First Report of Witches'-Broom Disease of Broussonetia papyrifera and Its Association with a Phytoplasma of Elm Yellows Group (16SrV).

Broussonetia papyrifera, commonly known as paper mulberry, is an ornamental tree that is native to northeastern Asia. Because of its fast-growing nature and tolerance of dust, smoke, and high temperatures, paper mulberry is an important component of the biological diversity in natural ecosystems as well as a favorable shade tree in the region. In September of 2003, a disease characterized by pronounced witches'-broom symptoms was observed in paper mulberry trees growing near a jujube (Ziziphus jujuba) orchard and in home gardens located in Taian, Shandong, China. The diseased trees developed d

Rice Orange Leaf Phytoplasma Imp Protein Blocks G-M Interaction by Binding Glycoprotein G to Suppress Rice Stripe Mosaic Virus Infection.

Plants frequently encounter multiple pathogens concurrently, yet the molecular bases of pathogen-pathogen interactions during mixed infections remain largely unresolved. Here we show that co-infection of rice by Rice orange leaf phytoplasma (ROLP) and Rice stripe mosaic virus (RSMV) markedly suppresses RSMV proliferation in both host plants and the insect vector. Electron microscopy and confocal imaging reveal that ROLP and RSMV co-inhabit individual rice phloem cells and undergo close membrane interaction, producing swollen RSMV virions with reduced infectivity. We further demonstrate that th

Molecular Variation and Phylogeny of Thymidylate Kinase Genes of Candidatus Phytoplasma ziziphi from Different Resistant and Susceptible Jujube Cultivars in China.

The thymidylate kinase (tmk) gene is indispensable for the proliferation and survival of phytoplasma. To reveal the molecular variation and phylogeny of the tmk genes of Candidatus phytoplasma ziziphi, in this study, the tmk genes of 50 phytoplasma strains infecting different resistant and susceptible jujube cultivars from different regions in China were amplified and analyzed. Two sequence types, tmk-x and tmk-y, were identified using clone-based sequencing. The JWB phytoplasma strains were classified into three types, type-X, type-Y, and type-XY, based on the sequencing chromatograms of the

Complete nucleotide sequence of the S10-spc operon of phytoplasma: gene organization and genetic code resemble those of Bacillus subtilis.

An 11.4-kbp region of genomic DNA containing the complete S10-spc operon was constructed by an integrative mapping technique with eight plasmid vectors carrying ribosomal protein sequences from onion yellows phytoplasma. Southern hybridization analysis indicated that phytoplasmal S10-spc is a single-copy operon. This is the first complete S10-spc operon of a phytoplasma to be reported, although only a part of six serial genes of the S10 operon is reported previously. The operon has a context of 5'-rps10, rpl3, rpl4, rpl23, rpl2, rps19, rpl22, rps3, rpl16, rpl29, rps17, rpl14, rpl24, rpl5, rps1

First Report of Witches'-Broom Disease in a Cannabis spp. in China and Its Association with a Phytoplasma of Elm Yellows Group (16SrV).

Hemp fiber plants (Cannabis spp.) spread naturally in almost every climate zone in China and have a long history of cultivation in the country (1). While hemp stalks provide high-quality fibers for making ropes, clothes, and paper products, hemp seeds are a rich source of edible oil. During the summer of 2004, a disease characterized by witches'-broom symptoms was observed in wild hemp fiber plants growing in suburban Taian, Shandong, China. The diseased plants developed clusters of highly proliferating branches with much shortened internodes and leaves on the affected branches were significan

First Report of Witches'-Broom Disease of Chinese Wingnut in China and its Association with a Phytoplasma of Aster Yellows Group (16SrI).

Pterocarya stenoptera C. DC., commonly known as Chinese wingnut, is a fast-growing deciduous tree with tough bark and attractive foliage. Because of its tolerance of compact and nutritionally poor soil, drought, and heat, Chinese wingnut is an important component of the biological diversity in natural ecosystems and is a favorable shade tree in China. Chinese wingnut has also been used as a rootstock for walnuts because of its high resistance to soilborne Phytophthora spp. In the spring of 2004, a disease characterized by witches'-broom symptoms was observed affecting Chinese wingnut trees gro

Flavescence dorée phytoplasma infection represses the transcription of the lignin pathway in Vitis vinifera cv. Pinot noir without compromising monolignol accumulation.

