A Dual-Reporter System for the Analysis of Phloem Structural and Signaling Responses In Vivo.

Plants have evolved an effective defense mechanism in the phloem to prevent the spread of pathogens and minimize the loss of phloem sap following injury. Specific structural phloem proteins known as P-proteins rapidly seal affected sieve elements by plugging the sieve plates, a phenomenon defined as sieve element occlusion. This chapter describes a live cell imaging method for the analysis of P-protein responses and signal propagation in vivo without tissue sectioning or mechanical manipulation. It is based on an Arabidopsis thaliana dual-reporter line where P-proteins are labeled with fluores

Stationary phloem proteins and their effects on viruses, aphids, and cyst nematodes in Arabidopsis.

The phloem is a specialized tissue that facilitates systemic transport of carbohydrates and signal molecules, making it a common target for viruses, phloem-feeding insects, and cyst nematodes. In Arabidopsis, the stationary phloem-associated SIEVE ELEMENT-LINING CHAPERONE1 (SLI1) and RESTRICTED TEV MOVEMENT (RTM) proteins restrict insect phloem-feeding and potyviruses systemic transport, respectively. However, their broader roles in plant-attacker interactions remain largely unexplored. We investigated the roles of SLI1, RTM1, RTM2, and RTM3 in tobacco etch virus (TEV) infection, as well as in

Determination of the Phloem Mass Flow Velocity and Calculation of the Volumetric Flow Rate.

Here, we describe a microscopic approach to measure the mass flow velocity and to calculate the volumetric flow rate in the midrib of leaves using a phloem-mobile fluorescent dye. The method requires measurements of the carboxyfluorescein (CF) fluorescence using a fluorescent microscope and the determination of the sieve element (SE) cross-sectional area. The translocated CF within the vascular system is tracked in the leaf using cross-sections and the mass flow velocity is deduced. Additionally, SE cross-sectional areas are measured in the center of the leaf at a high resolution to calculate

Experimental Evidence for Photosynthetic Dependency of Phloem Sap Generation in Minor Veins.

Factors controlling plant photosynthesis and primary production include parameters dictating photosynthetic activity together with export and allocation properties. In fact, phloem loading activity as well as phloem sap redistribution and velocity are considered as important players in plant carbon acquisition. But surprisingly, there is almost no information about how phloem sap properties such as sugar concentration and velocity, are controlled by photosynthetic rate. Here, we carried out gas exchange experiments coupled to isotopic labelling on sunflower leaves to monitor photosynthate expo

Ectopic Vascular Induction in Arabidopsis Cotyledons for Sequential Analysis of Phloem Differentiation.

Vascular system is vital for the transport of water and nutrients as well as for providing mechanical support in many land plants. Plant transcription factors play a central role in regulating vascular development downstream of hormones and peptide signaling pathways. Particularly, cell culture systems have contributed to isolating such key transcription factors for xylem differentiation. However, there had been no efficient systems that can mimic phloem differentiation in the model plant Arabidopsis, preventing the identification of phloem-related transcription factors. We have recently estab

Systemic response to aphid infestation by Myzus persicae in the phloem of Apium graveolens.

Little is known about the molecular processes involved in the phloem response to aphid feeding. We investigated molecular responses to aphid feeding on celery (Apium graveolenscv. Dulce) plants infested with the aphid Myzus persicae, as a means of identifying changes in phloem function. We used celery as our model species as it is easy to separate the phloem from the surrounding tissues in the petioles of mature leaves of this species. We generated a total of 1187 expressed sequence tags (ESTs), corresponding to 891 non-redundant genes. We analysed these ESTs in silico after cDNA macroarray hy

Pressurised liquid extraction of arylbutanoid and diarylheptanoid glycosides from Betula pubescens phloem.

Birch phloem (inner bark) is primarily burned as waste in the pulp and paper, forestry and wood processing industries. However, phloem represents a valuable source of biologically active plant secondary metabolites. In this study, we propose new optimised approach for the pressurised liquid extraction of arylbutanoid and diarylheptanoid glycosides from birch (Betula pubescens) phloem. The influence of solvent composition, extraction temperature, extraction time and plant-to-extractant on the extraction of analytes from birch phloem by the pressurised liquid extraction was studied. The optimal

Analysis of Phloem Sterols in Plants using EDTA-Facilitated Exudation and GC-MS.

