[Cerebrospinal fluid cAMP and cGMP in various peripheral and central nervous system diseases. Preliminary report].

In a group of 51 patients the concentrations of cAMP and cGMP and their ratio were determined in the cerebrospinal fluid in certain neurological diseases. Statistically significant differences were found in the values of the concentrations of the above metabolites in selected groups of neurological diseases. Generally speaking, apart from a group of multiple sclerosis cases the values of cAMP concentration were higher in central nervous system diseases than in peripheral nervous system diseases. The highest values of the cAMP/cGMP ratio were observed in the group of patients with inflammatory

[Involvement of the peripheral nervous system in systemic connective tissue diseases: report on clinical cases].

INTRODUCTION: The peripheral nervous system is usually involved in the majority of systemic connective tissue diseases, particularly in systemic lupus erythematosus, Sjögren's syndrome, vasculitis and systemic sclerosis. The pathogenesis of lesions in the peripheral nervous system associated with the autoimmune process is complex and it appears that two mechanisms, immunological and ischemic, are of greatest importance. Structures of the nervous system may be damaged by several autoantibodies (e.g. antineuronal, anti-nerve growth factor, anti-neurotrophins), by cytotoxic effects ofproinflammat

Peripheral nervous system lesion syndromes and the mechanisms of their formation in connective tissue diseases.

Systemic rheumatological diseases are often accompanied by the development of central and peripheral nervous system pathology. Data providing evidence of the high incidence of peripheral nervous system lesions in systemic lupus erythematosus and systemic scleroderma are presented. These diseases in particular are characterized by polyneuropathies and tunnel syndromes. Our own observations, along with published data, revealed the following major pathogenetic mechanisms of peripheral nervous system lesions in diffuse connective tissue diseases - ischemic, immunological, and metabolic. [Importance of a screening system in the prevention of diseases of the lumbosacral division of the peripheral nervous system].

An automated screening system for prophylactic examinations is offered. The system enables one to reveal early compensated forms of lumbar osteochondrosis, as well as persons with an increased risk of that disease. The system is based on a statistical analysis of the lumbar osteochondrosis risk factors revealed in a particular individual, and involves the use of a computer. The practical use of this system at large industrial plants has made it possible to reveal potential patients to whom individual therapeutic and prophylactic complexes were administered. It is shown that the size of the ris

Crosstalk between peripheral immunity and central nervous system in Alzheimer's disease.

The significance of peripheral immunity in the pathogenesis and progression of Alzheimer's diseases (AD) has been recognized. Brain-infiltrated peripheral immune components transporting across the blood-brain barrier (BBB) may reshape the central immune environment. However, mechanisms of how these components open the BBB for AD occurrence and development and correlations between peripheral and central immunity have not been fully explored. Herein, we formulate a hypothesis whereby peripheral immunity as a critical factor allows AD to progress. Peripheral central immune cell crosstalk is assoc

Peripheral nervous system hyperexcitability in VV2 sporadic Creutzfeldt-Jakob disease.

Creutzfeldt-Jakob disease (CJD) is a fatal, rapidly progressive neurodegenerative disease. Most cases are sporadic (sCJD). The pathogenesis of sCJD is associated with a conformational change in abnormal prion protein causing widespread neuronal degeneration, and clinical manifestations can be quite protean. Peripheral nerve hyperexcitability syndrome (PNHS) is rarely associated with CJD and is more commonly associated with autoimmune/paraneoplastic syndromes associated with antibodies against the voltage-gated potassium channel complex (VGKC-Abs). Reports of PNHS in CJD are rare. We report 2 p

Microglia in the crosstalk between peripheral and central nervous systems in Parkinson's disease.

Parkinson's disease (PD) is increasingly recognized as a multisystem disorder involving pathological α-synuclein (α-syn) accumulation and widespread neuroimmune dysregulation. Microglia, the resident immune cells in the central nervous system (CNS), are pivotal mediators of the bidirectional communication between the CNS and peripheral systems. In addition to sensing neuronal injury and α-syn pathology, microglia dynamically respond to peripheral immune signals, including circulating cytokines, immune cell infiltration, and microbial metabolites, through pattern recognition receptors such as T

Alpha-Synuclein Lesions in the Peripheral Nervous System of the Larynx in Parkinson's Disease.

Swallowing, speech and voice (SSV) disorders are very common in Parkinson's disease (PD). The aim of the present studies was to test our hypothesis that PD pathology affects the peripheral nervous system (PNS) of the larynx, thus possibly contributing to SSV deficits. Twenty-eight adult human larynges obtained from autopsied subjects with clinically diagnosed and neuropathologically confirmed PD (n = 20) and age-matched healthy controls (n = 8) were studied. Three laryngeal nerves (i.e., recurrent laryngeal nerve, RLN; external and internal superior laryngeal nerves, ESLN and ISLN), three musc

Peripheral Nervous System Involvement in Late-Onset Cobalamin C Disease?