Flavescence dorée phytoplasma (FDp) impairs shoot lignification in grapevines, but the underlying molecular mechanisms are unclear. This study examined how FDp infection affects the lignin/monolignol pathway in Vitis vinifera cv. Pinot Noir. Diseased (FDp positive) and healthy (FDp negative) shoots were sampled at three time points during the growing season. Metabolites were quantified by LC-MS/MS, key genes were analysed by RT-qPCR, and RNA sequencing was used to profile selected samples. FDp infection was found to reduce the expression of several lignin-biosynthetic genes and to broadly down

Phytoplasma Effector SJP8 Suppresses Host Immunity by Promoting the Degradation of ZjMYB15 and ZjMYB86-like to Perturb Jasmonic Acid and Hydrogen Peroxide Homeostasis in Jujube.

'Candidatus Phytoplasma ziziphi' infection causes severe damage to jujube (Ziziphus jujuba), inducing symptoms such as branch clustering, dwarfism, chlorosis and phyllody, leading to significant economic losses. However, the pathogenic mechanisms underlying jujube witches'-broom (JWB) disease remain largely unknown. In this study, we identified SJP8 as a key secreted effector protein of 'Ca. P. ziziphi'. Overexpression of SJP8 in jujube recapitulated the characteristic JWB-like symptoms, including dwarfism and leaf chlorosis. Transcriptome analysis revealed that SJP8 suppressed host immunity b

Dual biotic stressors shape volatile organic compound emission patterns in pome fruit trees.

Plants release volatile organic compounds (VOCs) as part of their defense mechanisms when attacked by phytopathogens and herbivores. These specific volatile emissions can help plants avoid damage. In this study, we investigated the effects of the brown marmorated stink bug (BMSB), Halyomorpha halys, and the phytopathogens "Candidatus phytoplasma mali", causing apple proliferation (AP) or "Ca. P. pyri", causing pear decline (PD) on the volatile emissions from apple (Malus domestica) or pear (Pyrus communis) trees. We analyzed VOCs emitted by uninfected and infected trees exposed to male and fem

A Bacterial Parasite Effector Mediates Insect Vector Attraction in Host Plants Independently of Developmental Changes.

Parasites can take over their hosts and trigger dramatic changes in host appearance and behavior that are typically interpreted as extended phenotypes that promote parasite survival and fitness. For example, Toxoplasma gondii is thought to manipulate the behaviors of infected rodents to aid transmission to cats and parasitic trematodes of the genus Ribeiroia alter limb development in their amphibian hosts to facilitate predation of the latter by birds. Plant parasites and pathogens also reprogram host development and morphology. However, whereas some parasite-induced morphological alterations

A unique virulence factor for proliferation and dwarfism in plants identified from a phytopathogenic bacterium.

One of the most important themes in agricultural science is the identification of virulence factors involved in plant disease. Here, we show that a single virulence factor, tengu-su inducer (TENGU), induces witches' broom and dwarfism and is a small secreted protein of the plant-pathogenic bacterium, phytoplasma. When tengu was expressed in Nicotiana benthamiana plants, these plants showed symptoms of witches' broom and dwarfism, which are typical of phytoplasma infection. Transgenic Arabidopsis thaliana lines expressing tengu exhibited similar symptoms, confirming the effects of tengu express

Hydrogen peroxide accumulation and transcriptional changes in grapevines recovered from flavescence dorée disease.

Flavescence dorée (FD) is considered one of the most severe phytoplasma diseases affecting grapevine. The spontaneous, complete, and stable remission of the symptoms of FD (recovery) is a phenomenon that may occur in infected grapevines. The molecular bases of this phenomenon are still unclear, although some works suggest that recovery could be linked to the accumulation of hydrogen peroxide (H2O2). Several genes coding for enzymes involved in H2O2 metabolism, in the ascorbate-glutathione cycle, defense responses, and the biosynthesis of hormones were identified. The H2O2 content was biochemic

The effector SJP3 interferes with pistil development by sustaining SHORT VEGETATIVE PHASE 3 expression in jujube.

