Adrenergic innervation of the monkey thalamus: an immunohistochemical study.

The distribution and function of the neurotransmitter adrenaline in the primate brain are poorly understood. Biochemical studies have shown the presence of adrenaline or its biosynthetic enzyme, phenylethanolamine-N-methyltransferase, in the rat and human thalamus. However, the distribution of the adrenergic fibres in the thalamus has only been demonstrated in rats. We study the adrenergic innervation of the macaque monkey thalamus using immunohistochemistry against phenyletanolamine-N-methyltransferase. The distribution of phenyletanolamine-N-methyltransferase-immunoreactive fibres is markedl

Catecholaminergic axonal varicosities appear to innervate growth hormone-releasing hormone-immunoreactive neurons in the human hypothalamus: the possible morphological substrate of the stress-suppressed growth.

CONTEXT: Stress is considered to be a major factor in the regulation of growth. Psychosocial dwarfism, characterized with short stature, del … ive juxtapositions between the catecholaminergic tyrosine hydroxylase (TH)-/dopamine-β-hydroxylase-/phenylethanolamine N-methyltransferase-IR and GHRH-IR neural elements in the human hypothalamus. To reveal these juxtapositions, double-label immunohistochemistry was used. RESULTS: Our findings revealed that the majority of the GHRH-IR perikarya formed intimate associations with TH-IR …

Evidence for retrograde degeneration of epinephrine neurons in Alzheimer's disease.

Alzheimer's disease (AD) is associated with a progressive loss of locus ceruleus neurons. These noradrenergic neurons receive a major afferent projection from epinephrine neurons in epinephrine cell groups in the brainstem. The epinephrine neurons have a specific enzymatic marker, phenylethanolamine N-methyltransferase (PNMT), which allows them to be identified chemically and immunohistochemically. We have previously reported a decrease in PNMT in brains of patients with AD. We now report that the decrease in PNMT activity in projections to the locus ceruleus is not due to the loss of epinephr…

Catecholamine synthesizing enzymes in brain stem and hypothalamus during the development of renovascular hypertension.

The activities of tyrosine hydroxylase (TH) and phenylethanolamine N-methyltransferase (PNMT) have been measured in brain stem and hypothalamic nuclei during the development of renovascular hypertension. TH activity fell at 72 h in the posterior hypothalamic and peri- and paraventricular nuclei of the hypothalamus, but had returned to control levels by 7 days. PNMT activity was raised in the nucleus of the solitary tract at 7 days and was also elevated in the nucleus of the solitary tract, parahypoglossal nucleus, locus coeruleus and cerebellar cortex at 4 weeks. No change in PNMT was noted in…

Stress and the adrenocortical control of epinephrine synthesis.

Psychologic states produced by environmental or physiologic stresses are usually associated with hypersecretion of adrenal hormones, particularly epinephrine and the glucocorticoids (hydrocortisone in humans or corticosterone in rats). A common mechanism links the secretion of these hormones, even though the adrenal medulla and cortex have different embryologic origins and biochemical properties and very different mechanisms controlling their secretory activities, ie, a cholinergic nervous input stimulates medullary secretion while a hormone, corticotropin (ACTH), activates secretion from the

Epinephrine synthesis by rat arteries.

Carotid artery and aorta homogenates synthesized epinephrine (E) from norepinephrine (NE) in the presence of S-adenosylmethionine. Aorta synthesized epinine by the N-methylation of dopamine (DA) about 3 times as well as it synthesized E from NE. In contrast, adrenal homogenates which contain phenylethanolamine N-methyltransferase (PNMT) methylated DA only 1% as well as NE. The PNMT inhibitor SKF 29661 had no significant effect on methylation of NE by aorta but inhibited adrenal PNMT by 88%. N-Methylating activity in arterial homogenates was increased by dexamethasone and following catecholamin…

[Adrenal catecholamines and their metabolism in the vitamin A deficient rat].

The adrenal catecholamines (norepinephrine and epinephrine) and their conversion enzymes (PNMT, COMT) were comparatively investigated in: (1) rats fed a diet deprived of vitamin A, inducing a rapid and severe deficiency; (2) rats fed another diet leading to a more progressive deficiency (rats supplemented with retinol served as controls for these two types of diet), and (3) rats fed a whole industrial diet. The first diet induced a strong accumulation of adrenal catecholamines and increased the activity of PNMT and COMT. The second diet only increased the adrenal content of norepinephrine and

Intracystic catecholamine variations in cystic pheochromocytoma.

PURPOSE: Approximately 20% of pheochromocytomas are cystic in nature. However, research on the correlation between catecholamine and metanephrine fractions in cystic fluid and peripheral blood is limited. This study investigated the relationship between catecholamine fractions and their metabolites in peripheral blood and cystic fluid. METHODS: Five patients with pathologically diagnosed cystic pheochromocytomas were included in the analysis. Catecholamine fractions and their metabolites in the cystic fluid were measured. The relationship between phenylethanolamine N-methyltransferase (PNMT) …

Possible mechanisms of sodium-dependent hypertension: volume expansion or vasoconstriction?

