[Features of the temporal structure of the respiratory cycle in the Tourette syndrome as a possible reflection of intensified dopaminergic processes].

The effect of dopamine blockers (chlorpromazine and haloperidol) and the dopaminomimetic piribedil on the structure of the breathing cycle was studied under conditions of the clinical employment of the drugs. It was found that piribedil increased the inspiration fraction (IF) in the respiratory cycle whereas neuroleptics diminished it. Twenty-six children with Tourette's syndrome (TS) and 24 with a temporal form of epilepsy were examined. Patients with TS showed two specific features of respiration regulation: a greater value of IF and a greater variability of respiratory parameters. Taking in

Reduced frequency of wheezing respiratory illness in infants with perinatal human immunodeficiency virus-type 1 infection: a model for immunologic and inflammatory mechanisms of airway obstruction?

INTRODUCTION: Clinicians switch from one dopamine agonist to another for various reasons. However, each change may inadvertently result in certain potential risks such as decreased medication efficacy or new side-effects. OBJECTIVE: We evaluated the tolerability of a switch of bromocriptine to piribedil using two conversion ratios as a primary outcome measure, with motor function as a secondary outcome measure, in patients with mild to moderate Parkinson's disease (PD). METHODS: Twenty consecutive patients with mild to moderate PD (Hoehn and Yahr, stage II-III) on treatment with stable doses

The effect of dopamine on shivering in the rat.

1. Dopamine (0.5--0.2 mumole) injected into a lateral cerebral ventricle inhibited shivering and lowered core temperature in rats at an ambient temperature of 0--5 degrees C. 2. These effects were inhibited by the dopamine antagonist, pimozide, and imitated by the dopamine agonists, piribedil and apomorphine. 3. Pimozide itself had no effect on shivering. 4. It is concluded that while there are inhibitory dopamine receptors on the cold sensor--shivering pathway in the rat they are not concerned in the normal function of this pathway.

Small Arms Fire-like noise: Effects on Hearing Loss, Gap Detection and the Influence of Preventive Treatment.

A noise-induced loss of inner hair cell (IHC) - auditory nerve synaptic connections has been suggested as a factor that can trigger the progression of maladaptive plastic changes leading to noise-induced tinnitus. The present study used a military relevant small arms fire (SAF)-like noise (50 biphasic impulses over 2.5 min at 152 dB SPL given unilaterally to the right ear) to induce loss (∼1/3) of IHC synaptic ribbons (associated with synapse loss) in rat cochleae with only minor (less than 10%) loss of outer hair cells. Approximately half of the noise-exposed rats showed poorer Gap Detection

Comparative effects of dopaminergic agonists on cardiovascular, renal, and renin-angiotensin systems in hypertensive patients.

The role of dopaminergic receptors on renal function has been extensively studied. Recently dopaminergic receptor has been classified in two subtypes D1 and D2, which seem to have different modulatory function. However, the role of dopaminergic receptors on cardiovascular function and more specifically the potential role of dopaminergic agonists as antihypertensive agents has not yet been clarified. Nine outpatients with mild and moderate hypertension were studied in the Cardiology Service of Vargas Hospital with a D1 agonist, piribedil, at 50-100 mg/day, orally, for 8 weeks, and with a D2 ago

Blockage of dopaminergic D(2) receptors produces decrease of REM but not of slow wave sleep in rats after REM sleep deprivation.

Dopamine (DA) has, as of late, become singled out from the profusion of other neurotransmitters as what could be called a key substance, in the regulation of the sleep-wake states. We have hypothesized that dopaminergic D(2) receptor blockage induced by haloperidol could generate a reduction or even an ablation of rapid eye movement (REM) sleep. Otherwise, the use of the selective D(2) agonist, piribedil, could potentiate REM sleep. Electrophysiological findings demonstrate that D(2) blockage produced a dramatic reduction of REM sleep during the rebound (REB) period after 96 h of REM sleep dep

[Neurochemical mechanisms of tremor in Parkinson's disease].

Tremor is one of the main motor symptoms of Parkinson's disease, and its pathophysiology remains largely unknown. The clinical and pathomorphological heterogeneity of tremor and the not always response to therapy complicate the task of researchers and clinicians. This review discusses the specific degeneration of neurotransmitter systems driving the development of tremor, and the influence of neurotransmitters on specific anatomical entities according to current models explaining tremor. It is discusses how changes in neurotransmitter systems may influence the clinical diversity of tremor and

Central regulation of adrenal tyrosine hydroxylase: effect of induction on catecholamine levels in the adrenal medulla and plasma.

