A new method to localise and quantify oxidative stress in live juvenile mussels.

Stress and survival of the juvenile New Zealand green-lipped mussel, Perna canaliculus, is a poorly understood bottleneck in the ecological and economic performance of a significant aquaculture crop. This species was therefore selected as a model organism for the development of a new method to quantify oxidative stress in whole individuals. An in vivo ROS-activated stain (CellROX™) was administered to anaesthetised, translucent juveniles that were subsequently formaldehyde fixed and then visualised using confocal microscopy. Subsequent application of image analysis to quantifying ROS-positive

Trophic transference of microplastics under a low exposure scenario: Insights on the likelihood of particle cascading along marine food-webs.

Microplastics are emergent pollutants in marine environments, whose risks along food-web still need to be understood. Within this knowledge gap, MPs transference and persistence along trophic levels are key processes. We assessed the potential occurrence of these processes considering a less extreme scenario of exposure than used previously, with microplastics present only in the hemolymph of prey (the mussel Perna perna) and absent in the gut cavity. Predators were the crab Callinectes ornatus and the puffer fish Spheoeroides greeleyi. Transference of microplastics occurred from prey to preda

Antifouling activity of twelve demosponges from Brazil.

Benthic marine organisms are constantly exposed to fouling, which is harmful to most host species. Thus, the production of secondary metabolites containing antifouling properties is an important ecological advantage for sessile organisms and may also provide leading compounds for the development of antifouling paints. High antifouling potential of sponges has been demonstrated in the Indian and Pacific oceans and in the Caribbean and Mediterranean seas. Brazilian sponges remain understudied concerning antifouling activities. Only two scientific articles reported this activity in sponges of Bra

Sub-lethal combined effects of illicit drug and decreased pH on marine mussels: A short-time exposure to crack cocaine in CO2 enrichment scenarios.

The increasing CO2-concentrations in the atmosphere promote ocean acidification. Seawater chemistry changes interact with contaminants, such as illicit drugs in the coastal zones. This work evaluates impacts of pH decrease and crack-cocaine exposure on the commercial mussel Perna perna through biomarker responses (lysosomal membrane stability, lipid peroxidation, and DNA strand breaks). The organisms were exposed to different crack-cocaine concentrations (0.5, 5.0, and 50 μg L-1) combined with different pH values (8.3, 8.0, 7.5, 7.0, 6.5, and 6.0) for 96 h. Crack-cocaine in the different acidi

Fighting for Resources: Who Started the Battle? Who Is Winning It?

Several vacuolar bacteria and parasites, such as Legionella, Chlamydia, and Toxoplasma, have been reported to grow associated with host mitochondria. The reason behind this phenomenon remains elusive. In this issue of Cell Metabolism, Pernas et al. (2018) propose that fusion of host mitochondria limits the availability of fatty acids needed for Toxoplasma gondii replication.

Antimicrobial resistance and genetic diversity of Escherichia coli isolated from marine bivalves.

Escherichia coli can contaminate the marine environment through sewage and accumulate in bivalve molluscs. We assessed antimicrobial resistance (AMR) mechanisms and genetic diversity of 69 E. coli isolates recovered from in natura, boiled, and purchased Perna perna mussels in Niterói city, Brazil. All isolates were sensitive to cefoxitin, imipenem, meropenem, and fosfomycin. In contrast, 61 (88.4%) isolates were not susceptible to streptomycin. We found ten (14.5%) multidrug-resistant (MDR) isolates. Extended-spectrum beta-lactamase (ESBL) production was detected in 30 (43.5%) isolates, with 1

Lepocreadioidea (Digenea: Trematoda) of southern Africa, including a new genus and species of Lepidapedidae infecting the galjoen, Dichistius capensis (Cuvier) (Centrarchiformes: Dichistiidae) from off Namibia.

Investigations into the digenean trematode fauna of southern African marine fishes revealed representatives of three families of the superfamily Lepocreadioidea Odhner, 1905. A new taxon, requiring the proposal of a new genus, Cutmoreia galjoen n. gen. et n. sp., is described from the intestine of the galjoen, Dichistius capensis (Cuvier) (Dichistiidae), collected from off the shore between Langstrand and Swakopmund, Namibia. A combination of morphology and molecular sequence data place this taxon in the Lepidapedidae Yamaguti, 1958, representing the first member of this family to be recorded

Dissolution of Phosphate and Precipitation of Carbonate in the Biomineralization of the Bivalve Shell Limnoperna fortunei.

The mantle of bivalves plays a fundamental role in shell formation and maintenance through biomineralization. Experiments performed on mantle-shell preparations using different experimental techniques revealed the presence of phosphorus (P)-containing compounds in the shell as the first mineral phase formed at the growing edge of the periostracum in the freshwater bivalve Limnoperna fortunei. The low P concentration in the shells suggests that phosphate is restricted to the growth regions and occurs at concentrations too low to be detected by techniques such as X-ray diffraction (XRD). Assessment of the health risks associated with the consumption of bivalve mollusks potentially contaminated with phycotoxins from the coastal ecosystem of the Ebrié lagoon, Côte d'Ivoire.

This work focused on assessing of the risk associated with the consumption of bivalve mollusks, potentially contaminated with phycotoxins. The studied phycotoxins are saxitoxin (STX), okadaic acid (OA), dinophysistoxins (DTXs), yessotoxins (YTXs), pectenotoxins (PTX), azaspiracids (AZAs), and domoic acid (DA). These toxins were investigated in three species of bivalve mollusks (Anadara senilis, Crassostrea gasar, and Perna perna), originating from the Ebrié lagoon. Chemical analyses were carried out by LC-MS/MS, HPLC-FLD, and HPLC-UV. The level of OA and DTXs, STX, and DA was 10.92 µg OA eq./k

Metabolomic and physiological insights to ameliorate post-harvest stress in cultured mussels.

