Nanomaterials at the forefront of antimicrobial therapy by photodynamic and photothermal strategies.

In the face of the increasing resistance of microorganisms to traditional antibiotics, the development of innovative treatment methods is becoming increasingly urgent. Nanophototherapy technology can precisely target the infected area and achieve synergistic antibacterial effects in multiple modes. This phototherapy method has shown significant efficacy in treating diseases caused by drug-resistant bacteria, especially in the elimination of biofilms, where it has demonstrated strong dissolution capabilities. PTT utilizes photothermal agents to convert near-infrared light into heat, effectively

Advances in Hollow Inorganic Nanomedicines for Photothermal-Based Therapies.

Nanotechnology has prompted the development of hollow inorganic nanomedicine. These medicines are now widely investigated as photothermal-based therapies for various diseases due to their high loading capacity, tuneable wavelength, relatively small size and low density. We begin this review with a brief introduction, followed by a summary of the development of imaging-guided photothermal therapy (PTT) for cancer treatment during the last three years (from 2017 to 2020). We then introduce the antibacterial effects of these medicines on some bacterial infections, in which the pathogenic bacteria

Photothermal Therapy for the Treatment of Glioblastoma: Potential and Preclinical Challenges.

Glioblastoma (GBM) is a very aggressive primary malignant brain tumor and finding effective therapies is a pharmaceutical challenge and an unmet medical need. Photothermal therapy may be a promising strategy for the treatment of GBM, as it allows the destruction of the tumor using heat as a non-chemical treatment for disease bypassing the GBM heterogeneity limitations, conventional drug resistance mechanisms and side effects on peripheral healthy tissues. However, its development is hampered by the distinctive features of this tumor. Photoabsorbing agents such as nanoparticles need to reach th

Dual-Functional Microreactor Enabling Hydrogen Therapy and Mild Photothermal Therapy for Osteoarthritis Treatment.

Osteoarthritis (OA) is a prevalent degenerative joint disorder. Current therapeutic approaches have yielded suboptimal clinical outcomes, primarily due to their inability to effectively eliminate reactive oxygen species (ROS), which are the key drivers of inflammatory pathology. Molecular hydrogen (H2) has emerged as a promising therapeutic agent owing to its selective scavenging of cytotoxic ROS, thereby exerting potent anti-inflammatory effects. In this study, we propose a spatiotemporally controlled, light-activated strategy for on-demand hydrogen generation to treat OA. To realize this, we

Recent Progress of Copper-Based Nanomaterials in Tumor-Targeted Photothermal Therapy/Photodynamic Therapy.

Nanotechnology, an emerging and promising therapeutic tool, may improve the effectiveness of phototherapy (PT) in antitumor therapy because of the development of nanomaterials (NMs) with light-absorbing properties. The tumor-targeted PTs, such as photothermal therapy (PTT) and photodynamic therapy (PDT), transform light energy into heat and produce reactive oxygen species (ROS) that accumulate at the tumor site. The increase in ROS levels induces oxidative stress (OS) during carcinogenesis and disease development. Because of the localized surface plasmon resonance (LSPR) feature of copper (Cu)

Dual functions of gold nanorods as photothermal agent and autofluorescence enhancer to track cell death during plasmonic photothermal therapy.

Gold nanorods have the potential to localize the treatment procedure by hyperthermia and influence the fluorescence. The longitudinal plasmon peak contributes to the photothermal effect by converting light to heat. When these nanorods are PEGylated, it not only makes it biocompatible but also acts as a spacer layer during fluorescence enhancement. When the PEGylated nanorods are internalized inside the cells through endocytosis, the transverse plasmonic peak combined with the enhanced absorption and scattering properties of the nanorods can enhance the autofluorescence emission intensity from

Nicotinamide-derived tumor-targeting carbon dots for Cancer Photothermal therapy.

The lack of tumor-specific targeting remains a major bottleneck in photothermal therapy. Here, we exploit the natural affinity of nicotinamide (NAM) for Ki67, a tumor proliferation marker, to design a targeted photothermal nanomaterial. NAM-derived carbon dots (NAM-CDs) were synthesized via a one-step solvothermal method, in which the carbon core serves as a scaffold for the multivalent presentation of NAM moieties. Dynamic molecular docking simulations revealed that NAM-CDs exhibited a markedly enhanced binding affinity toward Ki67, with binding energy of -7.7 kcal/molcompared with -3.8 kcal/

Preclinical advance in nanoliposome-mediated photothermal therapy in liver cancer.

Liver cancer is a highly lethal malignant tumor with a high incidence worldwide. Therefore, its treatment has long been a focus of medical research. Although traditional treatment methods such as surgery, radiotherapy, and chemotherapy have increased the survival rate of patients, their efficacy remains unsatisfactory owing to the nonspecific distribution of drugs, high toxicity, and drug resistance of tumor tissues. In recent years, the application of nanotechnology in the medical field has opened a new avenue for the treatment of liver cancer. Among these treatment methods, photothermal ther

Anticancer effects of DNA-functionalized gold Nanoparticle-Mediated photothermal therapy.

