Trace Element and Phytochemical Characterization of Pinus pinaster and Pinus halepensis Bark Extracts with Protective Effects Against Phenylhydrazine-induced Hemolytic Anemia in Rats.

Anemia, characterized by decreased hemoglobin levels and impaired oxygen-carrying capacity, remains a major global public health concern. Conventional therapies, such as blood transfusions and oral iron supplementation, are often associated with adverse effects and limited absorption, highlighting the need for safer and more effective alternatives. Natural products rich in polyphenols and essential minerals have attracted increasing attention due to their multifunctional therapeutic potential. In this context, the present study is the first to investigate the anti-anemic potential of Pinus pin

Niacin enhances erythropoiesis and attenuates oxidative stress and ferroptosis through the modulation of Nrf2/HO-1/GPX4 pathway in phenylhydrazine-induced hemolytic anemia in rats.

Anemia is a public health issue affecting people of all ages worldwide. 43% of children under five in Egypt are suffering from anemia. Hemolytic anemia (HA), characterized by the premature destruction of red blood cells, is often associated with oxidative stress. Phenylhydrazine (PHZ) is a chemical substance frequently used to induce HA in experimental animal models. Studies indicate that niacin (nicotinic acid) activates antioxidant pathways through nuclear factor erythroid 2-related factor 2 (Nrf2). This study aimed to evaluate the antianemic effects of niacin in PHZ-induced hemolytic anemia

Sappanwood extract modulates hepatic structure-function in hepatomegaly and hepcidin related iron regulatory pathways in a phenylhydrazine induced hemolytic anemia rat model relevant to thalassemia.

BACKGROUND: Hepatomegaly in hemolytic disorders, including in thalassemia, is a common complication, primarily due to hemosiderosis and activation of extramedullary hematopoiesis. Globin chain imbalance leads to chronic hemolysis and ineffective erythropoiesis, which are associated with suppression of hepcidin expression. Reduced hepcidin levels may increase iron absorption and accumulation, contributing to oxidative stress and liver tissue damage. Iron chelation therapy, such as deferipron, is effective but has side effects, so safer alternatives are needed. Sappanwood (Caesalpinia sappan L.)

Cyanide inhibition of porcine kidney diamine oxidase and bovine plasma amine oxidase: evidence for multiple interaction sites.

The interactions of cyanide and phenylhydrazine with porcine kidney diamine oxidase (PKDAO) and bovine plasma amine oxidase (BPAO) (EC 1.4.3.6) have been investigated. Cyanide displays mixed noncompetitive inhibition against amine substrates and also against O2. EPR spectroscopy shows that cyanide binds to an equatorial site on Cu(II) and can be displaced by chloride, which is not an inhibitor, without recovery of activity, indicating that Cu(II)-bound cyanide is not inhibitory. 14CN-. studies have shown that one cyanide in PKDAO and two in BPAO are covalently and irreversibly bound per enzyme

[The immunometaboic effects of benfotiamine and riboxine on hemolytic anemia].

Single (80 mg/kg) or multiply repeated (30 mg/kg) intramuscular injections of phenylhydrazine decrease the functional activity of mononuclear blood cells and the reduces immunological reactivity of the organism. Benfotiamine and riboxin decrease the extent of changes in the immunological response to a single administration of phenylhydrazine but do not significantly influence the immunological functions impaired by repeated injections of the hemolytic toxin.

Quantitative relationship between Heinz body formation and red blood cell deformability.

The ultimate cause of destruction of red blood cells (RBCs) after oxidative damage with Heinz body formation is not well understood. We correlated the changes in RBC morphology and membrane protein composition after oxidant treatment with the alterations in deformability of whole cells and cell membranes. The incubation of RBCs with phenylhydrazine concentrations of 0.3 to 100 mg/dL at 37 degrees C for one hour led to a dose-dependent formation of Heinz bodies, ranging from isolated Heinz bodies at 1 mg/dL to a confluent coating of the inner membrane surface at 100 mg/dL phenylhydrazine. Morphopathological Effect of Ferrihydrite Nanoparticles in an Experimental Model of Toxic Hemolytic Anemia.

We studied the effect of biogenic ferrihydrite nanoparticles synthesized as a result of the culturing of Klebsiella oxytoca on Wistar rats with experimental toxic hemolytic anemia. The pathology was simulated by single intraperitoneal injection of phenylhydrazine hydrochloride. On day 4, the functional parameters of erythrocytes in rats corresponded to the state of toxic hemolytic anemia. It is shown that ferrihydrite nanoparticles suspension has chronic toxicity and causes morphological changes in organs (mainly in the spleen), which are characterized by accumulation of nanoparticles. A new histochemical method for detection of sialic acids using a physical development procedure.

We have established an efficient histochemical method for demonstration of sialic acids in light microscopy. The method consists of a selective periodate oxidation-phenylhydrazine blockade and a thiocarbohydrazide-silver protein sequence followed by a physical development procedure. From the results obtained by the present experimental and control studies in tissue sections from a series of rat and mouse organs, such as stomach, duodenum, colon, liver, sublingual gland, lung, and kidney, the specificity and sensitivity of the method were sufficient. In the tissues tested, sialic acids were vis

Oxidative erythrocyte membrane damage in hereditary spherocytosis.

