Application of different methods for the diagnosis of experimental paratuberculosis in goats.

The diagnosis of subclinical paratuberculosis is still considered a major problem worldwide. As part of investigating diagnostic strategies for the paratuberculosis infection, sequential results of various diagnostic methods in a progressive experimental infection in goats were evaluated. Twenty-three goat kids were divided into three groups: the infected, contact and control, comprising 10, five and eight goats respectively. Animals of the infected group were orally inoculated on seven occasions with 5 ml of inoculum containing 2 x 10(9)Mycobacterium avium ssp. paratuberculosis per ml. Lympho

First report of paratuberculosis (Johne's disease) in sheep in Morocco.

Paratuberculosis (Johne's disease) was suspected in a flock of approximately 1000 sheep after weight losses and scouring had increased in adult animals despite repeated anthelmintic treatment. A total of 10 ewes showing clinical symptoms were submitted to laboratory examination. Gross pathological and histological examination of tissue samples rendered results compatible with Johne's disease in all 10 ewes. The diagnosis could only be confirmed by cultural isolation and identification of Mycobacterium paratuberculosis in the faeces of two ewes. This is considered the first documented isolation

Intestinal and mesenteric lymphangitis mimicking arteritis in a cow with paratuberculosis.

Paratuberculosis is a chronic infectious disease of ruminants caused by Mycobacterium avium subsp. paratuberculosis (MAP). MAP is transmitted primarily through ingestion of contaminated feces, milk, or water and establishes persistent infection within intestinal macrophages by evading host immune defenses. Typical lesions include granulomatous enteritis, lymphangitis, and lymphadenitis. Lymphangitis mimicking arteritis has not been reported in bovine paratuberculosis, to our knowledge. Here, we describe a case of paratuberculosis in a 2-y-old Angus cow that was found dead. Gross changes consis

Paratuberculosis in small ruminants in the Sudan: prevalence and risk factors.

BACKGROUND: Paratuberculosis (PTB), caused by Mycobacterium avium subsp. paratuberculosis (MAP), is a contagious and chronic enteric disease of ruminants and many non-ruminants leading to emaciation and death of the animal. PTB is poorly investigated in sheep and goats in Sudan, where these animals contribute significantly to food security and poverty alleviation as sources of income. They also play an important role in the national economy through animal exports. The objectives of this study were to investigate the prevalence of PTB and associated risk factors in small ruminants. Blood and fa

The role of Mce proteins in Mycobacterium avium paratuberculosis infection.

Mycobacterium avium subspecies paratuberculosis (MAP) is the causative agent of Johne's Disease, a chronic granulomatous enteritis of ruminants. MAP establishes an infection in the host via the small intestine. This requires the bacterium to adhere to, and be internalised by, cells of the intestinal tract. The effector molecules expressed by MAP for this purpose remain to be fully identified and understood. Mammalian cell entry (mce) proteins have been shown to enable other Mycobacterial species to attach to and invade host epithelial cells. Here, we have expressed Mce1A, Mce1D, Mce3C and Mce4

Pathways and Genes Associated with Immune Dysfunction in Sheep Paratuberculosis.

Multibacillary and paucibacillary paratuberculosis are both caused by Mycobacterium avium subspecies paratuberculosis. Multibacillary lesions are composed largely of infected epithelioid macrophages and paucibacillary lesions contain T cells but few bacteria. Multibacillary disease is similar to human lepromatous leprosy, with variable/high levels of antibody and a dysfunctional immune response. Animals with paucibacillary disease have high cell-mediated immunity and variable levels of antibody. This study aims to characterize the immunological dysfunction using TruSeq analysis of the ileocaec

Current strategies for eradication of paratuberculosis and issues in public health.

Paratuberculosis is a regional chronic enteritis caused by Mycobacterium avium subsp. paratuberculosis (MAP). The first complete description of this disease was made in 1895, but previous reports on clinical cases compatible with this entity can already be found in the literature of early 1800 s. Its obvious similarities with tuberculosis gave it the name of paratuberculosis, although it was clearly recognized it was not the same entity as that caused by mammal tuberculosis mycobacteria. Eradication has been considered the ideal goal for control of paratuberculosis, but the lack of efficient d

Effects of altered dietary iron intake in Mycobacterium paratuberculosis-infected dairy cattle: sequential observations on growth, iron and copper metabolism and development of paratuberculosis.

Twenty calves were orally inoculated with Mycobacterium paratuberculosis at six weeks old. At six months old, 10 of these, plus four uninfected controls were maintained on limited dietary copper and supplemented iron intake for a further 27 months. During this time all these animals, together with a further four untreated controls, were bred before being killed and examined for evidence of paratuberculosis. Despite significant reduction in weight gain, attributable to both iron supplementation and infection, no significant difference was found in the numbers of iron-supplemented and unsuppleme

Sarcoidosis, Mycobacterium paratuberculosis and Noncaseating Granulomas: Who Moved My Cheese.

