BACKGROUND: T lymphocytes play an important role in contact hypersensitivity. This study aims to explore the immunosuppressive activity of SBF-1, an analog of saponin OSW-1, against T lymphocytes in vitro and in vivo.
METHODS: Proliferation of T lymphocytes from lymph nodes of mice was determined by MTT assay. Flow cytometry analysis was performed to assess T cell activation and apoptosis. Levels of cytokines were determined by PCR and ELISA. BALB/c mice were sensitized and challenged with picryl chloride and thickness of left and right ears were measured.

AIM OF THE STUDY: Artemisia vestita is a common traditional Tibetan medicinal plant which has been used widely in China for treating various inflammatory diseases. The choice of the type of instrument to measure delayed-type hypersensitivity (DTH) in mice, as assayed by ear swelling reactions, influences the experimental results. Effects of water extract (UW-ext) from the leaf of Arctostaphylos uva-ursi (L.) Spreng (Bearberry leaf) on the antiallergic and antiinflammatory activities of dexamethasone ointment were investigated. A mouse model for assessment of the anti-inflammatory effect of topical glucocorticoids is described. Mice, sensitized to picryl chloride, had both ears painted with the sensitizer followed 2 hours later by the application of the topical steroid to one ear and the corresponding vehicle to the other. An immunosuppressant with fewer adverse effects is still urgently needed for increasing numbers of patients suffering from allergic contact dermatitis. The present experiment was performed to examine the effects of oxatomide on the four types of allergic reactions classified by Cooms and Gell. 1) type 1: Oxatomide administered p.o. showed inhibitory effects on the 48 hr homologous passive cutaneous anaphylaxis in rats and the passive anaphylactic bronchoconstriction in guinea pigs. 2) type 2: Oxatomide showed few effects on the complement-dependent cytolysis of sheep red blood cells, the Forssman shock in guinea pigs, and the reversed cutaneous anaphylaxis and the swelling of the footpad induced by rabbit antiserum against rat serum in rats.
