Pax6 induces ectopic eyes in a vertebrate.

We report here that misexpression of the transcription factor Pax6 in the vertebrate Xenopus laevis leads to the formation of differentiated ectopic eyes. Multiple molecular markers indicated the presence of mature lens fiber cells, ganglion cells, Müller cells, photoreceptors and retinal pigment epithelial cells in a spatial arrangement similar to that of endogenous eyes. Lineage tracing experiments showed that lens, retina and retinal pigment epithelium arose as a consequence of the cell-autonomous function of Pax6. These experiments also reveal that the cell autonomous activity of misexpres

Spatial control of rhabdom shedding in the lateral eye of the American horseshoe crab, Limulus polyphemus.

Membrane leaves the rhabdom of Limulusphotoreceptors either by transient shedding at dawn or throughout the day by light-driven shedding. We examined whether the light trigger for transient shedding and the light drive for light-driven shedding are localized properties of the illuminated photoreceptors or whether they are an array property of the retina. Four experiments were conducted during which the lateral eye was exposed to one of a variety of different illumination patterns for a day, fixed, dissected and cut into serial frozen sections. Serial electron microscopic reconstruction of the drosophila larval eye: Photoreceptors with a rudimentary rhabdomere of microvillar-like processes.

Photoreceptor cells (PRCs) across the animal kingdom are characterized by a stacking of apical membranes to accommodate the high abundance of photopigment. In arthropods and many other invertebrate phyla PRC membrane stacks adopt the shape of densely packed microvilli that form a structure called rhabdomere. PRCs and surrounding accessory cells, including pigment cells and lens-forming cells, are grouped in stereotyped units, the ommatidia. In larvae of holometabolan insects, eyes (called stemmata) are reduced in terms of number and composition of ommatidia. The stemma of Drosophila (Bolwig or

Membrane protein trafficking in Drosophila photoreceptor cells.

Membrane protein trafficking occurs throughout the lifetime of neurons and includes the initial protein synthesis and anterograde transport to the plasma membrane as well as internalization, degradation, and recycling of plasma membrane proteins. Defects in protein trafficking can result in neuronal degeneration and underlie blinding diseases such as retinitis pigmentosa as well as other neuronal disorders. Drosophila photoreceptor cells have emerged as a model system for identifying the components and mechanisms involved in membrane protein trafficking in neurons. Here we summarize the curren

Inhibition of phospholipase C activity in Drosophila photoreceptors by 1,2-bis(2-aminophenoxy)ethane N,N,N',N'-tetraacetic acid (BAPTA) and di-bromo BAPTA.

In vivo light-induced and basal hydrolysis of phosphatidyl inositol 4,5-bisphosphate (PIP2) by phospholipase C (PLC) were monitored in Drosophila photoreceptors using genetically targeted PIP2-sensitive ion channels (Kir2.1) as electrophysiological biosensors for PIP2. In cells loaded via patch pipettes with varying concentrations of Ca2+ buffered by 4 mM free BAPTA, light-induced PLC activity, showed an apparent bell-shaped dependence on free Ca2+ (maximum at "100 nM", approximately 10-fold inhibition at <10nM or approximately 1 microM). However, experiments where the total BAPTA concentratio

Tachykinins: Neuropeptides That Are Ancient, Diverse, Widespread and Functionally Pleiotropic.

Tachykinins (TKs) are ancient neuropeptides present throughout the bilaterians and are, with some exceptions, characterized by a conserved FX1GX2Ramide carboxy terminus among protostomes and FXGLMamide in deuterostomes. The best-known TK is the vertebrate substance P, which in mammals, together with other TKs, has been implicated in health and disease with important roles in pain, inflammation, cancer, depressive disorder, immune system, gut function, hematopoiesis, sensory processing, and hormone regulation. The invertebrate TKs are also known to have multiple functions in the central nervous

Photolysis of caged Ca2+ facilitates and inactivates but does not directly excite light-sensitive channels in Drosophila photoreceptors.

