safety and efficacy data for vaccines in HIV-infected individuals as the number of people living with HIV in the United States and globally continues to increase. The quadrivalent human papillomavirus vaccine and the herpes zoster vaccine are newly licensed in the general population, and several studies have recently been published on the safety and efficacy of these vaccines in HIV populations. This manuscript reviews r
