Comparative genomic analysis and phenazine production of Pseudomonas chlororaphis, a plant growth-promoting rhizobacterium.

Pseudomonas chlororaphis HT66, a plant growth-promoting rhizobacterium that produces phenazine-1-carboxamide with high yield, was compared with three genomic sequenced P. chlororaphis strains, GP72, 30-84 and O6. The genome sizes of four strains vary from 6.66 to 7.30 Mb. Comparisons of predicted coding sequences indicated 4833 conserved genes in 5869-6455 protein-encoding genes. Phylogenetic analysis showed that the four strains are closely related to each other. Its competitive colonization indicates that P. chlororaphis can adapt well to its environment. No virulence or virulence-related fa

Phenazine production by Pseudomonas sp. LBUM223 contributes to the biological control of potato common scab.

Common scab of potato is mainly caused by Streptomyces scabies. Currently, no method can efficiently control this economically important disease. We have previously determined that Pseudomonas sp. LBUM223 exhibits antagonistic properties toward S. scabies under in vitro conditions. Inhibition was mainly attributed to phenazine-1-carboxylic acid (PCA) production because an isogenic mutant of LBUM223 (phzC-), not producing PCA, was incapable of significantly reducing S. scabies growth. In order to understand the impact of PCA production by LBUM223 in controlling common scab under soil conditions

Environmental determinants of phenazine- and 2,4-diacetylphloroglucinol-producing Pseudomonas in the wheat rhizosphere across a precipitation gradient.

The Columbia Plateau of the Inland Pacific Northwest (IPNW) encompasses one of North America's largest dryland wheat production regions, where soil moisture strongly influences plant growth, health, and rhizosphere microbial communities. Two ecologically and genetically distinct groups of the Pseudomonas fluorescens complex naturally suppress soilborne wheat pathogens in IPNW fields by producing either phenazine-1-carboxylic acid (Phz+) or 2,4-diacetylphloroglucinol (Phl+). To determine how precipitation and associated soil factors shape the abundance of these bacteria, we analyzed wheat root

Environmental redox conditions and strain variation define phenazine-mediated antagonism in co-infecting bacteria.

Pseudomonas aeruginosa and Klebsiella pneumoniae are gram-negative opportunistic pathogens that frequently colonize the human body and are major causes of infection. These bacteria are often co-isolated in polymicrobial urinary tract and lung infections, the latter of which is associated with increased disease severity and worse clinical outcomes. Despite their overlapping niches and clinical relevance, little is known about how these two pathogens interact and how those interactions influence human health. Given the growing recognition that microbial interactions are key drivers of disease, w

Redox-Conjugated CoII-Phthalocyanine-Phenazine Polymer as a Robust Catalyst for Efficient Electrochemical CO2 Conversion.

This work reports the synthesis and electropolymerization of 4-aminophenoxy-substituted CoII-phthalocyanine to prepare a phenazine (PNZ)-bridged CoII-phthalocyanine polymer as a durable catalyst for heterogeneous electrochemical CO2 reduction (ECO2R). At -1.19 V versus normal hydrogen electrode, the ECO2R under catalysis of the target polymer effectively produces CO with 94% faradaic efficiency (FECO), CO partial current density of 5.5 mA cm-2, and turnover frequency of 1.1 s-1 over 2 h. Moreover, the polymeric system exhibits remarkably higher stability than the monomeric one by achieving ave

Novel Chalcone-Phenazine Hybrids Induced Ferroptosis in U87-MG Cells through Activating Ferritinophagy.

Thirty-seven novel chalcone-phenazine hybrid molecules (C1∼C13 and F1∼F24) with 1,2,3-triazole or ethyl group as linkers were designed and synthesized in this study. Some compounds exhibited selective cytotoxicity against U87-MG cancer cell lines in vitro, in which compound C4 were found to have the best antiproliferative activity. SAR study indicated 1,2,3-triazole group may be crucial for enhancing compounds' cytotoxicity. C4 was verified to induce ferroptosis in U87-MG cells by transcription, lipid peroxidation, lipid ROS assays. Furthermore, C4 was up-regulated LC3-II, degradated FTH1, and

3,11-Diaminodibenzo[a,j]phenazine: Synthesis, Properties, and Applications to Tröger's Base-Forming Ladder Polymerization.

A new class of diamino-substituted π-extended phenazine compound was synthesized, and its photophysical properties were investigated. The U-shaped diaminophenazine displayed photoluminescence in solution with moderate quantum yield. The diamino aromatic compound was found applicable to the poly-condensation with formaldehyde to form Tröger's base ladder polymer. The obtained microporous ladder polymer features high CO2 adsorption selectivity against N2 , most likely due to the presence of basic nitrogen atoms in the phenazine rings.

Synthesis, molecular modeling and anti-inflammatory activity of new benzo[a]pyrano[2,3-c]phenazine compounds.