Background: Cobalamin C (cblC) has a fundamental role in both central and peripheral nervous system function at any age. Neurologic manifestations may be the earliest and often the only manifestation of hereditary or acquired cblC defect. Peripheral neuropathy remains a classical but underdiagnosed complication of cblC defect, especially in late-onset cblC disease caused by mutations in the methylmalonic aciduria type C and homocysteinemia (MMACHC) gene. So the clinical, electrophysiological, and pathological characteristics of late-onset cblC disease are not well-known. Methods: A retrospecti

[Central and peripheral nervous system involvement in immunoglobulin G4-related disease].

Immunoglobulin G4-related disease (IgG4-RD) is a novel clinical disease entity characterized by elevated serum IgG4 concentration and tumefaction or tissue infiltration by IgG4-positive plasma cells. IgG4-RD can occur in various organs, including the pancreas, lacrimal gland, salivary gland, thyroid, lung, bile duct, liver, gastrointestinal tract, kidney, prostate, retroperitoneum, arteries, lymph nodes, skin, and breast. Steroid therapy is often effective. In the field of neurology, pachymeningitis (IgG4-related pachymeningitis) and hypophysitis (IgG4-related hypophysitis) are known to be rel

Relapsing demyelinating disease affecting both the central and peripheral nervous systems.

BACKGROUND: Clinical and electromyographic findings of chronic inflammatory demyelinating polyradiculopathy (CIDP) are occasionally observed in patients with multiple sclerosis (MS). OBJECTIVE: To define a new inflammatory demyelinating disease unlike MS or CIDP. RESULTS: This study reports on five patients with a demyelinating disease affecting the central nervous system (CNS) and peripheral nervous system (PNS). Each case presented a relapsing-remitting course in which CNS involvement preceded PNS involvement. All patients fulfilled Barkhof's criteria on MRI and the McDonald criteria for M

Central and peripheral nervous system dysfunction in the clinical variation of Salla disease.

OBJECTIVE: To evaluate the degree of possible peripheral nervous system (PNS) involvement in addition to CNS manifestations in Salla disease, a free sialic acid storage disorder leading to severe mental retardation with a wide clinical variation. BACKGROUND: Salla disease is a lysosomal storage disorder that affects the white matter of the CNS. MRI findings and recent 1H MRS study results provide evidence for delayed central myelination, but there is no previous evidence for PNS involvement in this disease. The gene coding for a presumptive sialic acid transport protein has recently been iden

A sequential study of the peripheral nervous system involvement in experimental Chagas' disease.

To search for the sequential compromise of the spinal cord, nerves, and skeletal muscle in mice chronically infected with Trypanosoma cruzi, animals were subjected to electromyographic investigation, end-plate recordings, and histological studies at 7, 15, 37, 60, 90, 120, 180, 270, and 360 days postinfection. Electromyographic studies showed signs of motor unit remodeling as early as 15 days postinfection, when diminished duration and amplitude of motor unit potentials pointing to a primary muscle involvement were found. Thereafter, certain features of denervation, reinnervation, and primary

Pathogenic Crosstalk Between the Peripheral and Central Nervous System in Rheumatic Diseases: Emerging Evidence and Clinical Implications.

Systemic autoimmune rheumatic diseases (SARDs), such as rheumatoid arthritis (RA), systemic lupus erythematosus (SLE), and Sjögren's syndrome (SS), are traditionally characterized by chronic inflammation and immune-mediated damage to joints and other tissues. However, many patients also experience symptoms such as widespread pain, persistent fatigue, cognitive dysfunction, and autonomic disturbances that cannot be attributed directly or entirely to peripheral inflammation or structural pathology. These conditions suggest the involvement of interactions between the nervous and immune systems, w

Severe allo-immune antibody-associated peripheral and central nervous system diseases after allogeneic hematopoietic stem cell transplantation.

Allogeneic hematopoietic stem cell transplantation (alloHSCT) is a curative treatment for hematologic malignancies. Acute and chronic graft-versus-host disease (GvHD) are the major immune-mediated complications after alloHSCT. However, there is controversy whether neurologic complications after alloHSCT might represent manifestations of GvHD. We report three patients who acquired distinct, severe immune-mediated peripheral or central nervous system diseases after alloHSCT without other, concomitant GvHD manifestations. One patient had been diagnosed with B-cell chronic lymphocytic leukemia and
