Adsorbate-Directed Surface Phase Separation in Cu-Ag Catalysts.

The determination of surface configuration is essential for understanding catalytic reactivity, yet these active states cannot be reliably inferred from bulk phase diagrams. In this work, we develop a machine-learning-accelerated molecular dynamics (ML-MD) framework on the Cu-Ag system to predict and control surface structure, identifying two competing descriptors: surface energy and adsorbate binding energy. We show that Ag's lower surface energy dominates in vacuum, forming a passive sheath, whereas a CO atmosphere reverses segregation and exposes active Cu sites through lateral phase separa

Liquid-Liquid Phase Separation in Viral Infection and Immunology.

Liquid-liquid phase separation (LLPS) has emerged as a fundamental physicochemical principle that organizes macromolecules into dynamic, membraneless condensates. These assemblies are increasingly recognized as critical regulators of diverse cellular processes. Notably, both viruses and their hosts exploit LLPS to optimize their respective strategies for replication and defense, forming a dynamic interplay centered around phase separation. However, a comprehensive mechanistic understanding of how LLPS modulates the dynamic viral-host battle, and how this knowledge can be leveraged for therapeu

Nuclear Phase Separation Drives NPM1-mutant Acute Myeloid Leukemia.

During cancer development, mutations promote gene expression changes that cause transformation. Leukemia is frequently associated with aberrant HOXA expression driven by translocations in nucleoporin genes or KMT2A, and mutations in NPM1. How disparate mutations converge on this regulatory pathway is not understood. Here we demonstrate that mutant NPM1 (NPM1c) forms nuclear condensates in multiple human cell lines, mouse models, and primary patient samples. We show NPM1c phase separation is necessary and sufficient to coordinate the recruitment of NUP98 and KMT2A to condensates. Through extens

Regulating Enzyme Activity via Microaggregates Mediated by Phase Separation.

Liquid-solid phase separation (LSPS) is a biomolecular segregation process forming solid-like aggregates that encapsulate and concentrate specific biomolecules, thus profoundly influencing biochemical reactions. Building on this phenomenon, we developed an enzyme-regulation strategy inducing LSPS of target enzymes to isolate them from substrates. As proof of concept, we created a novel D-Peptide mediated Microaggregate Degradation named DPMD to capture trypsin and chymotrypsin in the seroperitoneum during postoperative pancreatic fistula (POPF). DPMD is designed with cationic and hydrophobic r

Cell-guiding microporous hydrogels by photopolymerization-induced phase separation.

Microporous scaffolds facilitate solute transport and cell-material interactions, but materials allowing for spatiotemporally controlled pore formation in aqueous solutions are lacking. Here, we propose cell-guiding microporous hydrogels by photopolymerization-induced phase separation (PIPS) as instructive scaffolding materials for 3D cell culture. We formulate a series of PIPS resins consisting of two ionic polymers (norbornene-functionalized polyvinyl alcohol, dextran sulfate), di-thiol linker and water-soluble photoinitiator. Before PIPS, the polymers are miscible. Upon photocrosslinking, t

Intercalated Iron Chalcogenides: Phase Separation Phenomena and Superconducting Properties.

Organic molecule-intercalated layered iron-based monochalcogenides are presently the subject of intense research studies due to the linkage of their fascinating magnetic and superconducting properties to the chemical nature of guests present in the structure. Iron chalcogenides have the ability to host various organic species (i.e., solvates of alkali metals and the selected Lewis bases or long-chain alkylammonium cations) between the weakly bound inorganic layers, which opens up the possibility for fine tuning the magnetic and electrical properties of the intercalated phases by controlling bo

Microscale extraction and phase separation using a porous capillary.

We report the use of a porous polytetrafluoroethylene capillary for the inline separation of liquid-liquid segmented flows, based on the selective wetting and permeation of the porous capillary walls by one of the liquids. Insertion of a narrow flow restriction at the capillary outlet allows the back pressure to be tuned for multiple liquid-liquid combinations and flow conditions. In this way, efficient separation of aqueous-organic, aqueous-fluorous and organic-fluorous segmented flows can be readily achieved over a wide range of flow rates. The porous-capillary-separator enables the straight

Role of desolvation on biomolecular liquid-liquid phase separation.

Biomolecular condensates play essential roles in cellular organization and are implicated in diverse pathological processes. Their formation is driven by liquid-liquid phase separation (LLPS), a process that requires coordinated multistep desolvation of biomolecular chains and multivalent inter-chain interactions. Although coarse-grained (CG) models with implicit solvent are widely used to probe LLPS thermodynamics and kinetics, they typically neglect water-mediated desolvation effects, limiting their accuracy and mechanistic interpretability. Here, guided by all-atom simulations and experimen

Stereosynthesis of stationary phase with precision defined separation selectivity.

Chemical separation is based on molecules' differential interactions with a stationary phase and a mobile phase. To enable efficient thermodynamics and molecular kinetics, micrometer-size spheres with bonded chemistry have become the general choice for stationary phase materials. For half a century, the application of stationary-phase chemistry to microspheres has been a random, unordered and bulk process in the manufacture of chromatographic materials. Such a non-stereoresolving synthetic strategy results in the co-location of multiple functional groups, which share the same space, including

Farnesylation-driven KRAS phase separation promotes colon tumor growth.

