A biocompatible model for evaluation of the responses of rat periapical tissue to a new zinc oxide-eugenol sealer.

We aimed to establish an experimental animal model to evaluate materials for endodontic therapy. We focused on the biocompatibility of new paste-type zinc oxide-eugenol (ZOE) sealer. The results of this sealer were compared with those of conventional powder/liquid ZOE and eugenol-free sealers. The molars of Wistar rats were extracted and repositioned in the original socket after application of the sealers on the root apices. Mild inflammation occurred in the periapical tissue of the replanted teeth with both ZOE sealers on day 7, whereas the eugenol-free sealer induced severe inflammation. On

Radiographic evaluation of the effect of endotoxin (LPS) plus calcium hydroxide on apical and periapical tissues of dogs.

The aim of this study was the radiographic evaluation of the apical and periapical region of dog teeth submitted to intracanal bacterial endotoxin (lipopolysaccharide, LPS), associated or not with calcium hydroxide. After removal of the pulp, 60 premolars were divided into four groups and were filled with bacterial endotoxin (group 1), bacterial endotoxin plus calcium hydroxide (group 2), saline solution (group 3), or periapical lesions were induced with no treatment (group 4), for a period of 30 days. Similar periapical lesions were observed in groups 1 and 4. The lamina dura was intact in gr

A Follicle-Stimulating Hormone Exacerbates the Progression of Periapical Inflammation Through Modulating the Cytokine Release in Periodontal Tissue.

Menopause is directly related to a systemically low grade of inflammation, indicating that postmenopausal women might be more prone to the development of inflammation. The high levels of circulating follicle-stimulating hormone (FSH) may cause hypogonadal bone loss during postmenopausal osteoporosis independent of estrogen. Previous research revealed that FSH could aggravate alveolar bone loss during experimental periapical lesions in ovariectomized rats; however, the mechanisms for these effects remain unclear. In this study, we showed that FSH enhanced the expression and secretion of the pro

Gene-expression analysis of matrix metalloproteinases 1 and 2 and their tissue inhibitors in chronic periapical inflammatory lesions.

BACKGROUND: Periapical inflammatory lesions have been investigated previously, but understanding of pathogenesis of these lesions (granulomas and radicular cysts) at the molecular level is still questionable. Matrix metalloproteinases (MMPs) are enzymes involved in the development of periapical pathology, specifically inflammation and tissue destruction. To elucidate pathogenesis of periapical granulomas and radicular cysts, we undertook a detailed analysis of gene expression of MMP-1, MMP-2 and their tissue inhibitors, TIMP-1 and TIMP-2. METHODS: A total of 149 samples were analyzed using re

Guided tissue regeneration using bioresorbable membranes: what is the limit in the treatment of combined periapical and marginal lesions?

The maintenance of single teeth may often be of crucial importance for the prognosis of the total dentition. In such cases, as when a single tooth supports a fixed partial denture, a special effort should be made to maintain that tooth. This study reports the treatment of six such terminal cases. The results of a combination of local and systemic antibiotics and the use of guided tissue regeneration with resorbable membranes and grafting material is demonstrated. After defect debridement and root planing, the defects were filled with Biostite (Coletica), and Paroguide (Coletica) membranes were

Endodontic micro-resurgery and guided tissue regeneration of a periapical cyst associated to recurrent root perforation: a case report.

Although the success rates of microsurgery and micro-resurgery are very high, the influence of a recurrent perforation combined with radicular cyst remains unclear. A 21-year-old white female patient had a history of root perforation in a previously treated right maxillary lateral incisor. Analysis using cone-beam computed tomography (CBCT) revealed an extensive and well-defined periapical radiolucency, involving the buccal and palatal bone plate. The perforation was sealed with bioceramic material (Biodentine) in the pre-surgical phase. In the surgical phase, guided tissue regeneration (GTR)

Periapical tissue reactions to calcium hydroxide and MTA after external root resorption as a sequela of delayed tooth replantation.

Clinical experience has shown that most avulsed teeth are replanted after a long extra-alveolar time and dry or inadequate wet storage, causing necrosis of periodontal ligament cells. This condition invariably leads to development of external root resorption, leaving the filling material in contact with the periapical connective tissues. In this study, the periapical tissue reactions to calcium hydroxide (CH) and mineral trioxide aggregate (MTA) were evaluated after occurrence of external root resorption as an expected sequela of delayed tooth replantation. Twenty male Wistar rats (Rattus norv

Effect of calcium hydroxide-based materials on periapical tissue healing and orthodontic root resorption of endodontically treated teeth in dogs.

This study evaluated periapical tissue healing and orthodontic root resorption of endodontically treated teeth sealed with calcium hydroxide in dogs. The sample consisted of three contralateral pairs of maxillary incisors and two contralateral pairs of mandibular incisors in each of two dogs using a split mouth design. After biomechanical preparation of the teeth in the first group (n = 10), a Ca(OH)(2) dressing was placed for 14 days before root canal filling with Ca(OH)(2)-based sealer (Sealapex) and gutta-percha points. In the second group (n = 10), root canals were obturated immediately af

Vital pulp therapy of a dens evaginatus‒affected tooth with an immature apex and inflamed pulpal and periapical tissue: a case report.

