A Rare Recombination in the Phytophthora sojae Genome Allows the Redefinition of a Major Avirulence Gene.

Significant yield losses in soybean production are imputable to Phytophthora root and stem rot (PRR) caused by Phytophthora sojae. Resistance to PRR relies on resistance genes (Rps) that recognize key effectors from the pathogen, which are encoded by avirulence genes (Avr). Key Avr haplotypes can be monitored in natural P. sojae populations and exploited to predict which resistance gene can be deployed to provide protection to the soybean crop. In this study, we reassessed the identity of the avirulence gene whose protein product is recognized by Rps6. Following extensive soil sampling and sin

Performance of a tetracycline-responsive transactivator system for regulating transgenes in the oomycete Phytophthora infestans.

The oomycete genus Phytophthora includes many important plant pathogens for which extensive genome data exist, but lacking is an inducible expression system to study contributions of their genes to growth and pathogenicity. Here the adaptation of the reverse tetracycline transactivator (rtTA) system to P. infestans is described. Vectors were developed containing rtTA expressed from an oomycete promoter, and beta-glucuronidase (GUS) controlled by TetR binding sites fused to a minimal oomycete promoter. Transformants were obtained in which GUS was expressed in a dose-dependent manner by the rtTA

A novel method for extraction of high purity and high production Phytophthora sojae oospores.

BACKGROUND: Phytophthora sojae, a soil-borne oomycete pathogen, has been a yield limiting factor for more than 60 years on soybean. The resurgence of P. sojae (Phytophthora sojae) is primarily ascribed to the durable oospores found in soil and remnants of the disease. P. sojae is capable of infesting at any growth periods of the soybean, and the succeed infestation of P. sojae is predominantly attributed to long-lived oospores present in soil. Comprehending the molecular mechanisms that drive oospores formation and their significance in infestation is the key for effective management of the di

Chloroplasts alter their morphology and accumulate at the pathogen interface during infection by Phytophthora infestans.

Upon immune activation, chloroplasts switch off photosynthesis, produce antimicrobial compounds and associate with the nucleus through tubular extensions called stromules. Although it is well established that chloroplasts alter their position in response to light, little is known about the dynamics of chloroplast movement in response to pathogen attack. Here, we report that during infection with the Irish potato famine pathogen Phytophthora infestans, chloroplasts accumulate at the pathogen interface, associating with the specialized membrane that engulfs the pathogen haustorium. The chemical

Contribution of Hydrogen Cyanide to the Antagonistic Activity of Pseudomonas Strains Against Phytophthora infestans.

Plants face many biotic and abiotic challenges in nature; one of them is attack by disease-causing microbes. Phytophthora infestans, the causal agent of late blight is one of the most prominent pathogens of the potato responsible for multi-billion-dollar losses every year. We have previously reported that potato-associated Pseudomonas strains inhibited P. infestans at various developmental stages. A comparative genomics approach identified several factors putatively involved in this anti-oomycete activity, among which was the production of hydrogen cyanide (HCN). Here, we report the relative c

Evaluation of New Oomycota Fungicides for Management of Phytophthora Root Rot of Citrus in California.

Phytophthora root rot, caused by several species of Phytophthora, is an important disease of citrus in California and other growing regions. For chemical management, mefenoxam and potassium phosphite have been available for many years, and resistance in Phytophthora spp. has been reported for both compounds. We evaluated the efficacy of the new Oomycota fungicides ethaboxam, fluopicolide, mandipropamid, and oxathiapiprolin, each with a different mode of action, against Phytophthora root rot of citrus in field and greenhouse studies. Root balls of navel orange trees on 'Carrizo citrange' rootst

Identification of new polymorphic microsatellite markers in the NA1 and NA2 lineages of Phytophthora ramorum.