Substantial evidence implicates the phosphoinositide cascade in invertebrate phototransduction, but the final pathway of excitation remains obscure. In order to test the hypothesis that Ca2+ is the excitatory messenger rapid concentration jumps of cytosolic Ca2+ were achieved in dissociated Drosophila photoreceptors by flash photolysis of the caged Ca2+ compounds DM-nitrophen and nitr-5. Both compounds were introduced via patch pipettes used to record whole-cell currents. Calibrations using INDO-1 and Mag-INDO-1 indicated that photolysis of DM-nitrophen (5 mM loaded with 4 mM Ca2+), raised Ca,

Synaptic high-frequency jumping synchronises vision to high-speed behaviour.

During high-speed behaviour, animals must synchronise perception and action despite rapid environmental and self-generated motion. How neural systems achieve such precision remains unclear. Here we show how the housefly (Musca domestica) maintains visual accuracy during fast motion. Using intracellular and photomechanical recordings during saccade-like stimulation, we traced information flow from photoreceptors to large monopolar cells (LMCs). Visual neurons achieved record-high information sampling (~2500 bits·s-1) and synaptic transmission (~4100 bits·s-1), far exceeding previous estimates.

Exploring dose-dependent outcomes of photobiomodulation with two infrared wavelengths applied to injured Drosophila retinae.

The use of photobiomodulation as a therapeutic tool has attracted copious research over the past several years. However, given the many variables involved in the design of a treatment regimen, such as wavelength, irradiance, exposure duration, number of treatments, and total treatment period, many studies are inherently not comparable as they often differ in at least one of these parameters. Here, we assess dose and wavelength dependency of photobiomodulation in a model of light-induced retinal damage in Drosophila melanogaster. Light-damaged Drosophila were exposed to either 850 or 950 nm lig

Graded opsin co-expression along the butterfly retina fine tunes the spectral sensitivity of a colour-opponent cell across the visual field.

Compound eyes deliver a vast stream of information to the tiny insect brains. To maximize the information content and minimize the redundancy of neural signals, insect eyes are built so to encode the relevant and filter out the unimportant elements of the visual environment. Terrestrial habitats have a predictable spatio-spectral structure, which can be matched by the distribution of photoreceptors with different spectral sensitivities across the retina. Here, we investigate the retinal organization of the nymphalid butterfly Heliconius melpomene using single-cell recordings, immunohistochemis

Structures and molecules involved in generation and regulation of biological rhythms in vertebrates and invertebrates.

Melatonin from the retina and the pineal gland functions in neuroendocrine hierarchies. Photoreceptors--eyes and extraretinal--detect light. Oscillators--pineal and suprachiasmatic nuclei--act as pacemakers. Driven neuroendocrine rhythms carry temporal hormone signals throughout the body. Light controls melatonin: light sets the phase of the melatonin rhythm and determines the duration of melatonin synthesis. By these means, circadian rhythms (e.g., in locomotor activity and body temperature) and seasonal rhythms (e.g., in reproduction) are controlled.

Cryptochrome interaction networks across different tissues in Drosophila melanogaster.

BACKGROUND: Drosophila CRYPTOCHROME (dCRY) is a blue light-sensitive protein involved in various biological processes, including photoreception, circadian rhythm regulation, synaptic plasticity, and magnetoreception. Its circadian function is strictly connected with light: upon light exposure, dCRY undergoes a conformational change, becoming active and binding to various proteins. However, it can also form complexes in the absence of light, with its function varying depending on the cell type in which it is expressed. RESULTS: Here, we use an experimental approach based on co-immunoprecipitati

Glucose uptake in pigment glia suppresses Tau-induced inflammation and photoreceptor degeneration.

Brain inflammation contributes to the pathogenesis of neurodegenerative diseases, such as Alzheimer's disease (AD). Glucose hypometabolism and glial activation are pathological features seen in AD brains; however, the connection between the two is not fully understood. Using a Drosophila model of AD, we identified that glucose metabolism in glia plays a critical role in neuroinflammation under disease conditions. Expression of human MATP (hereafter referred to as Tau) in the retinal cells, including photoreceptor neurons and pigment glia, causes photoreceptor degeneration accompanied by the fo

Cell fate ratios are encoded by transcriptional dynamics in the Drosophila retina.