Dens evaginatus (DE) is a developmental anomaly presenting as an enamel-covered tubercle on the occlusal surface of a premolar, particularly found in people of Asian descent. This case report describes partial pulpotomy in a mandibular premolar with a fractured evaginatus tubercle and endodontic infection. A 10.5-year-old girl of Asian descent was referred for endodontic evaluation and treatment because of local swelling and pain. Clinical examination suggested the presence of DE in a noncarious mandibular right second premolar with a diagnosis of symptomatic irreversible pulpitis and symptoma

Comparative immunohistochemical identification and relative distribution of immunocompetent cells in sections of frozen or formalin-fixed tissue from human periapical inflammatory lesions.

The presence of immunocompetent cells (B, Th/i and Ts/c cells and macrophages) and the ratios of these cell populations in periapical lesions (radicular granulomas, radicular cysts and apical scars) were demonstrated immunohistochemically using paraffin and cryo-sections. Thirty-four human periapical lesions were examined for the presence of immunocompetent cells by monoclonal antibodies and the biotin-avidin-horseradish peroxidase method. The T/B cell ratio of radicular cysts was significantly higher than that of both radicular granulomas and apical scars, and the average number of Th/i cells

Differential localization of type I and type III procollagen messenger ribonucleic acids in inflamed periodontal and periapical connective tissues by in situ hybridization.

Inflammatory lesions of periodontal and periapical connective tissue were studied by in situ hybridization to detect cells responsible for type I and type III collagen production. Formalin-fixed and paraffin-embedded tissue specimens from patients with oral lesions of various stages of inflammation were hybridized with cDNA probes specific for human pro alpha 1(I) and pro alpha 1(III) collagen mRNAs, and with bacteriophage lambda DNA as a control probe. This technique permitted us to localize fibroblasts active in type I collagen synthesis in the vicinity of inflammatory infiltrates in all the

The interaction between third molars and surrounding periapical tissues in mandibular stress distribution during high-impact trauma: a finite element study.

BACKGROUND: The presence of mandibular third molars has been associated with the risk of mandibular fractures, highlighting the need for comprehensive studies considering the interaction with other mandibular structures. This study investigates how mandibular third molars and neighboring tissues can influence the structural fragility of the mandible using finite element analysis. MATERIAL AND METHODS: A finite element analysis study following the guidelines proposed by RIFEM 1.0 was performed using three previously created mandible models: Model A, without right and left third molars; Model B

Trauma-specific and time-dependent changes of the RANK/RANKL/OPG signaling axis in the periapical bone tissue after experimental traumatic dental injuries in rats.

The RANK (receptor activator of NF-κB)/RANKL (RANK ligand)/OPG (osteoprotegerin) system regulates osteoclast differentiation, activity, and survival, as well as inflammatory/immune responses, suggesting its potential involvement in the healing of traumatic dental injuries (TDIs). The aim was to characterize the molecular expression dynamics of the RANK/RANKLOPG axis in periapical bone tissue following TDIs in a rat model. TDIs were reproduced on the maxillary left first molar of rats (n = 18): tooth avulsion (TA); lateral luxation (LL); complicated crown fracture (CCF). Target protein levels i

[The role of natural medicinal factors in federal and regional recreational zones in the rehabilitation of patients following surgical treatment of pathological periapical tissues and oral cavity phlegmonas].

The necessity of considering the problem with which this paper is concerned arises from the current scientific discussion of the actions of physicians working in municipal healthcare facilities and sanatoria directed toward rehabilitation of patients after surgical treatment of periapical tissues and oral cavity phlegmonas. The study elucidated the mechanism of action of mineral waters (alkaline, sodium hydrocarbonate or boron-containing water with elevated fluorine and chlorine content) extracted from natural deposits of the Belgorod Region and Krasnodar Krai (Territory). The post-endodontic periapical lesion: histologic and etiopathogenic aspects.

Apical periodontitis is produced in the majority of cases by intraradicular infection. Treatment consists in the elimination of the infectious agents by endodontia. Even when carrying out a correct cleansing and filling of canals, it is possible that periapical periodontitis will persist in the form of an asymptomatic radiolucency, giving rise to the post-endodontic periapical lesion. The chronic inflammatory periapical lesion is the most common pathology found in relation to alveolar bone of the jaw. From the histological point of view, it can be classified as chronic periapical periodontitis

[Histopathology and etiopathogenesis of chronic apical periodontitis--periapical granuloma].

Periapical lesions are among the most frequently diagnosed apical odontogenic pathologies in human teeth. The condition is generally described as apical periodontitis. Apical periodontitis is a sequel to endodontic infection and manifests itself as the host defense response to microbial challenge emanating from the root canal system to the periapical tissue. It is viewed as a dynamic encounter between microbial factors and host defenses at the interface between infected radicular pulp and periodontal ligament that results in local inflammation, resorption of hard tissues, destruction of other
