Senile dementia of the Alzheimer type: is there a correlation between entorhinal cortex and dentate gyrus lesions?

Senile plaques (SP) are one of the neuropathological hallmarks of senile dementia of the Alzheimer type (SDAT). In 14 patients affected with SDAT (over 74 years of age), thioflavine S, Tau and acetylcholinesterase (AChE) stainings demonstrated an increased density of SP in the outer two thirds of the dentate gyrus molecular layer. However, a wide range of SP density was observed among the cases. The molecular layer of the dentate gyrus is one of the termination site of the perforant pathway that originates in layers II and III of the entorhinal cortex. We have found that the number of AChE-, t

Pathway interactions and synaptic plasticity in the dendritic tuft regions of CA1 pyramidal neurons.

Input comparison is thought to occur in many neuronal circuits, including the hippocampus, where functionally important interactions between the Schaffer collateral and perforant pathways have been hypothesized. We investigated this idea using multisite, whole-cell recordings and Ca2+ imaging and found that properly timed, repetitive stimulation of both pathways results in the generation of large plateau potentials in distal dendrites of CA1 pyramidal neurons. These dendritic plateau potentials produce widespread Ca2+ influx, large after-depolarizations, burst firing output, and long-term pote

Hemodynamic responses in the rat hippocampus are simultaneously controlled by at least two independently acting neurovascular coupling mechanisms.

We combined electrical perforant pathway stimulation with electrophysiological and fMRI recordings in the hippocampus to investigate the effects of neuronal afterdischarges (nAD) on subsequent fMRI BOLD signals in the presence of isoflurane and medetomidine. These two drugs already alter basal hemodynamics in the hippocampus, with isoflurane being mildly vasodilatory and medetomidine being mildly vasoconstrictive. The perforant pathway was stimulated once for 8 seconds with either continuous 20 Hz pulses (continuous stimulation) or 8 bursts of 20 high-frequency pulses (burst stimulation). Burs

Host cell perforation by listeriolysin O (LLO) activates a Ca2+-dependent cPKC/Rac1/Arp2/3 signaling pathway that promotes Listeria monocytogenes internalization independently of membrane resealing.

Host cell invasion is an indispensable step for a successful infection by intracellular pathogens. Recent studies identified pathogen-induced host cell plasma membrane perforation as a novel mechanism used by diverse pathogens (Trypanosoma cruzi, Listeria monocytogenes, and adenovirus) to promote their internalization into target cells. It was concluded that T. cruzi and adenovirus damage the host cell plasma membrane to hijack the endocytic-dependent membrane resealing machinery, thereby invading the host cell. We studied L. monocytogenes and its secreted pore-forming toxin listeriolysin O (L

Potent Anti-seizure Effects of Locked Nucleic Acid Antagomirs Targeting miR-134 in Multiple Mouse and Rat Models of Epilepsy.

Current anti-epileptic drugs (AEDs) act on a limited set of neuronal targets, are ineffective in a third of patients with epilepsy, and do not show disease-modifying properties. MicroRNAs are small noncoding RNAs that regulate levels of proteins by post-transcriptional control of mRNA stability and translation. MicroRNA-134 is involved in controlling neuronal microstructure and brain excitability and previous studies showed that intracerebroventricular injections of locked nucleic acid (LNA), cholesterol-tagged antagomirs targeting microRNA-134 (Ant-134) reduced evoked and spontaneous seizures

Synaptophysin immunoreactivity in Pick's disease: comparison with Alzheimer's disease and dementia with Lewy bodies.

Frontotemporal lobe atrophy is a hallmark of Pick's disease (PiD), however, the underlying pathobiology of the neuronal losses is unknown. Synaptic losses have been described in Alzheimer's disease (AD) and correlate with the severity of dementia, however few studies of synaptic integrity have been done to determine whether synaptic loss also contributes to symptoms in non-AD dementias. To begin to assess synaptic integrity in other types of dementia, we examined the site of termination of the hippocampal perforant pathway, the major source of afferent tracts to the hippocampus. We determined

Morphometric and dendritic analysis of fascia dentata granule cells in human aging and senile dementia.

