Pinealectomy does not affect the healing of experimental colonic anastomoses.

Gastrointestinal system anastomoses, especially colonic anastomoses, have significant morbidity and mortality despite recent technical improvements. Besides regulating the circadian rhythm, the pineal gland and its main neurohormone product melatonin have widespread actions in the organism. The purpose of this study was to investigate the effects of pinealectomy on the healing of colonic anastomoses. One hundred male albino Wistar rats were used in this study. The rats were separated into three groups: control, pinealectomy, and sham groups. In the control group, only colonic resection and ana

Influence of pinealectomy on levels of PACAP and cAMP in the chicken brain.

One of the recently found functions of pituitary adenylate cyclase activating polypeptide (PACAP) is the modulation of circadian rhythms. Widespread distribution of PACAP-containing neurons and receptors has been shown in the chicken. Recently, we have demonstrated that PACAP levels oscillate in a circadian manner in the chicken brain. Daily variation in PACAP levels might be influenced by several regulatory mechanisms. Among the structures that may regulate PACAP levels, one candidate is the pineal gland. Therefore, in the present study, we investigated the effect of pinealectomy on the level

Correlation between the age of pinealectomy and the development of scoliosis in chickens.

STUDY DESIGN: Pinealectomy induces experimental scoliosis in chickens. This study analyzed the correlation between the age at which pinealectomy was performed and the development of scoliosis in chickens. OBJECTIVE: To investigate the differences in the rate or magnitude of scoliosis and the type of curvature in chickens pinealectomized at different times after hatching. SUMMARY OF BACKGROUND DATA: Scoliosis develops in almost all chickens pinealectomized within 3 days after hatching, but there are no data on whether the condition will develop in chickens pinealectomized earlier or later aft

Effects of pinealectomy on baseline sleep and response to sleep deprivation.

STUDY OBJECTIVES: We have previously reported that older (24 mo.) Fischer rats manifest a diminished post-sleep deprivation increase in NREM and REM sleep. In order to examine whether this decline reflects an age-related change in pineal function, we are now reporting on baseline and recovery sleep parameters in pinealectomized 3-, 12-, and 24-month old rats following 24 hours of sleep deprivation using the disk-over-water method. DESIGN: Three independent age groups; within each group there were sequential measures of sleep under baseline conditions and during recovery from sleep deprivation

Melatonin, pinealectomy, and release of neurohypophysial hormones: in vitro studies.

The effect of melatonin as well as pinealectomy on the basal and K(+)-evoked release of vasopressin and oxytocin from the neurointermediate lobes in vitro was determined. Pineal removal resulted in a diminution of vasopressin and oxytocin release from the neurointermediate lobes in vitro. Melatonin (10(-3) or 10(-6) M/l) increased vasopressin and oxytocin release from neurointermediate lobes of sham-operated rats. Nevertheless, when pinealectomized rats served as donors of the neurointermediate lobes, melatonin (10(-3) or 10(-6) M/l) increased vasopressin release under basal conditions. For th

Effects of pinealectomy on the ultrastructure of the golden hamster parathyroid gland.

Ultrastructural changes of the parathyroid glands of pinealectomized golden hamsters were investigated. The main changes in the parathyroid glands 1 hour and 1 day after pinealectomy compared with the control and sham-operated groups were an increase of the Golgi complexes, cisternae of the granular endoplasmic reticulum and large vacuolar bodies. In addition, many chief cells contained numerous prosecretory granules in the Golgi areas and many secretory granules in the peripheral cytoplasm. The morphology of the parathyroid glands 7 and 30 days after pinealectomy resembled that of the control

Sleep disturbance after pinealectomy in patients with pineocytoma WHO°I.

BACKGROUND: Because the pineal gland produces melatonin, it is suggested to be involved in the regulation of sleep and circadian rhythm, though there is scant proof of this. Tumors of the pineal gland are rare and various in terms of histological and biological malignancy. We evaluated the occurrence of subjective sleep disturbances in nine patients who underwent a pinealectomy due to pineocytoma WHO°I without additional therapy. METHODS: Patients with intracranial low-grade lesions and patients without a craniotomy who underwent a microscopic lumbar discectomy were matched to our study group

Maternal pinealectomy increases depressive-like responses in Siberian hamster offspring.

This study investigated the effect of maternal pinealectomy and postnatal pinealectomy on affective responses. Siberian hamsters were born to either pinealectomized or sham-operated dams and then underwent pinealectomy or a sham operation. Maternal pinealectomy increased depressive-like responses of offspring in the forced swim test. Maternal pinealectomy increased rearing behaviour and postnatal pinealectomy increased locomotor behaviour in the open field test. These results suggest that prenatal melatonin organizes adult affective responses.

Experimental scoliosis in melatonin-deficient C57BL/6J mice without pinealectomy.

The etiology of idiopathic scoliosis is unknown. Scoliosis with many characteristics closely resembling those seen in idiopathic scoliosis has been produced in young chickens and bipedal rats after pinealectomy. In this study, we induced experimental scoliosis in C57BL/6J mice without pinealectomy and melatonin treatment suppressed the development of scoliosis. A total of 100 mice were divided into four groups: 20 quadrupedal mice served as controls; 30 mice underwent resection of two forelegs and tail at 3 wk of age (bipedal mice); the remaining 20 quadrupedal and 30 bipedal mice received int

Does pinealectomy affect the recovery rate after spinal cord injury?

