SSR-Based Molecular Identification and Population Structure Analysis for Forage Pea (Pisum sativum var. arvense L.) Landraces.

Plant genetic diversity has a significant role in providing traits that can help meet future challenges, such as the need to adapt crops to changing climatic conditions or outbreaks of disease. Our aim in this study was to evaluate the diversity of 61 forage pea specimens (P. sativum ssp. arvense L.) collected from the northeastern Anatolia region of Turkey using 28 simple sequence repeat (SSR) markers. These primers generated a total of 82 polymorphic bands. The number of observed alleles (Na) per primer varied from 2 to 4 with a mean of 2.89 alleles/locus. The mean value of expected heterozy

Possible roles for His 208 in the active-site region of chloroplast carbonic anhydrase from Pisum sativum.

His 208 of chloroplast pea carbonic anhydrase is conserved among the dicotyledonous carbonic anhydrases. This His was replaced by an Ala to test whether a histidine residue at this position, in analogy to His 64 of human carbonic anhydrase II, acts as an internal proton shuttle. Values of the kinetic parameters kcat and kcat/Km for the H208A mutant enzyme are high over the pH range 6-9 and of the same magnitude as those for the wild-type enzyme, indicating that this residue is not crucial for the catalytic event. The pH dependence of kcat/Km, reflecting the Zn-H2O ionization, is, however, simp

The auxin conjugate indole-3-acetyl-aspartate affects responses to cadmium and salt stress in Pisum sativum L.

The synthesis of IAA-amino acid conjugates is one of the crucial regulatory mechanisms for the control of auxin activity during physiological and pathophysiological responses. Indole-3-acetyl-aspartate (IAA-Asp) is a low molecular weight amide conjugate that predominates in pea (Pisum sativum L.) tissues. IAA-Asp acts as an intermediate during the auxin degradation pathway. However, some recent investigations suggest a direct signaling function of this conjugate in various processes. In this study, we examine the effect of 100 μM IAA-Asp alone and in combination with salt stress (160 mM NaCl) …

Non-host disease resistance response in pea (Pisum sativum) pods: Biochemical function of DRR206 and phytoalexin pathway localization.

Continually exposed to potential pathogens, vascular plants have evolved intricate defense mechanisms to recognize encroaching threats and defend themselves. They do so by inducing a set of defense responses that can help defeat and/or limit effects of invading pathogens, of which the non-host disease resistance response is the most common. In this regard, pea (Pisum sativum) pod tissue, when exposed to Fusarium solani f. sp. phaseoli spores, undergoes an inducible transcriptional activation of pathogenesis-related genes, and also produces (+)-pisatin, its major phytoalexin. One of the inducib…

Ultraviolet-B induced changes in morphological, physiological and biochemical parameters of two cultivars of pea (Pisum sativum L.).

Increase in perception of solar ultraviolet-B (UV-B) radiation on Earth's surface due to anthropogenic activities has potential in causing detrimental effects on plants. The present study was performed to evaluate the effect of elevated UV-B on Pisum sativum L., a leguminous plant with emphasis on nitrogen metabolism, flavonoids and hormonal changes. Elevated UV-B (ambient+7.2 kJ m(-2) day(-1)) negatively affected the growth, biomass, yield and its quality by generating oxidative stress directly or due to elevation of salicylic acid in two cultivars with higher magnitude being observed in HUP-…

Relationships of root conductivity and aquaporin gene expression in Pisum sativum: diurnal patterns and the response to HgCl2 and ABA.

Experiments were undertaken to test how aquaporins (AQPs) facilitate the uptake of water by roots of Pisum sativum. Changes in PsPIP2-1 gene expression and root hydraulic conductivity (Lpr) were measured in response to the time of day as well as treatment of the roots with a compound that reduced Lpr [i.e. mercuric chloride (HgCl2)] and one that was intended to increase Lpr [abscisic acid (ABA)]. There was a diurnal rhythm in PsPIP2-1 expression in lateral roots that was strongly correlated with diurnal changes in Lpr. Taproots also displayed a rhythm in PsPIP2-1 expression, but this was offse…

Effects of Pea (Pisum sativum) Prebiotics on Intestinal Iron-Related Proteins and Microbial Populations In Vivo (Gallus gallus).

Iron deficiency remains a public health challenge globally. Prebiotics have the potential to improve iron bioavailability by modulating intestinal bacterial population, increasing SCFA production, and stimulating expression of brush border membrane (BBM) iron transport proteins among iron-deficient populations. This study intended to investigate the potential effects of soluble extracts from the cotyledon and seed coat of three pea (Pisum sativum) varieties (CDC Striker, CDC Dakota, and CDC Meadow) on the expression of BBM iron-related proteins (DCYTB and DMT1) and populations of beneficial in…

Green Pea (Pisum sativum L.) Hull Polyphenol Extract Alleviates NAFLD through VB6/TLR4/NF-κB and PPAR Pathways.

Green pea hull is a processing byproduct of green pea and rich in polyphenols. Nonalcoholic fatty liver disease (NAFLD) is a chronic metabolic disease characterized by accumulation of lipids in the liver for which there are no effective treatment strategies. Here, a mouse model of NAFLD induced by a DSS+high-fat diet (HFD) was established to investigate the effect of green pea hull polyphenol extract (EGPH). The results show that EGPH relief of NAFLD was a combined effect, including reducing hepatic fat accumulation, improving antioxidant activity and blood lipid metabolism, and maintaining gl

Loss of galactan synthesis in pea (Pisum sativum) causes defects in organ expansion and is associated with increased extensin content.