Although transcription happens in bursts, it is unclear whether variation in the rate and pattern of bursting matters during animal development. We examined whether the amount and timing of transcription influence the ratio of cell types produced during stochastic patterning of the Drosophila retina. This system is balanced between 2 outcomes: ∼70% of R7 photoreceptors (PRs) express Rhodopsin 4 (Rh4), and the rest express Rhodopsin 3 (Rh3). Cell fate depends on the cell-intrinsic binary decision to express the transcription factor spineless (Ss). Changing the ss bursting pattern by replacing t

Regulated Alternative Splicing of Drosophila Dscam2 Is Necessary for Attaining the Appropriate Number of Photoreceptor Synapses.

How the brain makes trillions of synaptic connections using a genome of only 20,000 genes is a major question in modern neuroscience. Alternative splicing is one mechanism that can increase the number of proteins produced by each gene, but its role in regulating synapse formation is poorly understood. In Drosophila, photoreceptors form a synapse with multiple postsynaptic elements including lamina neurons L1 and L2. L1 and L2 express distinct isoforms of the homophilic repulsive protein Dscam2, and since these isoforms cannot bind to each other, cell-specific expression has been proposed to be

OTX2 and CRX rescue overlapping and photoreceptor-specific functions in the Drosophila eye.

BACKGROUND: Otd-related transcription factors are evolutionarily conserved to control anterior patterning and neurogenesis. In humans, two such factors, OTX2 and CRX, are expressed in all photoreceptors from early specification through adulthood and associate with several photoreceptor-specific retinopathies. It is not well understood how these factors function independently vs. redundantly, or how specific mutations lead to different disease outcomes. It is also unclear how OTX1 and OTX2 functionally overlap during other aspects of neurogenesis and ocular development. Drosophila encodes a sin

Chickens' Cry2: molecular analysis of an avian cryptochrome in retinal and pineal photoreceptors.

We have identified and characterized an ortholog of the putative mammalian clock gene cryptochrome 2 (Cry2) in the chicken, Gallus domesticus. Northern blot analysis of gCry2 mRNA indicates widespread distribution in central nervous and peripheral tissues, with very high expression in pineal and retina. In situ hybridization of chick brain and retina reveals expression in photoreceptors and in visual and circadian system structures. Expression is rhythmic; mRNA levels predominate in late subjective night. The present data suggests that gCry2 is a candidate avian clock gene and/or photopigment

Homeobox genes in the rodent pineal gland: roles in development and phenotype maintenance.

The pineal gland is a neuroendocrine gland responsible for nocturnal synthesis of melatonin. During early development of the rodent pineal gland from the roof of the diencephalon, homeobox genes of the orthodenticle homeobox (Otx)- and paired box (Pax)-families are expressed and are essential for normal pineal development consistent with the well-established role that homeobox genes play in developmental processes. However, the pineal gland appears to be unusual because strong homeobox gene expression persists in the pineal gland of the adult brain. Accordingly, in addition to developmental fu

Blue light induces a neuroprotective gene expression program in Drosophila photoreceptors.

BACKGROUND: Light exposure induces oxidative stress, which contributes to ocular diseases of aging. Blue light provides a model for light-induced oxidative stress, lipid peroxidation and retinal degeneration in Drosophila melanogaster. In contrast to mature adults, which undergo retinal degeneration when exposed to prolonged blue light, newly-eclosed flies are resistant to blue light-induced retinal degeneration. Here, we sought to characterize the gene expression programs induced by blue light in flies of different ages to identify neuroprotective pathways utilized by photoreceptors to cope w

Immunolocalization of Drosophila eye-specific diacylgylcerol kinase, rdgA, which is essential for the maintenance of the photoreceptor.

The Drosophila retinal degeneration A (rdgA) mutant has photoreceptor cells that degenerate within a week after eclosion. The degeneration starts with the disruption of the subrhabdomeric cisternae (SRC), which are the organelles essential for the transport of phospholipids to the photoreceptive membranes. Our previous biochemical and molecular studies suggested that the rdgA gene encodes an eye-specific diacylglycerol kinase (DGK). In this study, we show that retinal degeneration is prevented by the introduction of the eye-DGK gene in the rdgA mutant genome, suggesting that the DGK activity i