Phytophthora ramorum is a recently introduced pathogen in Europe and North America consisting of three clonal lineages. Due to the limited intralineage genetic variation, only a few polymorphic markers are available for use in studies involving the epidemiology and evolution of P. ramorum. A total of 159 primer pairs for candidate polymorphic SSR loci were tested with universal labeling. Four polymorphic microsatellite loci were identified within the NA1 lineage and one within the NA2 lineage, demonstrating the power and flexibility of the screening technique. The markers may significantly inc

TiO2-Ni Hybrid Suspension as Visible-Light-Responsive Antifungal Agents for Controlling Phytophthora palmivora.

Black pod disease caused by Phytophthora palmivora (P. palmivora) remains a major constraint in global cocoa production, leading to yie ld losses of up to 40 % and increased resistance to synthetic fungicides. In this study, a novel visible-light-responsive TiO2-Ni hybrid suspension system was synthesized via a modified sol-gel method and evaluated as an eco-friendly antifungal agent specifically against P. palmivora. Structural, morphological, and compositional analyses using FTIR, XRD, and SEM-EDX confirmed the successful incorporation of Ni into the TiO2 lattice, while UV-Vis DRS revealed a

Levan induced the resistance of tomato against Botrytis cinerea and pepper against Phytophthora capsici.

Pathogenic challenges constrain plant growth, and the application of pesticides remains the primary approach to control plant diseases worldwide. However, overuse of chemical fungicides can lead to pathogen resistance and environmental contamination. In recent years, compounds that act as induced resistance stimuli have gained increasing attention. Levan, a β-2,6-linked fructan, has been reported to induce disease resistance in several plant species, though its effect on solanaceous plants and the underlying defense mechanisms remain unclear. In this study, foliar application of levan reduced

Editing miR482b and miR482c Simultaneously by CRISPR/Cas9 Enhanced Tomato Resistance to Phytophthora infestans.

Late blight, caused by Phytophthora infestans, is severely damaging to the global tomato industry. Micro-RNAs (miRNAs) have been widely demonstrated to play vital roles in plant resistance by repressing their target genes. Recently, the clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) method has been continuously improved and extensively applied to edit plant genomes. However, editing multiplex miRNAs by CRISPR/Cas9 in tomato has not been studied yet. We knocked out miR482b and miR482c simultaneously in tomato through the multiplex CRISPR/Ca

Copper ions suppress abscisic acid biosynthesis to enhance defence against Phytophthora infestans in potato.

Copper-based antimicrobial compounds are widely and historically used to control plant diseases, such as late blight caused by Phytophthora infestans, which seriously affects the yield and quality of potato. We previously identified that copper ion (Cu2+ ) acts as an extremely sensitive elicitor to induce ethylene (ET)-dependent immunity in Arabidopsis. Here, we found that Cu2+ induces the defence response to P. infestans in potato. Cu2+ suppresses the transcription of the abscisic acid (ABA) biosynthetic genes StABA1 and StNCED1, resulting in decreased ABA content. Treatment with ABA or inhib

Phytophthora infestans RXLR effector PITG20303 targets a potato MKK1 protein to suppress plant immunity.

Pathogens secret a plethora of effectors into the host cell to modulate plant immunity. Analysing the role of effectors in altering the function of their host target proteins will reveal critical components of the plant immune system. Here we show that Phytophthora infestans RXLR effector PITG20303, a virulent variant of AVRblb2 (PITG20300) that escapes recognition by the resistance protein Rpi-blb2, suppresses PAMP-triggered immunity (PTI) and promotes pathogen colonization by targeting and stabilizing a potato MAPK cascade protein, StMKK1. Both PITG20300 and PITG20303 target StMKK1, as confi

Molecular Basis of Citrus sunki Susceptibility and Poncirus trifoliata Resistance Upon Phytophthora parasitica Attack.

Coevolution has shaped the molecular basis of an extensive number of defense mechanisms in plant-pathogen interactions. Phytophthora parasitica, a hemibiothrophic oomycete pathogen and the causal agent of citrus root rot and gummosis, interacts differently with Citrus sunki and Poncirus trifoliata, two commonly favored citrus rootstocks that are recognized as susceptible and resistant, respectively, to P. parasitica. The molecular core of these interactions remains elusive. Here, we provide evidence on the defense strategies employed by both susceptible and resistant citrus rootstocks, in para

Potato foliar infection with Phytophthora infestans drives strong, cultivar-specific shifts in rhizosphere communities.