Previous reports documented demonstrated that melatonin, a free radical scavenger, is important in protecting against oxidative stress-induced tissue damage after spinal cord injury (SCI). This study was undertaken to investigate the effects of pinealectomy (PX) and administration of exogenous melatonin after SCI in rats. These animals were randomized into six groups, each having 12 rats. Group 1 underwent laminectomy alone. Group 2 underwent laminectomy followed by SCI and received no medication. Group 3 underwent laminectomy followed by SCI and received melatonin. Group 4 underwent PX and la

Pup circadian rhythm entrainment--effect of maternal ganglionectomy or pinealectomy.

In rodents, during late embryonic and early neonatal development, circadian rhythms develop in synchrony with those of their mothers, which in turn are synchronized with the environmental photoperiod. This paper examines the effect of maternal ganglionectomy (pineal gland sympathetic denervation) or extirpation of the pineal gland on pups' drinking rhythms, a behavior that is continuously monitored in individual animals starting after weaning and studied up to 3 weeks later. Maternal ganglionectomy or pinealectomy performed on the 7th day of gestation significantly disrupts rat pups' drinking

Long-term pinealectomy alters hypothalamic serotonin metabolism in the rat.

In the present study, the effects of long-term pinealectomy on tryptophan, 5-hydroxytryptamine (5-HT or serotonin), 5-hydroxy-3-indoleacetic acid (5-HIAA), and tryptophan hydroxylase and monoamine oxidase activities were studied in preoptic area-anterior hypothalamus (POA-AH) and in the medial and posterior hypothalamus of the rat. After pinealectomy, 5-HT levels decreased significantly in medial hypothalamus but increased in the POA-AH. The levels of 5-HIAA decreased significantly in the POA-AH and medial hypothalamus. Tryptophan levels remained unchanged while tryptophan hydroxylase activity

Pinealectomy prevents short photoperiod inhibition of male hamster sexual behavior.

The role of the pineal gland in mediating photoperiodic influences on copulatory behavior (CB) of male hamsters (Mesocricetus auratus) was assessed in the presence and absence of testosterone (T). The results demonstrate that the pineal gland is necessary for short photoperiod exposure to alter CB. Sexually experienced males were exposed to either long (14L:10D; LP) or short (8L:16D; SP) photoperiods for 13 weeks; after the first 2 weeks of exposure, all animals were castrated and then either pinealectomized (PINX) or sham operated (SHAM PINX). CB tests over an 8-week period following surgery

Effects of Pinealectomy and Gonadectomy on Olfactory Bulb Dopaminergic Neurons in Rats.

BACKGROUND: Olfactory disorder is an early manifestation of Parkinson's disease (PD), likely to be associated with abnormalities of the dopaminergic neurons in the olfactory bulb (OB); however, the causes of olfactory disorder in PD are not entirely clear. Some studies showed that melatonin (MT) and androgens (mainly testosterone, T) might participate in the pathogenesis of PD. The research aimed to investigate effects of MT or T deficiency on OB dopaminergic neurons in rats. METHODS: One hundred and twenty normal male Wistar rats were randomly divided into the control, sham operation pineale

Effects of pinealectomy on the levels and the circadian rhythm of plasma homocysteine in rats.

Nonenzymatic glycation of proteins has been implicated in various diabetic complications and age-related disorders. Proteins undergo glycation at the N-terminus or at the epsilon-amino group of lysine residues. Glycation of proteins proceeds through the stages of Schiff base formation, conversion to ketoamine product and advanced glycation end products. Gramicidin S, which has two ornithine residues, was used as a model system to study the various stages of glycation of proteins using electrospray ionization mass spectrometry. The proximity of two ornithine residues in the peptide favors the g

Effects of pinealectomy on the circadian release pattern of leptin in male rat.

OBJECTIVES: Exogenous and endogenous melatonin decrease leptin release. It is not known whether melatonin also has an effect on circadian release pattern of leptin. So, this study was planned to investigate the possible changes in the circadian release of leptin following pinealectomy. METHODS: A group of rats were surgically pinealectomized while some others were exposed to sham operation. The animals were decapitated at 13.30 p.m. and 01.30 a.m. Serum leptin levels were measured by radioimmunoassay. RESULTS: Serum leptin levels at 13.30 p.m. were lower than the values at 01.30 a.m. in both

Pinealectomy reduces melatonin levels in the serum but not in the gastrointestinal tract of rats.

Melatonin levels were determined in serum and gastrointestinal tract (GIT) tissues of control (C), sham-pinelaectomized (SPx) and pinealectomized (Px) rats sacrificed in mid scotophase. Serum melatonin concentrations of Px rats exhibited the significantly lowest values (8.6 pg/ml), followed by SPx (20.1 pg/ml) and C (37.5 pg/ml) rats. In C, the ileum (542 pg/ml) and jejunum (531 pg/ml) exhibited the highest average GIT concentrations, followed by the colon (362 pg/ml), stomach (359 pg/ml) and cecum (164 pg/ml). However, only jejunum and ileum samples had significantly higher melatonin levels t

Effects of pinealectomy on SCN electrical firing rhythm in Djungarian hamsters.

Djungarian hamsters (Phodopus sungorus) from a long day photoperiod (16:8 h light-dark cycle) were either pinealectomized (PINX; n = 7) or sham-pinealectomized (SHAM; n = 6). One week after surgery coronal brain slices (500 microm), containing the suprachiasmatic nucleus (SCN), were prepared. The firing-rate rhythm of SCN neurons was recorded over at least a 22 h period. The amplitude of the firing rhythm for SHAM slices (3.42 +/- 0.26 Hz) was not significantly different from PINX slices (3.45 +/- 0.29 Hz). Maxima and minima of the firing rhythms also could not be distinguished statistically b
