Peste des petits ruminants virus (PPRV) is recognized as one of the most economically significant transboundary pathogens affecting small ruminants. However, despite the increasing availability of whole-genome sequences, comprehensive global analyses integrating evolutionary history, phylogeography, and genome-wide lineage-defining molecular markers remain limited. This study investigated the global genomic diversity, evolutionary history, and molecular characteristics of PPRV using whole-genome sequences. A total of 212 temporally informative whole-genome sequences, retrieved from GenBank (be

Macroautophagy/autophagy is an essential cellular response in the fight against intracellular pathogens. Although some viruses can escape from or utilize autophagy to ensure their own replication, the responses of autophagy pathways to viral invasion remain poorly documented. Here, we show that peste des petits ruminants virus (PPRV) infection induces successive autophagic signalling in host cells via distinct and uncoupled molecular pathways. Immediately upon invasion, PPRV induced a first transient wave of autophagy via a mechanism involving the cellular pathogen receptor NECTIN4 and an AKT-

Process intensification for Peste des Petites Ruminants Virus (PPRV) vaccine production in anchorage dependent Vero cells is challenging, involving substantial amount of bioprocess development. In this study, we describe the implementation of a new, scalable bioprocess for PPRV vaccine production in Vero cells using serum-free medium (SFM), microcarrier technology in stirred-tank bioreactors (STB), in-situ cell detachment from microcarriers and perfusion. Vero cells were successfully adapted to ProVero™-1 SFM, reaching growth rates similar to serum-containing cultures (0.030 1/h vs 0.026 1/h,

BACKGROUND: Peste des petits ruminants virus (PPRV) is a major transboundary viral pathogen of small ruminants and causes substantial economic losses in endemic regions. The hemagglutinin (H) protein mediates receptor recognition and host-cell attachment and is an important target for vaccine development. This study applied an integrated computational framework to identify conserved immunogenic regions within the PPRV H protein.
METHODS: A total of 64 unique PPRV H protein sequences were analyzed using multiple sequence alignment, entropy-based conservation profiling, conservation-aware epito

In December 2017, Peste des Petits Ruminants (PPR) emerged in Burundi (East Africa) and rapidly spread to five provinces (Gitega, Kirundo, Mwaro, Muramvya and Karuzi) in the country, causing severe disease and killing more than 4,000 goats in the province of Gitega alone. An initial outbreak investigation was conducted in December 2017 by the Burundi Government Veterinary Services and samples were collected for laboratory confirmation. A competitive Enzyme Linked Immuno-Sorbent Assay (cELISA: Chinese Patent No. ZL201210278970.9) supplied by the Lanzhou Veterinary Research Institute was used to

Peste des petits ruminants (PPR) is a contagious viral disease of sheep and goats common in Africa and Asia. Its high morbidity and mortality has a devastating impact on agriculture in developing countries. As an example, an Asian lineage IV strain of PPRV was responsible for mass fatalities among wild goats in Kurdistan in 2010/2011. In separate experiments, three sheep and three goats of German domestic breeds were subcutaneously inoculated with the Kurdish virus isolate; three uninfected sheep and goats were housed together with the inoculated animals. All inoculated animals, all in-contact

Peste des petits ruminants (PPR) is one of the highly contagious transboundary viral diseases of small ruminants. Host microRNA (miRNA) expression patterns may change in response to virus infection, and it mainly works as a post-transcriptional moderator in gene expression and affects viral pathogenesis and replication. In this study, the change of miRNA expression profile in peripheral blood lymphocyte (PBMC) from sheep inoculated with PPR vaccine virus in vivo as well as primary sheep testicular (ST) cells inoculated with PPR vaccine virus in vitro were determined via deep sequencing technol

Four goats were inoculated with an inactivated peste des petits ruminants virus (PPRV) vaccine. Three unvaccinated goats were kept as controls. After 36 days, the four goats were revaccinated. The immune response was monitored by virus neutralization test showing that two doses of the vaccine were able to stimulate strong immune response in all the vaccinated animals. The vaccinated goat and the controls were challenged with virulent PPRV intranasally. After PPRV challenge, the three control goats showed fever, viremia and virus excretion through mucosal surfaces, whereas the vaccinated goats

Peste des petits ruminants virus (PPRV) is a highly contagious pathogen that infects domestic and wild small ruminants, causing high mortality worldwide. PPRV has evolved multiple strategies to suppress the host antiviral immune response, thereby promoting viral replication. In this study, we demonstrate that caveolin-1 (CAV1) promotes PPRV replication. Mechanistically, CAV1 inhibits IFN-β production, ISG expression, and the IFN signaling pathway by promoting cholesterol synthesis. Importantly, we reveal that PPRV infection upregulates cellular CAV1 protein levels, and the viral nucleocapsid (

Peste des petits ruminants (PPR) is a vaccine-preventable transboundary animal disease of goats and sheep majorly, and is regarded as a major constraint to small ruminant production especially in developing countries like Nigeria. Despite different strategies that have been employed to control PPR in Nigeria, cases of the disease are still reported in PPR-vaccinated and unvaccinated small ruminant farms. In this study, molecular detection of field PPR virus (PPRV) strains was carried out to determine the presence of PPRV. A total of 135 samples (45 oculo-nasal swabs and 90 tissue samples) were

Peste des petits ruminants (PPR) is one of the highly contagious viral disease, characterized by fever, sore mouth, conjunctivitis, gastroenteritis, and pneumonia, primarily affecting sheep and goats. Reports suggested variable host response in goats and sheep and this host response vis-a-vis the expression of microRNAs (miRNAs) has not been investigated. Here, miRNAs were sequenced and proteomics data were generated to identify the role of differentially expressed miRNA (DEmiRNA) in PPR virus (PPRV) infected lung and spleen tissues of sheep and goats. In lungs, 67 and 37 DEmiRNAs have been id

Peste des petits ruminants (PPR) is a viral disease of sheep and goats that is spreading through many countries in the developing world. Work on the virus is often restricted to studies of attenuated vaccine strains or to work in laboratories that have high containment facilities. We have created a helper cell dependent form of PPR virus by removing the entire RNA polymerase gene and complementing it with polymerase made constitutively in a cell line. The resultant L-deleted virus grows efficiently in the L-expressing cell line but not in other cells. Virus made with this system is indistingui

BACKGROUND: Peste des petits ruminants virus (PPRV) is currently the only member of the Morbillivirus caprinae species within the genus Morbillivirus of the family Paramyoxviridae. PPRV causes a highly contagious disease in small ruminants, especially goats and sheep. Succinylation is a newly identified and conserved modification and plays an important role in host cell response to pathogen infection. However, the extent and function of succinylation in Vero cells during PPRV infection remains unknown.
RESULTS: In this study, a global profile of the succinylome in Vero cells infected with PPR

Peste des petits ruminants (PPR) mostly affects small ruminants. Vaccination with attenuated vaccines derived from PPR virus (PPRV) provides successful protection against the disease. However, current molecular methods are unable to distinguish vaccine strains from field strains. In this study, we used an EvaGreen-based high-resolution melting (HRM) assay to differentiate a PPRV vaccine strain (Nigeria 75/1) from Turkish field isolates of lineage IV based on melting peaks and melting profiles.