In this study the cellular morphology in the human fascia dentata of 5 very old demented cases (4 Alzheimer's disease and 1 multi-infarct dementia patients) was compared with 5 (very) old controls cases. The postmortem delay in fixation was for all cases within 3.5 h. In the demented group, a significant reduction in thickness of the molecular layer, density of dendritic spines in the middle third of the molecular layer and total dendritic length (+/- 30%) was found. The number of dendritic segments, indicative of the branching frequency showed no difference. In both the control and the dement

Hippocampal formation: anatomy and the patterns of pathology in Alzheimer's disease.

Anatomical studies of the primate brain have shown that the subicular and CA1 allocortices give rise to hippocampal efferents that course to numerous telencephalic and diencephalic targets including other parts of the cortex. The hippocampal formation is damaged heavily in Alzheimer's disease, and is a focal point for pathology. We examined the anteroposterior extent of the hippocampal formation in 52 cases of Alzheimer's disease, 6 cases of other types of dementia and 10 age-compatible controls, to determine the patterns of pathology. We have observed that only certain subfields of the hippoc

Amyloid deposition and advanced age fails to induce Alzheimer's type progression in a double knock-in mouse model.

It has been challenging to develop transgenic and gene-targeted mouse models that recapitulate all of the neuropathological features of Alzheimer's disease (AD). For example, in the APP/PS-1 double knock-in mutant mouse (DKI), frank neurodegeneration is not observed at middle age and synapse loss is pronounced only within amyloid plaques. Here, we investigated whether continued amyloid deposition and advanced age of 24-27 months lead to loss of neurons and synapses, tau hyperphosphorylation, and other pathological features of AD. We focused on the perforant pathway projection from entorhinal c

Homosynaptic and heterosynaptic changes in driving of dentate gyrus interneurons after brief tetanic stimulation in vivo.

This study addressed changes in interneuron driving in the dentate gyrus (DG) of urethane-anaesthetized rats in response to tetanic stimulation of the perforant path (PP) or the converging dentate commissural pathway (CP). Using an extracellular tungsten electrode, we recorded from putative interneurons in the DG that fired to stimulation of both the PP and the CP. Conditioning trains (400 Hz, 17.5 ms) were delivered to each pathway individually and to the two pathways together. The primary measure of synaptic drive was the latency of interneuron discharge. High-intensity PP tetany, CP tetany,

Effect of intrahippocampal microinjection of VU0155041, a positive allosteric modulator of mGluR4, on long term potentiation in a valproic acid-induced autistic male rat model.

The precise cause of autism spectrum disorder (ASD) is not fully understood. Despite the involvement of glutamatergic dysregulation in autism, the specific contribution of mGlu4 receptors to synaptic plasticity remains unclear. Using the positive allosteric modulator VU0155041, we aimed to restore long-term potentiation (LTP) in the perforant path-dentate gyrus (PP-DG) pathway in VPA-induced autistic rat model. High-frequency stimulation was applied to the PP-DG synapse to induce LTP, while the VU0155041 was administered into the DG. Unexpectedly, VU0155041 failed to alleviate the observed LTP

Long-term synaptic enhancement and short-term potentiation in rat fascia dentata act through different mechanisms.

1. The component processes contributing to post-activation change in synaptic efficacy in the perforant pathway to the fascia dentata were studied in rats under sodium pentobarbitone anaesthesia.2. With low stimulus strength, which activated only a relatively small number of perforant path fibres, repetitive stimulation led to effects which had very similar characteristics to those observed at neuromuscular synapses under similar conditions. Paired shocks resulted in a short ( approximately 100 ms) facilitation superimposed on a depression, possibly due to depletion of available transmitter, w

Early-stage dysfunction of hippocampal theta and gamma oscillations and its modulation of neural network in a transgenic 5xFAD mouse model.

Alzheimer's disease (AD) is pathologically characterized by amyloid-β (Aβ) accumulation, which induces Aβ-dependent neuronal dysfunctions. We focused on the early-stage disease progression and examined the neuronal network functioning in the 5xFAD mice. The simultaneous intracranial recordings were obtained from the hippocampal perforant path (PP) and the dentate gyrus (DG). Concomitant to Aβ accumulation, theta power was strongly attenuated in the PP and DG regions of 5xFAD mice compared to those in nontransgenic littermates. For either theta rhythm or gamma oscillation, the phase synchroniza

C1q and immunoglobulins mediate activity-dependent synapse loss in the adult brain.

