Prevalence of theileriosis in Red Hartebeest (Alcelaphus buselaphus caama) in Namibia.

A total of 23 blood samples from Red Hartebeest and 28 ticks of the subspecies Rhipicephalus evertsi were collected in Namibia during the summer 2003. For a Piroplasmida species, Theileria sp 95.7% of blood and 50.0% of tick samples (all R. e. mimeticus, none of R. e. evertsi were carrying the pathogen) were PCR positive. Sequencing showed infection from a re-emerging mild species of Theileria phylogenetically different from other African Theileria species such as Theileria parva, T. buffeli, T. annulata, T. taurotragi or T. mutans.

First molecular detection of Cytauxzoon felis and Babesia sp. in ticks collected from wildlife in Atwemonom bushmeat market of Ghana and their potential veterinary health implications.

Tick-borne protozoan parasites pose significant risks to veterinary health worldwide, affecting both companion animals and livestock. When wildlife and domestic animals come into contact due to habitat encroachment, the likelihood of pathogen transmission increases, with ticks serving as potential vectors. This study investigated the occurrence of tick-borne pathogens of veterinary concern in ticks collected from wildlife commonly hunted and sold at the Atwemonom Bushmeat Market in Kumasi, Ghana. Ticks removed from wildlife carcasses were identified morphologically and screened using primers t

Degrade to survive: the intricate world of piroplasmid proteases.

Piroplasmids of the genera Babesia, Theileria, and Cytauxzoon are tick-transmitted parasites with a high impact on animals and humans. They have complex life cycles in their definitive arthropod and intermediate vertebrate hosts involving numerous processes, including invasion of, and egress from, host cells, parasite growth, transformation, and migration. Like other parasitic protozoa, piroplasmids are equipped with different types of protease to fulfill many of such essential processes. Blockade of some key proteases, using inhibitors or antibodies, hinders piroplasmid growth, highlighting t

Babesiosis in cats: ABCD guidelines on prevention and management.

OVERVIEW: Babesiosis is a tick-borne protozoan disease caused by parasites of the genus Babesia that belong to the Piroplasmida. The disease is named after the Romanian bacteriologist Victor Babeş. Babesiosis is also known as piroplasmosis (from Latin pirum, meaning 'pear', and plasma, 'image, formation'). INFECTION: Babesiosis affects domestic and wild animals and humans worldwide. While the disease is recognised in dogs around the world, it is found only rarely in cats. HUMAN DISEASE: Babesia species are common blood parasites of mammals. Human babesiosis is uncommon, but more cases in peop

Babesia Life Cycle - When Phylogeny Meets Biology.

Although Babesia represents an important worldwide veterinary threat and an emerging risk to humans, this parasite has been poorly studied as compared to Plasmodium, its malaria-causing relative. In fact, Babesia employs highly specific survival strategies during its intraerythrocytic development and its intricate journey through the tick vector. This review introduces a substantially extended molecular phylogeny of the order Piroplasmida, challenging previous taxonomic classifications. The intriguing developmental proficiencies of Babesia are highlighted and compared with those of other haemo

Piroplasmid infection is not associated with clinicopathological and laboratory abnormalities in cats from Midwestern Brazil.

Feline piroplasmids include the genera Babesia spp., Cytauxzoon spp., and Theileria spp. In Brazil, there are few reports regarding these hemoprotozoans; however, clinicopathological and molecular data are scarce. This study aimed to characterize the clinical relevance of these parasites through hematological, biochemical, and molecular approaches. For this purpose, 166 cats from Brasilia, Federal District, Midwestern Brazil, were screened using a quantitative polymerase chain reaction (qPCR) for piroplasmids based on the LSU4 mitochondrial gene, which resulted in an overall prevalence of 36/1

Natural Infection of Wild Canids (Cerdocyon thous and Lycalopex gymnocercus) with the Intraendothelial Piroplasm Rangelia vitalii in Southern Brazil.

Rangelia vitalii is a piroplasm that infects canines, causing lesions typical of a hemolytic disorder. Two wild canids, a crab-eating fox (Cerdocyon thous) and a Pampas fox (Lycalopex gymnocercus), were presented for necropsy in Setor de Patologia Veterinária at the Universidade Federal do Rio Grande do Sul, Porto Alegre, Brazil. On gross examination, both animals had pale mucosae and moderate tick infestation (Amblyomma aureolatum). There was severe splenomegaly, and the liver had a diffusely orange-reddish lobular pattern. The mesenteric lymph nodes were brownish and slightly enlarged. Struc

Molecular evidence for piroplasms in wild Reeves' muntjac (Muntiacus reevesi) in China.