BACKGROUND: Potato is an important crop worldwide, yet its production is severely threatened by Phytophthora infestans, the causal agent of late blight. Alternatives to the current control strategies are needed, as these rely heavily on environmentally harmful treatments. The recruitment of beneficial microbes by plants upon stress ("cry-for-help" mechanism) may represent an opportunity to find new biocontrol agents but this has not yet been reported for potato. The aim of this study was to analyse whether foliar late blight infection induces shifts in the phyllosphere, rhizosphere and soil ba

Inhibition of Phytophthora infestans chitin synthase via cyclic peptide targeting for sustainable disease control.

Phytophthora spp. are devastating plant pathogens causing significant economic losses in agriculture, but unlike true fungi, oomycetes have cell walls composed predominantly of cellulose and glucans, with little to no detectable chitin. Despite this, one to two putative chitin synthase (Chs) genes are present in Phytophthora spp. suggesting a functional role in its biology. In this study, we characterized the single chitin synthase of P. infestans (PiChs) to better understand its function and explore its potential as a target for novel fungicidal treatments. Unlike typical chitin synthases tha

Tactical Diagnostic Strategies at Native California Ornamental Nurseries for Preventing Phytophthora Havocs in Wildlands.

Native plant restoration efforts in California are continuously challenged by Phytophthora-induced diseases, particularly through nursery introductions, resulting in significant losses and distribution of pathogens into restoration areas. Effective diagnostic strategies can help manage Phytophthora, yet determining the most suitable method, considering specificity, sampling time, resource availability, and expertise, remains uncertain. This study compares the effectiveness of five diagnostic methods: irrigation leachate baiting, ImmunoStrip® for Phytophthora, culture-based isolations, recombin

The limit of detection of a Phytophthora agathidicida LAMP assay is higher under alternative laboratory conditions.

Surveillance of Phytophthora agathidicida , the causal agent of kauri dieback, has included soil baiting followed by loop-mediated isothermal amplification (LAMP)-based detection. This approved assay was reported to detect 1 fg of DNA using OptiGene Isothermal Master Mix on a BioRanger instrument. We evaluated its analytical robustness using the same master mix on an alternative platform (QuantStudio 3), and separately tested an alternative NEB master mix. Both assays yielded a reliable detection limit of 1,000 fg, with variable detection at 100 fg and none below. Diagnostic laboratories shoul

Structure guided development of phenoxyethanol based N-aryloxyethyl heterocycles for the control of Phytophthora infestans.

BACKGROUND: Late blight caused by the oomycete Phytophthora infestans remains one of the most devastating diseases threatening global potato and tomato production. Overuse of a narrow range of fungicides has sped up resistance, making it urgent to develop new agents that work differently and are safer. RESULTS: In this study, a series of N-aryloxyethyl heterocycles were rationally designed by integrating the aryloxyethyl scaffold of phenoxyethanol with bioactive heterocyclic frameworks. Among the 40 synthesized derivatives, compound 8c exhibited the highest anti-oomycete activity against P. i

Inhibition mechanism of pectin-modified nano-selenium on Phytophthora capsici and the improvement of the resistance in pepper.

The principal disease impacting pepper production is Phytophthora blight, which nano‑selenium (nano-Se) controls well but has high storage requirements and low heat resistance. Varying pectin amounts yielded pectin-modified nano-Se (nano-Se@PT) with 0.1 %, 0.5 %, 1 %, and 2 % concentration. As pectin content grew, nano-Se@PT particle size reduced, zeta potential value rose, and thermogravimetric temperature climbed. The incorporation of nano-Se@PT can improve its stability and thermal resistance at 40 °C and 70 °C. Nano-Se@PT outperformed nano-Se in fungicidal and Phytophthora capsici inhibiti
