OBJECTIVE: Although periodontal disease is known to be associated with obstetric complications, evidence is limited regarding the relationship with placenta-mediated pregnancy complications (PMPCs), a major cause of maternal and fetal morbidity and mortality. This study investigated whether periodontal disease is a risk factor for PMPCs.
METHODS: We conducted a retrospective case-control study of inpatients with singleton pregnancies who underwent perinatal dental examinations at Nippon Medical School Tama Nagayama Hospital between September 2012 and August 2015. Medical records were reviewed

Autoimmune diseases (ADs), such as systemic lupus erythematosus (SLE), require multiple medications to ensure maternal-fetal health during pregnancy. These medications are often substrates for placental transporters that could cross over to the fetal compartment. However, the effects of ADs on placental transporters remain poorly understood. This study aimed to investigate the impact of ADs on placental transporters and key inflammatory cytokines. Human preterm and term placentas from AD-affected women (n = 28) and gestational age-matched controls (n = 38) were collected. The placentas were ex

Proteoglycan macromolecules play key roles in several physiological processes (e.g., adhesion, proliferation, migration, invasion, angiogenesis, and apoptosis), all of which are important for placentation and healthy pregnancy. However, their precise roles in human reproduction have not been clarified. To fill this gap, herein, we provide an overview of the proteoglycans' expression and role in the placenta, in trophoblast development, and in pregnancy complications (pre-eclampsia, fetal growth restriction), highlighting one of the most important members of this family, syndecan-1 (SDC1). One of the major immune checkpoints responsible for immune evasion in cancer cells is the interaction between programmed cell death-1 (PD-1) and its ligand (PD-L1). As human trophoblastic cells display many of the features of malignant cells such as the ability to invade normal tissue including blood vessels and are apparently not eradicated by the host immune system, we undertook the present study to determine whether PD-L1 was upregulated in different types of trophoblastic cells during normal pregnancy and in gestational trophoblastic diseases. Immunohistochemistry using an anti-PD-L1-speci

Although the mammalian embryo is well protected in the uterus, environmental chemicals, drugs, and maternal nutritional imbalances can interfere with regulatory pathways directing placental and embryonic development early in gestation. Embryonic cells are most susceptible to environmental influences during cellular specification and differentiation stages. Because biochemical differentiation precedes morphological outcome often by days, the period of susceptibility to environmental chemicals expectedly precedes visible morphogenic effects. The cellular mechanisms by which drugs and other envir

Placenta accreta refers to different grades of abnormal placental attachment to the uterine wall, which are characterised by invasion of trophoblast into the myometrium. Placenta accreta has only been described and studied by pathologists for less than a century. The fact that the first detailed description of a placenta accreta happened within a couple of decades of major changes in the caesarean surgical techniques is highly suggestive of a direct relationship between prior uterine surgery and abnormal placenta adherence. Several concepts have been proposed to explain the abnormal placentati

Behçet disease is an inflammatory disorder characterized by recurrent aphthous stomatitis, genital ulcers, uveitis, and skin lesions. There are occasional reports of neonatal Behçet disease suggesting a possible prenatal transmission. However, no placental Behçet disease lesion has been reported. Here, we report characteristic necrotizing inflammation in placentas and decidua from 2 patients with Behçet disease. The first patient had full-term delivery after uneventful medical therapy. The second patient underwent therapeutic abortion in the first trimester because of the fetal exposure to a p

Expression of proliferating cell nuclear antigen (PCNA), an auxiliary factor for the DNA polymerase delta, is closely related to cell proliferation. Using the monoclonal anti-PCNA antibody 19F4, we examined the distribution of PCNA in formalin-fixed, paraffin-embedded sections of mature and first-trimester placentas, 8 hydatidiform moles, and 7 choriocarcinomas. In normal placentas there was strong expression of PCNA in cytotrophoblastic nuclei, while the syncytium and the amnionic epithelium were PCNA unreactive. Staining of stromal cells was variable. These results are in complete agreement

BACKGROUND: Scant data on placental lymphatic vessels have pointed to the absence of lymphatic circulation. A recent study on mesenchymal dysplasia (MD), however, has identified pathologic lymphangiogenesis using the D2-40 lymphatic marker. These conflicting data have prompted us to investigate whether lymphatic vessels are present in normal developing placentas and in placental disorders characterized by cistern formation.
DESIGN: Seventeen human placentas without significant pathological abnormality ranging from 12 to 39 weeks of gestational age were studied. Cisternal placental disorders w