Kirsten Rat Sarcoma viral oncogene homolog (KRAS) is one of the most frequently activated driver genes across human cancers. We identified a regulatory mechanism where KRAS forms condensates in the cytoplasm through liquid-liquid phase separation (LLPS), driven by farnesylation at the C185 residue within its hypervariable region (HVR). These condensates are associated with advanced stages and poor outcomes in colon cancer. Functionally, KRAS condensates efficiently interact with Ras-converting enzyme 1 (RCE1), promoting RCE1 clustering, enhancing KRAS processing, and facilitating its transloca

Air-permeable hydrogels through viscoelastic phase separation of aerogels.

Hydrogels are widely used in biomedical interfaces, in which effective gas exchange (for example, O2, CO2) within a water-rich environment is essential. However, hydrogels show intrinsically limited air exchange efficiency, owing to the low solubility (C) and diffusivity (D) of non-polar gases in the polar water medium1. This limitation poses a substantial bottleneck in long-term applications, such as wearable health monitors2-7 and tissue engineering8-12. Existing methods13-16 to enhance air permeability suffer from poor robustness and/or an inherent trade-off between permeability and water c

Clathrin adaptors drive phase separation in endocytosis and trafficking.

Liquid-liquid phase separation (LLPS) underlies the formation of biomolecular condensates that organize processes, including endocytosis and membrane trafficking. Although LLPS has been implicated in clathrin-mediated pathways, the contribution of adaptor proteins to condensate formation and function remains unclear. Here we show that two yeast adaptors, Ent5 and Sla2, undergo LLPS in vitro and this property correlates with their roles in membrane trafficking and condensate recruitment in vivo. Ent5 condensation is driven by a dynamic helix within its disordered region that acts as a molecular

Protocol for in vitro phase separation of ARID3B proteins.

Liquid-liquid phase separation (LLPS) of ARID3B is critical in nonsyndromic cleft lip/palate pathogenesis. Here, we present a protocol to visualize ARID3B LLPS. We describe steps for regenerating LLPS with purified protein in vitro and detecting ARID3B condensates in mammalian cells. This protocol enables analysis of ARID3B phase separation dynamics and the study of other transcription factors that undergo LLPS. For complete details on the use and execution of this protocol, please refer to Li et al.1.

The Skin Barrier: A System Driven by Phase Separation.

The mammalian epidermis forms a critical barrier against environmental insults and water loss. The formation of its outermost layer, the stratum corneum, involves a rapid terminal differentiation process that has traditionally been explained by the "bricks and mortar" model. Recent advances reveal a more dynamic mechanism governed by intracellular liquid-liquid phase separation (LLPS). This review proposes that the lifecycle of the granular layer is orchestrated by LLPS. Evidence is synthesized showing that keratohyalin granules (KGs) are biomolecular condensates formed by the phase separation

Glycogen accumulation and phase separation drives liver tumor initiation.

Glucose consumption is generally increased in tumor cells to support tumor growth. Interestingly, we report that glycogen accumulation is a key initiating oncogenic event during liver malignant transformation. We found that glucose-6-phosphatase (G6PC) catalyzing the last step of glycogenolysis is frequently downregulated to augment glucose storage in pre-malignant cells. Accumulated glycogen undergoes liquid-liquid phase separation, which results in the assembly of the Laforin-Mst1/2 complex and consequently sequesters Hippo kinases Mst1/2 in glycogen liquid droplets to relieve their inhibiti

Phase separation in biology and disease-a symposium report.

Phase separation of multivalent protein and RNA molecules enables cells the formation of reversible nonstoichiometric, membraneless assemblies. These assemblies, referred to as biomolecular condensates, help with the spatial organization and compartmentalization of cellular matter. Each biomolecular condensate is defined by a distinct macromolecular composition. Distinct condensates have distinct preferential locations within cells, and they are associated with distinct biological functions, including DNA replication, RNA metabolism, signal transduction, synaptic transmission, and stress respo

Drug release characteristics of phase separation pHEMA sponge materials.

A number of phase separation pHEMA sponge hydrogels have been prepared based on variations in monomer contents, concentration of cross-linking agent, solvent mixture and temperature of polymerization. The loading levels and release profiles of the anti-inflammatory drug prednisolone were examined for each of the pHEMA sponge materials. An effective diffusion coefficient determined by an optimization approach based on the experimental data was used to measure their release characteristics. The effect of morphological variations, revealed by the environmental scanning electron microscopy, and po

The huntingtin inclusion is a dynamic phase-separated compartment.

Inclusions of disordered protein are a characteristic feature of most neurodegenerative diseases, including Huntington's disease. Huntington's disease is caused by expansion of a polyglutamine tract in the huntingtin protein; mutant huntingtin protein (mHtt) is unstable and accumulates in large intracellular inclusions both in affected individuals and when expressed in eukaryotic cells. Using mHtt-GFP expressed in Saccharomyces cerevisiae, we find that mHtt-GFP inclusions are dynamic, mobile, gel-like structures that concentrate mHtt together with the disaggregase Hsp104. Although inclusions m