The occurrence, in Hereditary Spherocytosis, of an oxidative damage to red blood cell membranes was studied by "in vitro" treatment of the erythrocytes with tert-butylhydroperoxide, methylene blue, or phenylhydrazine. Spherocytes were found to be more sensitive than normal erythrocytes to the action of these drugs. Tert-butylhydroperoxide caused a more intense lipid peroxidation as well as more extensive membrane protein alterations, namely spectrin degradation, formation of high molecular weight aggregates, and globin binding to the membrane. Marked spectrin degradation was also induced by me

An improved method for the inhibition of endogenous peroxidase non-deleterious to lymphocyte surface markers. Application to immunoperoxidase studies on eosinophil-rich tissue preparations.

The inhibitory effect of phenylhydrazine and azide combined with either pre-formed or nascent hydrogen peroxide H2O2 upon endogenous peroxidatic activity, expressed by tissue eosinophils in different disease states, was investigated. It was found that whilst endogenous peroxidatic activity due to eosinophils in a Hodgkin's disease and a histiocytosis X case were adequately inhibited by phenylhydrazine combines with pre-formed or nascent H2O2, the eosinophils in the Onchocerca volvulus nodule were either not at all or only partly inhibited by the two regimens. On the other hand, a combination o

Influence of some aldehyde blocking agents on staining of depurinized DNA with cationic dyes.

Rat liver, spleen and Walker carcinosarcoma imprints were subjected to depurinizing Feulgen hydrolysis and then treated with blocking agents of aldehyde groups. Such blockators as sodium bisulfite and hydroxylamine which multiplay additionally anionic groups in DNA and intensify the reactions with cationic dyes, ensuring anisotropic staining. Hydrazine lowers the binding of carionic dyes to DNA, instead phenylhydrazine, completely blocks both aldehyde and phosphate groups. When the imprints were treated with 2.4-dinitrophenylhydrazine, aldehyde and phosphate groups of apurinic acid were blocke

In vivo transferrin-iron receptor relationships in erythron of rats.

Quantitative measurements of transferrin receptors, tissue transferrin, tissue iron uptake, and erythroid cellularity have been carried out in rats with altered erythropoiesis and altered iron balance. Erythroid receptors increased with erythroid hyperplasia, with the increase in proportion to the increased number of red cell precursors in phenylhydrazine-treated rats. Receptors increased disproportionately in iron deficiency due to both erythroid hyperplasia and an increase in receptors in the individual cell. There was a ratio of 1:1 between cell-related transferrin and receptors in circulat

Effect of splenic congestion associated with hemolytic anemia on mortality of rats challenged with Haemophilus influenzae b.

The effect of splenic congestion associated with acute hemolytic anemia on susceptibility to bacterial infection was investigated in rats inoculated with Haemophilus influenzae b by intranasal (in) or intravenous (iv) challenge. The rats were made anemic by phenylhydrazine treatment and were challenged with 10(6) (in) or 5 X 10(7) (iv) H influenzae b. Forty-eight hours after in inoculation of bacteria, the number of bacteria in the blood was 10 times greater in the anemic rats with extensive splenic congestion than in controls. After iv inoculation of bacteria, a significantly (P less than 0.0

Oxidative hemoglobin breakdown induced by a rubber additive.

An antioxidant commonly present in black rubber, N-isopropyl-N'-phenyl-p-phenylenediamine (IPPD) caused very rapid oxidation and denaturation of purified hemoglobin. In the red cells, the reaction resulted in the formation of Heinz bodies, a decrease in reduced glutathione concentration, and stimulation of the hexose monophosphate shunt. IPPD appears therefore to have the potential to induce hemolysis in glucose-6-phosphate dehydrogenase deficiency. It would be expected to be more potent than most redox reagents of this type. It does not require further metabolism to be reactive, and its react

Erythropoietin in renal and hepatic lymph of conscious ewes.

Spontaneously flowing fistulae were established in the efferent lymphatics of popliteal, prescapular and prefemoral nodes and lumbar trunk or in the afferent lymphatics draining the kidney and liver of sheep. Lymph was collected from these sites over various time intervals and assayed for erythropoietin (Ep) content. The objective of the study was to establish the anatomic site(s) of Ep production. Normal lymph did not contain detectable titers of Ep, nor did renal lymph or blood plasma from a sheep systematically treated with cobaltous chloride. Renal lymph did contain measurable levels of Ep

Malaria impairs resistance to Salmonella through heme- and heme oxygenase-dependent dysfunctional granulocyte mobilization.

In sub-Saharan Africa, invasive nontyphoid Salmonella (NTS) infection is a common and often fatal complication of Plasmodium falciparum infection. Induction of heme oxygenase-1 (HO-1) mediates tolerance to the cytotoxic effects of heme during malarial hemolysis but might impair resistance to NTS by limiting production of bactericidal reactive oxygen species. We show that co-infection of mice with Plasmodium yoelii 17XNL (Py17XNL) and Salmonella enterica serovar Typhimurium 12023 (Salmonella typhimurium) causes acute, fatal bacteremia with high bacterial load, features reproduced by phenylhydra

Fused 1H-/2H-Indazole as Bioactive Privileged Scaffolds: Synthetic Strategies and Pharmaceutical Applications.

Bioheterocycles represent a pharmaceutically privileged class of compounds due to their broad spectrum of medicinal applications. Among them, indazole (benzopyrazole or isoindazole) is a fused nitrogen-containing heterocycle with significant importance in pharmaceuticals, bioactive molecules, and natural products. The indazole system exists in tautomeric forms 1H-, 2H-, and 3H-indazole, with 1H-indazole being the most prevalent and stable. Conventionally, indazoles are synthesized via the reaction of aromatic aldehydes or ketones with substituted phenylhydrazines. Naturally occurring alkaloids
