Enhanced lactic acid production from P2O5-pretreated biomass by domesticated Pediococcus pentosaceus without detoxification.

Expensive cellulase and complex detoxification procedures increase the cost of biomass lactic acid fermentation. Therefore, it is of great significance to develop a robust method to ferment lactic acid using biomass by avoiding cellulase and detoxification. This study demonstrates the advantage of combining mechanocatalytic P2O5 pre-treatment and strain domestication. We show that an enzyme-free mechanocatalytic saccharification process by combining mix-milling of P2O5 with biomass and successive hydrolysis produces a fermentable hydrolysate with much less inhibitory compounds than the hydroly

Bacteriocin activity against various pathogens produced by Pediococcus pentosaceus VJ13 isolated from Idly batter.

Bacteriocins, an antimicrobial peptide, is known to have wide spectrum antimicrobial activity against various pathogens. Because they are easily digested in the intestine, they are considered as safe and are widely used as food preservatives. Hence their purification and characterization have attracted considerable attraction, especially for those having activity against human pathogens. In this study, the bacteriocin produced by Pediococcus pentosaceus VJ13 was precipitated with cold acetone and purified by gel permeation chromatography and hydrophobic interaction chromatography. The bacterio

Effects of a Bacterial Hay Preservative (Pediococcus pentosaceus) on Hay under Experimental Storage Conditions.

The abundant growth of molds and thermophilic actinomycetes in stored hay decreases its quality and can be hazardous for the producer who inhales these contaminants when the moldy hay is fed in closed barns. These microbes are responsible for a respiratory disease called farmer's lung. Products, including bacterial cultures that can be inoculated in hay, are available to prevent hay deterioration by molds and bacteria. The aim of this study was to verify the effectiveness of Pediococcus pentosaceus (a bacterial inoculant) in preventing hay deterioration at different humidity levels in a labora

Membrane-acting bacteriocin purified from a soil isolate Pediococcus pentosaceus LB44 shows broad host-range.

Bacteriocin LB44 was purified from cell-free supernatant (CFS) of Pediococcus pentosaceus LB44 using activity-guided chromatography techniques. It was stable up to 121 °C, pH 2.0-6.0, sensitive to proteinase K, papain and trypsin, and retained complete activity in the presence of organic solvents tested. The molecular weight of bacteriocin was ∼6 kDa and initial ten amino acid residues (GECGMCXECG) suggested a new compound. The loss in viable cell count and K+ ion efflux of target cells of Micrococcus luteus suggested bactericidal activity. The cell membrane of bacteriocin-treated cells was fo

Influence of Lactobacillus plantarum and Pediococcus pentosaceus addition on the nutritional quality of Sourdough-based bread.

This study explored the nutritional benefits of using a mixed culture of Lactobacillus plantarum and Pediococcus pentosaceus in sourdough bread fermentation. Over a 120-h fermentation period, spontaneous microbial activity resulted in peak lactic acid bacteria populations of 516.29 × 106 cfu/g at 72 h in acha-based dough and 1082.78 × 106 cfu/g at 24 h in rice-based dough (P < .001). The fermentation process led to a steady reduction in pH alongside a rise in titratable acidity. Bread produced with the mixed LAB cultures showed a significant decrease in phytic acid content and a notable enhanc

Pediococcus pentosaceus DF29-derived extracellular vesicles ameliorate EPEC-induced intestinal injury via M2 macrophage polarization.

Enteropathogenic Escherichia coli (EPEC) infection remains a major cause of intestinal barrier dysfunction and diarrhea, necessitating the development of novel non-antibiotic therapies. While Pediococcus pentosaceus (P. pentosaceus) exhibits probiotic potential, the specific role of its postbiotic components, particularly extracellular vesicles (EVs), in host-pathogen interactions remains obscure. In this study, we systematically evaluated the protective efficacy and underlying mechanisms of P. pentosaceus DF29 (DF29) and its derived EVs (PpEVs) against EPEC-induced enteritis. Trehalose improves cold tolerance of Pediococcus pentosaceus OL77 and enhances low-temperature oat silage fermentation.

Low temperature remains a major bottleneck in silage fermentation, especially in cold regions and high-altitude areas. This study investigated the regulatory role of compatible solutes in enhancing the cold adaptation of Pediococcus pentosaceus OL77 and their synergistic application in oat silage under suboptimal temperatures (5°C-15°C). RT-qPCR analysis showed that expression of the cold shock protein gene CspP was sharply induced at 5°C, indicating its central role in cold-stress response. Exogenous trehalose and betaine significantly downregulated CspP expression, promoted bacterial growth,

Comprehensive safety assessment and therapeutic potential of Pediococcus acidilactici NMCC-B in attenuating arthritis progression.

Dysbiosis of gut microbiota and loss of gut-barrier integrity contribute to the development and severity of rheumatoid arthritis (RA). The available treatments pose a burden of major adverse effects and new treatment strategies are therefore the need of time. In this study, Pediococcus acidilactici NMCC-B (Probiotic) was evaluated for its safety and efficacy in complete Freund's adjuvant (CFA)-induced mice model of RA. Mice were treated with either Escherichia coli (1 × 109 CFU/ml) or P. acidilactici NMCC-B (1 × 109 CFU/ml, 2 × 109 CFU/ml) to assess acute, sub-acute, and chronic toxicities. In