Jujube witches' broom (JWB) is a phytoplasma disease that causes severe damage to jujube (Ziziphus jujuba) crops worldwide. Diseased jujube plants show enhanced vegetative growth after floral reversion, including leafy flower structures (phyllody) and the fourth whorl converting into a vegetative shoot. In previous research, secreted JWB protein 3 (SJP3) was identified as an inducer of phyllody. However, the molecular mechanisms of SJP3-mediated pistil reversion remain unknown. Here, the effector SJP3 was found to interact with the MADS-box protein SHORT VEGETATIVE PHASE 3 (ZjSVP3). ZjSVP3 was

A bacterial effector SJP3 induces leaf-like sepal formation by hijacking SHORT VEGETATIVE PHASE in plants.

Jujube (Ziziphus jujuba), the most economically important member of the Rhamnaceae family, is sensitive to phytoplasma-induced witches' broom disease. The SAP54/phyllogen-like effector SJP3 in jujube witches' broom (JWB) phytoplasmas interacts with and stabilizes the MADS-box protein SHORT VEGETATIVE PHASE 3 (ZjSVP3), resulting in pistil reversion. Here, we further demonstrated that SJP3 induced leaf-like sepal formation by activating the ZjSVP1-ZjSVP3 complex at multiple levels. Among the cascading events, SJP3-mediated ZjAP1 and ZjSEP3 degradation promoted ZjSVP1 expression. The miRNA396c-Zj

Structural insights into the interaction between phytoplasmal effector causing phyllody 1 and MADS transcription factors.

Phytoplasmas are bacterial plant pathogens which can induce severe symptoms including dwarfism, phyllody and virescence in an infected plant. Because phytoplasmas infect many important crops such as peanut and papaya they have caused serious agricultural losses. The phytoplasmal effector causing phyllody 1 (PHYL1) is an important phytoplasmal pathogenic factor which affects the biological function of MADS transcription factors by interacting with their K (keratin-like) domain, thus resulting in abnormal plant developments such as phyllody. Until now, lack of information on the structure of PHY

Jasmonate-mediated defence responses, unlike salicylate-mediated responses, are involved in the recovery of grapevine from bois noir disease.

BACKGROUND: Bois noir is an important disease of grapevine (Vitis vinifera L.), caused by phytoplasmas. An interesting, yet elusive aspect of the bois noir disease is "recovery", i.e., the spontaneous and unpredictable remission of symptoms and damage. Because conventional pest management is ineffective against bois noir, deciphering the molecular bases of recovery is beneficial. The present study aimed to understand whether salicylate- and jasmonate-defence pathways might have a role in the recovery from the bois noir disease of grapevine. RESULTS: Leaves from healthy, bois noir-diseased and

Genome analysis of Ceratobasidium theobromae and its causal association with cassava witches' broom disease in the Philippines.

Cassava witches' broom disease (CWBD) has been present in the Philippines from the early 2010s and has since been considered a phytoplasma-caused disease. This assumption has guided CWBD diagnostics and management, yielding mixed results. However, recent results in continental Southeast Asia (SEA) indicate that the disease is rather associated with the fungus Ceratobasidium theobromae, the causal agent of Vascular Streak Dieback (VSD) of cacao, another important disease occurring in the Philippines. Field surveys showed that CWBD was present in all cassava-growing regions, reaching field level

Genome-wide transcriptional analysis of 14-3-3 genes in Paulownia fortunei under abiotic and biotic stresses.

BACKGROUND: The 14-3-3 proteins are evolutionarily conserved regulators in eukaryotes and play pivotal roles in plant stress responses and immunity. Paulownia is a fast‑growing tree valued for its high‑quality timber and diverse industrial applications, but it encounters multiple abiotic and biotic stresses, including drought, salinity, and phytoplasma‑induced witches' broom disease. To better understand the molecular mechanisms underlying stress resilience in Paulownia fortunei, a comprehensive analysis of the 14-3-3 gene family was conducted. RESULTS: Genome‑wide identification revealed 14

The pathogenic effector PHYL1JWB interacts with ZjCURT1A to mediate jujube witches' broom-induced photosynthesis impairment of jujube trees.

Photosynthesis impairment and the accompanying leaf yellowing represent a prominent and physiologically critical symptom of phytoplasma infection. However, the underlying molecular mechanisms remain poorly understood. In this study, we identified the Jujube Witches' Broom pathogenic effector PHYL1JWB, which was previously reported to induce phyllody in jujube, as a key inducer of photosynthesis impairment. Overexpression of PHYL1JWB in sour jujube and Arabidopsis thaliana disrupted photosynthesis by causing severe disorganization of chloroplast ultrastructure. PHYL1JWB directly interacts with