INTRODUCTION: Survival and quality of Green-lipped mussels (Perna canaliculus) exported live could be further improved through enhanced post-harvest handling, aiming to reduce the physiological stress associated with transport out of water. Addressing these issues requires identifying treatments to reduce post-harvest stress and understanding underpinning molecular mechanisms. OBJECTIVE: This study aimed to evaluate treatments (low temperature and MgCl2 anaesthetic baths) to mitigate post-harvest handling stress in mussels. METHODS: We analysed metabolomic profiles using gas chromatography-m

Effects of dichlofluanid in tropical marine bivalves exposed to water and spiked sediments: an assessment of biomarker responses.

Antifouling paints have been used to control biofouling on submerged surfaces for decades. They contain biocidal compounds that may be released into the environment and harm aquatic ecosystems; however, their effects on tropical organisms are little known. Dichlofluanid, or N-dichlorofluoromethythio-N',N'-dimethyl-N-phenylsulfamide (C9H11Cl2FN2O2S2), is a biocide present in antifouling paints and its toxicity to tropical marine organisms is poorly understood. This study aimed to evaluate the effects of dichlofluanid at the cellular and biochemical levels in bivalves exposed to seawater and sed

Interactive effects of ocean acidification and benzophenone-3 on mussel physiology: A biomarker-based approach.

Benzophenone-3 (BP3), a widely used ultraviolet (UV) filter in personal care products and industrial applications, is an emerging concern in aquatic ecosystems due to its lipophilicity, bioaccumulation potential, and toxicity to marine biota. Its occurrence in marine environments coincides with ocean acidification driven by rising atmospheric CO2 emissions, which disrupts the bicarbonate buffer system and reduces pH. This study investigated the acute effects (96 h exposure) of BP3 (at realistic concentrations of 0.2 and 2 μg/L) under 3 pH conditions (8.1- the current South Atlantic condition-

Fatal bowel infarction and sepsis: an unusual complication of systemic strongyloidiasis.

A 58 year old Chinese male, one week after arriving in Canada from Hong Kong, presented with acute abdominal pain and diarrhoea which was rapidly followed by Escherichia coli infection causing septicaemia and meningitis. His past history revealed bronchial asthma for 15 years treated with steroids. At laparotomy, 7 days after the onset of symptoms, he was found to have extensive haemorrhagic infarction of the small bowel and right colon. Examination of the fibrosed mesenteric vessels revealed numerous filariform larvae of Strongyloides stercoralis, within the walls, and in all layers of bowel

A randomized controlled trial investigating the effects of PCSO-524, a patented oil extract of the New Zealand green lipped mussel (Perna canaliculus), on the behaviour, mood, cognition and neurophysiology of children and adolescents (aged 6-14 years) experiencing clinical and sub-clinical levels of hyperactivity and inattention: study protocol ACTRN12610000978066.

BACKGROUND: The prevalence rate of attention-deficit/hyperactivity disorder (ADHD) within Western cultures is between 5% and 12%, and is the most common psychiatric illness among school-aged children, with an estimated 50% of these children retaining ADHD symptoms for the rest of their lives. Children with ADHD have lower blood levels of long-chain Poly Unsaturated Fatty Acids (LC PUFAs) compared with children without ADHD, and following PUFA supplementation, have shown improvements in ADHD-related symptoms. One highly promising marine based LC PUFA preparation is the Omega-3-rich Lyprinol/Ome

Multi-species okadaic acid contamination and human poisoning during a massive bloom of Dinophysis acuminata complex in southern Brazil.

On June 2016, a major bloom of Dinophysis acuminata complex was noticed over the coast of Paraná State (PR), southern Brazil, an area unprotected by any official monitoring program. Here we report the results of an extensive sampling effort that ultimately led PR authorities to issue the first State shellfish-harvesting ban due to multi-species okadaic acid (OA) contamination. During its peak, the bloom covered an area of 201 km2 (∼2.0-3.5 × 54.0 km), attaining unprecedentedly high cell densities along the shallow (<15 m) continental shelf (mean 2.2 × 105, maximum 2.1 × 106 cells L-1) and adja

Accumulation and depuration of ciguatoxins in green-lipped mussels fed toxic Gambierdiscus polynesiensis: Evidence from controlled feeding and transcriptomic analysis.

Ciguatera poisoning (CP) was traditionally associated with fish consumption, but other marine invertebrates such as gastropods and bivalves can also serve as non-traditional vectors. In this study, ciguatoxins (CTXs) accumulation, tissue distribution, depuration kinetics, and associated transcriptomic responses were studied using controlled feeding trials on green-lipped mussels (Perna canaliculus), an important seafood species, fed Gambierdiscus polynesiensis. In two experiments, mussels accumulated CTXs rapidly, peaking at concentrations of 5.14 µg kg-1 (day 19, Experiment 1) and 4.57 µg kg-

Butyltin accumulation in marine bivalves under field conditions in Taiwan.

This study aimed to characterize the butyltin bioaccumulation in a simple food chain under varied conditions. Significant trophic level magnification factors of tributyltin (21.5-1546) were observed in two filter-feeders, oysters (Crassostrea gigas) and mussels (Perna viridis), in an environment with low tributyltin pollution levels (0.4-13.1 ng L(-1) as tin). Both of these bivalve species showed higher bioaccumulation factors (BAFs) of tributyltin under low pollution levels, while smaller magnification factors (5.4-6.4), an up-regulated tributyltin metabolism and smaller BAFs of tributyltin w