Vascular endothelial growth factor (VEGF)-targeted therapies are promising in inhibiting tumor angiogenesis but often face limitations due to resistance and incomplete efficacy. Combining VEGF inhibition with photothermal therapy (PTT), which uses nanoparticles such as gold nanoparticles to generate heat upon light exposure, offers a potential solution by targeting both the tumor vasculature and the tumor cells directly. However, the molecular mechanisms driving the synergy between these treatments remain unclear. In this study we employed quantitative proteomics to investigate the molecular a

Ultrasound-Driven Nitric Oxide Generation for Enhanced Sonodynamic-Photothermal Therapy.

Recently, green gas therapy based on nitric oxide (NO) has gained considerable attention in cancer treatment. The supplementation of exogenous NO and its controlled release represent promising strategies for adjuvant tumor therapy. In this study, we developed a novel ultrasound (US)-triggered NO generation and release nanoplatform that integrates NO therapy, sonodynamic therapy, and photothermal therapy (PTT) into a collaborative therapeutic modality. An environmentally friendly biomacromolecule, polydopamine, was employed to coload chlorin e6 (Ce6) and NO donor (BNN6), resulting in the nanoco

Apoptotic bodies encapsulating Ti2N nanosheets for synergistic chemo-photothermal therapy.

Extracellular vesicles (EVs) have great potential in oncology drug delivery because of their unique biological origin. Apoptotic bodies (ABs), as a member of the EV family, offer distinct advantages in terms of size, availability and membrane properties, but have been neglected for a long time. Here, using ABs and Ti2N nanosheets, we propose a novel drug delivery system (Ti2N-DOX@ABs), which exhibit a homologous targeting ability for dual-strategy tumor therapy with intrinsic biological property. The experimental results demonstrate that such a drug delivery system possesses a drug loading cap

Gas-Mediated Tumor Energy Remodeling for Sensitizing Mild Photothermal Therapy.

The metabolic reprogramming of tumors requires high levels of adenosine triphosphate (ATP) to maintain therapeutic resistance, posing a major challenge for photothermal therapy (PTT). Although raising the temperature helps in tumor ablation, it frequently leads to severe side effects. Therefore, improving the therapeutic response and promoting healing are critical considerations in the development of PTT. Here, we proposed a gas-mediated energy remodeling strategy to improve mild PTT efficacy while minimizing side effects. In the proof-of-concept study, a Food and Drug Administration (FDA)-app

Graphene family nanomaterials for application in cancer combination photothermal therapy.

Combining hyperthermia with other therapies holds a great potential for improving cancer treatment. In this approach, the increase in the body temperature can exert a therapeutic effect on cells and/or enhance the effectiveness of anticancer agents. However, the conventional methodologies available to induce hyperthermia cannot confine a high temperature increase to the tumor-site while maintaining healthy tissues unexposed and ensuring minimal invasiveness. To overcome these limitations, combination photothermal therapy (PTT) mediated by graphene family nanomaterials (GFN) has been showing pr

Photothermal-Responsive Soluble Microneedle Patches for Meibomian Gland Dysfunction Therapy.

Meibomian gland dysfunction (MGD) is a leading cause of evaporative dry eye disease, presenting a challenge for targeted treatment. Traditional topical ocular drug delivery methods often fail to effectively reach the meibomian glands (MGs). To address this, the study has developed a soluble microneedles (MN) patch comprising poly(vinyl alcohol), cyclodextrin modified polyacrylic acid, and new indocyanine green. This innovative MN patch facilitates the transdermal release of peroxisome proliferator-activated receptor gamma (PPAR-γ) agonists, such as rosiglitazone in response to near-infrared ra

Synthetic carbon-based lanthanide upconversion nanoparticles for enhanced photothermal therapy.

Photothermal therapy (PTT), a vanguard strategy in cancer/ocular neovascularization treatment, has attracted considerable attention owing to its precision, controllability, high efficacy, and minimal side effects. Nevertheless, its inherent limitations necessitate innovative solutions. One promising strategy is to develop reagents with enhanced photothermal conversion efficiency under long-wavelength laser irradiation. Carbon nanomaterials, known for their broad absorption spectra, are currently hindered by single-wavelength lasers in clinical treatments. In this study, we address this limitat

Mild Photothermal Therapy Prevents Posterior Capsule Opacification through Cytoskeletal Remodeling.

Cataract is the first leading cause of blindness in the world and posterior capsule opacification (PCO) is the most common long-term complication after surgery. The primary pathogenic processes contributing to PCO are the proliferation and migration of residual lens epithelial cells (LECs). This study aimed to explore the mild photothermal effect on LECs. Interestingly, this work finds that the mild photothermal effect significantly inhibited the proliferation and migration of LECs. The live cell fluorescence imaging reveals that the remodeling of the actin cytoskeleton and cell morphology att

Photothermal Nanoheaters-Modified Spores for Safe and Controllable Antitumor Therapy.

INTRODUCTION: To present a safer tumor therapy based on bacteria and identify in detail how the activation and infection behavior of spores can be controlled remotely by near-infrared light (NIR-irradiation) based on nanoheaters' modification. METHODS: Spores bring a better tolerance to surface modification. Transitive gold-nanorods-allied-nanoclusters-modified spores (Spore@NRs/NCs) were constructed by covalent glutaraldehyde crosslink. The photothermal properties of nanoheaters before and after attachment to spores were studied by recording temperature-irradiation time curves. The controlle
