[Progress in Newcastle disease virus against tumor].

Newcastle disease virus is paramyxoviridae, Avian mumps virus genus type I, and infects more than 250 species of birds, causing huge losses on poultry farming worldwide. Numerous experiments have demonstrated that Newcastle disease virus has oncolytic activity on tumor cells and can selectively replicate in cancer cells. Thus, Newcastle disease virus is a potential therapeutic agent for cancer treatment. Some human clinical trials achieved good results. In this review, we summarized research progress of the relationship between the structural protein of Newcastle disease virus and virulence, a

The origins and potential cross-species transmission of paramyxoviruses and other RNA viruses between native snakes and invasive Burmese pythons in the Florida Everglades.

UNLABELLED: During November-December 2020, a disease outbreak involving eastern mudsnakes (Farancia abacura abacura) was documented in the Greater Everglades Ecosystem, Florida, USA. Moribund snakes were observed writhing on riverbanks with concomitant frothy oral discharge. Pathological examination revealed necrotizing enteritis and pulmonary edema, and a reptilian ferlavirus phylogenetically related to anaconda paramyxoviruses was isolated. While ferlaviruses are well-described pathogens of snakes in zoological and private collections worldwide, this represented the first outbreak of ferlavi

CROSS-SPECIES TRANSMISSION OF A GENOGROUP C FERLAVIRUS IN A ZOOLOGICAL COLLECTION IN THE UNITED STATES.

Ferlaviruses have been associated with significant mortality events in squamates. Over a 5-month period in 2012, a mortality event at the Phoenix Zoo involved seven rattlesnakes (Crotalus spp.), a Sonoran gopher snake (Pituophis catenifer affinis), and a Gila monster (Heloderma suspectum). All individuals had been managed within the same group of enclosures at the zoo, and many of the affected individuals had previously been housed together. Clinical signs during the outbreak included sudden death, agonal behavior, anorexia, regurgitation, and loss of body condition. Histologic findings were s

[Molecular basis for negative-strand RNA virus entry and neutralization by antibodies].

Mononegaviruses are non-segmented negative-strand RNA viruses, and include measles, mumps, Marburg, and Ebola viruses. Measles virus and mumps virus, members of the family Paramyxoviridae, are immunotropic and neurotropic, respectively. Marburg virus and Ebola virus, members of the family Filoviridae, cause highly lethal hemorrhagic fever. In this paper, I summarize the recent structural and functional studies on the viral glycoproteins (GPs) of these viruses, which have shed light on virus entry and the humoral response. The structural and functional analyses of the interaction between viral

Lower Respiratory Tract Infection with Human Metapneumovirus: Chest CT Imaging Features and Comparison with Other Viruses.

PURPOSE: Human metapneumovirus has been increasingly identified as a cause of lower respiratory tract infection in adults worldwide. The CT imaging features of human metapneumovirus in adults have not been characterized. The purpose of this paper is to determine the imaging features of human metapneumovirus and to compare them with features of other viruses. METHODS: Two clinicians retrospectively reviewed the medical records of 104 adults with lower respiratory tract infection due to human metapneumovirus at four hospitals in the northeast USA over 32 months. CT images were evaluated by two

[Corneal manifestations of measles in the unvaccinated adult: two typical cases during an epidemic].

Measles is a disease due to morbillivirus, which belongs to the paramyxoviridae subfamily. It affects mostly young patients, and evolves through four phases: incubation, invasion, eruption and desquamation. Ophthalmic manifestations may occur during the invasive and eruptive phases. Conjunctivitis is the most common ophthalmologic manifestation and is often asymptomatic. Measles keratitis is the most concerning manifestation, with possible corneal ulcer, bacterial superinfection and corneal perforation. We report two cases of acute keratitis occurring during the eruptive phase of measles in tw

Detection and molecular epidemiology of ferlaviruses in farmed snakes with respiratory disease in Guangxi Province, China.

We screened 104 snakes with respiratory disease, collected from 52 snake farms in Guangxi Province, China, for pathogens. Ferlaviruses were detected in 70 of 104 lung samples by reverse-transcription PCR; 34 of 52 of the snake farms were positive for ferlaviruses. No reovirus, adenovirus, sunshine virus, or nidovirus was detected in any of the snakes. We obtained 96 bacterial isolates from snake organs, of which the most commonly isolated species were Salmonella (18) and Proteus (16). Sequence analysis, based on 27 partial RNA-dependent RNA polymerase gene (L) sequences, revealed that ferlavir

Molecular prevalence, genomic characterization, and zoonotic potential of novel paramyxovirus and hepacivirus in Alexandromys fortis, Republic of Korea.

Rodents are substantial reservoirs of zoonotic viruses with regular human exposure restricted to a limited number of species. Numerous rodent species have been shown to harbor emerging viruses, including paramyxoviruses and hepaciviruses. Reed voles (Alexandromys fortis), a rodent species that inhabits grasslands and riparian environments throughout East Asia, remain poorly characterized in terms of their viral diversity. In this study, 258 A. fortis specimens collected from rural areas in Gyeonggi Province, Republic of Korea (ROK) were screened for paramyxoviruses and subjected to metagenomic

Principles and practical applications of structure-based vaccine design.