BACKGROUND: Fetuses with congenital heart defects (CHD) show delayed neurodevelopment, fetal growth restriction (FGR) and placenta related complications. The neurodevelopmental delay may be, partly, attributed to placental factors.
AIM: As both placental development and fetal aortic flow/oxygenation influence neurodevelopment, placentas were compared within fetal CHD groups based on aortic oxygenation and flow, aiming to unravel the true effects in the developmental processes.
STUDY DESIGN: Placental tissues of pregnancies with fetal CHD and healthy controls were selected from biobanks of tw

Background: Chronic venous disease (CVD) is a prevalent lower limb venous pathology that especially affects women, who also show an increased risk of this disease during pregnancy. Studies have shown significant structural changes in the placentas of women with CVD and several markers of tissue damage have been also described. Patients and Methods: To try to understand the different placental pathologies, research efforts have focused on examining metabolomic profiles as indicators of the repercussions of these vascular disorders. This study examines changes produced in the metabolomic profile

Placenta growth factor (PlGF) is released by immature erythrocytes and is elevated in sickle cell disease (SCD). Previous data generated in vitro suggest that PlGF may play a role in the pathophysiology of SCD-associated pulmonary hypertension (PHT) by inducing the release of the vasoconstrictor, endothelin-1. In this cross-sectional study of 74 patients with SCD, we confirm that PlGF is significantly elevated in SCD compared with healthy control subjects. We found significantly higher levels of PlGF in SCD patients with PHT but observed no association of PlGF with the frequency of acute pain

Pre-eclampsia (PE) and eclampsia is the leading cause of maternal death in the UK and a major cause of perinatal morbidity and mortality. PE is predominantly a disorder of first pregnancy. There is a genetic component. PE is a syndrome without a defined cause. The variability with which it presents, and the speed with which it progresses, make it hard to diagnose. High blood pressure and proteinuria, with or without oedema, are classic symptoms but it may also present in an unusual way, eg as jaundice; as proteinuria with normal blood pressure; as intra-uterine growth retardation; as abdominal

In order to determination if an interdependence exists between the well-known maturational disturbances in placentas of diabetics and the quality of metabolic adjustment, a morphometric study covering 7,500 terminal villi of 50 placentas was performed. The diabetic mothers were divided into two groups according to satisfactory or unsatisfactory blood glucose values. Significant differences in placental retardation were observed between both diabetic groups and a normal control group. Although outcomes did not differ greatly between the two diabetic groups, macrosomia and the ratio of malformat

The placenta is a highly metabolic organ essential for fetal growth by mediating nutrient transport, hormone production, and immunological regulation. These functions depend on continuous and efficient adenosine triphosphate (ATP) supply, primarily generated through glycolysis and oxidative phosphorylation. However, due to high turnover of ATP and multi-step de novo synthesis, these pathways may not always meet the rapid and localized energy demands of trophoblast cells. The phosphagen system, comprising creatine kinase (CK), creatine (Cr), and phosphocreatine (PCr), provides a rapid ATP-buffe

A woman in her mid-20s with repaired congenital heart disease (CHD) delivered a 33-week neonate in 2024 following preterm labor and a 24-week neonate in 2025 for non-reassuring fetal heart tones. Placental examination in both cases revealed grossly abnormal, red-brown discolored membranes with a circumvallate insertion. Histologic evaluation demonstrated diffuse chorioamniotic hemosiderosis (DCH) in both cases. This report describes a novel case of recurrent DCH in a mother with CHD and proposes that Fontan physiology may contribute to impaired uteroplacental perfusion, increasing the risk for

OBJECTIVE: Concurrent development of the placenta and heart during early gestation suggests a shared biological basis for the co-occurrence of abnormal placentation and congenital heart disease (CHD). This study investigated the association between placental vascular pathology and CHD type.
METHODS: A retrospective study at two institutions included CHD (n = 521) and unaffected (n = 122) infants. CHDs were categorized into three groups (left and right ventricular outflow tract obstruction and mixed lesions) based on the anticipated effect of placental venous return streaming toward the fetal