Pectic galactan is a rhamnogalacturonan-I side chain, composed of β-(1→4)-linked D-galactose residues. Its multifunctional role in cell wall assembly and properties has been proposed in various developmental contexts. In this study, we investigated the role of galactan in pea (Pisum sativum syn. Lathyrus oleraceus). First, we used glycan profiling to examine stems with different auxin-induced elongation rates for changes in cell wall composition. The galactan epitopes, recognised with the LM5 monoclonal antibody, were more abundant in the convex, more elongated part of the pea stem. This findi…

Differential ozone-induced alterations in the photosynthetic apparatus and chlorophyll fluorescence dynamics of two pea cultivars (Pisum sativum L.).

The phytotoxic nature of tropospheric ozone (O3) has led to substantial declines in global crop productivity. Given their nutritional richness, legumes serve as vital dietary staples, valued for their high protein, oil, and fiber content. A field experiment was conducted to assess the physiological plasticity of two pea (Pisum sativum L.) cultivars, K Agaiti (O3-sensitive) and K Damini (O3-tolerant), under elevated O3 (ambient + 15 ppb). The Ball-Woodrow-Berry (BWB) model revealed a greater change in the intercept for K Agaiti compared to K Damini, indicating stronger stomatal disruption. Anal…

Biochemical and proteomic approaches to investigating effects of IAA-aspartate in pea (Pisum sativum L.) seedlings during osmotic shock.

Osmotic shock is the first step of high salt or drought action that involves biochemical and molecular changes during plant response to these unfavorable conditions. Indole-3-acetyl-aspartate (IAA-aspartate, IAA-Asp) is the main amide conjugate of auxin in pea (Pisum sativum L.) tissues. Although the exact molecular mechanism of the IAA-Asp action is unknown, this conjugate's indole-3-acetic acid (IAA)-independent biological activity has been observed during physiological and stress conditions. In this work, we investigated the effect of IAA-Asp alone, as well as in combination with NaCl or po…

Variation in precaecal amino acid and energy digestibility between pea (Pisum sativum) cultivars determined using a linear regression approach.

An experiment was conducted to study the variation in the precaecal amino acid (AA) and energy digestibility of 4 cultivars of white-flowering peas (Pisum sativum) for broiler chickens. The 4 cultivars were grown and harvested under the same agronomic and environmental conditions. One basal diet and 8 diets including each pea cultivar at inclusion rates of either 150 or 300 g/kg were used. Peas were included at the expense of starch. Hence, the differences in dietary crude protein and AA concentrations resulted only from the inclusion of peas. Titanium dioxide was included as an indigestible m…

The sulfhydryl groups of Cys 269 and Cys 272 are critical for the oligomeric state of chloroplast carbonic anhydrase from Pisum sativum.

Chloroplast carbonic anhydrase is dependent on a reducing environment to retain its catalytic activity. To investigate the properties of the three accessible cysteine residues of pea carbonic anhydrase, mutants were made in which Ala or Ser substituted for C165, C269, and C272. The mutants at position 165 were found to be spectroscopically similarly to the wild-type. They have a high catalytic activity, and are also sensitive to oxidation. In contrast, both C269 and C272 were found to be critical both for the structure and for the catalytic activity. All mutants with substitutions at either of

Fulvic-polyphosphate composite embedded in ZnO nanorods (FA-APP@ZnO) for efficient P/Zn nutrition for peas (Pisum sativum L.).

A nano-fertilizer (FA-APP@ZnO) was designed and prepared based on the copolymer of fulvic acid (FA) and ammonium polyphosphate (APP) with ZnO nanorods embedded, to tackle the antagonism between phosphorus (P) and zinc (Zn) in fertilization. FA-APP@ZnO was confirmed to revert the precipitability of H2PO4 - and Zn2+ into a synergistic performance, where FA and APP can disperse ZnO nanorods, and in return, ZnO catalyzes the hydrolysis of the absorbed APP. The hydrolysis rate constant of pyrophosphates consequently increased 8 times. The dry biomass of pea (Pisum sativum L.) under the FA-APP@ZnO h…

Solution structure of Pisum sativum defensin 1 by high resolution NMR: plant defensins, identical backbone with different mechanisms of action.

Pisum sativum defensin 1 (Psd1) is a 46 amino acid residue plant defensin isolated from seeds of pea. The three-dimensional structure in solution of Psd1 was determined by two-dimensional NMR data recorded at 600 MHz. Experimental restraints were used for structure calculation using CNS and torsion-angle molecular dynamics. The 20 lowest energy structures were selected and further subjected to minimization, giving a root-mean-square deviation of 0.78(+/- 0.22) A in the backbone and 1.91(+/-0.60) A for over all atoms of the molecule. The protein has a globular fold with a triple-stranded antipa…

[Time course of the content of folates and free amino acids in leaves of Pisum sativum L. after irradiation with ultraviolet C].

The effect of irradiation with UV-C on the time course of the content of total folates and free amino acids in leaves of pea (Pisum sativum L.) cultivar Neistoshchimyi was studied. It was shown that photolysis of folates is a rapid response to exposure to ultraviolet, as a result of which the plant produces a stable compound, pterin-6-carboxylic acid, with a relative fluorescence quantum yield approximately 2.0 at 20 degrees C (total value, 0.58). Presumably, this compound may be involved in the pterin-mediated photosensitization of singlet oxygen production. The kinetics of changes in the com…