Viral proteins fold into a variety of structures as they perform their functions. Structure-based vaccine design aims to exploit knowledge of an antigen's architecture to stabilize it in a vulnerable conformation. We summarize the general principles of structure-based vaccine design, with a focus on the major types of sequence modifications: proline, disulfide, cavity-filling, electrostatic and hydrogen-bond substitution, as well as domain deletion. We then review recent applications of these principles to vaccine-design efforts across five viral families: Coronaviridae, Orthomyxoviridae, Para

More than 1 240 species of bats are known; many of them are insectivorous, others are frugivorous and some american species are haematophagous. More than 100 diferent viruses are associated with these mammals, particularly Rhabdoviridae (Lyssavirus like rabies virus), Paramyxoviridae (like Nipah and Hendra viruses), Filoviridae (Ebola and Marburg viruses) and Coronavirida (viruses causing SARS and MERS). These infections are usually asymptomatic in bats but the mechanism of this tolerance is not yet understood. For those viruses, bats are efficient reservoirs and disseminators. So, they repres

Reemergence of mumps.

The mumps virus is a single-stranded, non-segmented, negative-sense RNA virus belonging to the Paramyxoviridae family. Mumps is characterized by bilateral or unilateral swelling of the parotid gland. Aseptic meningitis is a common complication, and orchitis is also common in adolescents and adult men. Diagnosis is based on clinical findings, but because of high vaccination coverage, clinical findings alone are not sufficient for diagnosis, and laboratory confirmation is needed. Mumps is preventable by vaccination, but despite high vaccination coverage, epidemics occur in several countries, inc

Immunization of macaques with formalin-inactivated human metapneumovirus induces hypersensitivity to hMPV infection.

Human metapneumovirus (hMPV), a member of the family Paramyxoviridae, is an important cause of acute respiratory tract disease. In the 1960s, vaccination with formalin-inactivated paramyxovirus preparations--respiratory syncytial virus (RSV) and measles virus (MV)--resulted in predisposition for enhanced disease upon natural infection. We have produced a formalin-inactivated hMPV preparation (FI-hMPV), which was used to immunize young cynomolgus macaques. Six days after challenge FI-hMPV-primed monkeys had developed eosinophilic bronchitis and bronchiolitis, indicative of a hypersensitivity re

The Fecal Virome of Children with Hand, Foot, and Mouth Disease that Tested PCR Negative for Pathogenic Enteroviruses.

Hand, foot, and mouth disease (HFMD) affects infant and young children. A viral metagenomic approach was used to identify the eukaryotic viruses in fecal samples from 29 Thai children with clinical diagnosis of HFMD collected during the 2012 outbreak. These children had previously tested negative by PCR for enterovirus 71 and coxsackievirus A16 and A6. Deep sequencing revealed nine virus families: Picornaviridae, Astroviridae, Parvoviridae, Caliciviridae, Paramyxoviridae, Adenoviridae, Reoviridae, Picobirnaviridae, and Polyomaviridae. The highest number of viral sequences belonged to human rhi

The new China virus - Human Metapneumovirus, its infectivity and rapid spreadis it a threat?

Human Metapneumovirus (HMPV) causes a highly infectious respiratory disease for all ages and is part of the Pneumoviridae family, but causes diseases for respiratory, milder to most severe; from pneumonia, to bronchiolitis, up to COPD. The virus is pleomorphic in shape and size, varying between 150 nm to 600 nm but very contagious and easily spread by droplets via coughing or sneezing. Children are more at risk giving it a name of kid's virus. People with a weak immune system as those with heart or lung diseases are also easy victims. The symptoms of HMPV include runny nose, nasal congestion,

[Current progress in reverse genetics technology of parainfluenza virus type 5 and its applications].

Parainfluenza virus type 5 (PIV5) is a negative-sense single-stranded RNA virus belonging to the genus Rubulavirus of the family Paramyxoviridae. The virus has a broad host range and can infect various animals, including ticks, horses, dogs, pigs, hamsters, guinea pigs, cattle, and giant pandas. With extremely low pathogenicity in humans and a favorable safety profile, PIV5 has garnered significant attention as a promising novel vaccine vector platform. This article outlines the pathogenic biological characteristics of PIV5, summarizes the advances in reverse genetics technologies for PIV5 and

[Nipah and Hendra virus infectious diseases].

Nipah and Hendra virus were first identified in mid 1990s in Australia and Malaysia, caus- ing epidemics with high mortality rate in affected animals and humans. Since their first emer- gence, they continued to re-emerge in Australia and South East Asia almost every year. Nipah and Hendra virus were classified in the new genus Henipavirus because of their un- common features amongst Paramyxoviridae. Henipaviruses are zoonotic paramyxoviruses with a broad tropism, and cause severe acute respiratory disease and encephalitis. Their high virulence and wide host range make them to be given Biosecur

The measles virus.

Measles is one of widely spread virus infections that is a major cause of deaths in some tropical areas. The measles virus is a member of the genus of Morbillivirus of the family of Paramyxoviridae. The virions contain six polypeptides, including one glycoprotein; two of them are surface proteins that possess hemagglutinating and hemolytic activities, one of them is polymerase. Replication of the measles virus is similar to that of other Paramyxoviruses. Besides the acute infection for measles virus a persistent infection is characteristic that affects central nervous system and inner organs.

Osteoclasts in human osteopetrosis contain viral-nucleocapsid-like nuclear inclusions.

We report the discovery of nuclear inclusions in the osteoclasts of three unrelated patients with benign osteopetrosis that resemble the osteoclast inclusions characteristic of Paget's disease of bone. These inclusions are morphologically and dimensionally identical to the nucleocapsids of a virus of the Paramyxoviridae family.