Phytosterols occur in the phloem of vascular plants as free, acylated, and glycosylated derivatives. The phloem is increasingly recognized as an active site of sterol synthesis and transport used to support plant metabolism, while also providing a potential nutritional source for phloem-feeding insects. This chapter describes a method for the collection and quantitative analysis of phloem sterols using an EDTA-facilitated exudation procedure followed by gas chromatography-mass spectrometry (GC-MS). Excised leaves are incubated in an EDTA solution to induce phloem exudation, and sterols from co

Phloem iron remodels root development in response to ammonium as the major nitrogen source.

Plants use nitrate and ammonium as major nitrogen (N) sources, each affecting root development through different mechanisms. However, the exact signaling pathways involved in root development are poorly understood. Here, we show that, in Arabidopsis thaliana, either disruption of the cell wall-localized ferroxidase LPR2 or a decrease in iron supplementation efficiently alleviates the growth inhibition of primary roots in response to NH4+ as the N source. Further study revealed that, compared with nitrate, ammonium led to excess iron accumulation in the apoplast of phloem in an LPR2-dependent m

VISUAL-PH and VISUAL-CC: Modified Culture Systems for Phloem Differentiation in Arabidopsis.

Vascular cell culture systems have served as powerful tools for identifying key regulators of cell differentiation and function. Vascular cell Induction culture System Using Arabidopsis Leaves (VISUAL) is a tissue culture system that can induce not only xylem vessel-like cells but also sieve element-like cells, thus providing a culture-based system for studying phloem cell differentiation. Presented here are two improved versions of the VISUAL assay: VISUAL-PH, which predominantly induces sieve element-like cells with high specificity, and VISUAL-CC, which enables the induction of companion ce

Phloem-exudate proteome analysis of response to insect brown plant-hopper in rice.

Brown plant-hopper (Nilaparvata lugens Stål, BPH), one of the most devastating agricultural insect pests of rice throughout Asia, ingests nutrients from rice sieve tubes and causes a dramatic yield loss. Planting resistant variety is an efficient and economical way to control this pest. Understanding the mechanisms of host resistance is extremely valuable for molecular design of resistant rice variety. Here, we used an iTRAQ-based quantitative proteomics approach to perform analysis of protein expression profiles in the phloem exudates of BPH-resistant and susceptible rice plants following BPH

Measuring Esculin Export from Leaves as Proxy for Sucrose Loading Rates in the Phloem.

Phloem loading of sucrose and the transport from source leaves to sink tissues is vital for plant growth and carbon allocation. Traditional methods to measure phloem loading are often time-consuming or require specialized equipment. Here, we introduce a rapid, cost-effective esculin-based fluorometric assay as a reliable proxy for sucrose loading. Esculin, a fluorescent coumarin glucoside, is specifically transported by sucrose transporters in plants with active apoplastic phloem loading. After application to source leaves, esculin fluorescence is measured in the extracted leaf sap, providing

The role of phloem sieve elements and laticifers in the biosynthesis and accumulation of alkaloids in opium poppy.

The benzylisoquinoline alkaloids of opium poppy, including the narcotic analgesics morphine and codeine, accumulate in the multinucleate cytoplasm of specialized laticifers that accompany vascular tissues throughout the plant. In mature opium poppy plants, immunofluorescence labeling using specific antibodies showed that four alkaloid biosynthetic enzymes, (S)-norcoclaurine 6-O-methyltransferase (6OMT), (S)-coclaurine N-methyltransferase (CNMT), (S)-3'-hydroxy-N-methylcoclaurine-4'-O-methyltransferase (4'OMT) and salutaridinol-7-O-acetyltransferase (SAT) were restricted to sieve elements of th

Phloem-specific overexpression of AtOPT6 alters glutathione, phytochelatin, and cadmium distribution in Arabidopsis thaliana.