Complement component 1q (C1q), the initiator of the classical complement cascade, mediates synaptic elimination in development and disease, yet the triggers for its deposition on synapses remain unclear. Using in vivo chemogenetics, we demonstrate that neuronal hyperactivity induces region-specific, C1q-dependent synapse loss in the adult hippocampus. Suppressing perforant pathway hyperactivity in a mouse model of Alzheimer's disease reduced local amyloid-β amounts and C1q deposition and partially rescued synapse loss. Combining spatial transcriptomics, live cell tracking, and super-resolution

Barbiturate effects on hippocampal excitatory synaptic responses are selective and pathway specific.

Barbiturate actions on excitatory synaptic responses in CA 1 and dentate regions of hippocampal slices were studied to determine whether different effects occur on anatomically distinct synaptic pathways. Pentobarbital facilitated transmission between stratum radiatum inputs and CA 1 neurons at low concentrations (0.02-0.08 mM) and produced postsynaptic depression at higher concentrations. Only depression was observed for stratum oriens inputs to CA 1 and perforant path inputs to dentate granulae neurons. The (+) isomer of pentobarbital was approximately four times more potent than the (-) iso

Coenzyme Q10 and exercise training reinstate middle cerebral artery occlusion-induced behavioral deficits and hippocampal long-term potentiation suppression in aging rats.

RATIONAL: Patients experience post-stroke cognitive impairment during aging. To date, no specific treatment solution has been reported for this disorder. OBJECTIVE: The purpose of this study was to evaluate the effects of exercise training and coenzyme Q10 supplementation on middle cerebral artery occlusion (MCAO) induced behavioral impairment, long-term potentiation inhibition and cerebral infarction size in aging rats. METHODS: Fifty aging male rats underwent MCAO surgery and were randomly distributed in to the following groups: 1-Sham, 2- control, 3- Coenzyme Q10, 4- Exercise training and

CDKL5 Deficiency Augments Inhibitory Input into the Dentate Gyrus That Can Be Reversed by Deep Brain Stimulation.

Cognitive impairment is a core feature of cyclin-dependent kinase-like 5 (CDKL5) deficiency, a neurodevelopmental disorder characterized by early epileptic seizures, intellectual disability, and autistic behaviors. Although loss of CDKL5 affects a number of molecular pathways, very little has been discovered about the physiological effects of these changes on the neural circuitry. We therefore studied synaptic plasticity and local circuit activity in the dentate gyrus of both Cdkl5-/y and Cdkl5+/- mutant mice. We found that CDKL5 haploinsufficiency in both male and female mice impairs hippocam

Prenatal exposure to moderate levels of ethanol can have long-lasting effects on hippocampal synaptic plasticity in adult offspring.

Prenatal ethanol exposure has been associated with long-lasting intellectual impairments in children. Previous studies using animal models of fetal ethanol exposure suggest that these deficits are, at least in part, linked to neurochemical abnormalities in the hippocampal formation. We explored whether prenatal exposure to moderate quantities of ethanol produced functional deficits at the entorhinal cortical perforant path-dentate granule cell connection by examining some electrophysiological properties, including the induction of long-term potentiation (LTP). Rat dams consumed one of three di

Hippocampal network activity changes during early epileptogenesis predict subsequent epilepsy.

OBJECTIVE: Despite decades of research, the circuit mechanisms that underlie focal epileptogenesis remain incompletely understood. In this study, we aimed to characterize the changes in hippocampal network activity induced by an epileptogenic insult. METHODS: In rats, prolonged electrical perforant pathway stimulation leads in a high percentage of animals to the development of temporal lobe epilepsy. However, a subset of animals remained apparently non-epileptic for unknown reasons. We monitored alterations of neuronal activity by chronically recording the local field potential (LFP) from the

CHCHD10S59L/+ mouse model: Behavioral and neuropathological features of frontotemporal dementia.

CHCHD10-related disease causes a spectrum of clinical presentations including mitochondrial myopathy, cardiomyopathy, amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). We generated a knock-in mouse model bearing the p.Ser59Leu (S59L) CHCHD10 variant. Chchd10S59L/+ mice have been shown to phenotypically replicate the disorders observed in patients: myopathy with mtDNA instability, cardiomyopathy and typical ALS features (protein aggregation, neuromuscular junction degeneration and spinal motor neuron loss). Here, we conducted a comprehensive behavioral, electrophysiological