A series of experiments was designed to explore the mechanisms contributing to hypertension caused by an acute or chronic sodium load. Acute salt-loading in totally or subtotally nephrectomized animals caused hypertension mediated partly through stimulation of excessive vasopressin release and partly through adrenergic stimulation. Chronic high-salt diet in rats submitted to partial nephrectomy, mineralocorticoid excess or one-kidney-one-clip renovascular hypertension caused blood pressure elevation mediated through a central neurogenic mechanism that could be reversed by administration of an

Catecholamine involvement in the control of growth hormone secretion in the domestic fowl.

A neuropharmacologic approach was utilized to investigate the catecholaminergic influence on the hypothalamic regulation of growth hormone (GH) secretion in young (6-week-old) male domestic fowl. The selective inhibition of norepinephrine (NE) and epinephrine (E) synthesis or activity by diethyldithiocarbamate (DDC), FLA63 (dopamine-beta-hydroxylase inhibitors), phenoxybenzamine (alpha 1 receptor blocker), and yohimbine (alpha 1 and alpha 2 receptor antagonist) was associated with a decline in circulating GH levels. Similarly inhibition of NE reuptake by imipramine or desmethylimipramine were

Upregulation of neuronal nitric oxide synthase mRNA and protein in adrenal medulla of water-deprived rats.

Experiments were performed to investigate whether adrenal neuronal nitric oxide synthase (nNOS) mRNA and protein expression are responsive to alterations in body volume. Using an RT-PCR technique, the relative quantities of nNOS mRNA as well as the tyrosine hydroxylase and phenylethanolamine N-methyltransferase mRNA in the adrenals of water-deprived rats significantly increased from 12 hr to 4 days. In situ hybridization and immunohistochemical study showed that water deprivation activated nNOS mRNA and protein expression in the adrenal medulla. Four days after water deprivation, nNOS protein …

The adrenal gland of Triturus carnifex after glucagon administration.

The present work was undertaken in order to investigate the influence of endocrine pancreas on the adrenal gland of Triturus carnifex. Our experiments aimed at studying the effects of intraperitoneal injections of glucagon on ultrastructural morphological and morphometrical features of steroidogenic and chromaffin tissues, as well as serum levels of aldosterone, corticosterone, norepinephrine (NE) and epinephrine (E). With regard to steroidogenic tissue, in January and November, glucagon decreased lipid droplet content in steroidogenic cells, that showed clear signs of increased activity. More

Resistance of central phenylethanolamine-n-methyl transferase containing neurons to 6-hydroxydopamine.

The effects of the catecholamine neurotoxin 6-hydroxydopamine on phenylethanolamine-N-methyl transferase (PNMT) in rat brain has been investigated by biochemical and immunohistochemical analysis. 6-Hydroxydopamine was administerered either intracisternally to adult rats or systemically to newborn rats, treatments known to affect markedly central noradrenaline neurons. None of these treatments had any significant effect on the PNMT activity in the hypothalamus, pons-medulla and the spinal cord, as measured in vitro using a radiometric assay. The neonatal 6-hydroxydopamine treatment, which produ

The Regulation of Catecholamine Biosynthesis by the Gas Transmitters Carbon Monoxide and Hydrogen Sulfide.

The gas transmitters nitric oxide (NO), carbon monoxide (CO), and hydrogen sulfide (H2S) play important roles in physiological regulation, including adrenal function. Among them, only NO has been directly implicated in controlling catecholamine biosynthesis. This study examined whether CO and H2S exert similar effects by treating PC12 cells with a CO donor (CORM-2) or an H2S donor (NaHS), with or without glucocorticoid stimulation. Gene expression of tyrosine hydroxylase (Th), dopamine β-hydroxylase (Dbh), and phenylethanolamine N-methyltransferase (Pnmt) was assessed by RT-qPCR, and catechola…

Immunohistochemical and histochemical evidence for the presence of noradrenaline, serotonin and gamma-aminobutyric acid in chief cells of the mouse carotid body.

The immunohistochemical study revealed tyrosine hydroxylase (TH), dopamine beta-hydroxylase (DBH), phenylethanolamine N-methyltransferase (PNMT), serotonin, glutamate decarboxylase (GAD) and gamma-aminobutyric acid (GABA) immunoreactivities in the mouse carotid body. TH and DBH immunoreactivities were found in almost all chief cells and a few ganglion cells, and in relatively numerous varicose nerve fibers of the carotid body. The histofluorescence microscopy showed catecholamine fluorescence in almost all chief cells. However, no PNMT immunoreactivity was observed in the carotid body. Seroton…