The effect of induction of adrenal tyrosine hydroxylase (TH) by various centrally acting drugs on catecholamine levels in adrenal and plasma was investigated in rats. All the drugs tested, namely oxotremorine, Piribedil, B-HT 920, and HA-966, produced significant increases in adrenal dopamine content and plasma epinephrine level. Denervation of the adrenal abolished the increase in adrenal dopamine as it did the induction of tyrosine hydroxylase. The results suggest that the induced increase of adrenal TH activity, as mediated by certain drugs, results in an elevation of the plasma epinephrine

Dopaminergic drugs antagonize the psychotomimetic effects of partial-agonist analgesics.

The psychotomimetic actions of the partial-agonist analgesic drugs cyclazocine and levallophan have been demonstrated using a quantitative behavioural test in young rats which measures lateral head movements and pivoting on the hind paws. This drug induced behaviour is antagonized by low doses of the dopaminergic agonists apomorphine and piribedil, the dopamine releasing drug amphetamine, the dopamine reuptake blocking agent benztropine and by large doses of the dopamine precursor L-Dopa. Naloxone antagonize the behaviour, but only at one hundred times the analgesic antagonist dose. These resu

Effects of dopamine agonists and antagonists in Tourette's disease.

The actions of haloperidol, dextroamphetamine sulfate, levamfetamine succinate, apomorphine, and piribedil were studied in two patients with Giles de la Tourette's disease in an attempt to clarify the catecholamine mechanisms involved in this condition. Both dextroamphetamine and levamfetamine increased the severity of the symptoms; dextroamphetamine was more potent. Haloperidol controlled the symptoms and also antagonized the effect of dextroamphetamine. Apomorphine injections reduced the severity of symptoms, even in the presence of dextroamphetamine. We conclude that dopamine rather than no

Adsorptive stripping voltammetric behaviour of azomethine group in pyrimidine-containing drugs.

The stripping voltammetric behaviour of buspirone hydrochloride (BUS) and piribedil (PIR), as models of pyrimidine-containing compounds, was studied using a hanging mercury drop electrode (HMDE). A sensitive adsorptive stripping voltammetric method for determination of such drugs is described. The voltammetric peaks were obtained at -1.23 and -1.22 V for BUS and PIR. respectively, which correspond to the reduction of the azomethine group of pyrimidine ring in Britton-Robinson buffer (pH 7). Pathophysiology and management of syncope in Kearns-Sayre syndrome.

A 20-year-old woman with known Kearns-Sayre syndrome was transferred to the emergency department due to syncopal episodes. The electrocardiogram on admission showed complete atrioventricular block. The diagnosis of mitochondrial encephalomyopathy was made when she was 14 years old. At the time of the initial diagnosis, she displayed a normal electrocardiogram pattern. At the age of 17, electrocardiogram recordings demonstrated right bundle branch block with left anterior fascicular block and a prolonged QTc interval of 485 milliseconds (Figure). She was taking coenzyme Q10, oral nicotinamide a

Influence of the cation in hypophosphite-mediated catalyst-free reductive amination.

Reducing agents with phosphorus-hydrogen bond, such as sodium hypophosphite, phosphite, and hypophosphorous acid are commercially available in bulk amounts, however, their usage is understudied in organic processes. While NaH2PO2 has proved to be an efficient four-electron reductant in the catalyst-free reductive amination, the influence of cation in hypophosphite salt has not been studied yet. This issue is a fundamentally important factor. In the present work, the reactivity of the hypophosphites of alkali metals (Li, K, Rb, and Cs) in reductive amination was explored for the first time. Climbing and stereotyped behaviours in mice require the stimulation of D-1 dopamine receptors.

Apomorphine-induced climbing and sniffing behaviours in mice were antagonize by low doses of SCH 23390 and metoclopramide. The selective D-2 dopamine receptor agonists, LY 171555 and RU 24926, and some other dopamine agonists (piribedil, lergotrile, bromocriptine) exerted only inhibitory effects on spontaneous behaviours. The selective D-1 dopamine receptor agonist, SK&F 38393, did not modify the climbing score but increased the sniffing and decreased the gnawing scores compared to the scores of control mice. Typical climbing and stereotyped behaviours were produced by the combination of SK&F

Clinical trials of dementia with Lewy bodies and Parkinson's disease dementia.

Despite the frequency and importance of dementia associated with Parkinson's disease (PDD) and dementia with Lewy bodies (DLB), there is relatively little evidence on which to base treatment. Evidence from meta-analysis suggests that rivastigmine can improve cognition and functioning in PDD and also reduce risk of falling. There is also evidence supporting its use in DLB. Recent evidence suggests that memantine may also be effective, particularly for PDD, although evidence is more conflicting. Memantine may also improve parkinsonism and dyskinesias. Few clinical trials of cognition in PD witho