Clinical and histological similarities between sarcoidosis and tuberculosis have driven repeated investigations looking for a mycobacterial cause of sarcoidosis. Over 50 years ago, "anonymous mycobacteria" were suggested to have a role in the etiology of sarcoidosis. Both tuberculosis and sarcoidosis have a predilection for lung involvement, though each can be found in any area of the body. A key histopathologic feature of both sarcoidosis and tuberculosis is the granuloma-while the tuberculous caseating granuloma has an area of caseous necrosis with a cheesy consistency; the non-caseating gra

Serologic reactivity against Mycobacterium paratuberculosis antigens in patients with sarcoidosis.

Although sarcoidosis has clinical and histopathologic similarities to some forms of tuberculosis and other mycobacterial infections, attempts to establish a mycobacterial etiology have not been successful. Using cytoplasmic antigens derived from a wild strain of Mycobacterium paratuberculosis in an enzyme-linked immunosorbent assay, patients with sarcoidosis were found to have immunoglobulin levels significantly higher than those found in a control population in the IgG, but not in IgA or IgM antibody classes. Results were comparable to those reported from patients with Crohn's disease, where

[Experimental paratuberculosis: pathogenicity of mycobactin-dependent mycobacteria strains to calves].

The pathogenicity for calves of two strains of wood-pigeon mycobacteria isolated in our laboratory, one strain isolated in the United Kingdom, one strain isolated from a Roe-deer in Denmark and three strains of M. paratuberculosis, were compared. Seven groups of four 3 to 4 week-old calves were infected by the intravenous route using 10(6) to 10(9) viable units. They were slaughtered one year after the exposure or earlier if their state required it. The macroscopic and microscopic lesions and cultures from the mesenteric lymph nodes, ileon and caecum were studied. At necropsy, the post-mortem

Molecular pathogenesis of bovine paratuberculosis and human inflammatory bowel diseases.

Paratuberculosis (Ptb), caused by Mycobacterium avium subsp. paratuberculosis (Map), is a chronic enteritis that affects many ruminants and other wild animals worldwide. Ptb is a great concern in animal health and in etiology of human Crohn's disease (CD). In the present study, we detected Map-specific insertion sequence IS900 of DNA in tissue sections surgically removed from lesions of patients with CD (29 samples), ulcerative colitis (UC) (17 samples), and non-inflammatory bowel disease (IBD) (20 samples). We then compared the histopathological findings of 29 CD and 17 UC cases with those of

How accurately can we detect Mycobacterium avium subsp. paratuberculosis infection?

Mycobacteria have thwarted detection by scientists for centuries. Mycobacterium paratuberculosis is one of the most fastidious of the Mycobacteriaceae, and has been implicated in both animal and human diseases. In domestic livestock, M. paratuberculosis has been associated with Johne's disease, which given its increasing incidence, is currently a cause for concern, due to the potential for M. paratuberculosis to enter our food chain. In addition, a tenuous link has been reported between M. paratuberculosis and Crohn's disease, however evidence to support this link is hampered by the lack of ac

Detection of Mycobacterium avium ss. Paratuberculosis in Blau Syndrome Tissues.

Background and Aim of the Work. Blau syndrome is an inherited granulomatous inflammatory disorder with clinical findings of uveitis, arthritis, and dermatitis. Although rare, Blau syndrome shares features with the more common diseases sarcoidosis and Crohn's disease. The clinical findings of Blau syndrome are indistinguishable from juvenile sarcoidosis; the mutations of Blau syndrome are on the same gene of chromosome 16 (CARD15) that confers susceptibility to Crohn's disease. The product of this gene is part of the innate immune system. Mycobacterium avium ss. paratuberculosis (MAP) is the pu

Mycobacterium avium subspecies paratuberculosis diagnosis and geno-typing: genomic insights.

Effective control of paratuberculosis and investigations of potential link to Crohn's disease have been hampered by the lack of effective assays for easy and accurate diagnosis of Mycobacterium avium subspecies paratuberculosis (Map). Map is extremely fastidious and depends on iron chelator (Mycobactin). Map strains from humans and sheep are very difficult to isolate and may require years to emerge. Therefore, small numbers of Map isolates have been maintained in available collections. This situation has limited the study of biodiversity of Map. Though, much is known about environmental and ho

Mycobacterium avium subspecies paratuberculosis (MAP), the causative agent of Johne disease in cattle and other ruminants, is proposed to be at least one of the causes of Crohn disease in humans. MAP and Mycobacterium avium subspecies avium, a closely related opportunistic environmental bacterium, share 95% of their genes and exhibit homologies of more than 99% between these genes. The identification of molecules specific for MAP is essential for understanding its pathogenicity and for development of useful diagnostic tools.