DNA from liver samples of 17 free-ranging wild Reeves' muntjac (Muntiacus reevesi) was used for PCR amplification of piropalsm 18S rRNA gene. Of 17 samples, 14 (82.4%) showed a specific PCR product which were cloned and sequenced. BLAST analysis of the sequences obtained showed similarities to Babesia sp., Theileria capreoli, Theileria uilenbergi and Theileria sp. BO302-SE. Phylogenetic analysis showed that the Babesia sp. detected in the present study was distantly separated from known Babesia species of wild and domestic animals. Six sequences showed 100% similarity to T. capreoli while five

Identification and phylogenetic analysis of Babesia parasites in domestic dogs in Nigeria.

The present study examined the presence of Babesia parasites in 104 domestic dogs in Nigeria. Sequentially, Babesia parasites infecting domestic dogs underwent genetic and phylogenetic analyses. The results of nested PCR based on the Piroplasmida 18S rRNA gene illustrated that 13.5% (14/104) of the samples were positive. The obtained positive samples determined the nucleotide sequences of the 18S rRNA genes. In the genetic and phylogenetic analyses, four of five nucleotide sequences were similar to Babesia canis rossi, and one sample exhibited a close similarity to a Babesia sp. isolated from

Development of a loop-mediated isothermal amplification (LAMP) assay for rapid diagnosis of Babesia canis infections.

Vector-borne diseases are rising in interest due to global warming, which is believed to impact on the distribution of vectors into new areas thus influencing the occurrence and epidemiology of vector-borne pathogens. Babesia canis belongs to the Piroplasmidae and there are three described subspecies, namely B. canis canis, B. canis rossi and B. canis vogeli. They are each transmitted by a different tick-species, Dermacentor reticulatus, Haemaphysalis leachi and Rhipicephalus sanguineus, respectively. There are also differences in the geographical distribution and pathogenicity to dogs of each

[From piroplasmosis to babesiosis--problems with classification of Babesia protozoa isolated from dogs].

Babesia and Theileria are the tick-borne parasites belonging to the order Piroplasmida. Both of them cause a severe disease with symptoms of hemolytic anemia. The main etiological factors of canine piroplasmosis are protozoa Babesia. There are two species of this pathogen infective for dogs: Babesia canis and Babesia gibsoni. Based on the results of molecular biology techniques it is possible to distinguish them as well as many subspecies or strains. It is known since quite some time that there are substantial differences in vector specificity, cross-immunity and pathogenicity between isolates

Morphological characteristics and developmental changes of the ovary in the tick Haemaphysalis longicornis Neumann.

Haemaphysalis longicornis (Ixodida: Ixodidae) is an important vector of transovarially transmitted parasites of the genus Babesia (Piroplasmida: Babesiidae). In the present study, we investigated the morphological characteristics and developmental changes of the ovary of H. longicornis. We show that the ovary of H. longicornis has a single tubular structure and is surrounded by a tunica propria. There is a longitudinal groove along one side of the ovary. During feeding and after engorgement, great changes can be observed in the ovary of H. longicornis and two rapid growth phases can be detecte

Transmission of Cytauxzoon felis by injection of Amblyomma americanum salivary glands.

BACKGROUND: Cytauxzoonosis is a life-threatening disease of cats, caused by the tick-borne piroplasmid hemoparasite, Cytauxzoon felis. Current experimental models for cytauxzoonosis rely on either tick transmission or direct injection of infected cat tissues. These models require researchers to directly work with infected ticks or use cats with acute cytauxzoonosis. To improve the feasibility and accessibility, there is a need to establish sharable resources among researchers. In related piroplasmid parasites, sporozoite-based inoculums are routinely produced from tick salivary glands, cryopre

Molecular pathogen screening of louse flies (Diptera: Hippoboscidae) from domestic and wild ruminants in Austria.

BACKGROUND: Hippoboscid flies (Diptera: Hippoboscidae), also known as louse flies or keds, are obligate blood-sucking ectoparasites of animals, and accidentally of humans. The potential role of hippoboscids as vectors of human and veterinary pathogens is being increasingly investigated, but the presence and distribution of infectious agents in louse flies is still unknown in parts of Europe. Here, we report the use of molecular genetics to detect and characterize vector-borne pathogens in hippoboscid flies infesting domestic and wild animals in Austria.