The Arabidopsis oligopeptide transporter AtOPT6 is reportedly involved in the long-distance transport of thiol compounds into sink organs. In the present study, transgenic Arabidopsis lines overexpressing AtOPT6 under the control of a phloem-specific promoter, sucrose-proton symporter 2 (pSUC2), were analyzed for thiol and cadmium (Cd) distribution during the reproductive stage, both with and without Cd exposure. Phloem specific AtOPT6-overexpressing lines did not exhibit an evident impact on bolting time. In the absence of Cd exposure, these transgenic lines showed significantly enhanced tran

Integrated Phloem Sap mRNA and Protein Expression Analysis Reveals Phytoplasma-infection Responses in Mulberry.

To gain insight into the response of mulberry to phytoplasma-infection, the expression profiles of mRNAs and proteins in mulberry phloem sap were examined. A total of 955 unigenes and 136 proteins were found to be differentially expressed between the healthy and infected phloem sap. These differentially expressed mRNAs and proteins are involved in signaling, hormone metabolism, stress responses, etc. Interestingly, we found that both the mRNA and protein levels of the major latex protein-like 329 (MuMLPL329) gene were increased in the infected phloem saps. Expression of the MuMLPL329 gene was

The potential of silicon in crop protection against phloem feeding and chewing insect pests: a review.

As future crop production is threatened by the increasing incidence of insect pests caused by changing climate, it is crucial to focus on searching for alternative methods of insect pest management. This review summarizes the current knowledge on the effect of silicon (Si) on the incidence, growth, development, longevity, survival, feeding efficiency, and reproduction parameters of phloem sap-feeding and chewing insects of plants. While Si may negatively impact the biological parameters of both insect guilds, the effect is generally more pronounced in chewing insects and often varies under dif

Microbiome and metabolome dynamics in phloem and rhizosphere of Pinus tabuliformis against Dendroctonus valens infestation.

Microbial communities play essential roles in mediating plant defenses against insect pests. However, how host-associated microbiota and metabolites jointly respond to bark beetle infestation remains largely unexplored. Here, we integrated microbiome and metabolome profiling to elucidate how Pinus tabuliformis regulates its phloem and rhizosphere responses under varying levels of Dendroctonus valens infestation. Both bacterial and fungal diversity, as well as the relative abundance of dominant taxa such as Erwinia and Pseudoxanthomonas, shifted significantly with infestation intensity. Concurr

Suppression of host salicylic acid defense by a phloem-colonizing pathogen effector in citrus Huanglongbing.

Citrus Huanglongbing (HLB), caused by Candidatus Liberibacter asiaticus (CLas), severely threatens citrus production. Here, we identify Sec-delivered effector 10 (SDE10), a key effector from CLas, whose ectopic expression in citrus mimics HLB symptoms. Genetic and biochemical analyses demonstrate that SDE10 suppresses salicylic acid (SA)-mediated defense by inhibiting xylem cysteine peptidase 2 (CsXCP2). CsXCP2 positively regulates SA homeostasis through two activity-dependent mechanisms. First, it processes pathogenesis-related 1 (CsPR1) to generate cysteine-rich secretory proteins, antigen 5

Impact of Iron Deficiency on the Arabidopsis thaliana Phloem Sap Proteome, a Key Role for bHLH121.

Iron (Fe) is an essential micronutrient for plant growth and development whose homeostasis must be tightly regulated to avoid deficiency or excess that could be detrimental to the cells. In Arabidopsis thaliana, this mechanism is regulated by a series of transcription factors that act in an intricate regulatory network among which URI/bHLH121 (UPSTREAM REGULATOR OF IRT1) plays a predominant role. Tremendous efforts were deployed to decipher the molecular mechanisms that regulate iron homeostasis in plants. Nonetheless, the nature of the long-distance signal that conveys, via the phloem sap, in

The Small RNA Component of Arabidopsis thaliana Phloem Sap and Its Response to Iron Deficiency.

In order to discover sRNA that might function during iron deficiency stress, RNA was prepared from phloem exudates of Arabidopsis thaliana, and used for RNA-seq. Bioanalyzer results indicate that abundant RNA from phloem is small in size-less than 200 nt. Moreover, typical rRNA bands were not observed. Sequencing of eight independent phloem RNA samples indicated that tRNA-derived fragments, specifically 5' tRFs and 5' tRNA halves, are highly abundant in phloem sap, comprising about 46% of all reads. In addition, a set of miRNAs that are present in phloem sap was defined, and several miRNAs and