A convenient microwave-assisted three-component synthetic approach was developed for the synthesis of new benzo[a]pyrano[2,3-c]phenazine derivatives starting from benzo[a]phenazin-5-ol, benzaldehyde derivatives, and enaminone derivatives. Sixteen new derivatives were successfully synthesized and identified by spectroscopic data analysis. To analyse the potent anti-inflammatory activity of these new derivatives, the Griess assay and the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay were used to assess their effects on NO production in lipopolysaccharide (LPS)-stimulat

Zinc-Enhanced Activity of an Antimicrobial Halogenated Phenazine Against Streptococcus mutans and Other Gram-positive Bacteria.

Halogenated phenazine (HP) compounds have shown promise as antimicrobial agents, particularly against biofilm-associated Gram-positive pathogens. Among these compounds, HP-29 demonstrates potent activity against methicillin-resistant Staphylococcus aureus by inducing rapid iron starvation. As maintenance of trace metals homeostasis is critical for the survival of Streptococcus mutans, this study investigated the antimicrobial efficacy of HP-29 and the impact of metal supplementation on this major oral and occasional systemic pathogen. As anticipated, HP-29 inhibited S. mutans growth in a dose-

Novel phenazine-based microporous organic network for selective and sensitive determination of trace sulfonamides in milk samples.

BACKGROUND: Sulfonamide (SA) residues in food of animal origin possess a potential threat to human health and environment. However, due to the polar and ionic characteristics and trace level of SAs and the complexity of food matrices, direct measurement of SAs in these samples is still very difficult. Development of efficient sample pretreatment method for sensitive and selective extraction of trace SAs is of great significance and urgently desired. Therefore, rational design and synthesizing advanced and selective extractants is quite important. RESULTS: In this work, a novel phenazine-based

Inhibition of Three Potato Pathogens by Phenazine-Producing Pseudomonas spp. Is Associated with Multiple Biocontrol-Related Traits.

Phenazine-producing Pseudomonas spp. are effective biocontrol agents that aggressively colonize the rhizosphere and suppress numerous plant diseases. In this study, we compared the ability of 63 plant-beneficial phenazine-producing Pseudomonas strains representative of the worldwide diversity to inhibit the growth of three major potato pathogens: the oomycete Phytophthora infestans, the Gram-positive bacterium Streptomyces scabies, and the ascomycete Verticillium dahliae. The 63 Pseudomonas strains are distributed among four different subgroups within the P. fluorescens species complex and pro

Efficient synthesis of glycosylated phenazine natural products and analogs with DISAL (methyl 3,5-dinitrosalicylate) glycosyl donors.

Inspired by the occurrence and function of phenazines in natural products, new glycosylated analogs were designed and synthesized. DISAL (methyl 3,5-dinitrosalicylate) glycosyl donors were used in an efficient and easily-handled glycosylation protocol compatible with combinatorial chemistry. Benzoylated D-glucose, D-galactose and L-quinovose DISAL glycosyl donors were synthesized in high yields and used under mild conditions to glycosylate methyl saphenate and 2-hydroxyphenazine. The glycosides were screened for biological activity and one compound showed inhibitory activity towards topoisomer

RhlR quorum-sensing receptor ligand sensitivity regulates the differential expression of phenazine genes in Pseudomonas aeruginosa.

Quorum sensing enables bacteria to coordinate gene expression in response to population density through the detection of small-molecule signals. In gram-negative bacteria, LuxR-type transcription factors bind acyl-homoserine lactones to regulate collective behaviors, yet how ligand sensitivity is tuned to shape transcriptional outcomes remains poorly defined. In Pseudomonas aeruginosa, the quorum-sensing receptor RhlR responds to N-butyryl-L-homoserine lactone (C4HSL) and controls late-stage quorum-sensing behaviors, including phenazine biosynthesis. Here, we use a chemical-genetic and structu

Phosphorus stress and spatial confinement lower the quorum-sensing activation threshold for phenazine production in Pseudomonas synxantha.

Bacteria often coordinate collective behaviors such as biofilm formation and secondary metabolite production through quorum sensing (QS), a regulatory system traditionally linked to high cell density. However, in environments such as soil, where microbial populations are spatially fragmented, sparse, nutrient-limited, and subject to mass transport, the mechanisms that enable QS-dependent processes remain incompletely understood. Here, we investigate the regulation of a secreted redox-active metabolite, phenazine-1-carboxylic acid (PCA), in Pseudomonas synxantha 2-79, a model rhizobacterium, un

Among them, phenazine-1-carboxylic acid (PCA)-a registered biopesticide-stands out for its broad-spectrum antifungal efficacy, low environmental toxicity and potential as a lead structure for developing anti-tumor and antituberculosis agents. However, the chemical synthesis of PCA derivatives is often hindered by harsh reaction conditions, toxic byproducts and poor selectivity, limiting their sustainable production. In contr
